Effects of Dietary Copper on Ruminal Fermentation, Nutrient Digestibility and Fibre Characteristics in Cashmere Goats

  • Thirty-six 1.5 year-old Inner Mongolian White Cashmere wether goats (body weight $28.14{\pm}1.33$ kg) were used to determine the effects of dietary copper (Cu) concentration on ruminal fermentation, nutrient digestibility and cashmere fibre characteristics. Wethers were fed a basal diet (containing 7.46 mg Cu/kg DM) that was supplemented with either 0 (control), 10, 20 or 30 mg Cu/kg DM. To ensure full consumption, animals were fed restrictedly with 0.75 kg feed (DM) in two equal allotments per day. The results indicated that: (1) supplemental 10 mg Cu/kg DM in the basal diet significantly (p<0.05) decreased ruminal fluid pH value and total VFA concentrations were significantly (p<0.05) increased on all Cu treatment groups. (2) Cu supplementation had no influence on DM intake and digestibility of DM, CP and ADF (p>0.05); however, NDF digestibility of groups supplemented with 10 and 20 mg Cu/kg DM were significantly higher than that of the control group (p<0.05). Apparent absorption and retention of copper were decreased with increasing level of supplementation. (3) 20 mg Cu/kg DM treatment significantly (p<0.05) improved cashmere growth rate, but cashmere diameter was not affected by Cu supplementation (p>0.05). In conclusion, supplementation of cashmere goats with Cu at the rate of 10 to 20 mg/kg DM in the basal diet resulted in some changed rumen fermentation and was beneficial for NDF digestibility, while supplementation of 20 mg Cu/kg DM improved cashmere growth. Collectively, the optimal supplemental Cu level for cashmere goats during the fibre growing period was 20 mg/kg DM (a total dietary Cu level of 27.46 mg/kg DM).

Effects of Feeding Different Chelated Copper and Zinc Sources on Growth Performance and Fecal Excretions of Weanling Pigs

  • Two feeding trials were conducted to study the effects of different chelated copper and zinc compounds on the performance and fecal excretions of weanling pigs. In Exp. 1, 150 weanling pigs ($L{\times}Y{\times}D$, $12.30{\pm}2.07kg$) were randomly assigned to five dietary treatments: 170 ppm Cu from $CuSO_4$, 85 ppm Cu from Cu amino-chelate (CAC), 170 ppm Cu from CAC, 85 ppm Cu from Cu-Lysine (CL), and 170 ppm Cu from CL. In Exp. 2, 150 weanling pigs ($L{\times}Y{\times}D$, $12.52{\pm}1.80kg$) were randomly assigned to five dietary treatments: 120 ppm Zn from $ZnSO_4$, 60 ppm Zn from Zn-amino-chelate (ZAC), 120 ppm Zn from ZAC, 60 ppm Zn from Zn-Methionine (ZM), and 120 ppm Zn from ZM. In both experiments, pigs were randomly distributed to the treatments following a randomized complete block design on the basis of body weight as the blocking variable. Each experiment was conducted for 28 days. Blood and fecal samples were collected to determine mineral contents as affected by the dietary treatments. There was no difference (p>0.05) in ADG and ADFI among treatments, but F/G was improved (p<0.05) in pigs fed diet with 170 ppm CAC than 85 ppm CL but not different (p>0.05) to the control (170 ppm $CuSO_4$). Regardless of copper source, concentration of Cu in serum and feces were higher in pigs fed diet with 170 ppm Cu than pigs fed diet with 85 ppm Cu (Exp 1). In Exp 2 the ADG was higher (p<0.05) in pigs fed diet with 120 ppm ZM than in pigs fed diets with 120 ppm $ZnSO_4$ and 60 ppm ZAC and ZM. The serum zinc concentration was generally higher (p<0.05) in pigs fed diet with organic source than the control group ($ZnSO_4$). Also, there was a trend towards a decrease in fecal excretions of zinc when dietary zinc level was low. The efficacy of the two chelated copper and zinc sources is similar in terms of growth performance. The fecal excretions for Cu and Zn could be reduced in pigs fed low level of these minerals using organic sources.

친환경 항균성 구리의 수질 내 생물학적 오염원 제거 가능성 연구 (The Studies on the Removal of Harmful Microorganisms in Water by Using Antimicrobial Copper)

  • 기후변화와 도시화에 따른 오염으로 인하여 대기 및 토양이 오염되었고, 이들 오염물질이 점오염원과 비점오염원을 통하여 수질 내로 흘러 들어가면서 수질오염이 심각한 수준에 이르렀다. 이러한 수질 내 생물학적 오염원 제거를 위하여 간편하면서도 효율이 높은 수질 정화용 필터의 개발이 요구되는 실정이며, 이를 위하여 항균력을 가지고 있는 구리를 이용하여 연구를 진행하였다. 본 연구에서는 수질오염의 기준으로 사용되는 대장균 외 주요 병원성 균 4종 (Bacillus subtilis, Staphylococcus aureus, Klebsiella pneumoniae, Pseudomonas aeruginosa)을 사용하였으며, 스트립 형태의 구리선 0.5g, 1g, 2g을 이용하여 직경 2cm의 구 형태로 제작하였다. 항균력 확인을 위하여 각 병원성 균을 접종한 후 구리공을 장착한 다음, 99% 이상의 항균력을 가지는 조건을 조사하였다. 연구 결과 0.5g의 구리선으로 만든 구리 공을 최소 20분간 반응시켰을 때 5종의 병원성균에 대해서 99% 이상의 생장 억제능이 나타나는 것을 확인하였다. 본 연구 결과를 바탕으로 세포 실험 등의 추가 실험이 진행된다면 수질 정화용 필터로 사용되기에 충분할 것으로 판단되며, 이를 통해 공장 및 도시 폐수 뿐만 아니라 축산 및 양식 산업 분야에 광범위하게 적용될 수 있을 것으로 기대된다.

콩 불마름병 방제를 위한 살균제 선발 (Selection of Bactericides for Controlling Soybean Bacterial Pustule)

  • Xanthomonas axonopodis pv. glycines에 의해 발병되는 콩 불마름병은 콩에 가장 많이 발생하는 중요한 세균성 병해 중 하나이다. 게다가 병원균은 강한 병원성을 가지고 한국 전역에 분포하고 있지만, 한국에서는 불마름병방제를 위한 약제나 저항성 품종이 개발되어 있지 못한 실정이다. 따라서 본 연구의 목적은 불마름병을 화학적 방법으로 방제할 수 있는 방법을 개발하기 위해 수행되었다. 콩 불마름병 병원균의 효과적인 생육억제 약제를 선발하기 위해 여러 가지 살세균제들(8개의 항생제, 2개의 구리화합물, 1개의 quinoline, 18개의 농약)을 이용하여 효과를 확인하였다. 실내 생장억제 효과조사 결과, tetracycline, streptomycin sulfate 처리에서 병원균의 생육이 크게 억제되었으며, 또한 oxolinic acid도 병원균의 생육억제 효과가 우수하였다. 하지만 vancomycin, polymyxin B sulfate and copper compounds 등은 불마름병 병원균의 생육억제효과가 확인되지 않았다. 18종의 농용약제 중에는 streptomycin sulfate + oxytetracycline(18.8 + 1.5%)WP, oxytetracycline(17%)WP, oxolinic acid(20%) 등이 실내에서 병원균의 생육억제 효과가 높은 것으로 조사되었다. 선발된 5종의 농용약제에 대하여 포장에서 방제효과 시험을 실시한 결과, streptomycin sulfate + oxytetracycline (18.8 + 1.5%) WP와 oxytetracycline(17%) WP가 자연 감염된 태광콩 포장에서 70% 이상의 높은 방제효과를 나타내었다. 그러므로 콩불마름병균에 대한 강한 억제효과를 가지는 이 약제들은 콩 불마름병의 화학적 방제에 실용성이 있을 것으로 생각된다.

구리 이온의 김치산패 억제작용에 관한 연구 (Effect of Copper Ions on Over-Acidification of kimchi)

  • 김치에 $Cu^{+}$$Cu^{2+}$ 을 1.0, 2.0, 3.0, 4.0 및 5.0 mM 농도로 첨가하여 2$0^{\circ}C$에서 김치의 발효경향을 살펴보고 또한 젖산균 생육에 미치는 구리 이온의 영향을 알아보았다. Cu$SO_4$와 CuCl의 첨가는 김치에 생육하는 총생균, 젖산균의 생육을 저해하고 김치의 산도를 유지하는데 효과가 있었다. 또한 Leu. mesenteroides의 경우 발효 10일째 대조군의 급격한 감소와는 다르게 발효 14일째에 $10^{5}$ ~$10^{6}$ CFU/ml를 유지하였다. 따라서 $Cu^{+}$$Cu^{2+}$의 첨가는 김치의 숙성에 관여하는 주 발효균인 Leu. mesenteroides의 생육기간을 연장시키고 과도한 산의 생성을 억제함으로써 김치의 산패를 지연시킬 수 있는 것으로 생각된다. $Fe^{2+}$$SO_4$$^{2-}$ 이온은 김치발효에 영향을 미치지 않는 것으로 나타났다. 또한 각각의 젖산균의 생육에 미치는 $Cu^{2+}$ 이온의 영향을 보기 위하여 각 젖산균마다 Cu$SO_4$를 농도별로 첨가한 후 흡광도를 측정한 결과 Leu. mesenteroides 6, Str. faecalis 12, Lac. plantarum 14, Lac. brevis 15, Leu. mesenteroides LA 10, Lac. plantarum LA 97의 6가지 젖산균은 $Cu^{2+}$ 첨가농도에 따른 균주마다 내성의 차이를 보였으나 각각의 균주 모두 $Cu^{2+}$ 첨가에 따라 균의 생육이 저해되었다.

Streptomyces albus와 Streptomyces globosus의 몇가지 생장생리적 특성에 관하여 (Some Physiological Properties in Relation to the Growth of the Antibiotics Producing Streptomyces spp.)

  • In previous paper, it was reported that antibiotic substance such as tetracycline and streptomycin were produced by S'. albus subsp. and S'. globosus. And increase of mycelial growth of two strains, antibiotic production, and changes of pH range are extended to approximately 110-130 hrs in fermenting medium, there-after they decreased with culture period exception of pH range. Two Streptomyces spp. required commonly 4-5% starch as carbon sources and 1.5-2.0% soybean meal as nitrogen sources. However, 0.005-0.01M potassium phosphate dibasic, calcium carbonate (6mg/ml in S.albus subsp. and 2mg/ml in S. globosus), 0.01-0.03M, magnesium sulgate and 0.01M ferric chloride showed as optimal concentration for the growth of 2 strains. Mineral compoments such as zinc, manganese, cobalt, sodium and copper at the level of 10/sup -4/ -10/sup -6/M were observed. Especially, zinc ion showed toxicity to the growth of 2 strains at 0.005M. In relation with pH, there is a little difference in mycelial growth with cultural initial pH.

다중회귀분석법에 의한 소나무, 곰솔 및 리기다소나무의 상대성장 비교 (Comparisons on Relative Growth of Red Pine, Black Pine and Pitch Pine by Means of Multiple Regression)

  • The purpose of this study is to compare the relative growth of annual ring width of red pine(Pinus densiflora), black pine(Pinus thunbergii) and pitch pine(Pinus rigida) by means of multiple regression method according to Graybill hypothesis. The obtained results are as follows. 1. The changes of rainfall have affected to tree growth during the periods of 1975 through 1978. 2. Among these pine trees, red pine was mostly influenced by environmental factors. 3. The growth of annual ring width was sensitively responded to the changes of rainfall and air temperature. 4. Among the heavy metals analyzed, the concentrations(ppm) of Lead(Pb) and Copper(Cu) were negatively effected on the growth of annual ring width of pine trees. 5. The analytical technique of annual ring width may be useful for estimation of the pollution in forest areas near industrial complexes.

  • Chlorella ellipsoidea cells were cultured in an iron, copper, zinc, manganese, molybdenum or boron-free medium. Physiological activities such as growth rate, reproduction, endogenous and glucose respiration, photosynthetic activity and biosythesis of chlorophyll of the micro-element definition cells were measured. It generally, growth rate, respiratory and photosynthetic activities, and biosynthesis of chlorophyll of the micro-element deficient cells decreased more or less, compared with those of the normal cells. The growth of the algal cells in an iron-free medium were retarded severely with the chlorosis, and the photosynthetic activity of the cells decreased remarkably even though the low content of chlorophyll in the cells owing to the iron-deficiency is considered. Therefore, it is deduced that iron takes part in the photosynthetic process itself, possibly by its participation in the photo phosphorylation coupled with electron transport. Respiratory activity of boron-deficient cells showed the most severe decrease whereas those of the molybdenum-deficient cells showed very slight decrease in spite of severe growth retardation.

Optimization of Culture Conditions for Production of Pneumococcal Capsular Polysaccharide Type IV

  • The Pneumococcus, Streptococcus pneumoniae, has an ample polysaccharide (PS) capsule that is highly antigenic and is the main virulence factor of the organism. The capsular PS is the source of PS vaccine. This investigation was undertaken to optimize the culture conditions for the production of capsular PS by type 4 pneumococcus. Among several culture media, brain heart infusion (BHI) and Casitone based medium were found to support luxuriant growth of pneumococcus type 4 at the same level. Therefore in this study, the Casitone based medium was used to study optimization of the culture condition because of BHI broth's high cost and complex nature. The phase of growth which accomodated maximum PS production was exponential phase. Concentrations of glucose greater than 0.8% did not enhance growth or PS production. Substitution of nitrogen sources with other resources or supplementation of various concentrations of metal ion (with the exception of calcium, copper, and magnesium ions) had adverse effects on growth and PS production. On the other hand, low level aeration and supplementation of 3 mg/l concentration of asparagine, phenylalanine, or threonine were beneficial for increased PS production. The synergistic effect of all the favorable conditions observed in pneumococcal growth assays provided a two-fold cumulative increase in capsular PS production.

등온시효에 따른 Sn-3.5Ag 솔더 접합부의 금속간 화합물 성장에 관한 연구 (Growth Kinetics of Intermetallic Compound on Sn-3.5Ag/Cu, Ni Pad Solder Joint with Isothermal Aging)

  • The growth kinetics of intermetallic compound layers formed between the eutectic Sn-3.5Ag solder and the Cu and Ni/Cu pad by solid stateisothermal aging were examined. The interfacial reaction between the eutectic Sn-3.5Ag solder and the Cu and Ni/Cu pad was investigated at 70, 120, 150, $170^{\circ}C$ for various times. The intermetallic compound layer was composed of two phase: $Cu_6Sn_5$(${\varepsilon}-phase$) adjacent to the solder and $Cu_6Sn_5$(${\varepsilon}-phase$) adjacent to the copper and on solder/Ni pad the intermetallic compound layer was $Ni_3Sn_4$. Because the values of time exponent(n) have approximately 0.5, the layer growth of the intermetallic compound was mainly controlled by volume diffusion over the temperature range studied. The apparent activation energy for layer growth of total Cu-Sn($Cu_6Sn_5 + Cu_6Sn$), $Cu_6Sn_5$, $Cu_3Sn$ and $Ni_3Sn_4$ intermetallic compound were 64.82kJ/mol, 48.53kJ/mol, 89.06kJ/mol and 71.08kJ/mol, respectively.
