국내에 시판되고 있는 유기농산물과 일반농산물의 부패미생물 분리 및 동정 (Isolation and Identification of Spoilage Bacteria on Organic and Conventional Fresh Produce in Korea)

  • 본 연구는 부패미생물의 종류와 차이를 알아보기 위해 유기농산물과 일반농산물(깻잎, 양배추, 상추)을 구입하여 $4^{\circ}C$에 14일 저장한 후 일반목적배지를 사용하여 미생물을 분리 및 16S rRNA sequencing방법을 이용하여 동정하였다. 유기농산물 깻잎, 양배추, 상추의 총균수는 $5-7{\log}_{10}CFU/g$ 수준이었고, 일반농산물의 총균수는 $5-8{\log}_{10}CFU/g$ 수준이었다. 유기농산물보다는 일반농산물이 총균수를 더 많이 내재하고 있으나 유의적인 차이는 나타나지 않았다(p > 0.05). 유기농산물 깻잎에서 발견된 주요 균은 Microbacterium sp.이었으며, 일반농산물 깻잎에서 발견된 주요균은 Stenotrophomonas maltophilia이었다. 또한, 유기농산물에서 부패를 일으킬 수 있는 균인 Klebsiella pneumonia가 발견되었으며, 일반농산물보다 유기농산물에서 발견된 균의 종류가 더 많은 것을 확인할 수 있었다. 이에 본 연구 결과는 부패미생물에 대한 이해를 높이고 농산물의 신선도 및 유통기한을 늘리기 위한 새로운 살균소독제 및 부패 억제제를 개발하는데 좋은 기초자료로 사용될 수 있을 것으로 사료된다.

신선편의식품 포장기술 (Packaging technology of fresh-cut produce)

  • Processing steps such as washing and cutting, involved in preparing fresh-cut produce causes tissue damage, leading to rapid quality deterioration. Major defects of fresh-cut produce are discoloration, softening, off-odor development, and microbial growth. Packaging of fresh-cut produce has been changed to reduce these quality problems. Flexible packaging film is widely used to pack fresh-cut produce. Vacuum packaging was the popular packaging method in the beginning of fresh-cut industry in Korea. Vacuum packaging creates high $CO_2$ and low $O_2$ levels to control browning of fresh-cut produce. However, these conditions induce some visual defects and off-odor development. Discoloration problem was also found when fresh-cut produce was packaged with conventional packaging film or plastic tray. Modified atmosphere (MA) packaging is effective for prolonging shelf-life of fresh-cut produce by decreasing $O_2$ and increasing $CO_2$ concentration in the package. Retail MA packaging using different oxygen transmission rate (OTR) film and micro-perforated film has started to be applied to fresh-cut produce in Korea. Proper MA package design that provides optimum range of $O_2$ and $CO_2$ partial pressures is one of the major challenges in the industry. An initial package flushing with $N_2$ or an low $O_2$/high $CO_2$ atmosphere is also used to more rapidly establish steady-state MA condition. Film OTR and $O_2$ flushing affects the fermentative volatile production, off-odor development, electrolyte leakage, discoloration, $CO_2$ injury, microbial population of fresh-cut produce. There is also a demand for convenient packaging to attract consumers. Rigid fresh-cut produce container for retail market has increased since the packaging provides excellent protection from physical damage during transport. Rigid tray used as actual serving vessel for the consumer is increasing in Korea. The tray with flexible lid to wrap or seal fresh-cut produce is more and more gaining popularity. Further practical technology to control quality change and microbial growth for each fresh-cut product has been studied since various fresh-cut items were required. The fresh-cut industry also focuses on searching for more convenient and environmentally friendly packaging.

신선편이 농식품 생산라인의 환경미생물 오염도 측정을 위한 ATP 검사법의 이용 (Application of ATP Bioluminescence Assay for Measurement of Microbial Contamination in Fresh-cut Produce Processing Lines)

  • 신선편이 농식품 시장의 빠른 성장과 함께 안전관리기준도 강화되고 있어 제조 시설 현장에서 신속하고 간편하게 위생상태를 판단할 수 있는 모니터링기법이 필요하게 되었다. 본 연구는 신선편이 농식품 생산업체 3개소를 대상으로 생산에 사용되는 도구와 기기에 대한 미생물수를 조사하였고 그에 대한 검증법인 ATP 검사법의 상관관계를 도출하고자 하였다.신선편이 농식품 생산업체 3개소의 신선편이 가공 시설, 장비에서 채취된 총 50개 시료의 ATP bioluminescence assay 값과 일반세균수와의 상관계수 (r)은 0.8772로 나타났다. ATP 검사법은 신속, 간편, 일관성을 바탕으로 신선편이 농식품 산업체에서 미생물 배양법을 대체하여 위생 환경 개선에 유용하게 쓸 수 있는 것으로 판단된다.

생표고버섯에 대한 마스터 포장 시스템의 현장 적용 (Application of Master Packaging System to Fresh Shiitake Mushroom Supply Chain on Semi-commercial Scale)

  • 농가로부터 판매처까지의 유통단계에서 변화하는 온도 조건에 노출된 생표고버섯의 저장성향상을 확보하기 위해 개별포장과 2차 포장을 결합시킨 마스터 포장 시스템을 적용하였다. 온도가 저온으로 유지되는 수송과 저장 단계에서는 1차와 2차 포장의 결합 형태로 유통되고, 온도가 높은 판매단계에서는 외부 2차 포장을 해체한 후 1차 포장 조건에서 적절한 가스농도를 확보할 수 있는 개념으로 설계하고, 2차 마스터 포장 내에는 $Ca(OH)_2$의 이산화탄소 흡수제를 포함시켰다. 유통흐름 중에서 포장의 온습도와 기체조성을 측정하고, 마지막 단계인 판매처에서 포장을 개봉하여 버섯의 품질을 측정하였다. 공기조성을 유지하는 관행적인 통기성 천공 포장을 대조구로 함께 유통흐름을 거치게 하여 저장 후 품질을 비교하였다. 초기의 수송 및 저장 단계에서 적절한 저온조건이 유지되지 못한 것으로 나타났으나, 유통의 전체 과정에서 버섯 주위에 산소 농도 0.4~4.2%, 이산화탄소 농도 0.7~1.7%로 품질보존에 도움이 되는 기체조성이 유지되었다. 포장 전 예건처리가 버섯의 품질보존에 효과가 있는 반면에, 마스터 포장 시스템의 긍정적 효과는 비예건 버섯에 뚜렷이 나타났다. 보다 효과적인 버섯 품질보존을 위해서 예건 처리 및 온도관리와 함께 조화된 마스터 포장 시스템의 설계가 추가적으로 이루지고 검토될 필요가 있는 것으로 생각된다.

시비수준이 땅두릅의 생육과 수량에 미치는 영향 (Effects of Fertilization Level on Growth and Yield of Aralia cordata Thunb)

  • 두릅나무과에 속하는 다년생 숙근식물인 땅두릅(Aralia cordata Thunb.)의 양질 다수확 재배를 위한 적정 시비수준을 구명하기 위하여 실험을 수행하였던 바 얻어진 결과를 요약하면 다음과 같다. 1. 시비수준에 따른 지상부의 초장은 관행인 17-9-17kg보다 27-15-27kg에서 28.3cm로 생육이 우세하였으며 이러한 경향은 연화장의 결과와 마찬가지로 관행 시비수준이 7.0cm인 것보다 10.3cm로 양호한 결과를 보였다. 2. 지하부의 생육특성중 주근장은 관행시비에서 10.5cm로 처리간에 가장 높은 수치를 보였다. 3. 시비수준에 따른 년차별 건근중은 재배년수가 길어질수록 건근중의 증가를 보였으며 평균적으로 볼 때 관행시비인 17-0-17kg이 272kg인 것과 비교해 보면 27-15-27kg에서 310kg으로 약 14%정도 건근중의 증수효과를 보이는 것으로 나타났다. 4. 땅두릅의 연화재배를 통한 연화장의 엽수량은 관행시비구읜 17-9-17kg에서 391kgdlsrjt 보다 27-15-27kg 시비구에서 415kg으로 나타난 지상부의 건근 수량과 마찬가지로 가장 높은 수익성을 보이는 것으로 나타났다. 5. 이상의 결과를 종합해 볼 때 지상부의 엽수량과 지하부의 건근수량의 양질 다수확 재배를 위해서는 시비수준을 10a당 27-15-27kg으로 시용하는 것이 유리한 것으로 사료되었다.

Optimal numbers of mature oocytes to produce at least one or multiple top-quality day-3 embryos in normal responders

  • Objective: We attempted to identify the optimal cutoff numbers of mature oocytes that would produce at least one or multiple top-quality (grade A) day-3 embryos in normal responders undergoing stimulated in vitro fertilization (IVF) cycles. Methods: We selected 210 fresh IVF cycles performed in 170 infertile women at a single center from January 2014 to November 2019. Four to 14 (total) oocytes were obtained in all cycles after conventional ovarian stimulation. A receiver operating characteristic curve analysis was performed to find the moderate and extreme cutoff numbers of mature oocytes that would produce ≥ 1, ≥ 2, ≥ 3, ≥ 4, and ≥ 5 top-quality embryos. Results: The cutoff number of mature oocytes was significantly correlated with the number of top-quality embryos (r = 0.467, p= 0.000). The moderate cutoff number of mature oocytes was ≥ 3, ≥ 5, ≥ 5, ≥ 6, and ≥ 6 for obtaining ≥ 1, ≥ 2, ≥ 3, ≥ 4, and ≥ 5 top-quality embryos, respectively. The extreme cutoff number of mature oocytes was ≥ 9, ≥ 9, ≥ 10, ≥ 10, and ≥ 11 for obtaining ≥ 1, ≥ 2, ≥ 3, ≥ 4, and ≥ 5 top-quality embryos, respectively. Conclusion: We present the optimal cutoff numbers of mature oocytes that would yield ≥ 1, ≥ 2, ≥ 3, ≥ 4, and ≥ 5 top-quality embryos with 95% specificity. Our findings could help infertility clinicians to set target mature oocyte numbers in women undergoing stimulated IVF cycles.

Short term bond shear stress and cracking control of reinforced self-compacting concrete one way slabs under flexural loading

  • Fibre-reinforced self-compacting concrete (FRSCC) is a high-performance building material that combines positive aspects of fresh properties of self-compacting concrete (SCC) with improved characteristics of hardened concrete as a result of fibre addition. To produce SCC, either the constituent materials or the corresponding mix proportions may notably differ from the conventional concrete (CC). These modifications besides enhance the concrete fresh properties affect the hardened properties of the concrete. Therefore, it is vital to investigate whether all the assumed hypotheses about CC are also valid for SCC structures. In the present paper, the experimental results of short-term flexural load tests on eight reinforced SCC and FRSCC specimens slabs are presented. For this purpose, four SCC mixes - two plain SCC, two steel, two polypropylene, and two hybrid FRSCC slab specimens - are considered in the test program. The tests are conducted to study the development of SCC and FRSCC flexural cracking under increasing short-term loads from first cracking through to flexural failure. The achieved experimental results give the SCC and FRSCC slabs bond shear stresses for short-term crack width calculation. Therefore, the adopted bond shear stress for each mix slab is presented in this study. Crack width, crack patterns, deflections at mid-span, steel strains and concrete surface strains at the steel levels were recorded at each load increment in the post-cracking range.

Experimental analysis and modeling of steel fiber reinforced SCC using central composite design

  • The emerging technology of self compacting concrete, fiber reinforcement together reduces vibration and substitute conventional reinforcement which help in improving the economic efficiency of the construction. The objective of this work is to find the regression model to determine the response surface of mix proportioning Steel Fiber Reinforced Self Compacting Concrete (SFSCC) using statistical investigation. A total of 30 mixtures were designed and analyzed based on Design of Experiment (DOE). The fresh properties of SCC and mechanical properties of concrete were studied using Response Surface Methodology (RSM). The results were analyzed by limited proportion of fly ash, fiber, volume combination ratio of two steel fibers with aspect ratio of 50/35: 60/30 and super plasticizer (SP) dosage. The center composite designs (CCD) have selected to produce the response in quadratic equation. The model responses included in the primary stage were flowing ability, filling ability, passing ability and segregation index whereas in harden stage of concrete, compressive strength, split tensile strength and flexural strength at 28 days were tested. In this paper, the regression model and the response surface plots have been discussed, and optimal results were found for all the responses.

과수육종에 있어 생명공학의 이용 전망 (The Prospectss and Utilization of Biotechnology for the Improvement of Fruit Breeding)

  • The major objectives of fruit breeding lie in improvement of cultivar, easy to be cultivated and of high quality, in order to produce unexpensive, delicious fruit both for fresh fruit market and processing. Recently, fruit breeding in Korea has contributed to breeding of several superior cultivars in major fruit crops, resulting in appreciable improvement in qualities such as skin color, taste and fruit-bearing habit concerned with productivity. In spite of accomplishments mentioned above, the need for both highly disease-resistant cultivars and long-keeping, physiological disorder-resistant cultivars to meet long distance transsportation in the temperate fruit crops of apples, oriental pears, stone fruits such as peaches, and grapes grown in Korea is rapidly pressing more than ever, as cultivars of today susceptible to pests and diseases and vulnerable to physiological disorders are very expensive and time-consuming in post-harvest handling and management. Thus, imporvements made in the above problems through breeding level will lead to the really enhanced productivity in fruit industry. The major impediments of tree size, the long length of juvenile period and the highly heterogeneous genetic composition to the improvement of fruit crops are responsible for the lower amount and rate of improvements of fruit crops as compared to annuals. Considering the expected limitations of the above problems to be solved through conventional breeding methods and strategy, a turning point of breeding a near perfect cultivar would be laid down if innovative breakthroughs in biological technology will be realized in applying some of the techniques of genetic manipulation at the molecular level to the cultivar improvement of fruit crops, such as the selective insertion of DNA carrying genes that govern desirable characteristics. More than anything else, those traits such as fruiting habit deciding productivity, elements of fruit qualities conditioned by monogene, and disease-and pest-resistance of vital importance for successful fruit growing are urgently desired to be improved by advancement of biotechnology for they are more than difficult and need long period to be attained through conventional breeding method.

건조방법에 따른 건고추의 품질평가 (Evaluation of Quality of Red Pepper with variations in Drying Methods)

  • In order to produce the high-quality of dried red pepper with respect to a color and a taste we developed a automatic drier equipped with combined several heat energies(ADCHE). and compared the quality of ADCHE-treated red pepper with that of the pepper treated with conventional dryings such as natural, hot-air, and far-infrared ray dryings. The results obtained were as follows : (1) The contents of capsanthin varied significantly with drying methods within the range of 1.7 to 6. 4mg/g dry weight. The capsanthin level of red pepper treated with far-infrared ray drying was higher than that of the pepper treated with ADCHE showed the highest at 51.46mg/g dry weight of three drying methods. (2) As a result of determination of color intensity of-red-pepper using-a colorimeter, the red color intensity of the peppers was affected by drying methods irrespective of drying temperature, and especially a value of red pepper treated with ADCHE was appeared to be 19. 1, indicated that this pepper have the most bright color intensity. (3) The contents of soluble browing subatances increased with a increase in drying temperature, but L* value of red pepper treated with ADCHE was appeared to be 0.187, indicated that this red pepper have the most clear red color (4) The changes of capsanthin level of red pepper according to drying methodes did not show significantly, but generally a decreasing rate of capsanthin levels were lower in that order ADCHE
