  • The first crustal refraction survey in the Korean Peninsula was carried out over the survey line connecting Seosan-Yeongdong-Kyeongiu on Dec. 15, 2002. The total length of the survey line was about 300 Km and 198 portable seismometers were deployed with approximately 1.5-km interval. The survey line itself was geologically important since it was almost normal to the so-called Sino-Korean structural trend. Two shots, one at Seosan (west end point) and the other at Yeongdong (mid-point), were exploded. They were 100-m deep drill well explosions. The Seosan shot consisted of a ton emulsion type explosive, while Youngdong consisted of 500 kg one. Both shots generated signals with good S/N ratios to the farthest receivers. Seismic signals were recorded by 195 receivers out of 198 ones. Although the originally planned Kyeongju shot (east end point) could not be exploded due to public discontent, the experiment was evaluated very successful. First breaks in all recorded traces were picked up and two preliminary analyses were carried out. The one is conventional flat layer analysis and the other was refraction tomographic analysis. The one resulted in average 32-km thick two-layer crust and the underlying mantle with 8.05-km/s P-velocity. The top crust layer with 3.86 kw/s P-velocity was 2.5-km thick and the lower crust layer with 6.0l km/s P-velocity was 29.5-km thick. The other resulted in a velocity cross-section. The confidence level of the velocity cross-section could not be evaluated at this time because only two shot were exploded. Detailed analyses such as surface wave dispersion are on going. Continuing crustal scale refraction surveys are planned in Korea.

  • Seismic refraction survey is a geophysical method that delineates subsurface velocity structure using direct wave and critically refracted wave. The generalized reciprocal method(GRM) is an inversion technique which uses travel-time data from several forward and reverse shots and which can provide the geometry of irregular inclined refractors and structures underlain by hidden layer such as low velocity zone and thin layer. In this study, a simple Excel-GRM routine was tested for fast mapping of the interface between weathering layer and bedrock during the survey, with employing a pair of forward and reverse shots. This routine was proved to control the maximum dip of approximately $30^{\circ}C$ and maximum velocity contrast of 0.6, based on the panel tests in terms of dipping angle and velocity contrast for the two-layer inclined models. In contrast with conventional operation of five to seven shots with sufficient offset distance and indoor data analysis thereafter, this routine was performed in the field shortly after data acquisition. Depth to the bedrock provided by Excel-GRM, during the field survey for Cheongju granite area, correlates well with the elevation of the surface of soft rock from the drill core and SPS logging data. This cost-effective routine developed for quickly delineating the bedrock surface in the field survey will be readily applicable to mapping of weathering zone in narrow zone with small variation of elevation of bedrock.

  • This experiment was carried out at National Crop Experiment Station in 1994 to obtain basic information of growth characters and yield of rice under various nitrogen split application methods on different soil textures in direct seeding on dry paddy. Hwaseongbyeo sown on April 27 by flat drill seeding, and irrigation was done at 3 leaf stage after seedling establishment. Number of seedling stand was 159~177 seedlings per $m^2$ regardless of soil texture and nitrogen application method. Number of panicle per unit area in loam was higher than in sandy loam, and it also was higher in top dressing plots, which were 3 times application at rate of 40-30-30% (3rd leaf stage -7th leaf stage -panicle initiation stage) and 4 times application 10-30-30-30%(basal-3rd leaf stage -7th leaf stage -panicle initiation stage), than in conventional method. Leaf colour, leaf area index and dry matter production at heading stage were highest at top dressing plots among the nitrogen application methods in both sandy loam and loam. Lodging index in top dressing plots increased by low breaking weight with long culm. There were field lodging of degree 3 in top dressing plots. Rice yield in sandy loam, loam increased by 7~9%, 6~9% in top dressing of nitrogen, respectively.

  • Soil profiling is one of the most important work among geotehnical engineering practice. Generally, soil profile is estimated from the observation of soil samples during subsurface exploration but such estimation also includes some experiencing aspects such as flushed water from the borehole, slime colour, boring speed and so on. In addition, since the capacity of hydraulic drill rig is significantly increased, thin layers might be easily missed. So, continuous soil profile is almost impossible over all depth to be bored from conventional subsurface exploration. While CPT or CPTu can serve continuous soil profile information over all depth generally in 5cm interval. Many charts or methods for soil profile from CPT result have been proposed during last several decades over the world. However they have not been verified in local ground condition in Korea. In this research, CPT results and soil classification results based on USCS were compiled from 17 sites over the Korea. Soil classification results by using 7 CPT soil classification charts were compared with those of USCS for the compiled database. Most proper CPT soil classification chart for Korean soil characteristics was evaluated and effective parameters for the soil classification from CPT were discussed. Finally interrelationship between CPT soil classification chart and USCS soil classification was evaluated.

  • Biodegradable barrier membrane has been demonstrated to have guided bone regeneration capacity on the animal study. The purpose of this study is to evaluate the effects of cultured calvarial cell inoculated on the biodegradable barrier membrane for the regeneration of the artificial bone defect. In this experiment 35 Sprague-Dawley male rats(mean BW 150gm) were used. 30 rats were divided into 3 groups. In group I, defects were covered periosteum without membrane. In group II, defects were repaired using biodegradable barrier membrane. In group III, the defects were repaired using biodegradable barrier membrane seeded with cultured calvarial cell. Every surgical procedure were performed under the general anesthesia by using with intravenous injection of Pentobarbital sodium(30mg/Kg). After anesthesia, 5 rats were sacrificed by decapitation to obtain the calvaria for bone cell culture. Calvarial cells were cultured with Dulbecco's Modified Essential Medium contained with 10% Fetal Bovine Serum under the conventional conditions. The number of cell inoculated on the membrane were $1{\times}10^6$ Cells/ml. The membrane were inserted on the artificial bone defect after 3 days of culture. A single 3-mm diameter full-thickness artificial calvarial defect was made in each animal by using with bone trephine drill. After the every surgical intervention of animal, all of the animals were sacrificed at 1, 2, 3 weeks after surgery by using of perfusion technique. For obtaining histological section, tissues were fixed in 2.5% Glutaraldehyde (0.1M cacodylate buffer, pH 7.2) and Karnovsky's fixative solution, and decalcified with 0.1M disodium ethylene diaminetetraacetate for 3 weeks. Tissue embeding was performed in paraffin and cut parallel to the surface of calvaria. Section in 7${\mu}m$ thickness of tissue was done and stained with Hematoxylin-Eosin. All the specimens were observed under the light microscopy. The following results were obtained. 1 . During the whole period of experiment, fibrous connective tissue was revealed at 1week after surgery which meant rapid soft tissue recovery. The healing rate of defected area into new bone formation of the test group was observed more rapid tendency than other two groups. 2 . The sequence of healing rate of bone defected area was as follows ; test group, positive control, negative control group. 3 . During the experiment, an osteoclastic cell around preexisted bone was not found. New bone formation was originated from the periphery of the remaing bone wall, and gradually extended into central portion of the bone defect. 4 . The biodegradable barrier membrane was observed favorable biocompatibility during this experimental period without any other noticeable foreign body reaction. And mineralization in the newly formed osteoid tissue revealed relatively more rapid than other group since early stage of the healing process. Conclusively, the cultured bone cell inoculated onto the biodegradable barrier membrane may have an important role of regeneration of artificial bone defects of alveolar bone. This study thus demonstrates a tissue-engineering the approach to the repair of bone defects, which may have clinical applications in clinical fields of the dentistry including periodontics.

  • This study was carried out to determine the promising method for the production of mother bulbs using garlic bulbs at the experimental field of Ange high school, Eusung in Korea from 1996 to 1998. The bulbs and bulbils were taken from a local cultivar 'Eusung' garlic. The growth of bulbil and yield potential were compared with different harvesting times and cultivation methods of bulbil. The harvesting time of bulbils which were capable of producing the mother bulbs was ranged from 15 days to 30 days after the emergence of flower stalks. The best results based on the yield and number of bulbs were obtained from the bulbils sampled at the just before garlic harvesting, 30 days after flower-stalk emergence. The mulching with transparent polyethylene(PE) film in bulbil cultivation was more effective for the plant growth and bulb formation than that with black PE film. The PE mulching from 30 days after seeding of bulbils to 30 days (late-April) before bulb harvesting was the moot effective for the increase of bulb yield. In the cultivation of garlic bulbils, application of lime (150 kg/10a) and compost (3,000 kg/10a) without any chemical fertilizers produced more bulbs in comparison with the standard level of fertilizer (N - $P_2O_5$ - $K_2O$ - compost - lime = 24.8 - 8.0 - 19.2 - 3,000 - 150 kg/10a). It was determined that the involucre seeding and assemble seeding of bulbils was effective and labour-saving methods for the production of mother bulbs from garlic bulbils. Input of labour force for the involucre seeding was decreased up to 61% as compared with the conventional drill seeding of bulbils. Mean yield from the cultivation of bulbil-derived single cloves was remarkably increased up to 95% in comparison with that of the common cloves. The larger size of single clove produced more yields, and the optimum planting density of single clove was $20{\times}10cm$. The cloves differentiated directly from bulbil showed to have higher yield potential than that from common cloves.

  • This study was performed to estimate the effects of cultured bone cell inoculated on porous type hydroxyaptite for the regeneration of the artificial alveolar bone defect. In this experiment 3 beagle dogs were used, and each of them were divided into right and left mandible. Every surgical intervention were performed under the general anesthesia by using with intravenous injection of Pentobarbital sodium(30mg/Kg). To reduce the gingival bleeding during surgery, operative site was injected with Lidocaine hydrochloride(l:80,000 Epinephrine) as local anesthesia. After surgery experimental animal were feeded with soft dietl Mighty dog, Frisies Co., U.S.A.) for 1 weeks to avoid irritaion to soft tissue by food. 2 months before surgery both side of mandibular 1st premolar were extracted and bone chips from mandibular body were obtained from all animals. Bone cells were cultured from bone chips obtained from mandible with Dulbecco's Modified Essential Medium contained with 10% Fetal Bovine Serum under the conventional conditions. Porous type hydroxyapatite were immerse into the high concentrated cell suspension solution, and put 4 hours for attachin the cells on the surface of hydroxyapatite. Graft material were inserted on the artificial bone defect after 3 days of culture. Before insertion of cellinoculated graft material, scanning electronic microscopic observation were performed to confirm the attachment and spreading of cell on the hydroxyapatite surface. 3 artificial bone defects were made with bone trephine drill on the both side of mandible of the experimental animal. First defect was designed without insertion of graft material as negative control, second was filled with porous replamineform hydroxyapatite inoculated with cultured bone marrow cells as expermiental site, and third was filled with graft materials only as positive control. The size of every artificial bone defect was 3mm in diameter and 3mm in depth. After the every surgical intervention of animals, oral hygiene program were performed with 1.0% chlorhexidine digluconate. All of the animals were sacrificed at 2, 4, 6 weeks after surgery. For obtaining histological section, tissus were fixed in 10% Buffered formalin and decalcified with Planko - Rycho Solution for 72hr. Tissue embeding was performed in paraffin and cut parallel to the surface of mandibular body. Section in 8um thickness of tissue was done and stained with Hematoxylin - Eosin. All the specimens were observed under the light microscopy. The following results were obtained : 1. In the case of control site which has no graft material, less inflammatory cell infiltration and rapid new bone forming tendency were revealed compared with experimental groups. But bone surface were observed depression pattern on defect area because of soft tissue invasion into the artificial bone defect during the experimental period. 2. In the porous hydroxyapatite only group, inflammatory cell infiltration was prominet and dense connective tissue were encapsulated around grafted materials. osteoblastic activity in the early stage after surgery was low to compared with grafted with bone cells. 3. In the case of porous hydroxyapatite inoculated with bone cell, less inflammatory cell infiltration and rapid new bone formation activity was revealed than hydroxyapatite only group. Active new bone formation were observed in the early stage of control group. 4. The origin of new bone forming was revealed not from the center of defected area but from the surface of preexisting bony wall on every specimen. 5. In this experiment, osteoclastic cell was not found around grafted materials, and fibrovascular invasion into regions with no noticeable foreign body reaction. Conclusively, the cultured bone cell inoculated onto the porous hydroxyapatite may have an important role of regeneration of artificial bone defects of alveolar bone.
