Urease and nitrification inhibitors with pig slurry effects on ammonia and nitrous oxide emissions, nitrate leaching, and nitrogen use efficiency in perennial ryegrass sward

  • Objective: The present study was conducted to assess the effect of urease inhibitor (hydroquinone [HQ]) and nitrification inhibitor (dicyandiamide [DCD]) on nitrogen (N) use efficiency of pig slurry for perennial ryegrass regrowth yield and its environmental impacts. Methods: A micro-plot experiment was conducted using pig slurry-urea 15N treated with HQ and/or DCD and applied at a rate of 200 kg N/ha. The flows of N derived from the pig slurry urea to herbage regrowth and soils as well as soil N mineralization were estimated by tracing pig slurry-urea 15N, and the N losses via ammonia (NH3), nitrous oxide (N2O) emission, and nitrate (NO3-) leaching were quantified for a 56 d regrowth of perennial ryegrass (Lolium perenne) sward. Results: Herbage dry matter at the final regrowth at 56 d was significantly higher in the HQ and/or DCD applied plots, with a 24.5% to 42.2% increase in 15N recovery by herbage compared with the control. Significant increases in soil 15N recovery were also observed in the plots applied with the inhibitors, accompanied by the increased N content converted to soil inorganic N (NH4++NO3-) (17.3% to 28.8% higher than that of the control). The estimated loss, which was not accounted for in the herbage-soil system, was lower in the plots applied with the inhibitors (25.6% on average) than that of control (38.0%). Positive effects of urease and/or nitrification inhibitors on reducing N losses to the environment were observed at the final regrowth (56 d), at which cumulative NH3 emission was reduced by 26.8% (on average 3 inhibitor treatments), N2O emission by 50.2% and NO3- leaching by 10.6% compared to those of the control. Conclusion: The proper application of urease and nitrification inhibitors would be an efficient strategy to improve the N use efficiency of pig slurry while mitigating hazardous environmental impacts.

전처리 방법에 따른 연근가루의 이화학적 특성 (Effect of Pretreatments on the Physicochemical Properties of Lotus Root Powder)

  • 연근의 활용도를 높이고 연근 가공식품 개발을 위한 기초 자료로 활용하고자 가공방법에 따라 연근가루를 제조한 후 분석한 결과는 다음과 같다. 색도를 측정한 결과에서 L값은 소금물처리 시료가 가장 높았고, a값과 b값은 식초물처리 시료에서 가장 높았다. 물결합력은 가공 처리한 소금물, 식초물, blanching처리 시료가 높았고, 아밀로오스 함량은 19.57-20.43%이었으나 시료간에 차이가 없었다. 팽윤력 및 용해도는 온도가 상승함에 따라 증가하였고, 팽윤력은 $65^{\circ}C$, 용해도에서는 $60^{\circ}C$까지 blanching처리 시료가 가장 높았으며, 그 이후부터는 대조구가 가장 높았다. X-ray 회절도에 의한 결정형은 회절각도($2{\Theta}$) $16.9^{\circ}$에서 강한 peak를 보였고, $14.6^{\circ}$, $22.2^{\circ}$, $23.8^{\circ}$에서 중간 peak를 보이는 B형의 결정 형태가 나타났다. 상대적 결정화도는 식초물처리 시료에서 가장 높았고, blanching처리 시료에서 가장 낮았다. DSC에 의한 호화온도 및 호화엔탈피는 식초물처리 시료에서 가장 높았고, blanching처리 시료에서 가장 낮았다. Amylogram에 의한 호화 특성에서 식초물처리 시료는 최고점도와 강하점도에서 가장 높았고, 총 치반점도에서 가장 낮아서 노화가 가장 느린 것으로 나타났다.

헬스케어 홈 서비스를 위한 데이터베이스 및 응용 서비스 구현 (Design and Implementation of Process Management Model applying Agent Technology)

  • 본 논문은 헬스케어 홈 환경에서 얻어진 정보를 헬스케어 데이터베이스로 구축하고, 이를 헬스케어 홈 서비스에 활용하는데 목적을 둔다. 특히, 본 논문에서 우리는 헬스케어 데이터베이스 스키마의 설계 및 이전에 우리가 개발한 헬스케어 통합 서비스를 지원하는 프레임워크(FSHIS)상에서 구축된 데이터베이스를 어떻게 사용할 것인가에 초점을 둔다. 헬스케어 정보는 다양한 센서로부터 수집한 데이터의 특정 저장타입에 맞추어 설계하였고, 헬스케어 홈 서비스의 사용목적에 따라 구축된 헬스케어 데이터베이스는 실제 스키마를 가진 기본 정보와 뷰 스키마로 제공되는 상황정보로 분류하였다. 첫 번째 기본 정보는 물리적 센서로부터 얻어지는 위치, 건강, 환경관련 가공없는 데이터와 개인 건강관련 프로파일 정보로 이루어진다. 두 번째의 상황정보는 기본 정보들을 이용하여 혼합 가공한 정보이다. 이 상황정보는 헬스케어 응용 서비스에 따라 다양한 뷰 스키마를 통해 얻어진다. 마지막으로, 구축된 헬스케어 데이터베이스의 실질적인 활용을 검증하기 위해, 우리는 본 연구실에서 개발된 FSHIS상에 구축된 데이터베이스와의 연동을 통해, 독거노인을 위해 생활 활동 영역으로 부터 필요한 기본정보와 상황정보를 이용하여 응급상황 호출 그리고 홈 가전 제어 등의 헬스케어 홈 모니터링 서비스를 보였다.

스마트 그리드를 위한 확장 홈 네트워크 기반의 AMI 시스템 설계 (A Design of an AMI System Based on an Extended Home Network for the Smart Grid)

  • 스마트 그리드란 기존의 전력망에 정보기술을 융합하여 에너지 효율을 최적화하는 차세대 전력망을 구성하는 기술의 하나이다. 본 논문에서는 스마트그리드를 효과적으로 구축하기 위하여 기존 홈 네트워크와 연동되고 효율적인 관리 기능을 제공하는 AMI 시스템을 제안 한다. 확장된 홈 네트워크 기반의 AMI 시스템은 스마트미터, 통신 모듈, 홈 게이트웨이, 보안 모듈, 미터 데이터 관리 시스템, 전력 응용 모듈 등으로 구성된다. 제안하는 홈 네트워크는 전력소모를 줄이고 효율적인 데이터 전송이 가능할 수 있도록 IEEE 802.15.4를 기반으로 설계하였다. 제안 홈 게이트웨이는 웹 서비스를 통해 외부 관리 시스템과 에너지 소비 정보 등을 실시간으로 교환할 수 있고, AMI 시스템은 인터넷을 통하여 홈 게이트웨이와 미터 데이터 관리 시스템 간의 양방향 통신이 가능하도록 설계되었다. 정보 전달의 안전성을 얻기 위하여 보안 알고리즘을 적용 하였으며 보안 알고리즘은 대칭적 블록 암호화 방식인 AES 알고리즘을 사용하였다. 제안 시스템을 사용하는 경우 본 연구에 제한적이기는 하지만 제어를 하지 않을 경우보다 전력 소비가 평균적으로 4~42%정도 줄어드는 것을 실험 결과에서 확인할 수 있었다.

Lactoferrin Protects Human Mesenchymal Stem Cells from Oxidative Stress-Induced Senescence and Apoptosis

  • Mesenchymal stem cells (MSCs) have been suggested as a primary candidate for cell therapy applications because they have self-renewal and differentiation capabilities. Although they can be expanded in ex vivo system, clinical application of these cells is still limited because they survive poorly and undergo senescence or apoptosis when transplanted and exposed to environmental factors such as oxidative stress. Thus, reducing oxidative stress is expected to improve the efficacy of MSC therapy. The milk protein lactoferrin is a multifunctional iron-binding glycoprotein that plays various roles, including reduction of oxidative stress. Thus, we explored the effect of lactoferrin on oxidative stress-induced senescence and apoptosis of human MSCs (hMSCs). Measurement of reactive oxygen species (ROS) revealed that lactoferrin inhibited the production of hydrogen peroxide-induced intracellular ROS, suggesting lactoferrin as a good candidate as an antioxidant in hMSCs. Pretreatment of lactoferrin suppressed hydrogen peroxide-induced senescence of hMSCs. In addition, lactoferrin reduced hydrogen peroxide-induced apoptosis via inhibition of caspase-3 and Akt activation. These results demonstrate that lactoferrin can be a promising factor to protect hMSCs from oxidative stress-induced senescence and apoptosis, thus increasing the efficacy of MSC therapy.

Spark Plasma Sintering 법으로 제조한 CoSb3 Skutterudite계 열전소재의 n형 첨가제 효과 (Effect of n-type Dopants on CoSb3 Skutterudite Thermoelectrics Sintered by Spark Plasma Sintering)

  • $CoSb_3$ Skutterudites materials have high potential for thermoelectric application at mid-temperature range because of their superior thermoelectric properties via control of charge carrier density and substitution of foreign atoms. Improvement of thermoelectric properties is expected for the ternary solid solution developed by substitution of foreign atoms having different valances into the $CoSb_3$ matrix. In this study, ternary solid solutions with a stoichiometry of $Co_{1-x}Ni_xSb_3$ x = 0.01, 0.05, 0.1, 0.2, $CoSb_{3-y}Te_y$, y = 0.1, 0.2, 0.3 were prepared by the Spark Plasma Sintering (SPS) system. Before the SPS synthesis, the ingots were synthesized by vacuum induction melting and followed by annealing. For phase analysis X-ray powder diffraction patterns were checked. All the samples were confirmed as single phase; however, with samples that were more doped than the solubility limit some secondary phases were detected. All the samples doped with Ni and Te atoms showed a negative Seebeck coefficient and their electrical conductivities increased with the doping amount up to the solubility limit. For the samples prepared by SPS the maximum value for dimensionless figure of merit reached 0.26, 0.42 for $Co_{0.9}Ni_{0.1}Sb_3$, $CoSb_{2.8}Te_{0.2}$ at 690 K, respectively. These results show that the SPS method is effective in this system and Ni/Te dopants are also effective for increasing thermoelectric properties of this system.

  • With the great advances in process understanding, sensor and instrumentation technology and modelling capability it is important to ask what if any practical benefits can the lake manager and operator look forward to. Here, we examine some of the more important problems facing operators of drinking reservoirs, hydro-lakes and lakes used predominantly for recreation and the environment. In drinking reservoirs the main problems originate from increased loadings of nutrients leading to increased biomass and biomass that may give rise to toxins, of anthropogenic chemicals such as metals and synthetic organics and of pathogens of different types. Hydro-lakes are predominantly plagued by problems arising from low oxygen levels in the hypolimnion and in recreational and environmentally sensitive lakes the biggest challenge for the operator is to maintain an existing or establish a new trophic hierarchy or protect the water body from foreign species. The control variables that are at an operator's disposal are the choice of lake water level, the modification of the water colunm stratification via a de-stratification system, the modification of the lake flow path with flow intervention. curtains, intervention in the catchments to modify the loadings flowing into a lake, manipulation of the trophic chain with introduction of new species and chemical dosing, the latter being of marginal use in a large lake. Each of these options is cost effective under certain-circumstances. We endeavour to provide a users guide for their application and show how, especially new instrumentation and modelling methodologies may be used to achieve an effective intervention.

Genetic Diversity of Multi-resistant Salmonella enterica Serotype Typhimurium Isolates from Animals and Humans

  • In this study, the genetic diversities of multi-resistant Salmonella typhimurium (ST) isolates were analyzed via the application of both pulsed field gel electrophoresis (PFGE) and Polymerase chain reaction (PCR) analysis methods, using 6 kinds of primers (REP, ERIC, SERE, BOX, P-1254 and OPB-17). And their discriminative abilities (DA) were also compared in order to determine the most effective and reliable analysis method. 118 S. typhimurium isolates, cultured from diverse animals and human patients in Korea beginning in 1993, were analyzed and subjected to a comparison of Simpson's index of diversity (SID), using both PFGE and PCR methods. PFGE by XbaI enzyme digestion allowed for discrimination into 9 pulsotypes, with high SID values (0.991) on the genomic DNA level. This shows that PFGE is a very discriminative genotypic tool, and also that multiple clones of S. typhimurium isolates had existed in domestic animals and humans in Korea since 1993. However, we could ultimately not to trace the definitive sources or animal reservoirs of specific S. typhimurium isolates examined in this study. Depending on the SID values, the combined method (7 kinds of method) was found to be the most discriminative method, followed by (in order) SERE-PCR, REP-PCR, ERIC-PCR, PFGE & OPB-17 (RAPD), P-1254 (RAPD), and BOX-PCR at the $80\%$ clone cut-off value. This finding suggests that the REP-PCR method (which utilizes 4 primer types) may be an alternative tool to PFGE for the genotyping of S. typhimurium isolates, with comparable cost, time, and labor requirement. The establishment of a highly reliable and discriminatory method for epidemiologic analysis is considered necessary in order for researchers to trace the sources of specific pathogens and, consequently, to control and prevent the spread of epidemic S. typhimurium isolates to humans.

동심축류가 유도되는 미세유체 소자 기반 Collagen Type I 미세섬유의 제작 (Fabrication of Collagen Type I Microfiber based on Co-axial Flow-induced Microfluidic Chip)

  • In this study, a co-axial flow induced microfluidic chip to fabricate pure collagen type I microfiber via the control of collagen type I and Na-alginate gelation process. The pure collagen type I microfiber was generated by selective degradation of Ca-alginate from 'Core-Shell' structured hydrogel microfiber. To make 'Core-Shell' structure, collagen type I solution was introduced into core channel and 1.5% Na-alginate solution was injected into side channel in microfluidic chip. To evaluatethe 'Core-Shell' structure, the red and green fluorescence substances were mixed into collagen type I and Na-alginate solution, respectively. The fluorescence substances were uniformly loaded into each fiber, and the different fluorescence images were dependent on their location. By immoblizing EpH4-Ras and C6 cells within collagen type I and Na-alginate solution, we sucessfully demonstrated the co-culture of EpH4-Ras and C6 cells with 'Core-Shell' like hydrogel microfiber for 5 days. Only to produce pure collagen type I hydrogel fiber, tri-sodium citrate solution was used to dissolve the shell-like Ca-alginate hydrogel fiber from 'Core-Shell' structured hydrogel microfiber, which is an excellent advantage when the fiber is employed in three-dimensional scaffold. This novel method could apply various application in tissue engineering and biomedical engineering.

Ground response of a gob-side gateroad suffering mining-induced stress in an extra thick coal seam

  • This paper presents an investigation of the ground response of a gob-side gateroad suffering mining stress induced by a 21 m-thick coal seam extraction. A field observation, including entry convergence and stress changes monitoring, was first conducted in the tailgate 8209. The observation results of entry convergence showed that, during the adjacent panel 8210 retreating period, the deformation of the gob-side gateroad experienced a continuous increase stage, subsequently, an accelerating increase stage, and finally, a slow increase stage. However, strong ground response, including roof bending deflection, rib extrusion and floor heave, occurred during the current panel 8209 retreating period, and the maximum floor heave reached 1530 mm. The stress changes within coal mass of the two ribs demonstrated that the gateroad was always located in the stress concentrated area, which responsible for the strong response of the tailgate 8209. Subsequently, a hydraulic fracture technique was proposed to pre-fracture the two hard roofs above the tailgate 8209, thus decreasing the induced disturbance on the tailgate. The validity of the above roof treatment was verified via field application. The finding of this study could be a reference for understanding the stability control of the gob-side gateroad in extra thick coal seams mining.
