Expression and phosphorylation analysis of soluble proteins and membrane-localised receptor-like kinases from Arabidopsis thaliana in Escherichia coli

  • Molecular and functional characterization of proteins and their levels is of great interest in understanding the mechanism of diverse cellular processes. In this study, we report on the convenient Escherichia coli-based protein expression system that allows recombinant of soluble proteins expression and cytosolic domain of membrane-localised kinases, followed by the detection of autophosphorylation activity in protein kinases. This approach is applied to regulatory proteins of Arabidopsis thaliana, including 14-3-3, calmodulin, calcium-dependent protein kinase, TERMINAL FLOWER 1(TFL1), FLOWERING LOCUS T (FT), receptor-like cytoplasmic kinase and cytoplasmic domain of leucine-rich repeat-receptor like kinase proteins. Our Western blot analysis which uses phospho-specific antibodies showed that five putative LRR-RLKs and two putative RLCKs have autophosphorylation activity in vitro on threonine and/or tyrosine residue(s), suggesting their potential role in signal transduction pathways. Our findings were also discussed in the broader context of recombinant expression and biochemical analysis of soluble and membrane-localised receptor kinases in microbial systems.

  • The size-dependent nonlinear thermomechanical vibration analysis of pre- and post-buckled tapered two-directional functionally graded (2D-FG) microbeams is presented in this study. In the context of the modified couple stress theory, the formulations are derived based on the parabolic shear deformation beam theory and von Karman nonlinear strains. Different thermomechanical material properties are assumed to be temperature-dependent and smoothly vary in both length and thickness directions using the power law and the physical neutral axis concept is employed. The nonlinear governing equations are derived using the Hamilton principle and the resulting variable coefficient equations of motion are solved using the differential quadrature method (DQM) and iterative Newton's method for clamped-clamped and simply supported boundary conditions. Comparison studies are presented to validate the derived model and solution procedure. The impacts of induced thermal moments, temperature power index, two gradient indices, nonuniform cross-section, and microstructure length scale parameter on the frequency-temperature configurations are explored for both clamped and simply supported microbeams.

On vibrations of functionally graded carbon nanotube (FGCNT) nanoplates under moving load

  • This article develops a nonclassical size dependent nanoplate model to study the dynamic response of functionally graded carbon nanotube (FGCNT) nanoplates under a moving load. Both nonlocal and microstructure effects are incorporated through the nonlocal strain gradient elasticity theory. To investigate the effect of reinforcement orientation of CNT, four different configurations are studied and analysed. The FGM gradation thorough the thickness direction is simulated using the power law. In the context of the first order shear deformation theory, the dynamic equations of motion and the associated boundary conditions are derived by Hamilton's principle. An analytical solution of the dynamic equations of motion is derived based on the Navier methodology. The proposed model is verified and compared with the available results in the literature and good agreement is found. The numerical results show that the dynamic performance of FGCNT nanoplates could be governed by the reinforcement pattern and volume fraction in addition to the non-classical parameters and the moving load dimensionless parameter. Obtained results are reassuring in design and analysis of nanoplates reinforced with CNTs.

A Proposal of Quality Assessment for System Model

  • Recently, the increased complexity of systems has made systems engineering necessary. It is very useful for system designers to understand the whole context of the concerned system based on systems engineering. A system model can be used to describe the outcome of a system design. A system model describes the system from the viewpoint of the stakeholder's needs using the mutually exclusive and collectively exhaustive principle. A system model can be used to smoothly design a large and complicated system based on the systems engineering development process. Many companies and countries are attempting to apply model-based systems engineering, and the significance of the system model quality is increasing as system models are referenced during system development. In this paper, we propose a quality assessment method for ontology which is one of system models by focusing on the system development process. First, in this process, a system developer should explicitly show the relationship between viewpoints. Then, the system developer should select dependent rather than independent viewpoints. With dependent viewpoints, each viewpoint used to describe the system has some logical relationship. The set of viewpoints makes it possible to show, not only tangible and physical system parts, but also conceptual system parts. In this paper, we develop an ontological system model of a Japanese weather observation system. By comparing some ontological system models, we verify the effectiveness of explicitly describing the relationships between viewpoints and select dependent viewpoints.

Analysis of the Apoptotic Mechanisms of Snake Venom Toxin on Inflammation-induced HaCaT Cell-line

  • Objectives : In this study, the roles of Interleukin (IL)-4 and Signal transducer and activator of transcription 6 (STAT6), which have been reported to play a role in the pathogenesis of inflammation and cancer, were evaluated in snake venom toxin (SVT)-induced apoptosis. Methods : Inflammation was induced in human HaCaT kerationocytes, by lipopolysaccharide (LPS; $1{\mu}g/mL$) or tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$), followed by treatment with SVT (0, 1, or $2{\mu}g/mL$). Cell viability was assessed by MTT assays after 24 h, and the expression of levels of IL-4, STAT6, and the apoptosis-related proteins p53, Bax, and Bcl-2 were evaluated by western blotting. Electro mobility shift assays (EMSAs) were performed to evaluate the DNA binding capacity of STAT6. Results : MTT assays showed that inflammation-induced growth of HaCaT cells following LPS or TNF-${\alpha}$ stimulation was inhibited by SVT. Western blot analysis showed that p53 and Bax, which promote apoptosis, were increased, whereas that of Bcl-2, an anti-apoptotic protein, was decreased in a concentration-dependent manner in LPS- or TNF-${\alpha}$-induced HaCaT cells following treatment with SVT. Moreover, following treatment of HaCaT cells with LPS, IL-4 concentrations were increased, and treatment with SVT further increased IL-4 expression in a concentration-dependent manner. Western blotting and EMSAs showed that the phosphorylated form of STAT6 was increased in HaCaT cells in the context of LPS- or TNF-${\alpha}$-induced inflammation in a concentration-dependent manner, concomitant with an increase in the DNA binding activity of STAT6. Conclusion : SVT can effectively promote apoptosis in HaCaT cells in the presence of inflammation through a pathway involving IL-4 and STAT6.

클래스 종속 반연속 HMM을 이용한 립싱크 시스템 최적화 (Lip-Synch System Optimization Using Class Dependent SCHMM)

  • 기존의 립싱크 시스템은 음소 분할 후, 각각의 음소를 인식하는 2단계의 과정을 거쳤다. 하지만, 정확한 음소 분할의 부재와 음성이 끊긴 분할 된 음소로 이루어진 훈련 데이터들은 시스템의 전체 성능을 크게 떨어뜨렸다. 이런 문제를 해결하기 위해 Head-Body-Tail (HBT) 모델을 이용한 단모음 연속어 인식 기술을 제안한다. 주로 소규모 어휘를 다루는데 적합한 HBT 모델은 Head 와 Tail 부분에 문맥 종속 정보를 포함하여 앞 뒤 문맥에 따른 조음효과를 최대한 반영한다. 또한, 7개의 단모음을 입모양이 비슷한 세 개의 클래스로 분류하여, 클래스에 종속적인 코드북 3개를 가진 반연속HMM (Hidden Markov Model)을 적용하여 시스템을 최적화하고, 변이 부분이 큰 단어의 처음과 끝은 연속HMM의 8 믹스쳐 가우시안 구조를 사용하여 모델링하였다. 제안한 방법은 HBT구조의 연속HW과 대등한 성능을 보이지만, 파라미터 수는 33.92% 감소하였다. 파라미터 감소는 계산 양을 줄여주므로, 시스템이 실시간으로 동작 가능하게 한다.

Sirtuins in Cancer: a Balancing Act between Genome Stability and Metabolism

  • Genomic instability and altered metabolism are key features of most cancers. Recent studies suggest that metabolic reprogramming is part of a systematic response to cellular DNA damage. Thus, defining the molecules that fine-tune metabolism in response to DNA damage will enhance our understanding of molecular mechanisms of tumorigenesis and have profound implications for the development of strategies for cancer therapy. Sirtuins have been established as critical regulators in cellular homeostasis and physiology. Here, we review the emerging data revealing a pivotal function of sirtuins in genome maintenance and cell metabolism, and highlight current advances about the phenotypic consequences of defects in these critical regulators in tumorigenesis. While many questions should be addressed about the regulation and context-dependent functions of sirtuins, it appears clear that sirtuins may provide a promising, exciting new avenue for cancer therapy.

Effect of nitrogen doping and hydrogen confinement on the electronic properties of a single walled carbon nanotube

  • This paper addresses the effect of dopants on the electronic properties of zigzag (8, 0) semiconducting single walled carbon nanotubes (SWCNTs), using extended Hückel theory combined with nonequilibrium Green’s function formalism. Through appropriate dopant concentrations, the electronic properties of SWCNTs can be modified. Within this context, we present our ongoing investigation on (8, 0) SWCNTs doped with nitrogen. Quantum confinement effects on the electronic properties of the SWCNTs have also been investigated. The obtained results reveal that the electronic properties of SWCNTs are strongly dependent on the dopant concentration and modification of electronic structures by hydrogen confinement.

Strongly coupling partitioned scheme for enhanced added mass computation in 2D fluid-structure interaction

  • A numerical model for fluid-structure interactions (abbr. FSI) is presented in the context of sloshing effects in movable, partially filled tanks to improve understanding of interactions between the fluid and the dynamics of a tank flexibly attached to a vehicle. The purpose of this model is to counteract the penalizing impact of the added mass effect on classical partitioned FSI coupling scheme: the proposed investigation is based on an added mass corrected version of the classical strongly coupled partitioned scheme presented in (Song et al. 2013). Results show that this corrected version systematically allows convergence to the coupled solution. In the rare cases where convergence is already obtained, the corrected version significantly reduces the number of iterations required. Finally, it is shown that the convergence limit imposed by added mass effect for the non-corrected coupling scheme, is directly dependent on the aspect ratio of the fluid domain and highly related to the precision order of the temporal discretization scheme.

The test object can be material, component, plant, earth, environment etc. Total qualify management, total productivity management, concurrent engineering and many other essential ingredients of success in plant engineering and manufacturing industry are dependent on NDE far success and good returns on investments.
