Variation of Pectin, Catechins and Caffein Contents in Korean Green Tea (Camellia sinensis L.) by Harvesting Time and Processing Recipe (채엽시기 및 제조법에 따른 한국산 녹차의 펙틴, 카테친, 카페인 함량 변이)

  • This study was conducted to find out the variation in some medicinal components of Korean green tea by processing recipe and harvesting time in Bongsan-ri(steamroasted green tea) and Buchun-ri(roasted green tea) area. Total chlorophyll content of green tea grown at Bongsan-ri and Buchun-ri was 293mg% and 275mg% and ratio of chlorophyll a to chlorophyll b was 70:30 and 67:33 respectively. The later harvested one had the higher chlorophyll content but the chlorophyll content of the last harvested green tea at Buchun-ri decreased more sharply. Water-soluble pectin of green tea produced at Bongsan-ri and Buchun-ri was 2.05% and 1.84%, respectively. It also increased at the later harvested ones. Total amino acids content of Bongsan-ri and Buchun-ri tea was 2,156mg% and 1,723mg%, respectively. The later harvested tea showed the remarkably decreased. Caffein and catechins of Bongsan-ri and Buchun-ri tea were 2.03%, 11.52% and 2.62%, 14.05%, respectively, Total free catechins content of Bongsan-ri and Buchun-ri tea was 35.59% and 42.73%, and ester type was 58.09% and 44.47%, respectively. Extracted catechins and caffeins of green tea were increased when extracting water temperature was raised from $65^{\circ}C$ to 85$^{\circ}C$ and leaching time was lengthened from 3min. to 9min. About 89% catechins and 97% caffeins of green tea was extracted by nine minutes soaking in 85$^{\circ}C$ water.

Physiological Responses of Chilling - Tolerant and Susceptible Rice Cultivars during Chilling Stress and Subsequent Recovery (저온스트레스와 회복기간중 저온 내성 및 감수성 벼 품종의 생리적 반응)

  • A chilling-tolerant japonica-type and a chilling-susceptible indica-type of rice (Oryza sativa L.) plants were compared with respect to various physiological parameters during chilling and subsequent recovery. The japonica-type and the indica-type of rice cultivars used were Ilpumbyeo and Taebaekbyeo, respectively. The two rice cultivars exhibited little or no differences in the changes of leaf fresh weight and chlorophyll content during chilling at $5^{\circ}C$ for 3 days. During subsequent recovery at $25^{\circ}C$, however, Ilpumbyeo restored its growth more rapidly than Taebaekbyeo. Since the changes of relative water content, malondialdehyde production, an estimate of lipid peroxidation, and chlorophyll fluorescence were significantly different in the two rice cultivars during the chilling and subsequent recovery, they were found to be more sensitive physiological parameters than fresh weight and chlorophyll content. However, the differences in relation to water content, malondialdehyde production, and chlorophyll fluorescence between the two rice cultivars were smaller during chilling than those during subsequent recovery. These results suggest that relative water content, malondialdehyde production, and chlorophyll fluorescence could widely be used as important physiological parameters for screening chilling-tolerant plants.

Effects of Rare Earth and Nitrogen Application on the Growth and Nitrate Content of Chicory (희토 및 질소시용이 치커리의 성장과 질산태질소 함량에 미치는 영향)

  • The effect of rare earth(RE) at three difference levels of nitrogen on the growth and nitrate content of chicory (Cichorium intybus L.) were studied in terms of leaf size, fresh and dry weight increase, and chlorophyll and nitrate content grown in plastic pots within glasshouse. Leaf size was enlarged as the level of nitrogen applied was high, and leaf within, length, and thickness of leaf were increased by RE treatment showing significant difference at high nitrogen(N+1) plot. As the level of nitrogen applied was high, as fresh and dry weight per plant was increased significantly(p<0.05), and sprinkling cabbage with RE increased fresh and dry matter yield to 2∼12, 4∼6.2% more, respectively. Dry matter content of Chinese cabbage was increased by RE sprinkling. At all levels of nitrogen fertilized chlorophyll contents were increased by RE treatment. As the level of nitrogen was high, as the content of chlorophyll was highly increased by RE. Nitrate was accumulated more at high level of nitrogen application, but nitrate was decreased by RE application, 40% decrease at very high nitrogen plot(N+2). RE stimulated the growth of chicory with high chlorophyll content, and showed the possibility producing high quality agricultural products low in nitrate content.

Effect of Macronutritional-element Deficiencies on the Growth and Photosynthetic Activity of Chlorella Cells (Chlorella의 다량원소 결핍에 따른 생장 및 광합성능의 변화)

  • Chlorella ellipsoidea cells were cultured in a phosphate, magnesium, sulfur or potassium-free medium. Some portions of cells were taken out at intervals during the culture, and physiological activities such as growth rate, reproduction, photosynthetic activity, and biosynthesis of cholorophyll of macro-element deficient cells were measured. 1) Generally, growth rate, reproduction, photosynthetic activities, and biosynthesis of chlorophyll of the macro-element deficient cells decreased more or less, compared with those of the normal cells. 2) The growth and reproduction of the algal cells in sulfur, or magnesium free medium were retarted severely with the chlorosis ; the photosynthetic activity and the content of chlorophyll showed the same tendency. 3) It is considered that the decrease in growth rate of macro-element deficient Chlorella cells is due to the decrease in photosynthetic activity owing to the decrease in chlorophyll content of the cells.

  • This study was performed to investigate changes of chlorophyll and carotene contents by cooking methods and parts of pumpkin(zucchini, green pumpkin, and sweet pumpkin). The hightest chlorophyll content of 3-parts(peel, cortex, and seed), all of pumpkin was peel part for chlorophyll contents by cooking methods of all kinds of pumpkin. It was the highest when cooked by non-batter frying. Carotene was relativity stable with cooking. But in case of sweet pumpkin, steaming cooking method was reduced carotene contents. β-carotene of zucchini, green pumpkin, and sweet pumpkin was higher than standard contents.

  • The relationship between storage stability and chlorophyll contents of dried laver(DL), roasted laver(RL) and roasted seasoned laver(RSL) according to packaging during storage at 20$\pm$0.5$^{\circ}C$ were investigated by measuring the acid value(AV) and the peroxide value(POV). There results are as follows; 1.The storage stability was decreased in the rank of 0 < 3< 6 < 9 months by storage term, RL < DL < RSL by the kind of laver products and PP/PE/AL/PE/LLDPE(Al) < PP/PE(PP) y packaging. 2. When the silica gel was added to RSL, the storage stability was improved. The more the addition level of silica gel was increased, the more the storage stability was improved. The rank order was 2g < 4g in RSL. 3. Reduction of the total chlorophyll content in RSL were deacreased to 10% when packed with Al and 24% when packed with PP.

Bilin and Bilinone Chlorophyll Catabolite Content in the Hamamelidaceae Autumnal Leaves

  • In order to facilitate the quantification in autumnal Hamamelidaceae leaves, a HPLC method was used for the determination of two chlorophyll catabolites and their isomers: bilin-type (1) and bilinone-type (2) ones. The separation was done on a RP-C4 column with a gradient solvent system of 0.1% trifluoroacetic acid aqueous-methanol at the flow-rate of 0.2 mL/min and detected at 244 nm. The quantity of bilin-type (1) and bilinone-type (2) chlorophyll catabolite isomers from ten species of Hamamelidaceae autumnal leaves methanol extracts: Corylopsis pauciflora, Corylopsis spicata, Forthergilla major, Hamamelis intermedia, Hamamelis japonicum, Hamamelis japonicum var. flavopurpurscens, Hamamelis virginiana, Parrotiopsis jacquemontiana, Parrotia persica and X Sycoparrotia semidecidua were from 0.85 mg/g ~ 57.50 mg/g for bilin-type isomers (1) and 3.40 mg/g ~ 49.30 mg/g for bilinone-type isomers (2). The results obtained gave insight in quantitative bilintype (1) and bilinone-type (2) chlorophyll catabolite composition of the Hamamelidaceae plant species autumnal leaves.

Effect of Blue, Red and Far-red Lights on Seeding Growth and Cotyledon Chlorophyll Content of Lagenaria siceraria Standl (광질처리에 따른 박 유묘의 생장 및 자엽의 엽록소 함량 변화)

  • Various types of the seeding of bottle gourd widely used as a rootstock of watermelon has been required to satisfy the farmers need. The study was done to determine the effect of light quality of blue, red and far-red lights treated with light emitting diodes on growth and morphology of bottle goured seeding and chlorophyll content of its cotyledons. The lights were treated in the growth chamber for 7 days to the seeding elapsed 8 days after sowing under natural condition, and 64 hole trays with commercial bedsoil. Plant height, length and diameter of hypocotyl, leaf area of cotyledon and first true leaf, its leaf length, number of true leaves, fraction and total dry weight were measured. Red light shortened and slenderized the hypocotyl, which lengthened by far-red light and thickened by blue light. Plant height was declined in order of far-red light treatment, blue and red lights mainly due to difference of hypocotyl length, Area and length of the first true leaf became smaller and shorter under far-red light than under the other lights. However, blue light increased leaf area of cotyledons. Two cultivars cv. Yongjadaemok and cv. Kunghap had different response to the light treatments in total seedling dry weight(dw); far-red and red light treatments showed the greatest and the least dw of hypocotyl, respectively, while blue and red lights did the greatest dw of the other organs. Among the ratio of each organ dw to total dw, those of hypocotyl and true leaves were different between the light treatments; the highest ratio of hypocotyl dw to total dw was observed in far-red light treatment but the lowest was in red light treatment. Those of the true leaves were the lowest in far-red light and similar response in blue or red light treatment. Chlorophyll content of cotyledons was decreased in order of red light treatment, blue and far-red lights, meaning that short period light treatment may influence photosynthesis of seeding and afterward its growth.

Influence of Nitrate Against Effect of Cadmium on Growth and Rubisco in Seedling of Tobacco. (담배 유식물의 생장과 Rubisco에 미치는 카드뮴의 효과에 대한 질산염의 영향)

  • This investigation was performed to study the influence of Cd and nitrate on growth, and chlorophyll and photosynthetic enzymes in seedling of tobacco. Growth inhibition by Cd was not recovered by nitrate. Chlorophyll levels were reduced by Cd. The combination of Cd and low concentration of nitrate decreased the chlorophyll content compared to that in plants exposed only to Cd. Activity and content of rubisco at Cd treatment was significantly lesser than in plants receiving no treatment, These data suggest that rubisco activity was associated with an amount of rubisco protein, and that the activation and synthesis of rubisco is inhibited by Cd. Both the activity and content of rubisco decreased by Cd were more decreased by nitrate. A similar change pattern was also observed in activity and content of rubisco activase. These results suggest that Cd- and nitrate-induced changes of rubisco could be correlated with rubisco activase, and that nitrate was concerned in not only the activation and synthesis of rubisco directly, but also rubisco activase leading to a large change in rubisco.
