Optimization of Cultivation and Extraction Conditions of Pupae-Cordyceps for Cordycepin Production

  • Cordycepin is a characteristic bioactive compound of Cordyceps militaris with various beneficial effects. Cordyceps grows on both grains and insects, and the content of cordycepin varies depending on the cultivation conditions. In this study, the effect of culture conditions on the cordycepin content was analyzed and the extraction conditions were optimized. Analysis of cordycepin content in Pupae-Cordyceps found that it was highly affected by temperature in culture conditions. In the case of mycelium, it grows well at 20 and 25 ℃, but not at 30 ℃. However, the content of cordycepin was highest at 30℃ and less at 20 ℃. The fruiting body also showed a similar tendency: growth was 20 ℃ > 25 ℃ > 30 ℃, but the cordycepin content was 30 ℃ > 25 ℃ > 20 ℃. The content of cordycepin decreased after the fruiting bodies were produced. Next, extraction conditions such as solvent and time were optimized for maximum cordycepin content using response surface methodology (RSM). There was a large difference in the content of cordycepin according to the content of ethanol and the extraction temperature. Through RSM, it was confirmed that the optimum condition for extraction of cordycepin was 48.9 ℃ using 49.0% ethanol, and 160.9 mg/g extract could be obtained under this condition. In conclusion, this study suggested the optimized conditions for the cultivation and extraction of Pupae-Cordyceps for maximizing the content of cordycepin, and this may be applied to the discovery of materials using cordycepin.

가상현실 기반의 치아발치 수기 훈련을 위한 교육콘텐츠 개발 (Development of Educational Content for Dental Extraction Skill Training Using Virtual Reality Technology)

  • 본 연구의 목적은 치의학 임상실습 장면에서 활용이 가능한 가상현실 기반의 치아발치 교육콘텐츠를 개발하는 것이다. 이러한 목적을 달성하기 위해 '학습내용 분석-설계모형 도출-개발-타당도 검증-수정 및 콘텐츠 완성'의 단계를 거쳐 치아발치 실습용 교육콘텐츠를 개발하였다. 연구결과, 3D모델링을 적용하여 환자의 안면해부학적 모델을 구축하고, 치아발치 수술과정 시나리오를 바탕으로 가상현실 기술을 적용한 단계별 수술장면을 에니메이션으로 구축하였다. 개발된 콘텐츠에 대한 전문가 평가를 실시하였으며, 인터페이스 타당도(M: 4.81, SD: 0.72)와 내용타당도(M:4.66, SD: 0.71)를 검증한 결과 타당하게 설계된 것으로 분석되었다. 따라서 개발된 치아발치 실습용 교육콘텐츠는 치의학 전공생의 임상수행 능력을 향상시키는데 적절한 도구로 판단된다. 본 연구를 토대로 의료 분야의 다양한 실습장면을 지원할 수 있는 가상현실 기술 기반의 교육용 콘텐츠 개발 연구가 활성화되기를 기대한다.

전탕 방법 및 전탕 시간에 따른 육미지황탕 전탕액 비교 연구 (The Comparative Study on Decoctions of Yukmijihwang-tang (Liuweidihuang-tang) Extracted by Different Extraction Method and Extraction Time)

  • Objectives : This study was performed to compare the differences between decoctions extracted by different extraction method and extraction time. Methods : Decoctions were prepared with pressed or non-pressed extraction conditions for 60, 120 and 180min. The yields of extracts, sugar contents, hydrogen ion concentrations(pH), the contents of reference compounds in Yukmijihwang-tang(Liuweidihuang-tang) were investigated. Results : The yields and the dissolved solid content containing sugar content of decoctions were more in pressed extraction method than unpressed extraction method, and they tended to be increased as extraction time increased. The pH values of decoctions methods did not show significant differences between pressed and unpressed extraction methods or extraction times. Most of reference compounds showed higher contents in pressed extraction method than unpressed extraction method and their contents were augmented according to increase of extraction time. The content of paeonol was decreased when extracted in more than 120min with pressed extraction method and tended to be decreased as extraction time increased in unpressed extraction method. Conclusions : The pressed extraction with long extraction time could be useful for decoction of Yukmijihwang-tang(Liuweidihuang-tang). However, another ingredients possible to decrease in such condition need to be considered to determine suitable extraction condition.

In Vitro Evaluation of Antioxidant Activity of Lycium barbarum Hot Water Extract and Optimization of Production Using Response Surface Methodology

  • This study is concerned with the optimization of the manufacturing process of a hot water extract containing antioxidant activity from Lycium barbarum, traditionally known to have various physiological activities. For the establishment of the optimization process, the central composite design of response surface methodology(RSM) was used. Thirteen extraction processes were performed by encoding the independent variables, extraction temperature (65.9℃-94.1℃) and extraction time (2.59 hr-5.41 hr). As a result of the experiment, the optimal manufacturing conditions for the extract were 340.0 mg/100 g of GAE at an extraction temperature of 94.1℃ and an extraction time of 5 hr. The maximum yield of flavonoids was 22.44 mg/100 g of HES at an extraction temperature of 94.1℃ and an extraction time of 4 hr. The conditions for producing the extract with the maximum antioxidant capacity (DPPH 92.12%) were 90℃ and 4.5 hr extraction time. Therefore, the optimal manufacturing process conditions for extracts containing total phenol content, flavonoid content, and DPPH radical scavenging activity, which are dependent variables, were extraction temperature of 90-95℃ and extraction time of 4 hr, which were not significantly different from the actual values. Therefore, Lycium barbarum extract rich in total phenol and flavonoid content related to antioxidant function is expected to be used as a functional food and cosmetic material.

Analysis of Anti-adipogenic Constituents of Cordyceps militaris Using High Performance Liquid Chromatography-Diode Array Detection in Different Samples: Comparison with Anti-adipogenic Activity

  • We previously isolated cordycepin, guanosine and tryptophan from Cordyceps militaris as antiadipogenic constituents. For the quality control of C. militaris for anti-adipogenic activity, simultaneous analytical method using high-performance liquid chromatography (HPLC)-diode array detection (DAD) was developed and validated. Quantitation of these compounds in various Cordyceps samples from different sources and various extraction methods were conducted using developed method. Our study shows that natural Cordyceps and host insect possess higher content than cultured ones and fruiting bodies, respectively. The content of cordycepin showed great difference in different C. militaris samples whereas trytophan content was similar in tested samples. Addition of water to extraction solvent greatly increased the yield of guanosine and tryptophan. High temperature and longer extraction time increased yield of guanosine, whereas the content of trytophan was decreased in high temperature during extraction with water. Extraction using ultrasonic apparatus slightly increased extraction efficiency. Cordycepin, however, has little variation in different extraction method tested. Strong anti-adipogenic activity was observed in the samples that contain all the three constituents. Taken together, quantitation of these compounds using developed analytical method might provide basic requirement for the anti-adipogenic activity of C. militaris.

Effect of Korean Red Ginseng extraction conditions on antioxidant activity, extraction yield, and ginsenoside Rg1 and phenolic content: optimization using response surface methodology

  • Background: Extraction conditions greatly affect composition, as well as biological activity. Therefore, optimization is essential for maximum efficacy. Methods: Korean Red Ginseng (KRG) was extracted under different conditions and antioxidant activity, extraction yield, and ginsenoside Rg1 and phenolic content evaluated. Optimized extraction conditions were suggested using response surface methodology for maximum antioxidant activity and extraction yield. Results: Analysis of KRG extraction conditions using response surface methodology showed a good fit of experimental data as demonstrated by regression analysis. Among extraction factors, such as extraction solvent and extraction time and temperature, ethanol concentration greatly affected antioxidant activity, extraction yield, and ginsenoside Rg1 and phenolic content. The optimal conditions for maximum antioxidant activity and extraction yield were an ethanol concentration of 48.8%, an extraction time 73.3 min, and an extraction temperature of $90^{\circ}C$. The antioxidant activity and extraction yield under optimal conditions were 43.7% and 23.2% of dried KRG, respectively. Conclusion: Ethanol concentration is an important extraction factor for KRG antioxidant activity and extraction yield. Optimized extraction conditions provide useful economic advantages in KRG development for functional products.

Main Content Extraction from Web Pages Based on Node Characteristics

  • Main content extraction of web pages is widely used in search engines, web content aggregation and mobile Internet browsing. However, a mass of irrelevant information such as advertisement, irrelevant navigation and trash information is included in web pages. Such irrelevant information reduces the efficiency of web content processing in content-based applications. The purpose of this paper is to propose an automatic main content extraction method of web pages. In this method, we use two indicators to describe characteristics of web pages: text density and hyperlink density. According to continuous distribution of similar content on a page, we use an estimation algorithm to judge if a node is a content node or a noisy node based on characteristics of the node and neighboring nodes. This algorithm enables us to filter advertisement nodes and irrelevant navigation. Experimental results on 10 news websites revealed that our algorithm could achieve a 96.34% average acceptable rate.

추출온도 및 함수율에 따른 오미자 종자유의 이화학적 특성 및 항산화활성 (Physicochemical Characteristics and Antioxidant Activities of Omija (Schizandra chinensis Baillon) Seed Oil Extracted at Different Temperatures and Moisture Contents)

  • Omija seed oil was extracted from Omija seeds, a by-product of Omija, using an eco-friendly pressed extraction method. Physicochemical properties and antioxidant activities of 12 extracts using different combinations of seed moisture content (5, 7.5, and 10%) and extraction temperature (25, 49, 75, and 100℃) were then investigated. The highest extraction yield was 31.33% at moisture of 5% and temperature of 75℃. The lowest acid value was 4.18±0.25 at moisture of 5% and temperature of 49℃or moisture of 7.5% and temperature of 25℃. Peroxide value, iodine value, and saponification value were the lowest at 0.64±0.56 meq/kg, 159.38± 6.03, and 57.60±9.40, respectively, at moisture of 5% and temperature of 25℃. The content of total polyphenolics was the highest at 4,413±125 mg TAE/100 g when the moisture content was 10% and the extraction temperature was 25℃. DPPH radical scavenging activities of oil extracts at 20~100 mg/mL were 28.68±7.30~87.65±2.20%. The maximum antioxidant activity and IC50 were 87.65±2.20% and <20 mg/mL, respectively, for extract obtained at moisture of 10% and temperature at 100℃. As a result, the extraction yield, acid value, peroxide value, iodine value, saponification value, and so on were excellent at moisture content of 5% and extraction temperature of 25℃ or 49℃. However, the content of total polyphenolic compounds and antioxidant activity were the highest at moisture of 10% and extraction temperature of 100℃. In conclusion, extracting Omija seed oil from Omija seeds, a by-product of Omija, is effective with a pressed extraction method.

추출조건에 따른 진달래 화분의 이화학적 특성 (Extraction Conditions for Rhododendron mucronulatum Pollen)

  • 진달래 화분의 다양한 이용성과 기능성에 관한 연구를 위한 기초연구의 일환으로 추출조건별 이화학적 특성을 조사하였다. 페놀성 화합물과 DPPH에 의한 전자공여능은 물보다 80%에탄올이 우수하였으며, 용매비에 따른 페놀성 화합물과 전자공여능은 용매비가 20배에서 높은 것으로 나타났다. 추출온도에 따른 페놀성 화합물은 $45^{\circ}C$에서 347.60 mg%로 높게 나왔으며, 전자공여능은 $25^{\circ}C$에서 67.93%로 높게 나타났다. 추출시간에 따른 페놀성 화합물의 추출물은 6시간동안 추출한 추출물이 312.63 mg%로 가장 높았으며, 전자공여능은 추출시간에 따라 크게 차이가 나지 않았으나, 바로 추출하였을 때 가장 높게 나타났다. 추출 횟수에 따른 페놀성 화합물의 단위당 추출물과 전자공여능은 2회 추출이 높은 것으로 나타났다. 따라서 진달래 화분의 추출물은 페놀성 화합물과 DPPH에 의한 전자공여 활성은 용매비 20배, 추출온도 $25{\sim}45^{\circ}C$, 추출횟수 2회, 추출시간 6시간에서 전반적으로 우수한 것으로 나타났다.

Compositional differences of Bojungikgi-tang decoctions using pressurized or non-pressurized extraction methods with variable extraction times

  • Objectives : In other to determine the optimal extraction conditions, the various Bojungikgi-tang (BJIGT) decoctions prepared by different pressure levels and different extraction times were compared and evaluated in terms of the extract yield and the total soluble solid content. Methods : Decoctions were prepared by the pressure levels of 0 (non-pressurized) and 1 $kgf/cm^2$ (pressurized) for 60, 120 and 180 min. The extract yield and the total soluble solids content of decoctions were measured, and the amounts of the reference compounds in decoctions were investigated by the analysis using high performance liquid chromatography. Results : The extract yield and the total soluble solid content were higher in decoctions extracted by the pressurized method than those from decoction with non-pressurized method. The patterns of yield and contents showed a proportional increase to the extraction time. In analysis of the linear regression for four reference compounds such as liquiritin, nodakenin, hesperidin, and glycyrrhizin, the good linearity with the correlation coefficient more than 0.9999 was observed. The highest contents for four reference compounds were observed at 180 min of both the pressurized method and the non-pressurized method. Conclusions : This study suggests that the pressure in extraction method and the extraction time affect the compositional constituents in BJIGT decoctions. The extraction time of 180 min could be chosen in both pressurized and non-pressurized method as optimal extraction condition.
