Comparison of Microbial Diversity of Korean Commercial Makgeolli Showing High ${\beta}$-Glucan Content and High Antihypertensive Activity, Respectively

  • We measured physiological functionalities, including antihypertensive angiotensin I-converting enzyme inhibitory activity and immun-stimulating ${\beta}$-glucan content for sixty kinds of Makgeolli that is commercially available from the market. As a result, we selected R-12 commercial raw Makgeolli, with a high content of immuno-stimulating ${\beta}$-glucan, and R-14 commercial raw Makgeolli, exhibiting high antihypertensive activity. Due to the similarities in their overall physicochemical properties and raw materials used for fermentation, we compared the microbial flora in order to investigate the reason for the differences in their functionalities. Nested PCR and denaturing gradient gel electrophoresis for yeasts and bacteria were performed for analysis of microbial diversity of two different kinds of Makgeolli (i.e., R-12, R-14), which showed immuno-stimulating ${\beta}$-glucan content and exhibited a very high level of antihypertensive activity, respectively. Analysis of the 18S rDNA amplicon revealed a major presence of the yeast strain Pichia burtonii in every Makgeolli sample. Analysis of the 16S rDNA amplicon revealed a predominance of lactic acid bacteria, and the most frequent lactic acid bacteria were Lactobacillus ingluviei, L. fermentum, and L. harbinensis, and Lactobacillus sp. Among these, L. harbinensis was detected only in R-12 and L. ingluviei was found only in R-14. Different functionalities from the individual commercially available Makgeolli may be attributed to actions of different microbial flora during fermentation.

Effects of Acidification on the Changes of Microbial Diversity in Aquatic Microcosms

  • In an artificial pH-gradient batch culture system, the effects of acidification on the species composition of a heterotrophic bacterial community were analyzed. As a result of this study, it was found that total bacteria numbers were not affected by acidification and that the population of hetero-trophic bacteria decreased as pH became lower. The heterotrophic bacteria isolated from the entire pH gradient were 12 genera and 22 species. Among them, 64% were gram negative and 36% were gram positive bacteria. As pH decreased, the distribution rate of gram negative bacteria increased while that of gram positive bacteria decreased. The diversity of genera decreased from 13 to 5 as pH decreased from 7 to 3. The G+C content of all of the 202 isolated strains varied from 22.8 to 77.0%, and increased in interspecies of same genus as pH decreased. As a result of clustering analysis, the diversity index of species ranged from 1.13 to 2.37, and it had lower indices as pH decreased. In order to evaluate the diversity of numbers of sample of different size, a rarefaction method was used to analyze the expected number of species appearance according to pH. The statistical significance of species diversity was verified by the fact that the number decreased at lower pH.

Effect of Glasswort (Salicornia herbacea L.) on Microbial Community Variations in the Vinegar-making Process and Vinegar Characteristics

  • Three types of nuruk were made from rice, wheat, and a rice-glasswort (6:4) mixture. Nuruk, makgeolli, and vinegar were manufactured with rice nuruk (RN), wheat nuruk (WN), and rice-glasswort nuruk (RGN). The variable region of 18S or 16S rDNA amplified with genomic DNA extracted directly from nuruk-, makgeolli-, and vinegar-making cultures was analyzed via temperature gradient gel electrophoresis (TGGE). The sequence of the 18S rDNA variable region extracted from the TGGE gel for nuruk was 99% homologous with Aspergillus sp. and that for the makgeolli-making culture was 99% homologous with Saccharomyces sp. and Saccharomycodes sp. The sequence of the 16S rDNA variable region extracted from TGGE gel for the vinegar-making culture was 98% homologous, primarily with the Acetobacter sp. The eukaryotic and prokaryotic diversities in the nuruk-, makgeolli-, and vinegar-making cultures was not significantly altered by the addition of glasswort. Prokaryotic diversity was higher than eukaryotic diversity in the nuruk, but eukaryotic diversity was higher than prokaryotic diversity in the makgeolli-making culture, on the basis of the TGGE patterns. No 18S rDNA was amplified from the DNA extracted from the vinegar-making culture. The diversity of the microbial community in the process from nuruk to vinegar was slightly affected by the type of raw material utilized for nuruk-making. The saccharifying activity and ethanol productivity of nuruk, polyphenol content in makgeolli, and acetic acid and polyphenol content in the vinegar were increased as a result of the addition of glasswort. In conclusion, the glasswort may be not simply an activator for the growth of microorganisms during the fermentation of nuruk, makgeolli, or vinegar, but also a nutritional supplement that improves the quality of vinegar.

한국 초등학교 급식 식단의 영양가 및 식품 구성의 다양성 평가 (Evaluation of Elementary School Foodservice Menus on Its Contents and Diversity of the Food Served.)

  • This study was conducted to evaluation the quality of meals served in elementary school based on nutrient contents and food diversity . A questionnaire was mailed to school foodservice dietitians requesting one week menus for April and October . Menus served in 388 schools from each province and from major cities in Korea were analyzed. Mean nutrient contents per meal of persons served, and food expenses per meal. Mean percentages of energy from fat was 20.2% , and was significantly l different by foodservice system. When nutrient contents were converted to amount per 1, 000kcal, all were higher than that calculated from RDA except calcium, iron, and Vitamin A. Most of these were not significantly different according to area, foodservice system, the number of persons served, and food expense per meal. Compared to 1/3 of RDA for children , many meals provided lower amounts of energy, Vit A, VitB2, and Ca. More than half of the meals provided less iron than 1/3 RDA for 10-12 years girls. Using the highest values of RDA for the elementary school age group as evaluation, criteria nutrient adequacy ratios(NAR) were lowest for Ca(0.61), Vit A(0.57), and iron(0.77). The mean adequacy ratio(MAR) was 0.86. Mean numbers of food items and dishes per meal were 15.3 and 5.1 , respectively. 66.3% of the meals provided three food groups of grain, meat, and vegetable. Except for the number of different food items served per meal, the aspect of food diversity was similar to that of nutrient contents. There was significant positive correlation between mean number of different food items served per meal and nutrient content . In summary , schools in small cities and rural areas than those in large cities, schools adapting commissary or joint management than those or density. Also the more food groups served per meal, the higher the nutrient content per meal . In summary , schools in small cities and rural areas than those in large cities, schools adapting commissary or joint management than those adapting conventional foodservice system, schools serving smaller number of persons, and schools with higher food expenses provided higher energy contents and food diversity.

Genetic diversity and relationship analyses of the Korea native black goat line using microsatellite markers

  • The aim of this study was to analyze the genetic diversity and distance of the Korean native black goat line. Thus far, this Korean native black goat line has not been studied intensively, especially in genetic diversity and relationship studies in comparison with other breeds. In total, eleven microsatellite (MS) markers were used to evaluate alleles from 391 Korean native black goats and foreign hybrid animals. The genetic diversity index was evaluated based on the allele distributions. Four Korean native black goat lines showed expected ranges of observed heterozygosity, expected heterozygosity, and polymorphism information content (PIC) values for use in genetic diversity research (0.509 - 0.643, 0.434 - 0.623 and 0.356 - 0.567). Lines from the Korean native black goat and foreign hybrid were clearly separated according to principal coordinates analysis (PCoA), phylogenetic tree and tended to be clustered in each Korean native black goat line. Thus, this study can be used for analyzing the genetic relationships between Korean native black goats and foreign breeds for line preservation and for fundamental information to determine breed improvement strategies.

  • 미디어 다양성은 글로벌 시장에서의 경쟁이라는 '환상의 짝꿍'과 더불어 한국 미디어 정책을 규정하는 화두로 등장했다. 미국식 기능 행태주의가 주도하는 미디어 정책은 다양성을 시장경쟁, 소비주체의 자유로운 선택이라는 좁은 울타리에 가둔다. 이에 본 논문은 다양성 그 자체를 하나의 완결한 정책목표로 보는 시각에 근본적인 문제제기를 하고자 한다. 다양성이 미디어 정책 담론의 전면에 등장하는 계기를 한국의 중첩적 정치 경제 맥락 속에서 파악하면서, 본 논문은 기존 계량주의 다양성 연구를 비판적으로 분석한다. 나아가 미디어 다양성이 '측정의 과학'에 과도하게 집착해 탈정치화하는 경향을 주목하며 문화연구자들이 미디어 다양성을 보다 진보적으로 전취하기 위한 몇 가지 제안을 하고자 한다. 핵심적인 주장은 미디어 다양성은 시장의 경쟁과 미디어 상품의 다양화라는 신자유주의의 경제논리를 넘어서야 한다는 것이다. 비판적 문화연구자들은 미디어 다양성을 한국 민주주의의 심화라는 보다 큰 정치 프로젝트 속에서 미디어 공공성의 강화, 독립 대안 미디어에 대한 실질적 지원, 급진적 정체성의 정치학 등의 전략과 함께 고려해야 한다.

Geostatistical Analysis of Soil Enzyme Activities in Mud Flat of Korea

  • Spatial variations of physicochemical and microbiological variables were examined to understand spatial heterogeneity of those variables in intertidal flat. Variograms were constructed for understanding spatial autocorrelations of variables by a geostatistical analysis and spatial correlations between two variables were evaluated by applications of a Cross-Mantel test with a Monte Carlo procedure (with 999 permutations). Water content, organic matter content, pH, nitrate, sulfate, chloride, dissolved organic carbon (DOC), four extracellular enzyme activities (${\beta}-glucosidase$, N-acetyl-glucosaminidase, phosphatase, arylsulfatase), and bacterial diversity in soil were measured along a transect perpendicular to shore line. Most variables showed strong spatial autocorrelation or no spatial structure except for DOC. It was suggested that complex interactions between physicochemical and microbiological properties in sediment might controls DOC. Intertidal flat sediment appeared to be spatially heterogeneous. Bacterial diversity was found to be spatially correlated with enzyme activities. Chloride and sulfate were spatially correlated with microbial properties indicating that salinity in coastal environment would influence spatial distributions of decomposition capacities mediated by microorganisms. Overall, it was suggested that considerations on the spatial distributions of physicochemical and microbiological properties in intertidal flat sediment should be included when sampling scheme is designed for decomposition processes in intertidal flat sediment.

  • Recently recommendation methods need personalization and diversity as well as accuracy whereas the traditional researches have been mainly focused on the accuracy of recommendation in terms of quality. The diversity of recommendation is also important to people in terms of quantity in addition to quality since people's desire for content consumption have been stronger rapidly than past. In this paper, we pay attention to similarity of data gathered simultaneously among different types of contents. With this motivation, we propose an enhanced recommendation method using correlation analysis with considering data similarity between two types of contents which are movie and music. Specifically, we regard folksonomy tags for music as correlated data of genres for movie even though they are different attributes depend on their contents. That is, we make result of new recommendation movie items through mapping music folksonomy tags to movie genres in addition to the recommendation items from the typical collaborative filtering. We evaluate effectiveness of our method by experiments with real data set. As the result of experimentation, we found that the diversity of recommendation could be extended by considering data similarity between music contents and movie contents.

Diversity of Culturable Soil Micro-fungi along Altitudinal Gradients of Eastern Himalayas

  • Very few studies have addressed the phylogenetic diversity of fungi from Northeast India under the Eastern Himalayan range. In the present study, an attempt has been made to study the phylogenetic diversity of culturable soil fungi along the altitudinal gradients of eastern Himalayas. Soil samples from 24 m above sea level to 2,000 m above sea level altitudes of North-East India were collected to investigate soil micro-fungal community structure and diversity. Molecular characterization of the isolates was done by PCR amplification of 18S rDNA using universal primers. Phylogenetic analysis using BLAST revealed variation in the distribution and richness of different fungal biodiversity over a wide range of altitudes. A total of 107 isolates were characterized belonging to the phyla Ascomycota and Zygomycota, corresponding to seven orders (Eurotiales, Hypocreales, Calosphaeriales, Capnodiales, Pleosporales, Mucorales, and Mortierellales) and Incertae sedis. The characterized isolates were analysed for richness, evenness and diversity indices. Fungal diversity had significant correlation with soil physico-chemical parameters and the altitude. Eurotiales and Hypocreales were most diverse and abundant group of fungi along the entire altitudinal stretch. Species of Penicillium (D=1.44) and Aspergillus (D=1.288) were found to have highest diversity index followed by Talaromyces (D=1.26) and Fusarium (D=1.26). Fungal distribution showed negative correlation with altitude and soil moisture content. Soil temperature, pH, humidity and ambient temperature showed positive correlation with fungal distribution.

통영 지역의 암반 조간대에서 배출수 유입으로 인한 해조 군집 구조와 엽체 내 질소 함량의 변화 (Changes of Tissue N Content and Community Structure of Macroalgae on Intertidal Rocky Shores in Tongyeong Area due to Sewage Discharge)

  • Enrichment in nutrients coming from urban sewage outfalls can lead to eutrophication in coastal areas, which can also change the species composition and community structure of macro algal communities. We investigated the structure of the macro algal community within three rocky shores in order to assess any possible differences in their characteristics. Site 1 was located near Tongyeong city's sewage outfall, Site 2 was located near a public beach area, and Site 3 faced open channel of the Ocean. All three sites were located within the same stretch of the coast, where Site 2 was located between sites 1 and 3. We measured the nutrient concentration in water and the tissue nitrogen content in macro algae samples. Nutrients in the water column surrounding site 1 were high in ammonium ($30.2\pm1.8{\mu}M$), nitrate ($26.2{\pm}0.1{\mu}M$), and phosphate ($2.7{\pm}0.1{\mu}M$) content, and were characterized by low numbers of macroalgal species and species and a low species diversity index. In contrast, site 3 exhibited relatively low nutrient concentration levels and a high number of macroalgal species and a high species diversity index. Comparative analysis showed that the tissue nitrogen content of macroalgae were significantly (P<0.05) affected by the nutrient concentration in the water column. The tissue nitrogen content of green algae within site 1 was higher than the others sites. However, the tissue nitrogen content of brown algae was similar at all three sites. Thus, the tissue nitrogen content of macro algae and the macro algal community structure of intertidal rocky shores were dependent on location and the performance of macroalgal communities was dependent on water quality.
