Development of Immuno-Analytical System for Microbial Cells by using Dot-Blotter (Dot-Blotter 진공 포획방식에 의한 미생물세포 면역분석시스템의 개발)

  • In order to eventually fabricate an analytical system for infectious microorganisms, we synthesized major immunochemical components, utilized them for the construction of model system, and investigated an assay concept for bacterial whole cells. For the preparation of system components, a polyclonal antibody, against Salmonella thompson as model analyte, purified by immuno-affinity chromatography was used to chemically link to streptavidin or an enzyme, horseradish peroxidase(HRP). The antibody and streptavidin was modified with sulfosuccinimidyl 4-[N-maleimidomethyl]cyclohexane-1-carboxylate and N-succinimidyl-3-[2-pyridyldithio]propionate(subsequently activated by dithiotheritol), respectively. The modified components were reacted to synthesize antibody-streptavidin conjugates which were then purified on a two-layer chromatography column of diaminobiotin gel and Sephadex G-100. For antibody-HRP conjugates, HRP molecules were activated by $NalO_4$ oxidation and then coupled to immunoglobulin. After stabilizing with ($NaCNBH_3$, the conjugates were purified by size exclusion chromatography on Biogel A5M column. To devise a model system, such produced components were combined with a dot-blotter in which a nitrocellulose membrane($12{\mu}m$ pre size) with immobilized biotin was already located. The analyte (S. thompson cells) was reacted with the both antibody conjugates in a liquid phase, and the complexes formed were captured on the membrane surfaces by applying vacuum in the bottom compartment of the blotter to invoke biotin-streptavidin reaction. Under optimal conditions, the system enabled to identify the analytical concept for bacterial whole cells, and the lower limit of detection was approximately $1{\mu}g/m{\ell}$($10^5-10^6$ cells/m$m{\ell}$). The controlling factors were the concentrations of each antibody conjugate that caused agglutination in the presence of analyte as they increased.

  • The planning stage of an architectural project has much more significant effects on the cost or outcome of the project than other stages of the project. In addition, the importance of architectural planning has been further increasing according to the recent trend of construction projects becoming larger in scale and more complex. In spite of this, the current situation is that the planning stage work is not being systematically managed. Accordingly, the purpose of this study was to develop a BIM-based simulation system for providing support during architectural planning stage such as spatial planning & review, cost review, project owner requirements management, etc. It is easy to review various alternatives using this system that allows not only the modeling of space object modeling but also the instantaneous review of spatial area & layout, cost, etc. based on object information. In addition, it can be used as a communication tool with the project owner as it provides the visualized information and quantitative data of the building model, and the information created through this system can be delivered to the following stage for usage. It is thought that using this system, the entire project work including the architectural planning stage can be supported and even contributing to the advancement of architectural process.

A Study on Deterioration of Stone Monuments by Acid Fog (산성안개에 의한 석조문화재 구성암석의 손상 연구)

  • In order to predict the deterioration of stone monument due to acid fog, an artificial fog test using pH4.0 and pH5.6 was applied to the Gyeongju Namsan granite, decite and marble. After the test had weathered Gyeongju Namsan granite a larger weight reduction due to acid fog than fresh one. Decite has shown the most significant changes among the tested rocks with about 0.005 % of weight reduction. Decite and weathered granite will have considerable weight reduction due to acid rain than the acid fog, whereas the marble was expected to show a weight reduction regardless of the phase of water. The porosity and water absorption rate of weathered granite had significantly increased. This result means that the weathered rock is predicted to be more susceptible to acid fog than the fresh rock. The absorption rate of the marble after the test had shown approximately 50 % increase. The color of the samples had slightly changed towards yellow, such tendency was greater shown in weathered rocks. The marble reacted with acid fog had an increased whiteness. A large amount of cation in the samples is caused mainly by the dissociation of minerals through the reaction with acid fog.

  • This study focused on mobile trend in diffusion of smart phone. Presently, it has been evolved from digital nomad to smart nomad. Smart nomad era is based on digital technology with mobility and being smart. Smart nomad that is made up high-end device and intellectual content has changed structure of industry and life style. Smart phone is a core key in smart nomad era. In 2010 smart phone has been distributed about 2.5 hundreds million which is 20% of whole mobile phone sales and the volume of smart phone sales will be getting bigger and bigger in near future. Although, there are various reasons for success of smart phone market, this research focused 4 reasons: first, development of mobile device, second, evolution of mobile content distribution, third, support of government system, fourth, success of Docomo i-mode and Apple i phone. Smart phone will grow up more rapidly. Smart phone eco-system will be more competitive in the world. In Korea, smart phone eco-system also will be very competitive. For success in the middle of this competitive environment, Korea should lead the competitive phase with development of ability of creation software and content, construction of various partnership and evolution of next generation technology and service.

A Study on the Treatment of Oil Contaminated Soils with Micro-nano Bubbles Soil Washing System (유류오염토양 처리를 위한 마이크로나노버블 토양세척에 관한 연구)

  • The objectives of this study are to examine the processing of oils contamination soil by means of using a micronano-bubble soil washing system, to investigate the various factors such as washing periods, the amount of micro-nano bubbles generated depending on the quantity of acid injection and quantity of air injection, to examine the features involved in the elimination of total petroleum hydrocarbons (TPHs) contained in the soil, and thus to evaluate the possibility of practical application on the field for the economic feasibility. The oils contaminated soil used in this study was collected from the 0~15 cm surface layer of an automobile junkyard located in U City. The collected soil was air-dried for 24 hours, and then the large particles and other substances contained in the soil were eliminated and filtered through sieve No.10 (2 mm) to secure consistency in the samples. The TPH concentration of the contaminated soil was found to be 4,914~5,998 mg/kg. The micronano-bubble soil washing system consists of the reactor, the flow equalization tank, the micronano- bubble generator, the pump and the strainer, and was manufactured with stainless material for withstanding acidic phase. When the injected air flow rate was fixed at 2 L/min, for each hydrogen peroxide concentrations (5, 10, 15%) the removal percents for TPH within the contaminated soil with retention times of 30 minutes were respectively identified as 4,931 mg/kg (18.9%), 4,678 mg/kg (18.9%) and, 4,513 mg/kg (17.7%). And when the injected air flow rate was fixed at 2 L/min, for each hydrogen peroxide concentrations (5, 10, 15%) the removal percents for TPH within the contaminated soil with retention times of 120 minutes were respectively identified as4,256 mg/kg (22.3%), 4,621 mg/kg (19.7%) and 4,268 mg/kg (25.9%).

Intertidal DEM Generation Using Satellite Radar Interferometry (인공위성 레이더 간섭기술을 이용한 조간대 지형도 작성에 관한 연구)

  • High resolution intertidal DEM is a basic material for science research like sedimentation/erosion by ocean current, and is invaluable in a monitoring of environmental changes and practical management of coastal wetland. Since the intertidal zone changes rapidly by the inflow of fluvial debris and tide condition, remote sensing is an effective tool for observing large areas in short time. Although radar interferometry is one of the well-known techniques for generating high resolution DEM, conventional repeat-pass interferometry has difficulty on acquiring enough coherence over tidal flat due to the limited exposure time and the rapid changes in surface condition. In order to overcome these constraints, we tested the feasibility of radar interferometry using Cosmo-SkyMed tandem-like one-day data and ERS-ENVISAT cross tandem data with very short revisit period compared to the conventional repeat pass data. Small temporal baseline combined with long perpendicular baseline allowed high coherence over most of the exposed tidal flat surface in both observations. However the interferometric phases acquired from Cosmo-SkyMed data suffer from atmospheric delay and changes in soil moisture contents. The ERS-ENVISAT pair, on the other hand, provides nice phase which agree well with the real topography, because the atmospheric effect in 30-minute gap is almost same to both images so that they are cancelled out in the interferometric process. Thus, the cross interferometry with very small temporal baseline and large perpendicular baseline is one of the most reliable solutions for the intertidal DEM construction which requires very accurate mapping of the elevation.

  • Toxicogenomics is now emerging as one of the most important genomics application because the toxicity test based on gene expression profiles is expected more precise and efficient than current histopathological approach in pre-clinical phase. One of the challenging points in Toxicogenomics is the construction of intelligent database management system which can deal with very heterogeneous and complex data from many different experimental and information sources. Here we present a new Toxicogenomics database developed as a part of 'Toxicogenomics for Efficient Safety Test (TEST) project'. The TEST database is especially focused on the connectivity of heterogeneous data and intelligent query system which enables users to get inspiration from the complex data sets. The database deals with four kinds of information; compound information, histopathological information, gene expression information, and annotation information. Currently, TEST database has Toxicogenomics information fer 12 molecules with 4 efficacy classes; anti cancer, antibiotic, hypotension, and gastric ulcer. Users can easily access all kinds of detailed information about there compounds and simultaneously, users can also check the confidence of retrieved information by browsing the quality of experimental data and toxicity grade of gene generated from our toxicology annotation system. Intelligent query system is designed for multiple comparisons of experimental data because the comparison of experimental data according to histopathological toxicity, compounds, efficacy, and individual variation is crucial to find common genetic characteristics .Our presented system can be a good information source for the study of toxicology mechanism in the genome-wide level and also can be utilized fur the design of toxicity test chip.

A Study on the Use of Fresh Root-chips in Slope Revegetation Works (비탈면 녹화에서 임목폐기물의 활용에 관한 연구)

  • In this study, we attempted to seek out the ways to recycle fresh root-chips in the slope revegetation works by breaking tree root wastes occurring during the construction works, also to review the applicability of fresh root-chips as the soil media in slope revegetation works. For this purpose, we organized test units in order to investigate on-site applicability of fresh root-chips (broken chips). In order to examine the desirable ration of combining fresh root-chips with the hydroseeding soil media on the cutting slopes, we organized test units depending on the amount of combination. The following is the main experimental results. 1. At first, we analyzed properties of hrdro-seeding soil media and soil of the experimental sites. The overall results demonstrate that all the test units show proper range for vegetation. 2. We believe that the physical properties of soils in the earlier phase of restoration works on the sloped sites are not greatly affected by the fact whether broken chips exist or not. However, as time elapses, broken chips needs to be investigated further on what kind of impact they have on the soil condition. 3. More species are found in the test unit combining broken chips and we believe that it will contribute to blossoming of green plants and ecological succession of neighboring plants. 4. We performed experiment on possibility for fresh root-chips as substitutes for the hydro-seeding soil-media. In the test unit that combines fresh root-chips, its mixture ratio tends to exceed that of the test unit that does not combine fresh root-chips by 5 %. In case of the Thick-Layer-Soil-Media Hydroseeding works, the mixture ratio of the test unit that combines fresh root-chips after 16 weeks exceeds that of the test unit that does not combine fresh root-chips by 75%. 5. From the result of our experiment, it is obvious that the ratio of mixture and the number of emerging species are higher for the test unit combining fresh root-chips than the test unit that does not combine them. In other words, we can replace the hydro-seeding-soil-media with some Fresh root-chips without affecting the physical property of soil.

Review of the installable area of multi-utility tunnel using feasibility assessment indexes (타당성 평가지표를 활용한 터널식 공동구 설치가능지역 검토)

  • In this paper, The study was conducted on the method to roughly determine the area and length of the multi-utility tunnel before the planning and design phase of the multi-utility tunnel construction. For this purpose, four feasibility indexes were used: traffic density, population density, disaster prevention index (number of workers), and urbanization rate, which reflect the regional characteristics. The installation criteria were set in consideration of the average value and minimum value of the feasibility indexes for the tunnel type among areas that can be installed in the multi-utility tunnel of Seoul. The analysis area included 200 areas based on 14 zones. The results of the analysis based on the minimum value of feasibility indexes indicated that the tunnel type of multi-utility tunnel is suitable for 39 areas with high traffic volume and population. On the other hand, the 'gun' area, etc., has a wider population than the 'si' and 'gu', suggesting that it is not suitable to install multi-utility tunnel. In addition, it can be seen that the larger the index value centered on the minimum value of each index, the smaller the tunnel type of multi-utility tunnel installation area.

Classifying and Identifying Asbestos and Non-Asbestos Fibers by a Rule Building Expert System (전문가시스템을 이용한 석면 및 비석면의 분류 및 확인)

  • Asbestos is the name of a group of minerals with long and thin fibers that originate naturally in the environment. Asbestos mainly affects lungs and the membrane that surrounds the lungs. In general, PCM (phase contrast microscopy) and PLM (polarized light microscopy) have been used to analyze asbestos fibers. However, these methods have often problems to over-estimate number concentration when counting real asbestos fibers. Moreover, there are many difficulties when separating and identifying various asbestos and non-asbestos fibers. In order to determine quantitative information on fibrous particles, source profiles for asbestos and non-asbestos fibers must be initially developed on the basis of their chemical compositions and physical parameters. In our study, a SEM/EDX was used to develop source profiles from known asbestos samples as reference samples. We could make the source profile matrix consisting of 6 types of asbestos fibers and 2 types of non-asbestos fibers by analyzing 380 fibers. Based on these profiles, a rule building expert system was developed by using the visual basic application (VBA). Various fibers were successfully classified by 2 simple rules in the EXCEL environment based on several visual steps such as inserting data, viewing results, and saving results. For a case study to test the expert system, samples from a construction materials and from various indoor environments such as a residental area, a preschool classroom, and an underground store were collected and analyzed. As a result of the survey, a total of 76 individual test fiber particles was well classified into 5 different types of particle classes; 9.3% of chrysotile, 15.4% of amosite, 0.8 of crocidolite, 4.2% of tremolite, 5.8% glass fiber, 21.1% of other fibers, and 43.5% of unknown fibers in terms of number concentration. Even though unknown portion was high, it will be decreased markedly when expanding fiber source profiles.
