Ripe rot caused by Botryosphaeria dothidea is one of the serious diseases of postharvest kiwifruit. In order to control ripe rot on Actinidia chinensis cultivar 'Zesy002', several commercial agrofungicides were selected by an antifungal test on an artificial medium. Furthermore, disease suppression by the selected fungicides was evaluated on the kiwifruit by inoculation with a conidial suspension of B. dothidea. On the artificial media containing boscalid + fludioxonil was shown to be the most effective antifungal activity. However, in the bio-test pyraclostrobin + boscalid and iminoctadinetris were the most effective agrochemicals on the fruit. On the other hand, the infection structures of B. dothidea on kiwifruit treated with pyraclostrobin + boscalid were observed with a fluorescent microscope. Most of the fungal conidia had not germinated on the kiwifruit treated with the agrochemicals whereas on the untreated fruit the fungal conidia had mostly germinated. Electron microscopy of the fine structures showed morphological changes to the conidia and branch of hyphae on the kiwifruit pre-treated with pyraclostrobin + boscalid, indicating its suppression effect on fungal growth. Based on this observation, it is suggested that ripe rot by B. dothidea may be suppressed through the inhibition of conidial germination on the kiwifruit treated with the agrochemicals.
Ampelomyces quisqualis 94013 (AQ94013), a hyperparasite, was selected as an effective biological control agent against cucumber powdery mildew. Optimal temperature for mycelial growth of AQ94013 was $26^{\circ}C$, and the optimal pH was 6.5. Conidia of AQ94013 were more Produced on potato dextrose agar (PDA) in darkness than under alternating cycles of 12 hr fluorescent light and 12 hr darkness. Temperature range for spore germination of the fungus was $10\~35^{\circ}C$, and optimal temperature was $20^{\circ}C$. Conidial germination of the fungus began 8 hr after incubation at $24^{\circ}C$. Germination rate of conidia at concentration of $5{\times}10^5\;spores/ml\;and\;5{\times}10^6\;spores/ml$ was higher than at concentration of $5{\times}10^7\;spores/ml$. The best source of carbon and nitrogen for mycelial growth of the fungus were dextrin and neopeptone, respectively.

This study was conducted to investigate agents inducing conidial germination of an entomopathogenic fungus, Beauveria bassiana KK5. Different chemicals including carbohydrates were mixed with conidia of B. bassiana and incubated on water agar for 12 hours. Fructose, mannose and skim milk were useful for spore germination compared to other chemicals. Bioassays against green peach aphids were conducted with the fungal conidia suspended in 1% fructose, mannose and skim milk. Of them, a mixture of skim milk plus conidia of B. bassiana KK5 showed the highest mortalities against $3^{rd}$ instar of green peach aphid.
Effects of silicon application on development of colonies of Sphaerotheca fuliginea were examined. Cucumber plants were applied with nutrient solutions amended with different concentrations of soluble silicon and selected leaves were inoculated with known concentrations of conidia of the pathogen. Colony number per leaf, colony area per leaf, and germination rate of conidia of S. fuliginea collected from the inoculated leaves were reduced as silicate concentrations in the nutrient solutions increased from 0.05 to 4.10 mM. The increase in resistance of plants to mildew infection was apparently due to silicate accumulation in leaves, and there was no correlation between cation or ionic strength effects and the silicate treatments. Silicate treatment in growth medium remarkably suppressed powdery mildew development on cucumber. Colonies of mildew fungus were visible with over approx. 38.3% of the mature leaf surface, while that of the leaves in high Si plants was 2.3% observed at 51 days after transplanting. No significant differences were observed between 1.7 mM and 3.4mM silicate treatments. Conidial germination rates were significantly reduced by increasing Si amendments. Conidial germination ranged from 14.7 to 20.3% for plants grown in low Si solution(<1.40 mM), and from 9.0 to 12.4% for plants grown in high Si solution(>1.8 mM). Foliar applications of Si with ${\geq}$ 17.0 mM decreased the number of powdery mildew colonies. Persistence of Si foliar sprays effects on cucumber demonstrated that the 17 mM Si spray applied 4 days before inoculation with S. fuliginea reduced mildew colony formation. The relationship was positive and linear.
Gamma irradiation and its convergence with nano-silver particles and sodium dichloroisocyanurate (NaDCC) were investigated to inhibit germination and mycelial growth of Botrytis elliptica, the pathogen of lily leaf blight. In addition, the same treatments were studied on the process of disease development with detached leaf of lily cv. Siberia. Spray inoculation, which is closer to natural infection than wound inoculation, can be a way to investigate infection ability of the treated pathogen. The irradiating dose required to reduce the population by 90%, $D_{10}$, was 526 Gy irradiating with 0-2000 Gy gamma ray on the conidial suspension as well as the growing mycelia. Even at 2000 Gy, the mycelium was not killed but just delayed its growth at 1-2 days behind. Convergent treatment with 40 mg/l of NaDCC just before 200 Gy gamma irradiation was the best way to decrease the conidial germination about 1/1000 times. The control values of gamma irradiation were 23% and 19.5% at wound inoculation and spray inoculation, respectively. On wound-inoculation, the control value of NaDCC only was 89%, and that of NaDCC convergent with 200 Gy gamma irradiation was 32%. On sprayinoculation, the highest control value was NaDCC at 50%, and that of NaDCC convergent with gamma irradiation was 24%.
The direct effects of acute $\textrm{O}_3$ on the growth, sporulation and infection of Pyricularia grisea, rece blast pathogen, were investigated to understand the interactions between ozone and the pathogen. Acute exposure of 200 nl $\textrm{l}^{-1}$ ozone for 8 h significantly reduced conidia germination on water atar. Ozone exposure of 200 nl $\textrm{l}^{-1}$ for 8h per day for 5 days had no effect on increase in colony diameter, but severely damaged actively growing aerial mycelia. However, the damage to mycalia was recovered during the following 16 h exposure of unpolluted air. Conidial production was also stimulated by the acute ozone exposure for 5 days. The conidia exposed to the acute ozone for 5 days normally germinates but slightly reduce appressoria formation on rice leaf. However, the conidia produced by artificial stimulation under the same ozone concentration for 10 days showed significant reduction in appressorea for mation on a hydrophobic film. This study suggests that the acute ozone could ingibit appressoria formation as well as vegetative growth of the pathogen, resulting in decrease in rece blast development in the field during summer when high ozone episodes could occur occasionally.
Septoria brown spot caused by Septoria glycines Hemmi is one of the serious fungal diseases in soybean. Since little has been known about the disease progress in the field, the present study was conducted to determine the factors affecting the disease progress in the soybean plant. Disease severity and pattern of the progress of the Septoria brown spot were different with varieties. Susceptibility of soybean plants increased with increase of plant age and leaf maturity in order from the primary leaf to the newly expanded leaf. It seemed to be related with conidial germination on the leaves. Germination and germtube elongation were more inhibited by the diffusates obtained from upper leaves than those from lower leaves and they were higher in a susceptible variety than in a moderately resistant one.

To study the control of postharvest decay caused by Colletotrichum gloeosporioides and Penicillium expansum, gamma irradiation alone or in combination with fumigation was evaluated to extend the shelf life of apples in South Korea. An irradiation dose of 2.0 kGy resulted in the maximum inhibition of C. gloeosporioides and P. expansum spore germination. The gamma irradiation dose required to reduce the spore germination by 90% was 0.22 and 0.35 kGy for C. gloeosporioides and P. expansum, respectively. Microscopic observations revealed that when the fungal spores were treated with gamma irradiation (4.0 kGy), conidial germination was stopped completely resulting in no germ tube formation in C. gloeosporioides. Treatment with the eco-friendly fumigant ethanedinitrile had a greater antifungal activity against C. gloeosporioides and P. expansum in comparison with the non-treated control under in vitro conditions. The in vitro antifungal effects of the gamma irradiation and fumigation treatments allowed us to further study the effects of the combined treatments to control postharvest decay on stored apples. Interestingly, when apples were treated with gamma irradiation in combined with fumigation, disease inhibition increased more at lower (< 0.4 kGy) than at higher doses of irradiation, suggesting that combined treatments reduced the necessary irradiation dose in phytosanitary irradiation processing under storage conditions.

TiO$_2$photocatalytic ozonation was attempted to disinfect fungal pathogens causing postharvest spoilage of kiwifruits and to decompose fungicide residuals on kiwifruits. TiO$_2$Photocatalytic ozonation process synergistically degraded organic compound and inhibited conidial germination of the fungal pathogen compared to single treatment of ozonation or photocatalysis. The efficient control of fungal spoilage and degradation of residual fungicide on kiwifruits indicate that TiO$_2$photocatalytic ozonation is a very attractive method for postharvest disease control of kiwifruits as an alternative to fungicides application.
Bacillus cereus 28-9 is a chitinolytic bacterium isolated from lily plant in Taiwan. This bacterium exhibited biocontrol potential on Botrytis leaf blight of lily as demonstrated by a detached leaf assay and dual culture assay. At least two chitinases (ChiCW and ChiCH) were excreted by B. cereus 28-9. The ChiCW-encoding gene was cloned and moderately expressed in Escherichia coli DH5$\alpha$. Near homogenous ChiCW was obtained from the periplasmic fraction of E. coli cells harboring chiCW by a purification procedure. An in vitro assay showed that the purified ChiCW had inhibitory activity on conidial germination of Botrytis elliptica, a major fungal pathogen of lily leaf blight.
