Forecasting the Pepper Gray Mold Rot to Predict the Initial Infection by Botrytis cinerea in Greenhouse Conditions

  • We determined threshold environmental factros to initiate infection of pepper plants by Botrytis cinerea, a fungal pathogen of pepper gray mold, in two greenhouse conditions. A new efficient spore-trapping method was developed to estimate population density of airborne conidia in the greenhouses, and spore release was measured using a Kerssies' selective medium. At a given day, spores were released greater during daytime (mostly from 7:30 am to 10:30 am and at 4:30 pm) than nighttime. Diurnal and nocturnal temperatures in the greenhouse-1 were about $25^{\circ}$ and $17^{\circ}$,and relative humidity was 100% for prolonged 24 h due to rain on December 17, 1997. Population density of air-borne conidia was 3.0$\times$103 conidia/ $0.5\textrm{m}^3$ after two days, and the initial infection occurred in ten days. During the same period of time in the greenhouse-2, diurnal temperature was about $25^{\circ}$ and nocturnal temperature was below $15^{\circ}$, and population density of air-borne conidia was 104 conidia/ $0.5\textrm{m}^3$. Under these conditions, the initial infection started in three days. This indicates that the early infection occurs under which diurnal temperature is approximately $25^{\circ}$, nocturnal temperature is maintained below $15^{\circ}$, and population density of air-borne conidia is 104 conidia/ $0.5\textrm{m}^3$ at saturated relative humidity condition.

Factors Affecting Sporulation of a Mycoherbicide, Epicoccosorus nematosporus, on the Lesion of Eleocharis kuroguwai

  • Effects of temperature and dew period on sporulation of a mycoherbicide, Epicoccosorus nematosporus, on the lesion of its host, Eleocharis kuroguwai were determined. Conidia formation was first observed after 10 days on plants incubated for either 12 or 16 h in a dew chamber at 28$^{\circ}C$; 16 h dew period resulted in more conidia formation. As the dew period was decreased to less than 8 h, fewer conidia formed. Conidial production was most abundant at 28$^{\circ}C$ and produced as much as 3.3$\times$10$^4$conidia per lesion, while 0.1$\times$10$^3$and 2.3$\times$10$^3$conidia per lesion were produced at 16$^{\circ}C$ and 36$^{\circ}C$, respectively. Alternating temperature regimes, i.e., 30/15, 30/20, 28/20, and 28/15$^{\circ}C$ (day/night) were much better than constant temperature, i.e., 30/30, 28/28/, and 20/2$0^{\circ}C$ for sporulation. In the second sporulation, there were as much as 3.1$\times$10$^4$conidia per lesion (ca. <50% of the first sporulation). Then, sporulation dropped sharply to 6.2$\times$10$^2$conidia per lesion in the third sporulation. Results of this study suggest that temperature combined with dew period is the primary limiting factor in the use of E. nematosporus as a mycoherbicide off, kuroguwai.

  • C. dematium 과 C. gloeosporioides의 포자발아 및 발아관 신장에 알맞는 온도는 $20{\sim}30^{\circ}C$이며 부착기는 $20^{\circ}C$에서 형성이 잘 되었으나 포자의 발아는 늦었다. $30^{\circ}C$에서는 포자발아와 발아관의 신장이 양호하였으나 부착기의 형성은 낮았다. C. acutatum의 포자발아에 알맞는 온도는 $20{\sim}30^{\circ}C$이었으나 발아관의 신장이 $25^{\circ}C$에서 촉진되었다. 분생포자는 격막이 생기고 발아관 대신 $1{\sim}2$개의 딸세포가 포자화되었으며, 제2차 포자를 형성하면서 수지상(樹枝狀)으로 되었다. 부착기는 포자의 발아에 의해서 드물게 형성되었으며 알맞는 온도는 $20{\sim}30^{\circ}C$이었다. 그러나 $30^{\circ}C$에서 발아된 포자는 부착기의 형성능력이 없었다. Colletotrichum acutatum, C. dematium 및 C. gloeosporioides 등의 분생포자 발아 및 부착기 형성에 미치는 온도의 영향에 관해서 논의(論議)하였다.

Aspergillus nidulans의 분생포자에 존재하는 $\beta$-Glucosidase에 관하여 (The conidial $\beta$-glucosidase of aspergillus nidulans)

  • Aspergillus nid$\mu$tans 의 분생포지에는 두 종류의 $\beta$.glucosidase, 즉 tightly-bound enzyme(TBE)과 loosely-bound enzyme(LBE)이 존재한다. 포자수와 효소활성도가 정비례함을 보아 이 효소의 존재을 확인할 수 있었으며, 배양조건 하에서 의 오랜기잔 방치에도 얼정 수준의 활성을 유치하는 높은 안정성을 보였다. 분생포자에 여러가지 물리.화학적 처리를 한 뒤에도 TBE의 환성도에는 큰 변화가 없었으며, TBE의 최적 pH 및 온도는 각각 6.0, $55^{\circ}C$였다.TBE의 열안정성은 균사체의 $\beta$-glucosidase보다 높았으며, $H^{2+}$이온에 대한 내성도 비교적 높은 수준을 보였다. LBE와 균사체 $\beta$-glucosidase의 전기영동 양상이 유사한 것으후 보아 율사체와 동일한 효소가 포자의 표면에 존재후L을 알 수있었다.

Genetic Control of Asexual Sporulation in Fusarium graminearum

  • Fusarium graminearum (teleomorph Gibberella zeae) is an important plant pathogen that causes head blight of major cereal crops such as wheat, barley, and rice, as well as causing ear and stalk rot on maize worldwide. Plant diseases caused by this fungus lead to severe yield losses and accumulation of harmful mycotoxins in infected cereals [1]. Fungi utilize spore production as a mean to rapidly avoid unfavorable environmental conditions and to amplify their population. Spores are produced sexually and asexually and their production is precisely controlled. Upstream developmental activators consist of fluffy genes have been known to orchestrate early induction of condiogenesis in a model filamentous fungus Aspergillus nidulans. To understand the molecular mechanisms underlying conidiogenesis in F. graminearum, we characterized functions of the F. graminearum fluffy gene homologs [2]. We found that FlbD is conserved regulatory function for conidiogenesis in both A. nidulans and F. graminearum among five fluffy gene homologs. flbD deletion abolished conidia and perithecia production, suggesting that FlbD have global roles in hyphal differentiation processes in F. graminearum. We further identified and functionally characterized the ortholog of AbaA, which is involved in differentiation from vegetative hyphae to conidia and known to be absent in F. graminearum [3]. Deletion of abaA did not affect vegetative growth, sexual development, or virulence, but conidium production was completely abolished and thin hyphae grew from abnormally shaped phialides in abaA deletion mutants. Overexpression of abaA resulted in pleiotropic defects such as impaired sexual and asexual development, retarded conidium germination, and reduced trichothecene production. AbaA localized to the nuclei of phialides and terminal cells of mature conidia. Successful interspecies complementation using A. nidulans AbaA and the conserved AbaA-WetA pathway demonstrated that the molecular mechanisms responsible for AbaA activity are conserved in F. graminearum as they are in A. nidulans. F. graminearum ortholog of Aspergillus nidulans wetA has been shown to be involved in conidiogenesis and conidium maturation [4]. Deletion of F. graminearum wetA did not alter mycelial growth, sexual development, or virulence, but the wetA deletion mutants produced longer conidia with fewer septa, and the conidia were sensitive to acute stresses, such as oxidative stress and heat stress. Furthermore, the survival rate of aged conidia from the F. graminearum wetA deletion mutants was reduced. The wetA deletion resulted in vigorous generation of single-celled conidia through autophagy-dependent microcycle conidiation, indicating that WetA functions to maintain conidia dormancy by suppressing microcycle conidiation in F. graminearum. In A. nidulans, FlbB physically interacts with FlbD and FlbE, and the resulting FlbB/FlbE and FlbB/FlbD complexes induce the expression of flbD and brlA, respectively. BrlA is an activator of the AbaA-WetA pathway. AbaA and WetA are required for phialide formation and conidia maturation, respectively [5]. In F. graminearum, the AbaA-WetA pathway is similar to that of A. nidulans, except a brlA ortholog does not exist. Amongst the fluffy genes, only fgflbD has a conserved role for regulation of the AbaA-WetA pathway.

Beauveria bassiana 대량배양을 위한 탄소원, 질소원 및 고체 기질 선발 (Selection of Carbon, Nitrogen Source and Carrier for Mass Production of Beauveria bassiana)

  • Two-phase fermentation을 통한 Beauveria bassiana 149균주의 대량 배양을 위한 탄소원, 질소원, 기질 그리고 배양 조건 선발 실험을 실시하였다. 1차 액체 배양을 위한 적정 탄소원으로는 옥수수 분말, 전분 그리고 쌀가루가, 질소원으로는 펩톤과 효모 분발을 첨가한 것에서 포자 생산량이 높았다. 포자 대량 생산을 위한 기질 및 첨가물로는 톱밥+밀기울+옥수수 분말, 톱밥+밀기울, 쌀겨+밀기울 혼합에서 포자 생산량이 높았다. B. bassiana 149 균주의 포자 대량 생산을 위한 고체 배양 결과, 포자 생산량은 $30^{\circ}C$ 배양에서 $20^{\circ}C$보다 2배 정도 높은 경향을 보였다. B. bassiana 149 균주의 포자 생산량은 $25^{\circ}C$, 수분함량 60%와 70%에서 배양 시 $26.9{\times}10^8conidia/g$$38.6{\times}10^8conidia/g$로 40%와 50%의 $13.9{\times}10^8conidia/g$$11.6{\times}10^8conidia/g$ 보다 상당히 높았다.

Evaluation of Strains of Metarhizium anisopliae and Beauveria bassiana against Spodoptera litura on the Basis of Their Virulence, Germination Rate, Conidia Production, Radial Growth and Enzyme Activity

  • Ten strains of the entomopathogenic fungi Metarhizium anisopliae and Beauveria bassiana were evaluated to find the most effective strain for optimization studies. The first criterion tested for strain selection was the mortality (> 50%) of Spodoptera litura larvae after inoculation of the fungus for 4 days. Results on several bioassays revealed that B. bassiana BNBCRC showed the most virulence on mortality S. litura larvae (80% mortality). B. bassiana BNBCRC also showed the highest germination rate (72.22%). However, its conidia yield ($7.2{\times}10^8$ conidia/mL) was lower than those of B. bassiana B 14841 ($8.3{\times}10^8$ conidia/mL) and M. anisopliae M6 ($8.2{\times}10^8$ conidia/mL). The highest accumulative radial growth was obtained from the strain B14841 (37.10 mm/day) while the strain BNBCRC showed moderate radial growth (24.40 mm/day). M. anisopliae M6 possessed the highest protease activity (145.00 mU/mL) while M. anisopliae M8 possessed the highest chitinase activity (20.00 mU/mL) during 96~144 hr cultivation. Amongst these criteria, selection based on virulence and germination rate lead to the selection of B. bassiana BNBCRC. B. bassiana B14841 would be selected if based on growth rate while M. anisopliae M6 and M8 possessed the highest enzyme activities.

  • 자연상태(自然狀態)에서 발현(發現)된 병반(病斑)을 이용(利用), 도열병균(稻熱病菌)의 포자형성량(胞子形成量)과 이탈량(離脫量)을 $1985{\sim}1986$년(年)에 조사(調査)하였다. 절취(切取)한 병반(病斑)의 포자형성량(胞子形成量)과 자연상태하(自然狀態下)에서의 포자이탈량(胞子離脫量)은 병반발현후(病斑發現後) $5{\sim}7$일(日) 사이에 최고치(最高値)에 달(達)했으며 그 수(數)는 각각(各各) 16,200개(個)와 15,900개(個)였다. 자연상태하(自然狀態下)에서의 포자이탈(胞子離脫)은 30일간(日間) 지속(持續)되었다.

골프장 잔디 및 농림해충 수종에 대한 곤충병원성 곰팡이 Metarhizium anisopliae J-22의 병원성 (Pathogenicities of Entomopathogenic Fungus, Metarhizium anisopliae J-22 against Turfgrass and Some Agro-forest Insect Pests)

  • Biological control of turfgrass insect pest Blitopertha orientalis, forest insect pests, Agelastica coerulea, Meganola melancholia, and Glyphodes perspectalis,vegetable insect pests, Plutella xylostella and Agrotis segetum were conducted with entonopathogenic fungus, Metarhizium anisopliae J-22 isolated from black pine forest soil in Cheju province. Mortality of B. orientalis larvae was 53.3% at the rate of 3.4 $\times$ 1O 7 conidia /ml. A. coerulea and M melancholia larvae showed 100% mortalities at 9.6 $\times$ 106 conidia /ml and 2.7 $\times$ 10 7 conidia /ml as well. However, G. perspectalis larvae were not dead even at 4$\times$ 1O 7 conidia /ml. On the other hand, M anisopliae J-22 was effective against P. xylostella larvae showing 100% mortality at 4 $\times$ 10 7 conidia /ml. Improved Method to Increase Conidia Production from Isolates of Different Pathotypes of Citrus Scab Pathogen Elsinoe spp.

  • Elsinoe fawcettii and E. australis are two currently recognized scab pathogens of citrus. E. fawcettii has at least six pathotypes while E. australis has at least two pathotypes. Colonies of E. fawcettii and E. australis do not sporulate in artificial media including potato dextrose agar (PDA). Whiteside's method has been widely used for preparing conidial inoculum in vitro. This study was carried out to develop efficient method for conidia production from artificial media. We developed a shaking method which included the following steps: 1) Colony grown on PDA was mashed with a steel spatula; 2) Mycelia fragments were cultured in 50 ml sterilized rain water in a rotary shaker-incubator (180 rpm) at $25^{\circ}C$ for 24 h: 3) The conidia suspension was filtered through two layers of cheesecloth. Average conidia production of all isolates tested using this shaking method was approximately 13.1 times higher than that from Whiteside's method in this study.
