A new Rockwell hardness (HRC) model using a volumetric parameter by a least square and fractal interpolation method is suggested. The results are also investigated in comparison to real measured hardness data. For this purpose, the measurement of an indented volume is performed using a confocal laser scanning microscope (CLSM), and the captured height encoded image (HEI) is used as an original surface for the calculation of the indented volume. After configuring the surface, the constructed volume is calculated and used as an independent variable for HRC hardness modeling. The hardness model is established using an experimental modeling technique involving a least square algorithm and fractal interpolating model, and this suggested model can be used to reliably predict the Rockwell hardness. These techniques can also be applied to the modeling of the Brinnell and Vickers hardnesses using a volumetric variable.
This study was to quantify fiber distributions in thickness direction of kenaf handsheets as a fiber distribution index (FDI) and to analyze the relationship between FDI and the handsheet properties. The images of fiber distribution in z-direction were obtained by Confocal Laser Scanning Microscope (CLSM) and analyzed by image analysis technique. The proposed FDI had a good correlation with high R2 vlaues with various properties of paper, such as apparent density, scattering coefficient , burst index, tear index, tensile index, and folding endurance. The proposed FDI was shown as a good index to quantify paper properties.
Motivation: Fibers as the extracellular filamentous structures determine the shape of the cytoskeletal structures. Their characterization and reconstruction from a 3D cellular image represent very useful quantitative information at the cellular level. In this paper, we presented a novel automatic method to extract fiber diameter distribution through a pipeline to reconstruct fibers from 3D fluorescence confocal images. The pipeline is composed of four steps: segmentation, skeletonization, template fitting and fiber tracking. Segmentation of fiber is achieved by defining an energy based on tensor voting framework. After skeletonizing segmented fibers, we fit a template for each seed point. Then, the fiber tracking step reconstructs fibers by finding the best match of the next fiber segment from the previous template. Thus, we define a fiber as a set of templates, based on which we calculate a diameter distribution of fibers.
This study was carried out to investigate the sheet formation properties of Morus Hanjis, made of bast and whole stalk pulps by different pulping methods, such as alkali, alkali-peroxide and sulfomethylated pulping. Two species of Morus, M. alba and M. lhou, were used. Effect of morphological properties of pulp stocks on the sheet formation and its gray levels based on optical property were evaluated using an Image analyzer. In addition, the effect of fiber distribution index(FDI) which was calculated from tile data of Confocal laser scanning microscopy(CLSM) on the sheet formation and optical properties of Morus Hanji were also discussed. On the sheet formation, Hanji from whole stalk pulp was superior than that of bast pulp. The more the sheet formation improved, the more paper opacity decreased. In the aspect of Hanji's surface characteristics analyzed by an Image analyzer, the average gray level and its standard deviation of Hanji from the whole stalk pulp were rather lower than those of bast pulp because of better sheet formation of the former. However, high brightness Hanji showed high value of gray level. The sheet formation and paper opacity were increased with the decrease of standard deviation of gray level. From these results, gray level measurement could be used to predict the paper opacity as well as sheet formation. Sheet formation of whole stalk Hanji and its FDI measured by CLSM were higher than those of bast fibers. In conclusion, the sheet formation and opacity of Hanji could be evaluated by standard deviation value of Hanji's gray level using an Image analyzer and by fiber distribution index using CLSM.
임상병리 검사분야에 있어서 환자로부터 유래된 조직이나 세포의 형태학적 변화, 세포 생리, 세포 내 분자의 추적 및 신호전달 체계 등의 임상검사 및 관련 연구를 위한 빼놓을 수 없는 주요한 진단과 연구장비로서 현미경이 가지는 의미는 크다고 할 수 있다. 현미경에 대한 포괄적인 지식과 이해를 바탕으로 현미경의 올바른 사용, 관리와 유지보수는 신뢰도 높은 이미지 획득과 그에 따른 정확한 데이터 분석을 통한 질병의 진단을 위해서 반드시 요구되는 부분이라고 할 수 있다. 광학현미경의 표준 운영 절차(standard operating procedure, SOP)는 현미경의 작동 절차와 함께 검사실 규모에 따른 현장 사용자의 체계적인 현미경 장해 해결 방안과 기계적 원리에 대한 핵심 정보가 함께 수록되어야 한다. 현미경 유지관리 업무에는 대물, 접안렌즈와 현미경 내부 광학필터의 청소, 광원의 교체와 교정, XY재물대 유지보수, 공초점 레이저 주사 현미경(confocal laser scanning microscope)에서의 점확산함수(point spread function, PSF) 측정, 형광현미경에서의 검사 품질관리(quality control, QC)와 체계적인 현미경 장해 해결방안 등이 포함되어야 한다. 본 종설에서는 국제적 기준에 따른 레이저의 위험도에 따라 일부 현미경에 장착된 레이저 광원에 대한 안전지침과 보호장구에 대한 내용을 함께 소개하였다. 현미경을 통해 획득된 이미지는 촬영된 시점의 검체에 대한 모든 정보를 제공한다고 할 수 있으며, 적절한 유지보수 프로그램과 그에 따라 적합하게 관리된 현미경만이 이미지 데이터를 통한 정보의 획득, 올바른 해석과 정확한 진단에 반드시 필요한 선제 조건들이라고 하겠다.
Motivation: Polymerized actin-based cytoskeletal structures are crucial in shape, dynamics, and resilience of a cell. For example, dynamical actin-containing ruffles are located at leading edges of cells and have a significant impact on cell motility. Other filamentous actin (F-actin) bundles, called stress fibers, are essential in cell attachment and detachment. For this reason, their mechanistic understanding provides crucial information to solve practical problems related to cell interactions with materials in tissue engineering. Detecting and counting actin-based structures in a cellular ensemble is a fundamental first step. In this research, we suggest a new method to characterize F-actin wrapping fibers from confocal fluorescence image stacks. As fluorescently labeled F-actin often envelope the fibers, we first propose to segment these fibers by diminishing an energy based on maximum flow and minimum cut algorithm. The actual actin is detected through the use of bilateral filtering followed by a thresholding step. Later, concave actin bundles are detected through a graph-based procedure that actually determines if the considered actin filament is enclosing the fiber.
The purpose of this study was to evaluate on the interfacial morphology between dentin and restorative materials. In this in vitro study, the cavity wall restorated with 3 different kinds of tooth colored restorative materials [resin-modified Glass Ionomer cement (Fuji II LC), composite resin (Z-100), compomer (Dyract)]. The thirty extracted human molar teeth without caries and/or restorations are used. The experimental teeth were randomly divided into three groups of ten teeth each. In each group, Wedge shaped cavities (width: 3mm, length: 2mm, depth: 1.5mm) were prepared at the cementoenamel junction on buccal and lingual surfaces. The adhesive of composite resin were mixed with rhodamine B. Primer of composite resin, Prime & Bond 2.1 of Dyract and liquid of Fuji II LC were mixed with fluorescein. In group 1, the cavity wall was treatment with dentin conditioner, and then restorated with Fuji II LC. In group 2, the cavity wall was treatment with Prime & Bond 2.1 and then restorated with Dyract. In group 3, the cavity wall was etching with 10% maleic acid, applied with primer and bonding agent and then restorated with Z-100. The interface between dentin and restorative materials was observed by fluoresence imaging with a confocal laser scanning microscope. The results were as follows : 1. In Glass ionomer group, adaptation of resin modified Glass-ionomer restoration against cavity wall is tight, but the crack formed inside of restoration were observed. 2. In Dyract group, the penetration of resin tag is shorter and the width of hybrid layer is narrower than composite resin group. 3. In Z-100 group, primer penetrated deeply through dentinal tubule. Also bonding agent was penetrated along the primer, but the penetration length is shorter than primer part, and in 3-D image, the resin tag is conical shape and lateral branch is observed.
The recent industrial application requires technical methods to get the cutting fluid droplet surfaces in particular from the viewpoint of topography and micro texture. To characterize the surface topography of droplet, the combination of the confocal laser scanning microscope (CLSM) and wavelet filtering is well suited for obtaining the droplet geometry encountered in tribological research. This technique indicates a better agreement in obtaining an appropriate droplet surface obtained by the CLSM over a detail range of surface accuracy (resolution: $2{\mu}m$). And the results allow an excellent accuracy in a measurement of a droplet surface. The combination of extended focal depth measurement configured and multi-scale wavelet filtering has proven that it can construct a droplet surface in a successive and accurate way. A multi-scale approach of wavelet filtering was developed based on the decomposition and reconstruction of droplet surface by 2D wavelet transform using db9 (a mother wavelet of daubechies). Also this technique can be extended to characterize the quantification of droplet properties and other field in a wide range of scales. Finally this method is verified to be a better droplet surface modeling in a micro scale arising in a mist machining.
The effects of Condebelt press drying on the densification of recycled sheets made from KOCC , AOCC UKP and BKP were examined using CLSM (Confocal Laser Scanning Microscope) and thermal image analysis techniques. It was shown that significant improvement in sheet density, compression strength, tensile strength, surface smoothness was obtained by Condebelt drying. Densification effect by press drying was most prominent for recycled KOCC sheets. And this beneficial effect of Condebelt drying of increasing sheet density and strength was shown to provide opportunities of reducing the utilization ratio of AOCC for cost saving.
마이켈슨 간섭계를 이용한 간섭계형 현미경을 구성하고 특성을 조사하였다. 본 마이켈슨 간섭계형 현미경은 시료 표면의 반사 신호와 거울의 반사 신호가 합쳐져서 일어난 간섭 신호를 이용해서 3차원 영상을 얻는다. He-Ne(λ=633 nm)레이저를 사용한 마이켈슨 간섭계는 반파장 (312.5 nm)의 경로차가 생길 때마다 같은 무늬가 반복해져 나타난다. 본 마이켈슨 간섭계형 현미경은 이러한 간섭 신호를 이용하기 때문에 white-light interferometer 수준의 수직 해상도를 얻을 수 있었다. 구성한 마이켈슨 간섭계형 현미경으로 얻은 영상을 공초점 현미경의 영상과 비교 분석하였다.
