Isolation of Rhodocyclus gelatinosus KUP-74 and its characteristic in ${\delta}-aminolevulinic$ acid production (Rhodocyclus gelatinosus KUP-74의 분리 및 ${\delta}-aminolevulinic$ acid 생산의 특성)

  • A photosynthetic bacterium strain KUP-74 producing high amount of S-amino-levulinic acid(ALA) was isolated from soils, which was identified as Rhodocyclus gelatinosus. After 10 days cultivation under anaerobic-light condition at $30^{\circ}C$, 4 Klux and pH 6.8, 5 mg/l of ALA was formed extracellularly. ALA productions were increased up to 8 mg/l and 12 mg/l in cell cultivations either by the addition of 0.5% glycerol (v/v) or 10 mM of glycine and succinic acid, respectively, using Lascelles basal medium eliminated L-glutamic acid. By cultivation in the presence of 30 mM each D,L-glutamic acids and D,L-glutamines the yield of ALA showing a late induction phenomenon was reached the maximum value of 21 mg/l. Different culture times were needed to generate maximum ALA yields by the addition of initial precursors of $C_4$ and $C_5$ pathways in basal medium, as being 107 h and 262 h, respectively. 40 mg/l yield of ALA was observed by cell cultivation with the basal medium containing each 10 mM levulinic acid(LA) and glycine simultaneously.

Isolation and Characterization of Plant Pathogen that Cause Soft Rot Disease in Napa Cabbage (배추무름병 원인균 분리 및 특성 연구)

  • In order to establish in vitro infection model for research of plant pathogen based on tissue softening disease in napa cabbage, eighty independent bacterial strains were isolated from the softened napa cabbage tissues. Eight bacterial isolates were primarily screened with the generation of reproducible tissue softening disease to fresh napa cabbages within 24${\sim}$48 hours after inoculation. Through various microbiological biochemical and morphological examinations, three Gram (-) isolates which harbor independent biological properties were finally chosen, and named as RBI, RB2 and RB6. Collective results obtained from API 20E test and analyses of VITEK 2 COMPACT and nucleotide sequences of 165 rRNA of each isolate proposed that isolates RBI and RB2 are close to the Erwinia carotovora subsp. odorifera, and RB6 is close to the Erwinia carotovora subsp. carotovora. These isolates grew optimally at $30^{\circ}C$ with neutral pH culture condition. The isolates caused softening tissue disease with dose-dependent manner regardless of pre-surface damages of napa cabbage. Minimum dose to cause soft rot disease for RBI, RB2 or RB6 were $8.0{\times}10^8$ CFU/mt $10^9$ CFU/ml or $4.7{\times}10^6$ CFU/ml respectively. These isolates caused tissue softening disease to eggplant, paprika and napa cabbage out of 14 different tested vegetables, indicating that these isolates damages specific plant tissues. The bacterial isolates obtained in this research and in vitro plant infection model will be adapted in the understanding of the mechanism of pathogenesis by plant pathogen.

Isolation and Characterization of Insoluble Phosphate-Solubilizing Bacteria with Antifungal Activity (항진균능을 가진 불용성 인산 가용화 세균의 분리 및 특성)

  • To develop multifunctional microbial inoculant, an insluble phosphate-solubilizing bacterium with antifungal activity was isolated from plant rhizospheric soil. On the basis of its morphological, cultural and physiological characteristics and Biolog analysis, this bacterium was identified as Pseudomonas fluorescens RAF15. P. fluorescens RAF15 showed antifungal activities against phytopathogenic fungi Botrytis cinerea and Rhizoctonia solani. The optimal medium composition and cultural conditions for the solubilization of insoluble phosphate by P. fluorescens RAF15 were 1.5% of glucose, 0.005% of urea, 0.3% $MgCl_2{\cdot}6H_2\;0.01%\;of\;MgSO_4{\cdot}7H_2O\;0.01%,\;of\;CaCl_2{\cdot}2H_2O$, and 0.05% of NaCl along with initial pH 7.0 at $30^{\circ}C$. The soluble phosphate production under optimum condition was 863 mg/L after 5 days of cultivation. The solubilization of insoluble phosphates was associated with a drop in the pH of the culture medium. P. fluorescens RAF15 showed resistance against different environmental stresses like $10-35^{\circ}C$ temperature, 1-4% salt concentration and pH 2-11 range. The strain produced soluble phosphate to the culture broth with the concentrations of 971-1121 mg/L against $CaHPO_4$, 791-908 mg/L against $Ca_3(PO_4){_2}$, and 844 mg/L against hydroxyapatite, respectively. However, the strain produced soluble phosphate to the culture broth with the concentrations of 15 mg/L against $FePO_4$, and 5 mg/L against $AlPO_4$, respectively.

  • Amelogenesis imperfecta (AI) occurs either in isolation or in association with other dental abnormalities and systemic disorder. A rare syndrome associating AI with nephrocalcinosis was named as Enamel Renal Syndrome (ERS; OMIM #204690). This syndrome is characterized by severe enamel hypoplasia, failed tooth eruption, intra pulpal calcifications, enlarged gingiva, and nephrocalcinosis. Nephrocalcinosis is a condition where calcium salts are deposited in renal tissue, and this may lead to critical kidney complications. This rare syndrome shows pathognomonic oral characteristics that are easily detectable at an early age, which proceeds the onset of renal involvement. Pediatric dentists are the first oral health practitioners whom ERS patients will meet at early age. The role of pediatric dentists is critically important for early diagnosis and referral of patients to both nephrologists for renal assessment and geneticists for identification of causative mutation and diagnosis. Early detection of renal involvement may provide chances to prevent further undesired renal complications.

Isolation and identification of soycurd forming lactic acid bacteria which produce GABA from kimchi (김치로부터 GABA를 생산하는 커드 형성 젖산균의 분리 및 동정)

  • Gamma amino butyric acid (GABA), known as a non-protein amino acid and major inhibitory neurotransmitter in the brain, has several functional properties such as neurotransmission, induction of hypotension, tranquilizer, and diuretic effects. The purpose of this study was to isolate and identify lactic acid bacteria, producing high GABA in fermented soy curd. Thirty-two strains of tofu-forming lactic acid bacteria were isolated from kimchi which a traditional Korean food fermented with many kind of microorganism. Among 32 strains, four strains (strain No. 10, 104, 214, 249) formed firm soycurd. In order to select lactic acid bacteria having high GABA producing potential, the isolated strains were cultured in the soymilk and fermented for 48 hr at $37^{\circ}C$. A strain No. 383, which showed highest GABA contents in fermented soycurd, was identified as L. sakei by 16S rDNA sequencing and API analysis, and named as L. sakei 383. L. sakei 383 showed optimal growth up to 24 hr at $35^{\circ}C$ in MRS broth. The optimal time and temperature for GABA production were 18 hr and $35^{\circ}C$ in soymilk. In the optimal condition time and temperature, GABA content of fermented soycurd by L. sakei 383 was 8.65 mg/100 g.

Influence on Efficacy of $\beta$-hemolytic Streptococcus iniae Vaccine by Mixed Infections with Edwardsiella tarda and Neoheterobothrium hirame in Cultured Olive flounder, Paralichthys olivaceus (양식 넙치에서 Edwardsiella tarda와 Neoheterobothrium hirame 혼합 감염이 $\beta$-용혈성 Streptococcus iniae 불활화백신의 효능에 미치는 영향)

  • We evaluated the efficacy of $\beta$-hemolytic Streptococcus(S.) iniae vaccine on cultured olive flounder. Three hundred flounders(weight $50{\pm}5$ g) were obtained from two farm at Wando and Taean in the southern and western coast of Korea at May and June 2007, respectively. Twenty of flounders moved in 0.5 tons aquaria in land-marine tank system of National Veterinary Research and Quarantine Service. Seawater was transported from the sea of Inchon in western Korea, and water temperature maintained to $22^{\circ}C$ and $25^{\circ}C$ during the vaccination and challenge test, respectively. We used the formalin-inactivated $\beta$-hemolytic S. iniae vaccine produced by domestic manufacturers. The vaccine was intraperitoneally administered to fish. The vaccinated and control group were challenged with intraperitoneal injection by virulent S. iniae SI-36 isolates with $5.0{\times}10^8$ CFU/fish at 3 weeks after vaccination. We evaluated the vaccine efficacy by calculating numbers of dead fish, and observing of clinical signs, exterior and gross lesions, and examining bacteria isolation and identification. Thirty-four(25.2%) of 135 control and vaccinated group fish were dead with serious anemia, abdominal extension, and hernia of intestine during 3 weeks post vaccination. We isolated Neoheterobothrium hirame from the buccal cavity and Edwardsiella tarda from kidney of dead and diseased fish. When infected fish with these agents were challenged with S. iniae SI-36 isolates, the cumulative mortality of control and vaccinated group were 86.7, and 46.7%, respectively. However, significant differences(p<0.05) were observed on cumulative mortality between control(20.0%) and vaccinated group(95.0%) at second trials with 40 healthy, and relative percent survival(RPS) was 78.0%. We confirmed that the efficacy of $\beta$-hemolytic S. iniae vaccine on olive flounder were impacted by health condition such as bacterial and parasitic diseases.

Factors Influencing Protoplast Isolation and Culture in Three Capsicum Species (세 가지 고추속 식물의 원형질체 분리 및 배양에 미치는 요인)

  • Protoplasts were isolated from cotyledons, hypocotyls, and mesophyll tissues of three species of Capsicum species (C. anuumm, C. bacatuum, and C. chacoense). Combination of Cellulysin (1%) and Macero-zyme (0.25%) in 0.65 M sorbitol was found to be the most effective for the digestion of cell wall, regardless of the Capsicum species. Antioxidant MES (2-[N-Morpholino]ethanesulfonic acid) in the enzyme solution helped protoplasts overcome browning. After 5 days of initial culture, Cell division occurred in modified K8p medium containing 1~5 mg/L zeatin, 0.5 mg/L IAA, 0.1~0.5 mg/L TDZ, and 1 mg/L 2,4-D under continuous dark condition at $25^{\circ}C$. Semi-solid agarose culture method was more effective than liquid culture, and it also protected the cells from browning caused by polyphenolic compound released during protoplast culture. A total of 4000 calli were obtained from protoplast culture of different capsicum species. All of these calli were transferred to the 100 combinations of regeneration media using various plant growth regulators; TDZ, IAA, 2ip, BAP, NAA, and zeatin. These calli derived from protoplast of three species of capsicum were, however, not differentiated into shoots.

The Psychological Structure and Characteristic of Hasteful Behaviors: Understanding the Relation between Hasteful Behaviors, Omission of Checking and Achievement Striving (서두름 행동의 심리적 구조 및 특성 파악: 서두름 행동, 확인생략행동, 성취욕구 간의 관계 이해)

  • Hasteful behavior means choosing the best suited methods while behaving fast and quickly. We can't conclude whether hasteful behavior is totally bad or good. Striving for achievement of own certain purpose reflects achievement motivation or need for achievement. However, this striving also has potentiality of missing confirmation, therefore the potentiality may cause errors. The aim of this study is to investigate the psychological structure and characteristic of the hasteful behavior. One hundred ninety-one students conducted Hasteful Behavior Questionnaire, Achievement Motivation Measuring Scale, and NEO Personality Inventory. We analyzed data of 188 respondents, because of missing value. The result of factor analysis showed that hasteful behavior consisted of 5 factors- 「Uncomfortableness」, 「Time Pressure」, 「Isolation」, 「Boring Condition」, and 「Expecting Rewards」. According to correlations among the hasteful behavior factors and the relationship between hasteful behavior and achievement motivation, we found that hasteful behavior had two aspects, one was "Missing Confirmation(MC)" and the other was "Need for Achievement(NA)". We also found that 「Time Pressure」 was related to the both aspects. MC had a positive relation to Neuroticism, whereas MC correlated negatively with Conscientiousness. On the other hand, NA had a positive relationship with Extraversion and Achievement Striving.

Changes in Sensitivity Levels of Botrytis spp. Population Isolated from Lily to Fungicides and Control under Field Condition (나리에서 분리한 잎마름병균의 살균제에 대한 감수성 변화와 포장 방제)

  • Forty eight isolates of Botrytis elliptica and 23 isolates of B. cinerea from several locations in Korea were tested for resistance to fungicides used in the farmer's fields. Isolation frequency of B. elliptica having $EC_{50}$ (effective concentration of 50%) value $500-1000{\mu}g/ml$ to benomyl and mancozeb appeared highly, suggesting that the two fungicides are not effective in controlling leaf blight of lily in the field. The isolates were tested for resistance to fungicides procymidone and iprodione which were most commonly used in the farmer's fields. The rates of $EC_{50}$ value $5-50{\mu}g/ml$ to procymidome and iprodione were 93.7% and 100%, respectively, and those of $0-0.1{\mu}g/ml$ to diethofencarb+carbendazim and fludioxonil were 98.0% and 93.8%, respectively. In the rain-protected cultivation, control of leaf blight of lily was the most effective when iprodine, diethofencarb+carbendazim, and fludioxonil were sprayed alternately four times during the growing season.

Shipboard Active Phased Array Antenna System for Satellite Communications (위성 통신용 선박 탑재 능동 위상배열 안테나 시스템)

  • In this paper, the novel shipboard Active Phased Array Antenna(APAA) system for maritime mobile satellite communications is introduced. The antenna uses novel technologies like wide range hybrid tracking, single antenna elements with both of Rx and Tx, asymmetrical array structure, interference isolation between Rx and Tx, and error correction method from frequency scan effect. The antenna has single aperture for both of Rx and Tx with 32 $\times$ 4 two-dimensional array. The antenna has two beams. Its frequencies are 7.25 ~ 7.75 GHz for Rx and 7.9 ~ 8.4 GHz for Tx. The antenna gains are 35.4 dBi for Rx and 35.7 dBi for Tx, those are 54 % of efficiency. The electrically steering ranges are $\pm$35$^{\circ}$ of elevation direction and $\pm$4$^{\circ}$ of azimuth direction. The mechanical control ranges at hybrid tracking capability are continuous 360$^{\circ}$ of azimuth direction and $\pm$10$^{\circ}$ of elevation direction. The antenna has 2.2$^{\circ}$ of 3 dB beamwidth, -14 dB of sidelobe level, and 21 dB of cross-pol suppression. The antenna performance was measured by near field measurement set. Its system performance was tested on the ship motion simulator and with the satellite transponder simulator. The test result showed that its tracking error was within -3 dB from its peak gain under motion condition. The antenna system was tested by real modulated Direct Broadcasting Satellite(DBS) signals to check its communication processing function.
