Enhanced Nasal Absorption of Ketoconazole by Inclusion with Cyclodextrin (시클로덱스트린과의 포접에 의한 케토코나졸의 비점막 흡수증가)

  • Inclusion complexes of ketoconazole(KT) with ${\alpha}^_$, ${\beta}^_$cyclodextrin(CD) and $dimethy1-{\beta}-cyclodextrin$ (CD) and $dimethy1-{\beta}-cyclodextrin(DM{\beta}CD)$ as nasal absorption enhancer were prepared in 1: 2 molar ratios by freeze-drying and solvent evaporation methods. In order to compare with the intrinsic absorptivity of KT in the jejunum(J) and the nasal cavity(N), the in situ simultaneous perfusion method was employed. The in situ recirculation study revealed that KT-CD inclusion complexes with the greater stability constant and the faster dissolution rate proportionally increased the absorption of KT in the J and N of rats. The rank order of apparent KT permeability$(P_{app}\;:\;cm/sec\;{\time}\;1O^{-5}{\pm}S.E.)$, corrected by surface area of absorption, was $5.10{\pm}0.3(N,\; KT-DM{\beta}CD)$ )> $4.13{\pm}0.4(N,\;KT-{\beta}-CD)$ )> $3.52{\pm}0.2(N,\;KT-{\alpha}-CD)$ )> $2.76{\pm}0.3(J,\; KT-DM{\beta}CD)$ )> $2.61{\pm}0.5(J,\;KT-{\beta}-CD)$ )> $2.42{\pm}0.4(J,\;KT-{\alpha}-CD)$ at pH 4.0. The in crease in permeability of $KT-DM{\beta}CD$ inclusion complex was 2.6 folds in the J and 4.5 folds in the N when the perfusing solution was changed from the buffer(pH 4.0) to saline. The absorption rate of $KT-DM{\beta}CD$ inclusion complex after nasal administration was more rapid than those of ketoconazole alone and $KT-DM{\beta}CD$ inclusion complex after oral administration to rats. In comparision with an oral administration of ketoconazole suspension in corn oil, the relative bioavailability was calculated 137.3% for the oral and 195.0% for nasal $KT-DM{\beta}CD$ inclusion complex in rats. The present results suggest that $KT-DM{\beta}CD$ inclusion complex may serve as a potential nasal absorption enhancer for the nasal delivery of ketoconazole.

Effects of Mitochondrial Reactive Oxygen Species on Neuronal Excitability in Rat Spinal Substantia Gelatinosa Neurons

  • Recent studies indicate that reactive oxygen species (ROS) are critically involved in persistent pain primarily through spinal mechanisms, and that mitochondria are the main source of ROS in the spinal dorsal horn. To investigate whether mitochondrial ROS can induce changes in membrane excitability on spinal substantia gelatonosa (SG) neurons, we examined the effects of mitochondrial electron transport complex (ETC) substrates and inhibitors on the membrane potential of SG neurons in spinal slices. Application of ETC inhibitors, rotenone or antimycin A, resulted in a slowly developing and slight membrane depolarization in SG neurons. Also, application of both malate, a complex I substrate, and succinate, a complex II substrate, caused reversible membrane depolarization and enhanced firing activity. Changes in membrane potential after malate exposure were more prominent than succinate exposure. When slices were pretreated with ROS scavengers such as phenyl-N-tert-buthylnitrone (PBN), catalase and 4- hydroxy-2,2,6,6-tetramethylpiperidine-1-oxyl (TEMPOL), malate-induced depolarization was significantly decreased. Intracellular calcium above $100{\mu}M$ increased malateinduced depolarization, witch was suppressed by cyclosporin A, a mitochondrial permeability transition (MPT) inhibitor. These results suggest that enhanced production of spinal mitochondrial ROS can induce nociception through central sensitization.

Effects of Hydrological Condition on the Coupled Thermal-Hydrological-Mechanical Behavior of Rock Mass Surrounding Cavern Thermal Energy Storage (암반 공동 열에너지저장소 주변 암반의 수리적 조건에 따른 열-수리-역학적 연계거동 분석)

  • The thermal-hydrological-mechanical (T-H-M) behavior of rock mass surrounding a large-scale high-temperature cavern thermal energy storage (CTES) at a shallow depth has been investigated, and the effects of hydrological conditions such as water table and rock permeability on the behavior have been examined. The liquid saturation of ground water around a storage cavern may have a small impact on the overall heat transfer and mechanical behavior of surrounding rock mass for a relatively low rock permeability of $10^{-17}m^2$. In terms of the distributions of temperature, stress and displacement of the surrounding rock mass, the results expected from the simulation with the cavern below the water table were almost identical to that obtained from the simulation with the cavern in the unsaturated zone. The heat transfer in the rock mass with reasonable permeability ${\leq}10^{-15}m^2$ was dominated by the conduction. In the simulation with rock permeability of $10^{-12}m^2$, however, the convective heat transfer by ground-water was dominant, accompanying the upward heat flow to near-ground surface. The temperature and pressure around a storage cavern showed different distributions according to the rock permeability, as a result of the complex coupled processes such as the heat transfer by multi-phase flow and the evaporation of ground-water.

  • Recently sewage facilities mainly consisted of concrete structures are being deteriorated seriously by biodeterioration originated from sulfur-oxidizing bacteria. In this study, to prevent biochemical corrosion of the sewer concrete, antibiotics which prevent growth of sulfur-oxidizing bacteria were developed and antimicrobial performance of it was investigated. After that, to consider applicability of antibiotics to concrete, physical properties such as bond strength, resistance to abraision, water absorption and air permeability of concrete covered with inorganic and complex antibiotics were investigated.

Effects of Annealing on Electromagnetic Wave Absorption Characteristics in FeSiCr Flakes/Polymer Composite Sheets (FeSiCr 박편/폴리머 복합 시트의 전자파 흡수 특성에 미치는 합금 어닐링 효과)

  • The soft magnetic Fe-Si-Cr flakes with the thickness of about 1 ${\mu}m$ were annealed at 500 and $700^{\circ}C$ for 1 h, and the composite sheets for electromagnetic wave noise absorber available for quasi-microwave band were fabricated by using these annealed flakes and polymer. Further the power loss characteristics of the composite sheets was investigated to clarify the annealing effect on electromagnetic wave absorption properties. The power loss decreased in the frequency range of several GHz when the annealed flakes were used as compared to the sheet using the as-milled FeSiCr alloy flakes. Moreover the sheets using annealed flakes exhibited lower value of real and imaginary part of complex permeability. These inferior electromagnetic wave absorption properties of the composite sheets using annealed alloy flakes were considered to be obtained by the enhanced eddy current effect upon annealing-induced recovery of microstructure and resulted low complex permeability.

Absorption Properties of Thin Permalloy-Rubber Absorbers in Mobile Telecommunication Frequency Bands (Permalloy를 이용한 이동통신주파수 대역용 박형 전파흡수체의 제조 및 특성 연구)

  • The raw materials of permalloy were processed the ball-mill for 30 hours and the shape of permalloy particles was changed from sphere to flake type, which was observed using scanning electron microscope. The complex permittivity and permeability spectra and reflection loss of permalloy-rubber composite was measured using Network Analyzer in order to investigate the relationship between the microwave absorption and the material constants. The flake type permalloy-rubber composite showed high reflection loss, which was due to the high complex permittivity and permeability. Also, the matching frequency shifted toward lower frequency range with microwave absorber thickness, and the maximum reflection loss of -6.1 dB was observed in $1.65\;GHz{\sim}1.86\;GHz$ for a 1.3 mm thickness.

  • Pervaporation has become important as a method to separate liquid mixtures, sepecially azeotropic and close boiling-point mixtures. Especially, water-alcohol separations have been carried out a lot because of the practical interests in the industries. However, outstanding membranes with high selectivity and high permeability have not been available in common use yet. In order to separate selectively out the water mixed with alcohols with an aim of the purification of the alcohols, a membrane has tO have excellent affinity to water. Among the hydrophilic polymers, polyacrylic acid and poly(vinyl alcohol) are used widely. In recent years, ionomers and polyion complexes, better hydrophitic materials, start to be used. The polyion complex membranes, consisting of polyacrylic acid (PAA) and polycation, showed excellent permeation rates and selectivities. It was known that among the polycations, ioneries, which have quaternary ammonium groups in the backbone chain, were more effective in giving membranes of higher permselectivities. On this base, syntheses and characterizations of the polycations, with different chemical structures from the published ones, for the polyion complex membrane formation were studied in this paper.

  • The antibacterial activity of chitosan-alginate-Fe(II) complex (CAFC) against Escherichia coli and Staphylococcus aureus, and an opportunistic pathogen, Candida albicans, was investigated. A concentration of 1 mg/1 was needed to inhibit the growth of S. aureus and E. coli, while 100 mg/liter was sufficient for the growth inhibition of Candida albicans. The ion leakage of potassium and phosphate from E. coli cell and the penetration of ethidium bromide dye into it indicate that CAFC might be able to increase the cell permeability and consequently cellular leakage, thus leading to cell plasmolysis. Scanning electronmicroscope showed that E. coli cells treated with CAFC became irregular, swelling and expanded. In a field trial, control piglets showed average mortality of up to 60% within 3 days after the onset of diarrhea. In contrast, CAFC-treated groups without mortality was decreased to average 56% on the 1 st day after the treatment, and average 7% on the 3rd day. After then, piglets with diarrhea was not found.

Electromagnetic Wave Absorption Characteristics of Nanocrystalline FeCuNbSiB Alloy Flakes/Polymer Composite Sheets with Different Flake Thickness

  • This study examined the effects of a decrease in thickness of magnetic alloy flakes on the electromagnetic wave absorption characteristics of nanocrystalline $Fe_{73.5}Cu_1Nb_3Si_{15.5}B_7$ (at.%) alloy flakes/polymer composite sheets available for a quasi-microwave band. The thickness of FeCuNbSiB alloy flakes decreased to 1-2 $\mu$m with increasing milling time up to 24 h, and the composite sheet including alloy flakes milled for 24 h exhibited considerably enhanced power loss properties in the GHz range compared to the sheets having non-milled alloy powders. Although a considerable increase in loss factor upon milling was observed in the narrow frequency range of 4-6 GHz, there was no correlation between the complex permeability and flake thickness. However, the complex permittivity increased with increasing milling time, and there was good agreement between the milling time and the frequency dependences of the complex permittivity and power loss.

  • The effect of aqueous and alcoholic extract of seeds of Nigella sativa (N. sativa) was studied on the spontaneous movements of the whole worm (w.w) preparation and nerve muscle (n.m) complex of Setaria cervi (S. cervi). Both the extracts caused inhibition of spontaneous movements of the w.w and n.m complex characterized by initial stimulation followed by irreversible paralysis, with the exception that aqueous extract produced a partially reversible paralysis of w.w preparation. Lesser concentration of both the extracts was required to inhibit the movements of n.m complex than the w.w. Suggesting a cuticular permeability barrier. The lethal concentration $50\;(LC_{50})$ and lethal concentration $90\;(LC_{90})$ were 30 and 55 ng/ml for aqueous and 45 and 60 ng/ml for alcoholic extracts respectively.
