Controlled Release of 2,4-D(2,4-Dichlorophenoxy Acetic Acid ) from the Complex of Rice Husk Lignin and 2,4-D-IV.   • Inactivation in soil absorption, translocation of 2, 4-D by plants vary depending upon soil environments and herbicide formulations. Experiment was conducted in a glasshouse using rectangular pots($1350cm^2$) to evaluate the growth responses of barnyardgrass (Echinochloa crus-galli) and Indian jointvetch (Aesehyrcomene indica) to two formulations of 2, 4-D. The formulations used were 40% 2, 4-D amin salt (2, 4-D/AS) and 19.7% complex of rice husk lignin and 2, 4-D (2, 4-D/LG) which were applied at 200g ai/ha. Soil environments included fertilizer levels, soil pH, organic matter contents, and soil textures, Each treatment was replicated three times. The herbicidal activity of 2.4-D increased and lasted with increased levels of fertilizer. The activity also increased and lasted with low soil pH and decreased content of organic matter. Generally 2, 4-D/LG showed higher and longer herbicidal activity than 2. 4-D/AS for both test plants under all conditions applied. However, the herbicidal activity was influenced by the formulations more than by soil textures. It was thought that 2, 4-D/AS was released in a short time and inactivated readily while 2, 4-D/LG was slowly released and gave an opportunity of absorption by plants for a long period.

Effect of Amino Acids and Organic Nitrogen Sources on Cyclosporin A Fermentation by Tolypocladium inflatum (Tolypocladium inflatum을 이용한 Cyclosporin A 발효에서 아미노산과 유기질소원의 영향)

  • Cyclosporin, an immunosuppressant, is a representative group of biologically active secondary metabolites produced by the fungus Tolypocladium inflatum. The amount and ratio of cyclosporin derivatives in the culture broth are an important factors for the production of cyclosporin A and the purification in the industrial process. Therefore, we studied the effect of amino acids and complex organic nitrogen sources using Tolypocladium inflatum mutants on the productivity of cyclosporin A and the ratio of cyclosporin derivatives. Overproducing mutant YHC-004 having seven times higher productivity than mother strain's could be obtained through the artificial mutation by UV irradiation. The concentration and kind of organic nitrogens and amino acids shows the profound effect on the productivity of cyclosporin A and ratio of cyclosporin derivatives. As a result, it was possible to raise the productivity and the ratio of cyclosporin A up to 3,430 mg/L and 93% respectively, but on the other hand the other cyclosporin derivatives decreased less than 2% in the culture broth.

Electrochemical Degradation of Phenol by Using Reticulated Vitreous Carbon Immobilized Horseradish Peroxidase (Horseradish Peroxidase가 고정화된 다공성 탄소 전극을 이용한 페놀의 전기화학적 분해)

  • Horseradish peroxidase, had the phenol degradation rate of 95% in aqueous phase, was covalently immobilized on the surface of reticulated vitreous carbon(RVC) and the degradation of phenol was performed with in situ generated $H_2O_2$-immobilized HRP complex in an electrochemical reactor. The incorporation of carboxylic group on the RVC surface was confirmed by FT/IR spectrometry and 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide hydrochloride(EDC) was used for peptide bonds between the carboxylic groups on the RVC surface and amine groups from HRP. The optimal conditions of in situ $H_2O_2$ generation such as concentration($10{\sim}200$ mM) and pH($5.0{\sim}8.0$) of electrolyte, supply of $O_2(10{\sim}50$ mL/min) and applied voltage($-0.2{\sim}-0.8$ volt, vs. Ag/AgCl) from potentiostat/galvanostat were determined by concentration of hydrogen peroxide and current efficiency. It was observed that the RVC immobilized HRP was stable maintaining 89% of the initial activity during 4 weeks. The phenol degradation rate of 86% was attained under the optimal condition of in situ $H_2O_2$ generation.

  • Demand of glass fiber reinforced composites (GFRC) increased with developing aircraft and defense industries using resin transfer molding (RTM) process to produce complex product. In this research, wetting, interfacial, and mechanical properties were evaluated with different Cross-linking Density by Molecular Weight of Hardener. Epoxy resin as matrices was used bisphenol-A type and amine-type hardeners with different molecular weight. Specimens were manufactured via RTM and wetting property of resin and glass fiber (GF) mat was evaluated to viscosity of epoxy and injection time of epoxy matrix. Mechanical property of GFRC was determined via flexural strength whereas interfacial properties were determined by interlaminar shear strength (ILSS) and interfacial shear strength (IFSS). The difference in mechanical property depends upon the fiber weight fraction (wt %) of GFRC by RTM as well as the different Molecular Weight of Hardener.

Heterocyclic Amines Removal by Binding Ability of Lactic Acid Bacteria Isolated from Soybean Paste (된장에서 분리된 유산균의 결합력에 의한 Heterocyclic Amines 제거)

  • The objective of the this study was to investigate the binding capacity and removal ability of lactic acid bacterial strains obtained from Korean soybean paste for mutagenic heterocyclic amines (HCAs) formed during cooking of protein-rich food at high temperature. Among 19 strains identified by carbohydrate fermentation and 16S rRNA sequencing, the live cell or cell-free culture supernatant of Lactobacillus acidophilus D11, Enterococcus faecium D12, Pediococcus acidilactici D19, L. acidophilus D38, Lactobacillus sakei D44, Enterococcus faecalis D66, and Lactobacillus plantarum D70 inhibited the mutagenesis caused by either 3-amino-1,4-dimethyl-5H-pyrido[4,3-b] indole (Trp-P-1) or 3-amino-1-methyl-5H-pyrido[4,3-b] indole (Trp-P-2) in Salmonella typhimurium TA98 and TA100. The bacterial cells of the isolated strains showed greater binding activity than the pure cell wall, exopolysaccharide, and pepetidoglycan. The carbohydrate moieties of the cell wall or protein molecules on the cell surface have a significant role in binding Trp-P-1 and Trp-P-2, since protease, heating, sodium metaperiodate, or acidic pH treatments significantly (P<0.05) reduced the binding efficacy of the tested bacteria. Addition of metal ions or sodium dodecyl sulfate decreased the binding ability of E. faecium D12, L. acidophilus D38, and E. faecalis D66. Therefore, the binding mechanisms of these strains may consist of ion-exchange and hydrophobic bonds. Especially, the high mutagen binding by L. acidophilus D38 and L. plantarum D70 may reduce the accumulation or absorption of Trp-P-1 and Trp-P-2 in the small intestine via increased excretion of a mutagen-bacteria complex.

Effects of Probiotics as an Alternative for Antibiotics on Growth Performance, Nutrient Digestibility, Noxious Gas Emission and Fecal Microbial Population in Growing Piglets (항생제 대체 생균제가 자돈의 생산성,영양소 이용률, 유해가스 발생량 및 분내 미생물 수에 미치는 영향)

  • This study was conducted to determine the effect of probiotics as an alternative for antibiotics on growth performance, nutrient digestibility, noxious gas emission and fecal microbial population in growing piglets. A total of 96 piglets ($22.5{\pm}1.3$kg average body weight) were allotted to 3 different treatment groups and replicated 4 times with 8 piglets per replicate in randomized complete block design. Treatments were T1) (Control, basal diet+0.2% antibiotics), T2) 0.2% probiotics complex and T3) 0.3% Bacillus probiotics. During the whole experiment period, there were no differences (p>0.05) in average daily gain (ADG), average daily feed intake (ADFI) and feed efficiency. However, digestibility of dry matter, crude protein, ether extract, nitrogen free extract and crude ash were showed higher in probiotics groups (T2 and T3) than those of control. In noxious gas emission, ammonia, amine, hydrogen sulfide and mercaptan were significantly (p<0.05) reduced in T2 and T3 treatments compared to those in control. Moisture content of feces was not significantly different among treatments. The colony forming units (CFU) of total bacteria, E. coli and thermoduric bacteria in feces were significantly different among treatments. The CFU of total bacteria, E. coli and thermoduric bacteria in T3 treatment were reduced by feeding probiotics B. From this study, we suggest that probiotics A and B are likely able to improve the growth performance and nutrients digestibility, reduce noxious gas emission and change the fecal microbial composition in growing piglets.

Determination of Heterocyclic Amines in Roasted Fish and Shellfish by Liquid Chromatography-Electrospray Ionization/Mass Spectrometry (Liquid chromatography-mass spectrometry를 이용한 가열 조리된 어패류에서의 heterocyclic amines 함량 분석)

  • Heterocyclic aromatic amines (HCAs) are mutagenic and carcinogenic substances that are formed during the heating of protein-rich foods. HCAs are generally found at low amounts in a complex matrix, which requires sophisticated analysis. In this study, HCAs were extracted from lyophilized fish and shellfish samples using solid-phase extraction (SPE) and determined by liquid chromatography-electrospray ionization/mass spectrometry (LC-ESI-MS). The HCA recoveries in the fish and shellfish ranged from 15.7 to 74.7% with standard deviations from 0.2 to 7.63%. And HCA concentrations ranged from 0.8 to 1,117.7 $ng/g^{-1}$ in cooked food samples. 1-methyl-9H-pyrido[3,4-b]indole (Harman), 9H-pyrido[3,4-b]indole (Norharman), and 2-amino-1-methyl-6-phenylimidazo[4,5-b]pyridine (PhIP) were the most abundant HCAs formed in the muscle of fried mackerel, at levels of 1,117.7, 926.6, and 133.7 ng/g, respectively. 3-amino-1-methyl-5H-pyrido[4,3-b]indole (Trp-P-2), 2-aminodipiryrido[1,2-a:3,2-d]imidazole(Glu-P-2), 2-amino-9H-pyrido[2,3-b]indole(A${\alpha}$C), 2-amino-3methyl-9H-pyrido [1,2-a:3,2-d]imidazole(MeA${\alpha}$C), 2-amino-3,4,7,8-tetramethylimidazo[4,5-f]quinoxaline (TriMeIQx), 2-amino-3,7,8-trimethylimidazo [4,5-f]quinoxaline(7,8-DiMeIQx), and 2-amino-3,8-dimethylimidazo[4,5-f]quinoxaline (MeIQx) were only detected by small quantities ranged from 1.5 to 98.6 ng/g. Overall, this study provides useful information on HCA levels in fish and shellfish products consumed in Korea.
