클로닉신과 시클로덱스트린과의 포접복합체 형성 (Inclusion Complexation of Clonixin with Cyclodextrins)

  • The aim of this study is to increase the solubility and dissolution rate of clonixin by inclusion complex formation. Inclusion complexes of clonixin, a non-steroidal antiinflammatory drug, with ${\beta]-cyclodextrin$ were $2-hydrolrypropyl-{\beta]-cyclodextrin$ were prepared by freeze drying method. Inclusion complex formation of clonixin with cyclodextrins was determined by UV, IR and DSC. The apparent stability constants were calculated from the phase solubility diagrams. Dissolution rate and solubility of clonixin in water markedly increased by the complex formation.

오메프라졸과 $\gamma$-시클로덱스티린과의 복합체 형성 및 제제학적 특성 (Interaction between Omeprazole and $\gamma$-Cyclodextrin)

  • The interaction of omeprazole(OMP) with $\gamma$-cyclodextrin($\gamma$-CyD) was investigated by solubility study and the complexation was confirmed by means of UV/VIS spectrophotometer, circular dichroism, differential scanning calorimeter, and $^{1}$H nuclear magnetic resonance spectra. The stability, dissolution rate, and partition coefficient of the complex were measured. The results present that the benzimidazole moiety and a part of pyridine ring containing sulfur atom of OMP might be included into the cavity of $\gamma$-CyD and the formation type of inclusion complex appeared to be B$_{s}$. The stoichiometric ratio of OMP to $\gamma$-CyD in the complex was found to be 1:1 and the stability constant of the complex found to be 97.1 M$^{-1}$. And the dissolution rate of OMP was markedly increased by inclusion complex formation with $\gamma$-CyD, and so it was above 90% in 5 min. from solid complex. Oil to water partition coefficient of OMP-$\gamma$-CyD complex was 60, which is significantly higher than that of OMP itself, 36.4. The degradation rate constant of OMP were greater than OMP-$\gamma$-CyD complex in aqueous solutions of various pHs, and the half lives of OMP and OMP-$\gamma$-CyD at pH 9 were 279.2 and 509.9 days, respectively, showing that the complex was more stable than OMP, therefore it was thought that OMP was stabilized by inclusion formation with $\gamma$-CyD.

푸로세미드의 안정성 및 생체내 이용율에 미치는 ${\beta}-$시클로덱스트린의 영향 (Influence of ${\beta}-Cyclodextrin$ on Stability and Bioavailability Of Furosemide)

  • Inclusion complex formation of furosemide with ${\beta}-cyclodextrin({\beta}-CyD)$ in solid state was confirmed by X-ray diffractometry, IR spectroscopy and differential scanning calorimetry (DSC). The solid complexes of ${\beta}-CyD$ with furosemide in molar ratio of 2 : 1 were prepared by solvent evaporation method. The photodegradation of furosemide in alkaline solution under the light and the hydrolysis of furosemide in acidic solution were not inhibited by complex formation with ${\beta}-CyD$. However, the bioavailability of furosemide was improved by complex formation with ${\beta}-CyD$ after oral administration to rats.

  • 各 種 아민(triethylamine, diethylamine, pyridine, diphenylamine, dimethylaniline)과 各種 Electron Acceptor($CCl_4,\;ICl,\;I_2$)를 Hexane 溶媒 存在下에서 作用시킨 結果 各各 Charge Transfer Complex를 形成하였으나 그 形成能은 아민의 Basicity에 比例하였고 Electron Donor의 Polarity의 差가 클 때에는 다른 形式의 Complex를 形成함을 紫外線 吸收 分光法으로 確認하였다. Complex 形成能과 아민의 Basicity 및 Electron Acceptor의 Polarity의 關係를 論하였다.

Furfuryl유도체와 하라이드 사이의 Complex형성능 (Formation of a complex between furfuryl derivatives and halides)

  • The tendency of forming a charge transfer complex between furfuryl derivatives (2-methyl furan, furfuryl acetate, and Ethyl 2-furoate) and halides(Iodine, Iodine monochloride, and Trichloro bromo methane) was studied by means of ultra violet spectrophotometry. In case of furfuryl acetate the formation of the complex could not be distinctly detected by this method. Iodine and trichloro bromo methane could show a distinct formation of charge transfer complex in the U.V. region, whereas iodine monochloride shows a possibility of forming an addition compound rather than the charge transfer complex itself. The results were discussed in conjunction with the stability of the furfuryl ring.

비이온성 고분자의 Iodine 착물형성에 대한 계면활성제의 영향 (Influence of Surfactant on the Iodine Complex Formation of Some Non-ionic Polymers)

  • 수용성 비이온고분자인 Polyvinylalcohol (PVA), Polyvinylpyrrolidone (PVP), Hydroxypropyl cellusoe (HPC)와 iodine과의 착물 형성에 대한 계면활성제의 영향을 알아보기 위해 Sodiumdodecylsulfate을 포함하는 수용액에서 이들 사이의 반응을 수행하였다. PVP와 HPC에서 tri-iodide band의 적색 이동에 의하여 착물이 만들어졌다는 것을 알게되었고, PVA-iodine 착물에서는 500 nm 부근에서 고유의 특색있는 띠를 나타내었다. SDS 계면활성제의 존재는 PVA-iodine 착물의 파괴를 가져왔고, 고유의 푸른색도 사라지게 만들었다. 그러나 SDS 단량체는 PVP, HPC와 iodine의 착물 형성을 도와주는 경향을 나타내었다. 고분자 용액에서 겔이 만들어지는 것을 방해하는 n-propanol은 고분자-iodine 착물이 형성되는 것을 도와주었다. SDS가 있을 때와 없을 경우의 영향을 알아보기 위해 순수한 HPC와 HPC-iodine 착물을 만들고 이들의 성질을 조사하였다.

인체 간 조직의 Cytochrome P450 3A4의 활성에 대한 Troleandomycin의 작용기전 (Reaction Mechanism of Troleandomycin on the Activity of Human Liver Microsomal Cytochrome P450 3A4)

  • Incubation of aflatoxin $B_1$ $(AFB_1)$ with microsomes isolated from human liver number 110 yielded two metabolite peaks which were aflatoxin $Q_1$ $(AFQ_1)$ and $(AFB_1)$-exo-8, 9-epoxide (exo-epoxide) in high performance liquid chromatography. Production ratio of $AFQ_1$ to exo-epoxide was 2.43$\pm $0.04. Metabolism of $(AFB_1)$ to $(AFQ_1)$ and exo-epoxide was inhibited by troleandomycin in a same degree although troleandomycin was not activated as a mechanism-based inhibitor. The inhibitory effect was dependent upon either the incubation time with $(AFB_1)$ or the preincubation time before the addition of $(AFB_1)$. Incubation of troleandomycin and NADPH by the microsomes resulted in the formation of a cytochrome P 450 (P450)-metabollc intermediate (MI) complex and the level was approximately 80% of total P450 3A4 in the microsomes. This figure was similar to that of the inhibitory effect of troleandomycin on $AFB_1$ metabolism. Glutathione which was reported that it prevented the formation of MI complex in rat liver microsomes did not inhibit the formation of MI complex in human liver microsomes. These results suggested that the inhibitory effect of troleandomycin on $AFB_1$ metabolism is due to the formation of MI complex with P450 3A4. And the reaction mechanism of troleandomycin by human liver microsomes might be dlfferent from that one by rat liver microsomes.

Rifampicin과 Arginine간의 가용성 Complex 형성에 관한 연구 (Soluble Complex Formation of Rifampicin with Arginine)

  • Rifampicin-arginine complex was prepared to increase the solubility and dissolution rate of rifampicin. Solvation method was applied to make the complex and its formation was identified by the solubility method, powder x-ray diffractometry, differential thermal analysis and spectroscopic determination. The complex was formed in the molar ratio of 1 : 1 which was proved by the slope ratio method and continuous variation method. The complex was a non-crystalline form determined by the x-ray powder diffractometric analysis. The solubility of complex in water was significantly higher than that of rifampicin itself. The stability constant and thermodynamical properties of the complex were determined to investigate the solubilization phenomena. The thermodynamic data showed that the complexation between rifampicin and arginine was an exothermic and spontaneous reaction. Simulated absorption studies carried out through the artificial lipid barrier in artificial gastric and intestinal juices. The results showed that the complex had an enhanced absorption rate of rifampicin nearly twice compared with that of rifampicin itself.

Synthesis and Solution Properties of La(III)-N-ethylmorpholine Complex

  • The complex of the composition LaL(N$O_3)_3\;(H_2O)_2$ is prepared by the reaction of La($NO_3)_3{\cdot}6H_2O$ with Nethylmorpholine in aqueous medium. The ligand is involved in the complex as a neutral species where the chelation occurs via the oxygen of the ligand moiety and the nitrate groups as bidentate ligand. The chemical structure of the studied complex is confirmed through IR, XRD, and thermal analysis data. The complexation equilibria of La(III) with N-ethylmopholine is studied in aqueous medium at ionic strength I = 0.1 mol${\cdot}dm^{-3}\;KNO_3$ and at 25, 35 and 45 ${^{\circ}C}$, respectively. The thermodynamic parameters $\Delta$G, $\Delta$H and $\Delta$S values were calculated to prove the association with the complex formation. It is clearly observed that the process is accompanied by absorption of heat, i.e. endothermic process, while the entropy does not change greatly attributed to the release of constant number of water molecules during chelate formation.

Two Types of Complex Predicate Formation:Japanese Passive and Potential Verbs

  • This paper deals with the complex verb formation of passive and potential predicates and syntactic structures projected by these verbs. Though both predicates are formed with the suffix -rare which has been assumed to originate from the same stem, they show significantly different syntactic behaviors. We propose two kinds of concatenation of base verbs and auxiliaries; passive verbs are lexically formed with the most restrictive mode of combination, while potential verbs are formed syntactically via more flexible combinatory operations of function composition. The difference in the mode of complex verb formation has significant consequences for their syntactic structures and semantic interpretations, including different combination with the honorific morphemes and subjectivization of arguments/adjuncts of base verbs. We also consider the case alternation phenomena and their implications for scope construals found in potential sentences, which can be accounted for in a unified manner in terms of the optional application of function composition.
