Effects of Multi-extracts of Mori Folium and of Exercise on Serum Lipid Profiles and Tissue Differentiation in Streptozotocin-induced Diabetic Rats

  • This research was performed to investigate the effects of the supplementation of multi-extracts of mori folium (MF) and of exercise on blood lipid profiles and tissue differentiation in streptozotocin (STZ)-induced diabetic rats. The animal groups consisted of a normal-control group, a STZ-control group, three STZ-induced diabetic groups supplemented ad libitum with various amounts of MF extracts (MF-720, MF-360, and MF-180 groups), and a STZ-induced diabetic group supplemented with MF-360 combined with exercise; eight male Sprague-Dawley rats, 4 weeks old, were assigned to each experimental group and were raised in the laboratory for a 10 week experimental period. The MF supplementation group showed a significant reduction in levels of serum total cholesterol and triglyceride compared to the STZ-control group. HDL-cholesterol levels were significantly increased in the MF supplementation group compared to STZ-control group. The ratio of HDL-cholesterol to total cholesterol was significantly higher in MF supplementation group compared to the STZ-control group. The Atherogenic Index (AI) values in the MF supplementation groups were found to be significantly lower than in the STZ-control group. Serum AST and ALT levels were significantly reduced in the MF-supplementation groups compared to the STZ-control group. Total cholesterol level in the liver tissue was significantly decreased in the MF-360 group and in the MF-360 + exercise group compared to the STZ-control group. In immunohistochemical staining of the pancreatic islets of the MF-supplemented groups, a significantly higher number of insulin-immunoreactive cells were observed compared to the STZ-control group. In the MF supplementation groups, Bowman's capsules were clearly observed as hypertrophy of the glomeruli was not obvious. In the MF supplementation groups, a relative reduction in the hypertrophy of the basement membrane of the glomeruli and a significant reduction in the mesangium were observed compared to the STZ-control group. The results of this study suggest that supplementation of MF has beneficial effect in improving plasma lipid and tissue metabolism in streptozotocin-induced rats.

Acupuncture Effects of Fatty Mice Induced by High Fat Diet (침 자극이 고지방식이로 유발된 비만 백서에 미치는 영향)

  • Objectives The aim of this study was to investigate the effects of acupuncture on weight loss, food intake, lipid metabolism, adipogenesis. Methods Four week-old C57BL/6J mice acclimatized to the laboratory environment for 1 week were allocated into four groups of 6~8 mice each: normal diet group, high fat diet group, high fat diet group and treated with acupuncture at HT7, high fat diet group and treated with acupuncture at ST36 for 90days. High fat diet group was used to control group. Results 1) Body weight, food intake of the ST36, HT7 groups decreased compared with those of control group. ST36 group was more effective with body weight decrease, and HT7 group was more effective with food intake decrease. 2) NEFA, TG, TC, ALT, AST of the ST36, HT7 groups decreased compared with those of control group. ST36 group was more effective. 3) The expression of SREP-1c, SREBP-2 of the ST36 group decreased compared with those of control group. These decreased rates were statistically significant. SREBP-2 of the HT7 group decreased significantly compared to the control group. 4) LXR, FAS, SCD of the ST36 group decreased significantly compared with control group. 5) p-ACC, HMGCR of the ST36 group decreased significantly compared with those of control group. HMGCR of the HT7 group decreased significantly compared with control group. Conclusions These results suggest that ST36, HT7 acupuncture may be used prevent or treat the obesity induced by high fat diet.

The Experimental Study on Anti-bacterial Potency of Coptidis rhizoma extract compare with quantity on Staphylococcus aureus (황련의 증량에 따른 전탕액이 Staphylococcus aureus에 미치는 항균효과에 대한 실험적 연구)

  • Objective This experimental study was performed to investigate the effect of Coptidis rhizoma extract compared with quantity on Staphylococcus aureus that induce keratitis. Methods Minimal inhibitory concentration(MIC) was measured by dropping to $50{\mu}l$ according to density Coptidis rhizoma extract(100%, 10%, 1%, 0.1%) compared with quantity(40g, 80g, 160g). Anti-bacterial potency was measured by the size of inhibition zone with change of volume. Results 1. MIC on Staphylococcus aureus in Coptidis rhizoma extract was showed anti-bacterial potency compared with quantity and density in 100% and 10% of all samples(40g, 80g, 160g). 2. MIC on Staphylococcus aureus in Coptidis rhizoma extract(40g, 80g, 160g) was showed anti-bacterial potency compared with quantity all samples($20{\mu}l,\;30{\mu}l,\;40{\mu}l,\;50{\mu}l$) in 100% density. 3. MIC on Staphylococcus aureus in Coptidis rhizoma extract(40g, 80g, 160g) was showed anti-bacterial potency compared with quantity all samples ($20{\mu}l,\;30{\mu}l,\;40{\mu}l,\;50{\mu}l$) in 100% density. Anti-bacterial potency of 80g Coptidis rhizoma extract decreased compared with 40g. Anti-bacterial potency of 160g Coptidis rhizoma extract decreased compared with 40g in $20{\mu}l,\;30{\mu}l$. Conclusions Coptidis rhizoma extract was showed anti-bacterial potency compare with quantity and density. In herbal drug, antibacterial potency compare with quantity and density must be studied.

Effects of Silk Fibroin in Oxdative Stress and Membrane Fluidity in the Liver of SD Rats (Rat 간장의 산화적 스트레스 및 세포막 유동성에 미치는 실크 피브로인의 영향)

  • This study was designed to investigate the effects of silk fibroin powder (Mw 500) on oxidative stress and membrane fluidity in liver membranes of rats. Sprague-Dawley (SD) male rats (160$\pm$10g) were fed basic diet (control group), and experimental diets (SEP-2.5 and SFP-5.0 groups) added 2.5 and 5.0 g/kg BW/day for 6 weeks. Cholesterol levels resulted in a significant decrease (12.1% and 9.0%, respectively) in the liver mitochondria and microsomes of SEP-5.0 group compared with control group. Membrane fluidity as significantly increased (16.1% and 16.5%, 5.8% and 17.4%) in the liver mitochondria and microsomes were significantly inhibited (16.1% and 18.3%, 8.1% and 15.1%, respectively) at the SFP-2.5 and SEP-5.0 groups compared with control group. Induced oxygen radicals (BOR) in liver mitochondria and microsomes were significantly inhibited (16.1% and 18.3%, 8.1% and 15.1%, respectively) at the SFP-2.5 and SEP-5.0 groups compared with control group. Induced oxygen radicals (IOR) in liver microsomes were significantly inhibited (17.0% and 26.6%, respectively) at the SFP-2.5 and SFP-5.0 groups compared with control group, but IOR in liver mitochondria was significantly inhibited about 12.3% at the SWP-400 group only compared with control group. Lipid peroxide (LPO) levels were significantly decreased (8.3% and 18.0%, 13.4% and 18.4%, respectively) in the liver mitochondria and microsomes of SFP-2.5 and SFP-5.0 groups compared with control group. Oxidized protein (OP) levels were dose-dependently decreased (5.4% and 11.6%, 19.0% and 24.4%, respaectively) in the iver mitochondria and microsomes of SFP-2.5 and SFP-5.0 groups compared with control group. These results suggest that administration of SFP may play an effective role in attenuating an oxidative stress and increasing a membrane fluidity in liver membranes.

Studies on Self-Perceived Health and Appearance, Health-Related Lifestyles and Dietary Behaviors of Korean College Students Attending Web Class

  • The purpose of this study was to investigate self-perception and practices of health and appearance, health-related lifestyles and dietary behaviors of college students(137 male, 115 female) attending web class via the Internet. This cross-sectional survey was conducted by a self-administered questionnaire and data was analyzed by SPSS program. The average height, weight and BMI of the male and female college students were 174.4cm, 67.8kg, 22.3kg/㎡ and 162.3cm, 52.1kg, 19.8kg/㎡, respectively. Male collage students perceived their health statuses significantly better compared to female students. As for self-evaluation of anemia, female college students perceived significantly more anemia symptoms compared to male college students. Female college students perceived their body image to be obese compared to male college students. Underweight college students perceived significantly less healthy statuses, more anemia symptoms, and worse hair conditions compared to other college students. The rate of male students who exercise was significantly higher compared to that of female students. Male students reported significantly more smoking and drinking of alcohol compared to female students. The rate of female college students skipping meals was significantly higher than that of male college students. Frequency of eating out in female college students was significantly higher compared to that in male students. These results may provide some basic information in developing a nutrition education program for Korean college students using Internet.

The Effect of Bee Venom on COX-2, P38, ERK and JNK in RAW 264.7 Cells (봉약침액(蜂藥鍼液_이 RAW 264.7 세포의 COX-2, P38, ERK 및 JNK에 미치는 영향(影響))

  • The purpose of this study was to investigate the effect of Bee Venom on the lipopolysaccharide(LPS), sodium nitroprusside(SNP), hydrogen peroxide($H_2O_2$)-induced expressions of cyclooxygenase-2(COX-2), p38, jun N-terminal Kinase(JNK) and extra-signal response kinase(ERK) in RAW 264.7 cells, a murine macrophage cell line. Method : The expressions of COX-2, p38, JNK and ERK were determined by western blotting with corresponding antibodies. Results : 1. The 0.5, 1 and $5\;{\mu}g/ml$ of bee venom inhibited significantly LPS and SNP-induced expression of COX-2 compared with control, respectively. The 0.5, 1 and $5\;{\mu}g/ml$ of bee venom inhibited insignificantly $H_2O_2$-induced expression of COX-2 compared with control, respectively. 2. The 0.5, 1 and $5\;{\mu}g/ml$ of bee venom inhibited significantly LPS, SNP and $H_2O_2$-induced expression of p38 compared with control, respectively. 3. The 1 and $5\;{\mu}g/ml$ of bee venom inhibited significantly SNP-induced expression of JNK compared with control, respectively. All of bee venom inhibited insignificantly LPS and $H_2O_2$-induced expression of JNK compared with control, respectively. 4. The $5\;{\mu}g/ml$ of bee venom inhibited significantly SNP-induced expression of ERK, the $0.5\;{\mu}g/ml$ of bee venom increased significantly $H_2O_2$-induced expression of ERK compared with control. The 0.5, 1 and $5\;{\mu}g/ml$ of bee venom inhibited insignificantly LPS-induced expression of ERK compared with control, respectively.

The Role of Ascorbic Acid on the Redox Status and the Concentration of Malondialdehyde in Streptozotocin-Induced Diabetic Rats

  • We investigated the role of ascorbic acid on the redox status in streptozotocin-induced diabetic rats. In the plasma of diabetic rats, the ratio of reduced/total ascorbic acid was significantly decreased as compared with normal control. Ascorbic acid supplementation increased the reduced and total ascorbic acid contents as compared with diabetic control. In the rutintreatment group, reduced and total contents of ascorbic acid were significantly decreased, however, the ratio of reduced/total contents of ascorbic acid had no difference as compared with diabetic rats. In the insulin-treatment group, this ratio is not significantly different as compared with diabetic control. However, in the insulin plus ascorbic acid treatment group, reduced form and the ratio of reduced/total ascorbic acid were significantly increased as compared with diabetic control. In addition, we measured the contents of malondialdehyde (MDA) in the plasma of diabetic rats. The contents of MDA was increased as compared with normal control, however, in insulin-treatment group, the contents of MDA was decreased as compared with diabetic rats. Ascorbic acid had no effects on the increases of MDA in diabetic rats. In conclusion, plasma ascorbic acid level and its reduced/total ratio reflects the status of the oxidative stress in the diabetic rats. Supplement of ascorbic acid did not correct the ratio of the reduced/total ascorbic acid. However, supplement of insulin and ascorbic acid corrected the ratio of reduced/total ascorbic acid.

The effects of acupuncture on NADPH-diaphorase and nNOS in the brain stem and cerebellum of SHR (침자극이 흰쥐 뇌줄기 및 소뇌의 NADPH-diaphorase와 nNOS 신경세포에 미치는 영향)

  • Objective : This study was to investigate the effect of acupuncture on NADPH-diaphorase and nNOS in the brain stem and cerebellum of spontaneously hypertensive rats. Methods : The experimental groups were divided into four groups : Normal, Choksamni(ST36), Kokchi(LI11), arbitrary group. Thereafter we evaluated changes in NADPH-diaphorase positive neurons histochemically and changes in nNOS neurons immunohistochemically. Results : 1. The optical densities of NADPH-diaphorase positive neurons of all the Choksamni & Kokchi groups were significantly different in SuG, DLPAG, IP, Pr, Gi areas of brain stem and cerebellum as compared to normal & arbitrary groups. In PPTg only Choksamni group was significantly different as compared to normal and arbitrary groups. 2. The optical densities of nNOS-positive neurons of Choksamni & Kokchi groups were significantly different in SuG, DLPAG areas of brain stem as compared to normal group. In IP, Pr only Kokchi group was significantly different as compared to normal group. The optical densities of nNOS-positive neurons of Choksamni & Kokchi groups were significantly different in SuG, DLPAG, PPTg, Pr, Gi areas of brain stem as compared to arbitrary group. In IP, Pr only Kokchi group was significantly different as compared to arbitrary group. 3. The optical densities of nNOS-positive neurons of all the Choksamni & Kokchi groups were not significantly different in cerebellum as compared to normal & arbitrary groups. Conclusions : We found out that acupuncture have effects on NADPH-diaphorase and nNOS in the brain stem and cerebellum of spontaneously hypertensive rats.

Comparison research of HRV between Postpartum Women and Normal Women (산후 여성의 심박변이도 특성 연구)

  • Objective : This research was designed to study the characteristics of heart rate variability in postpartum women compared with normal women. Method : HRV data of postpartum women were gathered from 255 women who was hospitalized for oriental postpartum treatments(Age : 25-35). HRV data of comparison group were gathered from 327 women who visited hospital to check up their health(Age : 25-35). The SPSS 12.0 for windows was used to analyze the date and the independent samples t-test was used to verify the result. Result : 1. Mean-RR and SDNN of postpartum women group significantly decreased compared with that of normal women group. But, Heart Rate of postpartum women group significantly increased compared with that of normal women group. 2. HRV-Index, RMSSD and SDSD of postpartum women group significantly decreased compared with that of normal women group. pNN50 of postpartum women group significantly increased compared with that of normal women group. 3. Ln(TP), Ln(VLF), Ln(LF) and Ln(HF) of postpartum women group significantly increased compared with that of normal women group. 4. There were no significant differences in Normal LF, Normal HF and LF/DF Ratio between postpartum women group and normal women group. Conclusion : The result suggest that the function of heart of postpartum women group significantly decreased compared with that of normal women group. Futhermore although the ANS maintained the balance in the range of normality, the sympathetic nervous system frequently revitalized which caused increasing the heart of pulsation.

Effects of Silkworm(Bombyx mori L.) Power on Oxidative Stress and Membrane Fluidity in Brain of SD Rats (뇌조직의 산화적 스트레스 및 세포막 유동성에 미치는 누에분말의 영향)

  • This study was designed to investigate the effects of silkworm(Bombyx moril L.) powder on oxidative stress and membrane fluidity in brain membranes of rats. Sprague-Dawley(SD) male rats(160$\pm$10 g) were fed basic diet(control group), and experimental diets(SWP-200 and SWP-400 groups) added 200 and 400mg/kg BW/day for 6 weeks. There were no significant differences in cholesterol levels of brain memberanes by administration of silkworm powder (SWP). Membrane fluidities were significantly increased(21.5% and 30.8%, respectively) in brain mitochondria of SWP-200 and SWP-400 groups compared with control group, but significant difference between brain microsomes could not obtained. Basal oxygen radicals (BORs) in brain mitochondria and mircrosomes were significantly inhibited(8.5% and 16.5%, 16.8%and 24.8%, respectively) by SWP-200 and SEP-400 groups compared with control group. Induced oxygen radicals(IORs) in brain mitochondria were significantly inhibited(16.6% and 21.4%, respectively)by sWP-200 and SWP-400 groups compared with control group, but IOR in brain microsome were significantly inhibited about 16.0% by SWP-400 groups only compared with control group. Lipid peroxide(LPO) levels were significantly decreaed(14.8%and 22.4%, respectively) in brain mitochondria of SWP-200 and SWP-400 groups compared with control group, but LPO level was significantly decreased about 16.0% in brain microsome of SWP-400 group only. Oxidized protein(OP) levels were remarkably decreased(about 14.8% and 16.5%, respectively) in brain mitochondria of SWP-200 and SWP-400 groups, but OP level was significantly decreased about 13.0% in brain microsome of SWP-400 group only compared with control group, Theses results suggest that administration of in brain microsome of SWP-400 group only compared with control group. These results suggest that administration of SWP may play effective role in attenuating an oxidative stress and increasing a membrane fluidity in brain membranes.
