Food component characteristics and antioxidant activities of commercial shiitake mushrooms (Lentinula edodes) (시판 표고의 식품성분 특성 및 항산화 활성)

  • This study analyzed food component characteristics and antioxidant activities in five kinds of domestic commercial shiitake mushrooms (Lentinula edodes). Moisture content (79.17-82.90 g/100 g) showed little difference, but there was a significant difference in crude protein content. Total mineral content showed a significant difference, and potassium (K) content was the highest. Total and essential amino acid contents were higher in mushroom A than in the other samples. Nonetheless, the ratios of essential amino acids, aspartic acid, and glutamic acid to total amino acids were similar in all samples. 𝛽-Glucan content was in the range of 10.28-5.68 g/100 g, exhibiting a significant difference. The color intensity and overall texture of the mushrooms showed no remarkable difference. Total phenol and flavonoid contents were in the range of 19.92-30.77 mg/100 g and 6.95-10.39 mg/100 g, respectively; the ratio of flavonoids to total phenols ranged from 31.53% to 38.21%. DPPH and ABTS radical scavenging activities were 74.92-79.04% and 80.47-84.97%, respectively, and showed little difference. However, the reducing power varied between 195.23 𝜇M and 317.85 𝜇M, displaying a significant difference in all samples. Therefore, this study observed similar trends in the food component characteristics and antioxidant activities of domestic commercial shiitake mushrooms.

Application of Master Packaging System to Fresh Shiitake Mushroom Supply Chain on Semi-commercial Scale (생표고버섯에 대한 마스터 포장 시스템의 현장 적용)

  • Master packaging system is a technology combining primary and secondary packaging to preserve the fresh produce in the supply chain. Master packaging system with tailor-designed gas transfer and $CO_2$ absorber of $Ca(OH)_2$ was applied to fresh shiitake mushroom in its supply from farm to retail store. The temperature, humidity and package atmosphere were monitored through the distribution and/or storage until the packages were opened to measure the mushroom quality. Conventional perforated individual packages without secondary master pack were prepared and travelled the same path for comparison purpose. While high temperature history was observed in some initial period of actual practice of the mushroom transportation and storage unexpectedly, the package atmosphere around the produce in the master packaging system was maintained at modified atmosphere consisting of $O_2$ concentration of 0.4 to 4.2% and $CO_2$ concentration of 0.7 to 1.7%, which is known to be beneficial for the mushroom preservation. While curing the mushrooms with precooled drying was effective for quality preservation, positive effect of master packaging system could be apparent for the uncured mushroom. Harmonized combination of curing treatment, master packaging system and temperature management was suggested for the best quality preservation of the fresh shiitake mushroom.

Damage and biological control of dark winged fungus gnats, Lycoriella ingenua (Diptera: Sciaridae) in a shiitake cultivation (표고 톱밥재배에서 검정날개버섯파리 Lycoriella ingenua (Diptera: Sciaridae)의 피해와 생물적 방제)

  • Monitorings and management experiment of adult Lycoriella ingenua (Diptera: Sciaridae) in artificial sawdust grown shiitake mushroom cultivation were carried out by using yellow sticky traps and Hypoaspis aculeifer (Acari: Laelapidae) at Hwaseong, Buyeo and Cheongwon in 2012. The larvae of L. ingenua reduced commercial value of the mushroom by direct eating or retarding the growth of mushroom by spongifying the sawdust medium. The number of L. ingenua caught by traps showed the highest peak in late september resulting in 127.5~1,025.7, 87.4~743.6 and 133.7~650.4 individuals per trap in Hwaseong, Buyeo and Cheongwon, respectively. Damage rate of fruiting bodies in shiitake mushroom by L. ingenua were 7.7~30.3% in Hwaseong, 6.7~25.3% in Buyeo and 5.3~26.0% in Cheongwon and showed the highest peak in late september. L. ingenua were effectively controlled when 30.3 individuals of H. aculeifer per $m^2$ inoculated for three time with 7 to 14 days of interval. In Hwaseong, Buyeo and Cheongwon, density of L. ingenua which were caught on the yellow sticky traps were 168.2, 126.1, 132.5 individuals, respectively. And, damage rate of fruiting bodies by L. ingenua were 5.9%, 5.5% and 5.4% respectively. Both of the L. ingenua density and damage rate of mushroom reduced more than 60% in all experimental site in contrast to the control treatment.

Occurrence of dsRNA Mycovirus (LeV-FMRI0339) in the Edible Mushroom Lentinula edodes and Meiotic Stability of LeV-FMRI0339 among Monokaryotic Progeny

  • dsRNA was found in malformed cultures of Lentinula edodes strain FMRI0339, one of the three most popular sawdust cultivated commercial strains of shiitake, and was also found in healthy-looking fruiting bodies and actively growing mycelia. Cloning of the partial genome of the dsRNA revealed the presence of the RdRp sequence of a novel L. edodes mycovirus (LeV), and sequence comparison of the cloned amplicon showed identical sequences sequence to known RNA-dependent RNA polymerase genes of LeV found in strain HKA. The meiotic stability of dsRNA was examined by measuring the ratio of the presence of dsRNA among sexual monokaryotic progeny. More than 40% of the monokaryotic progeny still contained the dsRNA, indicating the persistence of dsRNA during sexual reproduction. Comparing the mycelia growth of monokaryotic progeny suggested that there appeared to be a tendency toward a lower frequency of virus incidence in actively growing progeny.

Physico-chemical, Nutritional, and Enzymatic Characteristics of Shiitake Spent Mushroom Substrate (SMS) (표고버섯 수확 후 배지의 이화학적, 영양적, 효소적 특성)

  • In Korea, edible mushrooms are produced largely on commercial artificial media, so the annual production of spent mushroom substrate (SMS), as a by-product of the mushroom industry, is estimated at over 200 million tons. This SMS is assumed to contain abundant fungal mycelia and pre-fruiting bodies, as well as various nutritive and bioactive compounds that are presently discarded. This study examined the physico-chemical, nutritional, and enzymatic characteristics of uninoculated sterilized medium (USM) and SMS of shiitake mushrooms with the aim of developing a high-value added product from SMS. The contents of crude protein, crude lipid, and ash were higher after the third SMS harvest ($SMS-A-3^{rd}$) than in USM or $SMS-A-1^{st}$. The contents of Ca, Mg, and P in $SMS-A-3^{rd}$ were 2.95, 2.35, and 2.1-fold higher compared than in USM. No As or Cd was detected in USM or SMS. The pH, Brix, and acidity were 4.6, 20.0, and 1.4, respectively in $SMS-A-3^{rd}$, but 5.6, 6.0, and 0.0, respectively, in USM. These results suggest a highly active production of soluble components and organic acids in $SMS-A-3^{rd}$. The distinct color differences noted for USM, $SMS-A-1^{st}$, and $SMS-A-3^{rd}$ could be used as a mycelial growth indicator. Enzyme activity assays using the APIZYM system showed that SMS is a potent source of hydrolysis-related enzymes, especially esterase (C4) and ${\beta}$-glucuronidase. Our results suggested that the SMS of shiitake has a high potential for use in environmental, agricultural, and stock-breeding industries, for example, as active ingredients for sewage treatment, waste-polymer degradation, and feed additives.

Study of Viral Effects of the Mycovirus (LeV) and Virus-Free Commercial Line in the Edible Mushroom Lentinula edodes

This study attempted to cure the edible mushroom L. edodes strain FMRI0339 of the L. edodes mycovirus (LeV) in order to obtain an isogenic virus-free fungal strain as well as a virus-infected strain for comparison. Mycelial fragmentation, followed by being spread on a plate with serial dilutions resulted in a virus-free colony. Viral absence was confirmed with gel electrophoresis after dsRNA-specific virus purification, Northern blot analysis, and PCR using reverse transcriptase (RT-PCR). Once cured, all of fungal cultures remained virus-free over the next two years. Interestingly, the viral titer of LeV varied depending on the culture condition. The titer from the plate culture showed at least a 20-fold higher concentration than that grown in the liquid culture. However, the reduced virus titer in the liquid culture was recovered by transferring the mycelia to a plate containing the same medium. In addition, oxygen-depleted culture conditions resulted in a significant decrease of viral concentration, but not to the extent seen in the submerged liquid culture. Although no $discernable phenotypic changes in colony morphology were observed, virus-cured strains showed significantly higher growth rates and mycelial mass than virus-infected strains. We were also explored effects of LeV on fruiting body formation and mushroom yield. The fruiting body formation yield of virus-free L. edodes was larger than virus-infected L. edodes. These results indicate that LeV infection has a deleterious effect on mycelial growth and fruiting body formation. In addition, we have been investigated host-parasite interaction between L. edodes and its mycovirus interaction to study viral mechanism by establishment of proteomics.

Comparison of optimal culture and growth characteristics of the Lentinula edodes fruit body 'Sanjo701ho', 'Nongjingo' (표고 '산조701호', '농진고'의 최적 배양 및 자실체 특성 비교)

  • This study investigated the characteristics of different cultivars of Lentinula edodes in sawdust culture at different cultivation days. Between cultivation days 30-120, the color of 'Sanjo701ho' displayed a reduction in L value (brightness) from $83.8{\pm}2.5$ to $45.7{\pm}2.3$, values a and b increased, but hardness decreased from $9.4{\pm}0.9g/mm$ to $2.6{\pm}0.2g/mm$. Between cultivation days 30-120, 'Nongjingo' displayed a reduction in L value from $86.2{\pm}2.1$ to $53.4{\pm}1.3$. Values a and b increased with longer cultivation; however, hardness decreased from $4.8{\pm}0.7g/mm$ on day 30 to $3.8{\pm}1.0g/mm$ on day 120. 'Sanjo701ho' was first harvested at 46 days after a 30-, 89 days after a 60-, 8 days after a 90- (the shortest), and 9 days after a 120-day cultivation. The average fruit body weight was the highest on day 90 of cultivation, at 48.3 g, when the diameter and thickness of the mushroom cap also appeared highest. However, the colorimetric results showed that fruiting bodies produced in the culture medium for 120 cultivation days showed the most excellent commercial properties. 'Nongjingo' was first harvested at 22 days after a 30-, 18 days after a 60-, 8 days after a 90- (the shortest), and 9 days after a 120-day cultivation. Therefore, this study determined that a stable quantity of mushrooms with high commerciality can be produced with 120 cultivation days, considering the shiitake culture and the characteristics of the fruit body.
