A promising approach to the development of a new single-step vaccine, which would eliminate the requirement for multiple injections, involves the encapsulation of antigens into microspheres. Biodegradable poly(lactide-co-glycolide) (PLGA) microspheres gave us a bright insight for controling antigen release in a pulsatile fashion, thereby mimicking two or tree boosting injections. However, in spite of the above merits, the level of immunization induced by a single-shot vaccination is often lower tan two doses of alum-adsorbed antigen. Therefore, optima modification of the microsphere is essential for the development of single-step vaccines. In the review, we discuss the stability of antigen in microsphere, safety and non-toxic in human and encapsulation technology. Also, we attempted to outline relevant physicochemical properties on the immunogenicity of microsphere vaccine and attainment of pulsatile release pater by combination of different microsphere, as well as to analyze immunological data associated with antigen delivery by microsphere. Although a lot of variables are related to the optimized microsphere formulation, we could conclude that judicious choice of proper polymer type, adjustment of particles size, and appropriate immunization protocol along with a suitable adjuvant might be a crucial factor for the generation of long-lasting immune response from a single-step vaccine formulation employing PLGA microsphere.
Multi-drug resistance (MDR) has been known as a major hurdle in cancer chemotherapy. One of the most clinically significant causes of MDR was the efflux of anticancer agents mediated by p-glycoprotein (p-gp) over-expressed in MDR cancer cells. To overcome MDR, there have been several strategies such as co-administration with p-gp inhibitors and encapsulation of anticancer drugs into drug delivery systems. In the present study, curcumin was evaluated for its potential as p-gp inhibitor and MDR reversal activity when combined with paclitaxel incorporated into lipid nanoparticles (PTX/LN). Western blot assay showed curcumin did not modulate the level of p-gp expression in MCF-7/ADR which is a MDR variant of human breast cancer cell line, MCF-7, and over-expresses p-gp. However, curcumin inhibited p-gp-mediated efflux of calcein in a dose-dependent manner even though it showed lower activity compared to verapamil, a well-known p-gp inhibitor. Incorporation of paclitaxel into lipid nanoparticles partially recovered the anticancer activity of paclitaxel in MCF-7/ADR. The combined use of curcumin and PTX/LN exhibited further full reversal of MDR, suggesting susceptibility of PTX/LN to the efflux system. In conclusion, combined approach of using p-gp inhibitors and incorporation of the anticancer agents into nano-delivery systems would be an efficient strategy to overcome MDR.
Aspirin or acetylsalicylic acid, a member of the salicylate family, is frequently used as an analgesic, antipyretic, anti-inflammatory and antiplatelet drug. Because aspirin is chemically unstable in water and heat for tablet formulation, additives including lubricants are used in preparing aspirin tablets, using a dry-granulation process. Aspirin tablets are produced by a number of manufacturers which usually use their own unique combination of additives during the manufacturing process. In this study, we employed an NMR based metabolomics technique to identify the manufacturers of various aspirin tablets. Aspirin tablets from six different companies were analyzed by 1H 400 MHz NMR. The acquired data was then integrated and processed by principal component analysis (PCA). Based on the NMR data, we were able to identify peaks corresponding to acetylsalicylic acid in all of the six samples, whereas different NMR patterns were found in the aromatic and aliphatic regions depending on the unique additive used. These observations led to the conclusion that the differences in the NMR patterns among the different aspirin tablets were due to the presence of additives.
Recently, model-based iterative reconstruction (MBIR) has played an important role in transmission tomography by significantly improving the quality of reconstructed images for low-dose scans. MBIR is based on the penalized-likelihood (PL) approach, where the penalty term (also known as the regularizer) stabilizes the unstable likelihood term, thereby suppressing the noise. In this work we further improve MBIR by using a more expressive regularizer which can restore the underlying image more accurately. Here we used a spline regularizer derived from a linear combination of the two-dimensional splines with first- and second-order spatial derivatives and applied it to a non-quadratic convex penalty function. To derive a PL algorithm with the spline regularizer, we used a separable paraboloidal surrogates algorithm for convex optimization. The experimental results demonstrate that our regularization method improves reconstruction accuracy in terms of both regional percentage error and contrast recovery coefficient by restoring smooth edges as well as sharp edges more accurately.
In order to approach information system through smart device and pc, user has to authenticate him or herself via user authentication. At that time when user tries reaching the system, well-used user authentication technologies are ID/PW base, OTP, certificate, security card, fingerprint, etc. The ID/PWbased method is familiar to users, however, it is vulnerable to brute force cracking, keylogging, dictionary attack. so as to protect these attacks, user has to change the passwords periodically as per password combination instructions. In this paper, we designed and implemented a user authentication system using smartphone's USIM without using password while enhancing security than existing ID / PW based authentication technology.
Controlling the residual glucose concentration is important for improving productivity in $\text\tiny{L}$-threonine fermentation. In this study, we developed a procedure to automatically control the feeding quantity of glucose solution as a function of ammonia-water consumption rate. The feeding ratio ($R_{C/N}$) of glucose and ammonia water was predetermined via a stoichiometric approach, on the basis of glucose-ammonia water consumption rates. In a 5-L fermenter, 102 g/l $\text\tiny{L}$-threonine was obtained using our glucose-ammonia water combined feeding strategy, which was then successfully applied in a 500-L fermenter (89 g/l). Therefore, we conclude that an automatic combination feeding strategy is suitable for improving $\text\tiny{L}$-threonine production.
Daily monitoring of the heart rate can facilitate detection of heart-related diseases in their early stages. Therefore, providing an easy-to-use and noninvasive heart rate monitoring system has been a very popular research topic in the field of healthcare. One of good candidate methods is to use commonly available cameras and extract information that can help to estimate heart rate from a human face. Generally, such information can be retrieved using two different approaches: photoplethysmography (PPG) and ballistocardiography (BCG). PPG exploits slight color changes caused by blood volume variations during heartbeats; thus, it tends to be vulnerable to unstable lighting conditions. BCG exploits subtle head motions caused by pumped blood travelling through the carotid artery during heartbeats; thus, it is vulnerable to the voluntary head movements that are not related to heartbeats. Nevertheless, most related works use either to estimate the heart rate. In this paper, we propose to combine two approaches to be robust to challenging conditions. Specifically, we explore possible ways to combine raw signals obtained from two approaches and verify that the proposed combination shows better accuracies under challenging conditions, such as voluntary head movements and ambient lighting changes.
Simultaneous precision positioning and vibration suppression of a reciprocating flexible manipulator is investigated in this paper. The flexible manipulator is driven by a multifunctional active strut with fuzzy logic controllers. The multifunctional active strut is a combination of a motor assembly and a piezoelectric stack actuator to simultaneously provide precision positioning and wide frequency bandwidth vibration suppression capabilities. First, the multifunctional active strut and the flexible manipulator are introduced, and their dynamic models are derived. A control strategy is then proposed, which includes a position controller and a vibration controller to achieve simultaneous precision positioning and vibration suppression of the flexible manipulator. Next, fuzzy logic control approach is presented to design a fuzzy logic position controller and a fuzzy logic vibration controller. Finally, experiments are conducted for the fuzzy logic controllers and the experimental results are compared with those from a PID control scheme consisting of a PID position controller and a PID vibration control. The comparison indicates that the fuzzy logic controller can easily handle the non-linearity in the strut and provide higher position accuracy and better vibration reduction with less control power consumption.
Optimization of the concrete mixture design is a process of search for a mixture for which the sum of the cost of the ingredients is the lowest, yet satisfying the required performance of concrete. In this study, a statistical model was carried out to model a cost effective optimal mix proportioning of high strength self-compacting concrete (HSSCC) using the Response Surface Methodology (RSM). The effect of five key mixture parameters such as water-binder ratio, cement content, fine aggregate percentage, fly ash content and superplasticizer content on the properties and performance of HSSCC like compressive strength, passing ability, segregation resistance and manufacturing cost were investigated. To demonstrate the responses of model in quadratic manner Central Composite Design (CCD) was chosen. The statistical model showed the adjusted correlation coefficient R2adj values were 92.55%, 93.49%, 92.33%, and 100% for each performance which establish the adequacy of the model. The optimum combination was determined to be $439.4kg/m^3$ cement content, 35.5% W/B ratio, 50.0% fine aggregate, $49.85kg/m^3$ fly ash, and $7.76kg/m^3$ superplasticizer within the interest region using desirability function. Finally, it is concluded that multiobjective optimization method based on desirability function of the proposed response model offers an efficient approach regarding the HSSCC mixture optimization.
The effect of earthquake induced torsion, due to mass eccentricities, is investigated with the objective of providing practical design guidelines for minimizing the torsional response of building structures. Current code provisions recommend performing three dimensional static or dynamic analyses, which involve shifting the centers of the floor masses from their nominal positions to what is called an accidental eccentricity. This procedure however may significantly increase the design cost of multistory buildings, due to the numerous possible spatial combinations of mass eccentricities and it is doubtful whether such a cost would be justifiable. This paper addresses this issue on a theoretical basis and investigates the torsional response of asymmetric multistory buildings in relation to their behavior when all floor masses lie on the same vertical line. This approach provides an insight on the overall seismic response of buildings and reveals how the torsional response of a structure is influenced by an arbitrary spatial combination of mass eccentricities. It also provides practical guidelines of how a structural configuration may be designed to sustain minor torsion, which is the main objective of any practicing engineer. A parametric study is presented on 9-story common building types having a mixed-type lateral load resisting system (frames, walls, coupled wall bents) and representative heightwise variations of accidental eccentricities.
