Characterization of Sporulation-Specific Glucoamylase of Saccharomyces diastaticus (Saccharomyces diastaticus의 포자형성 특이 글루코아밀라제의 특성)

  • The yeast strains of Saccharomyces diastaticus produce one of three isozymes of an extracellular glucoamylase I, II or III, a type of exo-enzyme which can hydrolyse starch to generate glucose molecules from non-reducing ends. These enzymes are encoded by the STA1, STA2 and STA3 genes. Another gene, sporulation-specific glucoamylase (SGA), also exists in the genus Saccharomyces which is very homologous to the STA genes. The SGA has been known to be produced in the cytosol during sporulation. However, we hypothesized that the SGA is capable of being secreted to the extracellular region because of about 20 hydrophobic amino acid residues at the N-terminus which can function as a signal peptide. We expressed the cloned SGA gene in S. diastaticus YIY345. In order to compare the biochemical properties of the extracellular glucoamylase and the SGA, the SGA was purified from the culture supernatant through ammonium sulfate precipitation, DEAE-Sephadex A-50, CM-Sephadex C-50 and Sephadex G-200 chromatography. The molecular weight of the intact SGA was estimated to be about 130 kDa by gel filtration chromatography with high performance liquid chromatography (HPLC) column. Sodium dedecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) analysis showed it was composed of two heterogeneous subunits, 63 kDa and 68 kDa. The deglycosylation of the SGA generated a new 59 kDa band on the SDS-PAGE analysis, indicating that two subunits are glycosylated but the extent of glycosylation is different between them. The optimum pH and temperature of the SGA were 5.5 and $45^{\circ}C$, respectively, whereas those for the extracellular glucoamylase were 5.0 and $50^{\circ}C$. The SGA were more sensitive to heat and SDS than the extracellular glucoamylase.

Ecological Health Assessments on Stream Order in Southern Han River Watershed and Physical Habitat Assessments (남한강 수계에서 하천차수에 따른 생태건강도 평가 및 지점별 물리적 서식지 평가)

  • The ecological health, based on the Index of Biological Integrity (IBI) and Qualitative Habitat Evaluation Index (QHEI) was evaluated in 10 stream sites of Southern Han River. Eleven parameters of 12 parameters (Karr 1981) were modified for the application of regional Korean circumstance. The ecological health, based on IBI grade, was in "good condition" and the IBI score ranged from 33 to 47. Nine parameters of the original 12-parameter metrics in QHEI model (Plafkin et al. 1989) were applied in the habitat assessment. The mean QHEI model values were judged as "partially supporting" and ranged from 75 (non-supporting) to 109 (supporting). Comparative analyses revealed that values of IBI and QHEI models were greater in Gj stream than Ig- and Dn streams. The analysis of fish compositions showed that the proportions of insectivore, omnivore, and carnivore were 61.9%, 19%, and 9.5%, respectively. According to tolerance guild analysis, sensitive species and tolerant species were 76.1% and 4.7%, respectively, indicating a healthy trophic state in terms of food chain. The analysis by habitat guild type indicated that riffle benthic species dominated (57.1%) when compared to water column species (28.5%). The introduced species and individuals with diseases or external abnormality were not observed. Overall, the model values of IBI and QHEI suggested that the ecological health was maintained well in this upstream region.

Spatial and Temporal Distribution Characteristics of Zooplankton Appeared on Early Construction of Pumped Storage Power Plant Dam (양수발전댐의 건설 초기에 발생한 동물플랑크톤군집의 시.공간적인 분포특성)

  • Spatial and temporal distributions of zooplankton were measured in an oligotrophic pumped storage-type hydroelectric reservoir which was composed of two reservoirs exchanging water daily, with water going up at night and going down during the day. Repetitive diel disturbance of the water column can be a unique feature of this reservoir system. Chl-${\alpha}$ concentration was highest in the early winter season. Phytoplankton density was lower in summer monsoon due to high flushing rate on rainy days. The zooplankton density was higher in the smaller upper reservoir possibly due to lower fish density in the upper reservoir. In the seasonal variation a time gap was observed between the phytoplankton bloom and the zooplankton bloom (particularly a rotifer, Keratella cochlearis). It is likely to that Keratella production is partially supported by heterotrophic food sources than phytoplankton. The dominance of a mixotrophic dinoflagellate (Peridinium bipes f. ocultatum) might have complicated the trophic relationship between phytoplankton and zooplankton. Our results provide some ecological information of zooplankton community in a highly disturbed alpine reservoir ecosystem relying on mostly allochthonous organic matter.

The Analysis of Economic Contribution of Character Industry in China (산업연관분석에 의한 중국 캐릭터 산업의 경제적 효과 분석)

  • Due to the lack of national consensus on the importance and value of the character industry and the lack of recognition of value as a national strategic industry, the development of the character industry is experiencing great difficulties. The purpose of this study is to analyze the economic effects of character industry in China to help establish policies and strategies for the character industry in the future. To this end, this study utilized the China 2017 Industrial Association Table. The analysis results are as follows. China's character industry has a lower production inducement effect than other industries with a column total of 3.45514, and a row total of 1.30015. This shows that China's character industry is still being produced by small and medium-sized companies with a low equity ratio. Second, in the character industry, the index of the sensitivity of dispersion representing the forward linkage effect is 0.01426 and the impact factor is 0.03790, which are all less than 1. Therefore, it can be said to be the final demand manufacturing type.Third, in China character industry's income induction is 0.47690 and the production tax induction effect is -0.04912. It can be seen that the character industry has less income induction and tax burden generated every time the final demand increases by one unit in the entire industry than in other industries.Despite the quantitative growth of the character industry in China, the impact on other industries is low and it is not playing a role as an income-generating industry. Structural improvement is needed for the qualitative development of China's character industry.

A Study on the Anti-Pollution Effect of Kombucha Fraction through AhR Expression (AhR 발현을 통한 콤부차 분획물의 안티폴루션 효과 연구)

  • In this study, Kombucha, a fermented drink by adding beneficial bacteria to green tea or black tea with sugar, was classified using column chromatography, and the presence or absence of quercetin glycoside, a type of flavonoids, was confirmed through TLC. In addition, the anti-pollution effect of protecting and improving the skin from pollution was confirmed using the fraction. Keratinocytes were treated with K-QG to confirm the cell viability, showing a survival rate of 90% or more up to a concentration of 100 ㎍/mL, and the cell viability by benzo[e]pyrene and fine dust stimulation to see the anti-pollution effect was measured. At 100 ㎍/mL, the improvement rates were about 68.79% and 50.68%, respectively. In addition, as a result of confirming the expression of AhR activated by benzo[a]pyrene through Western blot, it showed an inhibition rate of about 31.08% at 100 ㎍/mL compared to the control group. Through the results of this study, K-QG is thought to be valuable as an anti-pollution functional material, protecting and improving skin irritated from benzopyrene and fine dust.

Optimal Design Standard and Application of Low Cost, High Performance Scrubber for Absorbing Hazardous Gas (유해가스 흡수처리를 위한 저비용 고효율 스크러버의 최적 설계기준 및 현장적용 방안)

  • Scrubbers that treat hazardous materials at workplaces have high treatment efficiency; however, the design is complex, and pumps need to be operated 24 hours a day, which can be costly. Therefore, to minimize the operating costs, small businesses do not install scrubbers, or operate them while circulation pumps are suspended. Hence, this study investigated the application of low-cost, high-performance scrubbers that can be used economically in small businesses. Low-cost, high-efficiency scrubbers are applied to bubble columns to utilize devices for hazardous chemical absorption treatment purposes, and for the development of these scrubbers, absorption performance was reviewed and the optimal application method was studied under certain conditions. The changes in the absorption performance of hazardous gas were studied in certain environments by varying the physical conditions, and the optimal application methods were analyzed. The results showed that, while it was possible to treat some of the gas flowing into the low cost, high performance scrubber, the treatment capacity was reduced. Performance degradation was prevented by supplying an absorption liquid, and a certain level of absorption was maintained depending on the amount of circulation. Based on this, three types of site application methods of low cost, high performance scrubbers were presented. In addition, the appropriate timing of circulation and anti freezing measures were also discussed.

A Study on the Seismic Performance Improvement of Mid and Low-Rise RC Grid Structures Using Steel Slab Hysteretic Damper (강재 슬래브 이력형 댐퍼(SSHD)를 이용한 중·저층 RC 격자 구조물의 내진성능 향상에 관한 연구)

  • Purpose: After analyzing the seismic capability of low-rise RC grid structures with insufficient seismic performance, the purpose of the project is to install steel slab hysteretic dampers (SSHD) to improve the seismic performance of beams and columns, and to suggest measures to minimize damage to the structure and human damage when an earthquake occurs. Method: The evaluation of the seismic performance of a structure is reviewed based on the assumption that the seismic performance is identified for the grid-type subway systems that are not designed to be seismic resistant and the installation of an SSHD system, a method that minimizes construction period, if insufficient, is required. Result: After the application and reinforce of structure with SSHD, and the results of eigenvalue analysis are as follows. The natural periodicity of longitudinal direction was 0.55s and that of vertical direction was 0.58s. Conclusion: As results of cyclic load test of structure with SSHD, the shear rigidity of damper is 101%, the energy dissipation rate is 108% and, plastic rotation angle of all column and beam is satisfied for $I_o$ level and therefore it is judged that the reinforce effect is sufficient.

3-Dimensional Numerical Analysis of Air Flow inside OWC Type WEC Equipped with Channel of Seawater Exchange and Wave Characteristics around Its Structure (in Case of Irregular Waves) (해수소통구를 구비한 진동수주형 파력발전구조물 내 공기흐름과 구조물 주변에서 파랑특성에 관한 3차원수치해석(불규칙파의 경우))

  • Oscillating Water Column (OWC) Wave Energy Converters (WEC) harness electricity through a Power-Take-Off (PTO) system from the induced-airflow by seawater oscillating inside a chamber. In general, an air chamber with a relatively small cross-sectional area is required compared to seawater chamber to obtain high-velocity air in the PTO system, and in order to simulate an accurate air flow rate in the air chamber, a three-dimensional study is required. In this study, the dynamic response of OWC-WEC that is equipped with the channel of seawater exchange for the case of irregular waves has been numerically studied. The open source CFD software, OLAFLOW for the simulation of wave dynamics to the openFOAM and FOAM-extend communities, was used to simulate the interaction between the device and irregular waves. Based on the numerical simulation results, we discussed the fluctuation characteristics of three dimensional air flow in the air-chamber, wave deformation around the structure and the seawater flow inside the channel of seawater exchange. The numerical results the maximum air flow velocity in the air-chamber increases as the Ursell value of the significant wave increases, and the velocity of airflow flowing out from the inside of air chamber to the outside is greater than the speed of flowing into the air chamber from the outside.

Recombinant Production and Antimicrobial Activity of an Antimicrobial Model Peptide (Uu-ilys-CF) Derived from Spoon Worm Lysozyme, Uu-ilys (개불 라이소자임 유래 항균성 모델 펩타이드(Uu-ilys-CF)의 재조합 단백질 생산 및 항균 활성)

  • Uu-ilys, an i-type lysozyme from spoon worm (Urechis unicinctus), is an innate immune factor that plays an important role in the defense against pathogens. It also possesses non-enzymatic antibacterial activity. Thus, there is a possibility to develop an antimicrobial model peptide from Uu-ilys. In this study, we report the design, production, and antibacterial activity of an Uu-ilys analog that exhibits antibacterial activity. The Uu-ilys structure was fragmented according to its secondary structures to predict the regions with antimicrobial activity using antimicrobial peptide (AMP) prediction tools from different AMP databases. A peptide containing the C-terminal fragment was predicted to exert antimicrobial activity. The chosen fragment was designated as an Uu-ilys analog containing the C-terminal fragment, Uu-ilys-CF. To examine the possibility of developing an AMP using the sequence of Uu-ilys-CF, recombinant fusion protein (TrxA-Uu-ilys-CF) was produced in an expression system that was heterologous. The produced fusion protein was cleaved after methionine leaving Uu-ilys-CF free from the fusion protein. This was then isolated through high performance liquid chromatography and reverse phase column, CapCell-Pak C18. The antibacterial activity of Uu-ilys-CF against different microbial strains (four gram-positive, six gram-negative, and one fungal strain) were assessed through the ultrasensitive radial diffusion assay (URDA). Among the bacterial strains tested, Salmonella enterica was the most susceptible. While the fungal strain tested was not susceptible to Uu-ilys-CF, broad spectrum antibacterial activity was observed.

Evaluation of the Soil-Structure Interaction of a MDOF Column Type Structure on Group Piles Based on the Large Scale 1g Shaking Table Test and the Numerical Analysis (대형 진동대 실험 및 수치해석을 이용한 다자유도 기둥 구조물과 군말뚝 기초의 지반-구조물 상호작용 평가)

  • Many 1g shaking table tests with an SDOF structure supported by a single pile were performed to evaluate the soil-structure interaction (SSI) effect. Since most structures supported by group piles are MDOF structures with columns, the SSI effect is simulated using a large 1g shaking table test and numerical analysis. According to the results, the movement in the piles tends to increase with input acceleration and when the input frequency is similar to the natural frequency. Furthermore, the slope of the dynamic p-y curve remains constant regardless of the variation of acceleration and input frequency. According to the results of the dynamic p-y backbone curve and the moment of group piles, a center pile with a leading pile has more soil resistance than side piles with a trailing pile, and the effect of group piles is observed above the 7D center to center pile distance.
