  • In order to manufacture a precision mold/die by machining, users need high speed & precision mold/die machining center. So, for development of this machine, we intend to use linear motor that is instead of ball-screw, servo-motor and coupling, high-speed spindle of pressurized air journal bearing and composite materials. In this paper we research column moving type and table moving type. The former is mainly piling 3 axes on one moving body, the latter is consist of two independent carriages. Both types are available to high speed & precision machine, but we finally draw a conclusion column moving type due to an advantage of high-speed control of linear motor.

  • To increase detection sensitivity for multi-DDT residues (o,p-/p,p-DDT, o,p-/p,p-DDE, o,o-/o,p-DDD) analysis, a highly selective sample clean-up method was introduced prior to GC/MS analysis using immunoaffinity column. The immunoaffinity matrix was prepared by coupling IgG fraction of DDT antiserum to cyanogens bromide activated Sepharose 4B. Three DDT antisera (DDA-1, DDHP-2, DDCP-3) were test for affinity column ligand that obtained by imunizing respective DDT immunogen to rabbits, and IgG was purified using protein A affinity purification.   • 본 연구에서는 비선형 전단변형을 고려할 수 있는 Timoshenko보 이론을 정식화 하였다. 제안된 모델은 전단변형을 고려하므로서 짧은 기둥이나 전단 지배 기둥에서 일반적인 Bernoulli보 이론 보다 합리적인 결과를 보여준다. 단면은 층상화 모델을 이용하였으며, 층상화 단면 모델은 단면을 분활하여 소성화 진행과정을 관찰할 수 있으며 축력과 모멘트의 상호작용을 알 수 있다. 정식화한 요소는 일반적인 철근 콘크리트 부재의 해석을 위해 유한요소 프로그램에 적용하였다. 철근콘크리트 기둥의 해석을 실험결과와 비교하였고, 철근콘크리트 기둥에 대한 거동특성을 분석하였다.

  • A model was developed to describe the microbial decontamination of diesel contaminated soil in a soil column. The biodegradation rate of diesel in nature depends on temperature and the pH of soil, availability of nutrients, oxygen and water. The soil moisture content is one of the essential factors because it characterizes the availability not only of water to microorganisms but also of oxygen and nutrient dissolved in soil. In this work, the rate of biodegradation was modeled by coupling Michaelis-Menten kinetics for the aqueous-phase solute with adsoption-desoption equation for diesel sorption and desorption from soil.

Plastic hinge length for coupled and hybrid-coupled shear walls

  • A coupled wall consists of two or more reinforced concrete (RC) shear walls (SWs) connected by RC coupling beams (CBs) or steel CBs (hybrid-coupled walls). To fill the gap in the literature on the plastic hinge length of coupled walls, including coupled and hybrid-coupled shear walls, a parametric study using experimentally validated numerical models was conducted considering the axial stress ratio (ASR) and coupling ratio (CR) as the study variables. A total of sixty numerical models, including both coupled and hybrid-coupled SWs, have been developed by varying the ASR and CR within the ranges of 0.027-0.25 and 0.2-0.5, respectively. A detailed analysis was conducted in order to estimate the ultimate drift, ultimate capacity, curvature profile, yielding height, and plastic hinge length of the models. Compared to hybrid-coupled SWs, coupled SWs possess a relatively higher capacity and curvature. Moreover, increasing the ASR changes the walls' behavior to a column-like member which decreases the walls' ultimate drift, ductility, curvature, and plastic hinge length. Increasing the CR of the coupled SWs increases the walls' capacity and the risk of abrupt shear failure but decreases the walls' ductility, ultimate drift and plastic hinge length. However, CR has a negligible effect on hybrid-coupled walls' ultimate drift and moment, curvature profile, yielding height and plastic hinge length. Lastly, using the obtained results two equations were derived as a function of CR and ASR for calculating the plastic hinge length of coupled and hybrid-coupled SWs.

Cloning and Expression of Cyclodextrin Glycosyltransferase Gene from Paenibacillus sp. T16 Isolated from Hot Spring Soil in Northern Thailand

  • Gene encoding cyclodextrin glycosyltransferase (CGTase), from thermotolerant Paenibacillus sp. T16 isolated from hot spring area in northern Thailand, was cloned and expressed in E. coli (JM109). The nucleotide sequences of both wild type and transformed CGTases consisted of 2139 bp open reading frame, 713 deduced amino acids residues with difference of 4 amino acid residues. The recombinant cells required 24 h culture time and a neutral pH for culture medium to produce compatible amount of CGTase compared to 72 h culture time and pH 10 for wild type. The recombinant and wild-type CGTases were purified by starch adsorption and phenyl sepharose column chromatography and characterized in parallel. Both enzymes showed molecular weight of 77 kDa and similar optimum pHs and temperatures with recombinant enzyme showing broader range. There were some significant difference in pH, temperature stability and kinetic parameters. The presence of high starch concentration resulted in higher thermostability in recombinant enzyme than the wild type. The recombinant enzyme was more stable at higher temperature and lower pH, with lower $K_m$ for coupling reaction using cellobiose and cyclodextrins as substrates.

간극공기압을 고려하는 불포화토의 유한요소해석 (Finite Element Analysis of Partially Saturated Soil Considering Pore-air Pressure)

  • 불포화 지반을 이루고 있는 흙의 알갱이, 물과 공기의 연계해석이 전체 지반의 변형에 미치는 영향을 알아보고자, 흙 알갱이와 물의 두 가지 요소로 해석되어지는 기존 관점과의 차이를 분석하고 그에 대한 원인을 비교하였다. 열역학적 가정을 토대로 평형방정식을 이용하여 불포화토 거동을 지배하는 물과 공기의 흐름 관계를 유한요속해석을 위한 수학적인 알고리즘을 유도하여 배수시험 수치해석을 통해 기존 문헌과 비교 검증하였다. 3상으로 이루어진 불포화토 거동을 통해서 흙의 알갱이와 물과의 상호작용에서 공기압의 역할을 분석하여 불포화 지반 내에 물과 공기의 두가지 흐름에 대해 지반공학적인 적용성을 알아보았다.

밀기울배지를 이용한 Bacillus macerans의 Cyclodextrin Glucanotransferase 생산과 효소특성 (Purification and Enzymatic Properties of Cyclodextrin Glucanotransferase from Bacillus macerans Cultivated in Wheat-bran Medium)

  • Bacillus macerans cyclodextrin glucanotrans­f ferase(CGTase)를 전분흡착법과 DEAE--cellulose 칼럼 크로마토그래피로 정제하였다. 효소의 분자량 은 67,000이였고 monomer였다. 정제된 효소는 전 분을${\alpha}$-, ${\beta}$-, ${\gamma}$-CD 로 전환시켰으며, CD생성비율은 각각 1 : 1.68: 0.32였다. ${\alpha}$-CD와 D-glucose의 coupling반응 초기에는 maltohexose가 주로 생성되었고, 그 후에 다른 oligosaccharide들이 생성되었다 .. a­C CD의 가수분해반응 초기에는 주로 maltotetrose가 생성되였고 그 이후에는 소량의 다른 이Igosa­C ccharide들이 생성되었다. 정제된 효소의 좋은 기질인 maltotriose 로부터 maltosyl 이나 D-glucopy ranosyl group이 전이될 수 있는데, 본 연구에서는 D정lucosyl transfer가 우세하였다.

Effect of rigid connection to an asymmetric building on the random seismic response

  • Connection of adjacent buildings with stiff links is an efficient approach for seismic pounding mitigation. However, use of highly rigid links might alter the torsional response in asymmetric plans and although this was mentioned in the literature, no quantitative study has been done before to investigate the condition numerically. In this paper, the effect of rigid coupling on the elastic lateral-torsional response of two adjacent one-story column-type buildings has been studied by comparison to uncoupled structures. Three cases are considered, including two similar asymmetric structures, two adjacent asymmetric structures with different dynamic properties and a symmetric system adjacent to an adjacent asymmetric one. After an acceptable validation against the actual earthquake, the traditional random vibration method has been utilized for dynamic analysis under Ideal white noise input. Results demonstrate that rigid coupling may increase or decrease the rotational response, depending on eccentricities, torsional-to-lateral stiffness ratios and relative uncoupled lateral stiffness of adjacent buildings. Results are also discussed for the case of using identical cross section for all columns supporting eachplan. In contrast to symmetric systems, base shear increase in the stiffer building may be avoided when the buildings lateral stiffness ratio is less than 2. However, the eccentricity increases the rotation of the plans for high rotational stiffness of the buildings.
