본 연구에서는 단 삼(Salvia miltiorrhiza)의 추출물이 인체 결장암세포인 HT-29, 간암세포인 HepG2 및 직장 암세포인 HRT-18의 증식에 미치는 영향을 in vitro에서 확인하였다. 암세포의 배양액에서 단삼의 수용성 추출물과 에탄올 추출물의 암세포 증식억제효과는 농도에 비례하여 에탄올 추출물에서 더 효과적이었다. 이를 토대로 단삼의 에탄올 추출물의 급성독성, 수명연장 및 고형암형 성억제를 살펴보기 위해 in vivo 실험을 하였다. 급성독성실험결과 대조군의 15일 째의 평균체중은 32.3 g 이었고 실험군은 31.6 g으로 정상적인 상태를 유지하였으며, 흰쥐의 육종암세포인 sarcoma-180를 접종한 mouse의 수명연장실험결과 대조군에 비해 61%의 수 명연장 효과가 있음을 관찰하였다. 고형암억제 실험 결과에서도 대조군에 비해 에찬올 추출 물 처리군이 35%의 고형암 형성억제능을 나타내었다. 따라서 단삼의 에탄올 추출물 중에는 in vitro와 in vivo 실험을 통해 항암활성을 갖는 성분이 존재하며, 이성분은 유효한 항암제 로 개발될 수 있으리라 사료된다.
Despite a considerable improvement in antiemetic chemotherapy and the use of antiemetic prophylaxis for decades, there are still numerous cancer patients suffering from anorexia and nausea after chemotherapy, which forces them to postpone or refuse treatment. This case report is about a 66-year-old female patient diagnosed with malignant lymphoma of the small intestine who underwent right hemicolectomy. After the tumor and the colon adjacent to it were surgically removed, the patient continued to undergo chemotherapy. Chemotherapy-induced anorexia and nausea appeared but were well treated with administration of Dokhwaljihwang-tang-gamibang. During the treatment, no adverse effect related to this Korean medicine was reported. The outcome was evaluated by the functional assessment of anorexia/cachexia therapy (FAACT); the total score increased from 85 to 130 within 12 days, and the symptoms were alleviated. This improvement might suggest the feasibility of Dokhwaljihwang-tang-gamibang to help cancer patients suffering from chemotherapy-induced anorexia and nausea.
Soamsan is known as an anti-cancer remedy in the traditional Korean Medicine. To enhance the synergic effects of anti-cancer activity of Soamsan, this study reconstituted the original components of Soamsan with a slight modification and produced a novel herbal remedy, namely Gamisoamsan. Extracts of Gamisoamsan inhibited the growth of cultured CT-26 cells, mouse colon adenocarcinoma, in a dose-dependent manner $(1\;to\;50{\mu}g/ml)$, and $ID_{50}$ was estimated approximately $16.7{\mu}g/ml$. Using tumor-bearing mouse model, in which was produced by subcutaneous injection of CT-26 cells ($1{\times}10^5$cells). the effects of Gamisoamsan on tumor growth and host survival were examined by evaluating tumor volume and increase in life span. When Gamisoamsan extracts in variable doses of 100, 200 and 500mg/kg body weight per day were orally administered to tumor-bearing mice, following results were obtained: Improvement in the hematological parameters following Gamisoamsan treatment such as hemoglobin contents, red blood cells and white blood cells of the tumor-bearing mice have been observed. Gamisoamsan treatment also showed a prolongation of life span and a reduction of tumor volume in the CT-26 tumor hosts. The results of the present study suggest that Gamisoamsan extracts has a potential anti-tumor activity and may be an useful remedy to prevent and/or treat cancer.
Colorectal cancer (CRC) is the leading cause of common malignant neoplasm worldwide. Many studies have analyzed compositions of gut microbiota associated with various diseases such as inflammatory bowel diseases (IBD) and colon cancer. One of the most representative bacteria involved in CRC is enterotoxigenic Bacteroides fragilis (ETBF), a species belonging to phylum Bacteroidetes. We used ETBF colonized mice with azoxymethane (AOM)/dextran sulphate sodium (DSS) and zerumbone, a compound with anti-bacterial effect, to determine whether zerumbone could restore intestinal microbiota composition. Four experimental groups of mice were used: sham, ETBF colonized AOM/DSS group, ETBF colonized AOM/DSS group zerumbone 60 mg kg-1 (ETBF/AOM/DSS + Z (60)), and only zerumbone (60 mg kg-1)-treated group. We performed reversible dye terminators-based analysis of 16S rRNA gene region V3-V4 for group comparison. Microbiota compositions of ETBF/AOM/DSS + Z (60) group and ETBF colonized AOM/DSS group not given zerumbone were significantly different. There were more Bacteroides in ETBF/AOM/DSS + Z (60) group than those in ETBF colonized AOM/DSS group, suggesting that B. fragilis could be a normal flora activated by zerumbone. In addition, based on linear discriminant analysis of effect size (LEfSe) analysis, microbial diversity decreased significantly in the ETBF colonized AOM/DSS group. However, after given zerumbone, the taxonomic relative abundance was increased. These findings suggest that zerumbone not only influenced the microbial diversity and richness, but also could be helpful for enhancing the balance of gut microbial composition. In this work, we demonstrate that zerumbone could restore the composition of intestinal microbiota.
In this study, potato kimchi was prepared by applying heat to raw potatoes, and then the physico-chemical properties and anti-cancer effects of the kimchi were analyzed. The texture results indicated the potato kimchi had very good hardness and springiness attributes. During th late storage period, total vitamin C content of the kimchi slowly increased. In addition, the potato kimchi had non-volatile organic acid changes that promoted early aging; however, after the complete aging period, it was comparatively similar to other types of kimchi. Using the methanol extracts of various kimchi samples, the potato kimchi(solid 100%) showed the highest anti-carcinogenic effects in terms of anti-tumor activity in tumor bearing Balb/c mice with sarcoma-180 cells. In addition, the effects of the methanol extracts on hepatic glutathione S-transferase content were $289.76\;{\mu}mol/mg$ protein/min, $250.97\;{\mu}mol/mg$ protein/min, $251.20\;{\mu}mol/mg$ protein/min, $219.53\;{\mu}mol/mg$ protein/min, $183.79\;{\mu}mol/mg$ protein/min, for control kimchi, mul kimchi, and two potato kimchis [(solid 100%) and(solid 60%+kimchi juice 40%)], respectively. The in vivo anti-cancer effects of the potato kimchi were investigated using AGS human gastric adenocarcionoma cells and HT-29 human colon adenocarcionoma cells. Overall, an MTT assay revealed that the methanol extract of the potato kimchi showed the highest anti-carcinogenic effects.
Picropodophyllotoxin (PPT), an epimer of podophyllotoxin, is derived from the roots of Podophyllum hexandrum and exerts various biological effects, including anti-proliferation activity. However, the effect of PPT on colorectal cancer cells and the associated cellular mechanisms have not been studied. In the present study, we explored the anticancer activity of PPT and its underlying mechanisms in HCT116 cells. The 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay was used to monitor cell viability. Flow cytometry was used to evaluate cell cycle distribution, the induction of apoptosis, the level of reactive oxygen species (ROS), assess the mitochondrial membrane potential (Δψm), and multi-caspase activity. Western blot assays were performed to detect the expression of cell cycle regulatory proteins, apoptosis-related proteins, and p38 MAPK (mitogen-activated protein kinase). We found that PPT induced apoptosis, cell cycle arrest at the G1 phase, and ROS in the HCT116 cell line. In addition, PPT enhanced the phosphorylation of p38 MAPK, which regulates apoptosis and PPT-induced apoptosis. The phosphorylation of p38 MAPK was inhibited by an antioxidant agent (N-acetyl-L-cysteine, NAC) and a p38 inhibitor (SB203580). PPT induced depolarization of the mitochondrial inner membrane and caspase-dependent apoptosis, which was attenuated by exposure to Z-VAD-FMK. Overall, these data indicate that PPT induced G1 arrest and apoptosis via ROS generation and activation of the p38 MAPK signaling pathway.
Liver colonization is initiated through the interplay between tumor cells and adhesion molecules present in liver sinusoidal endothelial cells (LSECs). This crosstalk stimulates tumor COX-2 upregulation and PGE2 secretion. To elucidate the role of the LSEC intercellular adhesion molecule-1 (ICAM-1) in the prometastatic response exerted by tumor and stromal COX-2, we utilized celecoxib (CLX) as a COX-2 inhibitory agent. We analyzed the in vitro proliferative and secretory responses of murine C26 colorectal cancer (CRC) cells to soluble ICAM-1 (sICAM-1), cultured alone or with LSECs, and their effect on LSEC and hepatic stellate cell (HSC) migration and in vivo liver metastasis. CLX reduced sICAM-1-stimulated COX-2 activation and PGE2 secretion in C26 cells cultured alone or cocultured with LSECs. Moreover, CLX abrogated sICAM-1-induced C26 cell proliferation and C26 secretion of promigratory factors for LSECs and HSCs. Interestingly, CLX reduced the protumoral response of HSC, reducing their migratory potential when stimulated with C26 secretomes and impairing their secretion of chemotactic factors for LSECs and C26 cells and proliferative factors for C26 cells. In vivo, CLX abrogated the prometastatic ability of sICAM-1-activated C26 cells while reducing liver metastasis. COX-2 inhibition blocked the creation of a favorable tumor microenvironment (TME) by hindering the intratumoral recruitment of activated HSCs and macrophages in addition to the accumulation of fibrillar collagen. These results point to COX-2 being a key modulator of processes initiated by host ICAM-1 during tumor cell/LSEC/HSC crosstalk, leading to the creation of a prometastatic TME in the liver.
목적: 최근 들어 $[^{11}C]$choline은 양전자방출단층촬영을 통한 각종 종양을 진단하는데 유용한 방사성의약품으로 보고되고 있다. 그러나 C-11의 반감기가 너무 짧은 단점 때문에 일상적인 작업이 쉽지않고, 잦은 합성이 필요하다. $[^{11}C]$Choline의 단점은 반감기가 적당한 F-18이 치환된 $[^{18}F]$fluorocholine을 합성하여 해결할 수 있다. 대상 및 방법: $[^{11}C]$Choline은 $[^{11}C]CH_3I$와- N,N-dimethylaminoethanol을 반응시켜 합성하였고, $[^{18}F]$fluorocholine은 $CH_2Br[^{18}F]$F과 N,N-dimethylaminoethanol을 반응시켜 합성하였다. 방사화학적 순도는 고성능액체크로바토그래프를 이용하여 확인하였다. 두 화합물의 체내동태는 balb/c mouse를 이용하여 5, 20, 40, 80분에 측정하였다. 세포섭취 실험은 glioma (9L)과 colon adenocarcinoma (SW620)를 사용하였다. 결과: 정제후 방사화학적 순도는 98% 이상이었다. 간에서의 섭취는 시간에 따라 변하지 않았고 $[^{11}C]$choline은 약 20 %ID/g, $[^{18}F]$fluorocholine은 약 13 %ID/g 섭취되었다. 신장에서의 섭취는 시간에 따라 감소하였고, 주사 80분 후에 각각 15 %ID/g, 20 %ID/g 섭취되었다. 9L과 SW620세포섭취 실험에 있어서 $[^{11}C]$choline은 각각 4.93, 18.69 %ID/g, $[^{18}F]$fluorocholine은 각각 1.77, 2.77 %ID/g이 섭취된 것을 확인하였다. 결론: $[^{11}C]$Choline과 $[^{18}F]$fluorocholine은 종양 세포주에 따라 다른 섭취 경향을 나타내는 것을 확인하였다. 이상의 결과들로 보아 다른 종양 세포주에 있어서 두 화합물의 섭취 경향에 관한 연구가 필요하다.
마늘(Allium sativum)의 주요 생리활성 성분들은 다양한 종류의 암에 대해 항암효과가 보고되고 있다. 본 연구에서는 활성추적분리방법(activity-guided purification)을 이용하여 마늘의 항암성분을 발굴하고자 하였다. 마늘 에탄올 추출물을 칼럼크로마토그래피로 얻은 각각의 분획물에 대해서 AGS세포의 증식 억제율을 검증하여 가장 효과가 좋은 분획물로부터 물질을 순수분리하여 구조를 동정한 결과 N-benzyl-N-methyldecan-1-amine (NBNMA)로 밝혀졌다. NBNMA의 암생장 억제효능을 검증하기 위해서 CT-26, AGS, HepG2, HCT-116, MCF7, B16F10 및 Sarcoma-180 세포에 대한 in vitro 효과와 CT-26 결장암 세포를 마우스에 이식한 다음 in vivo 효과를 조사하였다. NBNMA는 Bcl-2의 down-regulation과 Bad의 up-regulation을 유도하여 CT-26 세포의 세포사멸 촉진시켰다. 또한, NBNMA는 세포사멸의 외적 및 내적 경로에서 caspases 억제자인 caspase 3과 caspase 9의 활성을 약간 증가시켰다. CT-26세포를 이식한 쥐에 $19.13{\mu}M/kg$의 NBNMA를 21일 동안 경구투여한 결과 암종의 크기가 43% 감소하였다. NBNMA는 in vitro 및 in vivo에서 항암 효과를 나타내었는데, 이러한 결과는 마늘로부터 순수분리한 NBNMA가 대장암치료를 위한 항암제 후보물질로서 활용 가능성이 있을 것으로 기대된다.
연구배경: 악성종양에 대한 진단 및 치료방법의 발전으로 악성종양 환자의 생존기간이 연장되고, 생존율 또한 과거보다 높아졌다. 또한 수술적 절제술로 악성종양을 제거한 환자 중 약 10% 정도는 약 10년이내 속발성 원발성 종양이 생긴다고 알려지고 있다. 그래서 다발성 원발성 악성종양의 원인과 위험 인자 및 조기 진단은 매우 중요한 일이다. 저자들은 다발성 원발성 악성종양에 병발한 폐암과 원발성 폐암의 임상적 특징과 p53 종양 억제 유전자 변이에 의한 변이형 p53 발현율을 조사하여 두 질환사이의 차이를 비교해 보았다. 방법: Moertel의 다발성 원발성 악성 종양의 분류에 적합한 환자 중 폐암이 동반된 20례(비소세포암 16례, 소세포암 4례)와 비교군으로 원발성 비소세포암 26례를 대상으로 변이형 p53 발현율을 면역조직화학 염색법을 이용하여 조사하였다. 곁과: 폐암과 동반된 악성종양은 위암이 8례로 가장 많았고, 식도암, 폐암, 대장암이 각각 2례 이었으며, 후두암, 방광암, 간암, 유방암, 부신암, 소장암이 각각 1례 이었다. 다발성 원발성 악성종양과 병발한 비소세포암과 원발성 비소세포암의 남녀비는 각각 7:1, 2.7:1 이었고, 흡연력은 각각 16례 중 15례, 26례 중 19례가 있었다. 다발성 원발성 악성종양에 병발한 비소세포암은 원발성 비소세포암에 비해 임상적 병기가 비교적 균등하였으며(p<0.05), 조직학적 분화도는 나쁜 경우가 많은 경향이었다(p>0.05). 다발성 원발성 악성종양에 병발한 비소세포암과 원발성 비소세포암의 변이형 p53 각각 62.5%, 76.7%로 유의한 차이는 없었다(p>0.05). 결론: 다발성 원발성 악성종양에 병발한 비소세포암의 변이형 p53 단백 발현율은 차이가 없었다. 변이형 p53 단백 염색법은 다발성 원발성 악성종양에 병발한 비소세포암과 원발성 비소세포암을 구별하는데 유용성을 인정할 수 없었다.
