Antiproliferation Effects of Germinated-Korean Rough Rice Extract on Human Cancer Cells (한국산 발아 벼 추출물의 여러 가지 암세포주에 대한 증식 억제 효과 비교)

  • This study was conducted to investigate the effects of 70% ethanol extracts of various germinated-rough rice cultivars: ('Ilpum', 'Goami2', 'Keunnun', 'Sulgaeng', 'Baegjinju', and 'Heugkwang') on proliferation of human cancer cell lines (MKN-45, HCT116 and NCI-H460). Antiproliferation effects of rough rice on different cancer cell lines were higher in after germination than before germination. The viability of HCT-116 colon cancer cells was lowest at 18.89% in after germination of 'Heugkwang' at 1.0 mg/mL. The cell viability of MKN-45 lung cancer cells and MKN-45 stomach cancer cells were in the range of 5~10% in after germination of 'Ilpum', 'Goami2', 'Baegjinju', and 'Sulgaeng', and 'Heugkwang' at 1.0 mg/mL. These results suggest that germinated rough rice might have a potential preventive effect on human cancer cells.

  • Coffee enema was started by American doctor Max Gerson for cancer patients in 1920s. Coffee enema is known to remove waste material and toxins in the colon, and increase the enzymatic activity in the liver. In alternative medicine, It is to be used for fecal stasis removal, detoxification, obesity, pain control etc. In recently, The Society of Korean Medicine for Obesity Research make strenuous efforts to spread a new treatment about obesity. And Obesity is already known as one of the main causes of adult diseases. If we use the Korean medical treatment, and at the same time use coffee enema, maybe we will have a good or better clinical result about obesity and metabolic syndrome. In conclusion, we introduce a method that can be easily coffee enema. Thus, we hope that the reference to the teaching of patients.

Ameliorating Activity of Aspalactone on Cisplatin Induced Nephrotoxicity (백금항암제 Cisplatin의 신장독성에 대한 Aspalactone의 경감작용)

  • Cisplatin is an inorganic complex formed by a central atom of platinum surrounded by chlorine and ammonia atoms in the cis position in the horizontal plane, Cisplatin is one of the most effective anticancer drug, widely used against various tumor such as testicular tumor, brain tumor, ovary tumor, bladder carcinoma, colon cancer etc. However its clinical use has been limited by nephrotoxicity, ototoxicity , gastrointestinal disturbances, myeloscrppression and allergic reactions. In these toxicities, dose related and cumulative nephrotoxicity is the major dose limit factor. So, to evaluate the protective effect of aspalactone on cisplatin nephrotoxicity in rats, both compounds were given intraperitoneally, Protective effects of aspalactone against nephrotoxicity of cisplatin were observed when aspalactone was administered to rats 1hr beforecisplatin injection. Hepatotoxicity induced by combination treatment of cisplatin and aspalactone was not observed. The present results indicate that aspalactone may provide protection against cisplatin nephrotoxicity, when it is given 1hr before cisplatin injection.

Cytotoxicity of the ethanol extracts of mulberry leaves, branches and silkworm feces (상엽, 상지 및 잠분 에탄올 추출물의 품종별 세포독성 효과)

  • The cytotoxicity of the ethanol extracts of varietal samples of mulberry leaves and branches and silkworm feces was measured using CT-26 cells originated from murine metastatic colon cancer, using dye uptake assay in order to find potential anticancer agents. Two ethanol extracts (varietal mulberry leaves and mulberry branches) were prepared from 16 varietal mulberries and used as partial extract materials for the activity assay. Among these, the ethanol extracts from Shinkwangppong leaves showed a little anticancer activity, and those from Sugaeppong, Cheongunppong and Gumsulppong branches showed some anticancer activity as well as cytotoxicity. In contrast, ethanol extracts from freeze-dried, the 3rd day of 5th instar feces showed more potent anticancer activity than that of other mulberry leaves, mulberry branches and other 5th silkworm instar larva feces on the basis of high UV absorbance at 665 nm.

Antioxidative Effects of Purple Sweet Potato Extracts (자색고구마 추출물의 항산화 효과)

  • The colored sweet potato, particularly purple sweet potato, has been well known to contain anthocyanins abundantly. This study was conducted to examine the antioxidant properties of purple sweet potato. The chopped purple sweet potato was extracted 2 times with water or acetone for 18 hours at $28^{\circ}C$. The antioxidative potential of each solvent extract was assessed by DPPH free radical scavenging activity assay, FRAP assay, and total phenolic contents. The results showed that both extracts had not only high DPPH free radical scavenging activity but had high level of total phenolic compounds. Furthermore, both solvent extracts were found to have antioxidative effects in human colon cancer cells (HCT 116, HT 29) in DCFDA assay. The notable antioxidant activity of purple sweet potato suggests its significant health benefit and deserves further study to develop into functional food ingredient.

  • As part of a drug discovery program to develope more effective platinum-based anticancer drugs, a series of platinum complexes trans -diaminocyclohexane platinum bi sdiphenylphosphino -ethane (KHPC-002) cis-diaminocyclohexane platinum bisdiphenylphosphino-ethane (KHPC-006) has been evaluated in vitro against 4 human carcinoma cell lines with those of cisplatin using a tetrazolium-based colorimetric assay (MTT assay). The cell lines were two human bladder carcinoma cell lines, HT-1197 and HT-1376, human colon carcinoma cell line, HCT-116, and prostate cancer cell line DU-145.

  • Two known isoquinoline alkaloids, (+)-chelidonine (1) and (-)-stylopine (2), were isolated from $CHCl_3$-soluble fraction of whole plants of Chelidonium majus L. var. asiaticum, and their structures were identified by spectroscopic methods and X-ray crystallographic analysis. Two isolates (1 and 2) were examined for their in vitro cytotoxic activities against five human cancer cell lines including DU-145 (prostate), MCF (breast), A549 (lung), HePG2 (liver), and HT-29 (colon) by sulphorhodamine B (SRB) assay.

  • The effects of Bifidobacterium longum HY8001 supplement intake on the fecal microflora and fecal bacterial enzyme activity were studied in ten healthy human volunteers, before, during and after intake (respectively for 3 weeks). During intake of B. longum HY8001 supplement, fecal, $\beta$-glucuronidase and nitroreductase activities significantly decreased 44.6%(p<0.005) and 32.3%(p<0.01), respectively. Although numbers of major bacterial groups of fecal microflora were not affected by B. longum HY8001 intake for 3 weeks, the number of Bifidobacterium was significantly increased (p<0.05). This result indicates that intake of B. longum HY8001 might be potentially beneficial for the prevention and inhibition of colon cancer and improvement of human intestinal microflora composition.

  • Treatment of patients with esophageal carcinoma remains disappointing. Fewer than 20% *of patients with esophageal carcinoma treated with curative intent by surgery and radiation therapy will be alive at 5 years. Fifty five patients underwent treatment for carcinoma of the esophagus and cardia which retrospective clinical study done between January 1983, and December, 1989 in Department of Thoracic and Cardiovascular Surgery, National Medical Center, Seoul, Korea. There were 48 men. 7 women and the mean age was 56.2 years[range, 38 to 78 years]. The most frequent preoperative findings included dysphagia[46 cases], weight loss[33 cases] and substernal pain[19 cases]. 19 cases[34.5%] were underwent resectional surgery, which was total esophagectomy with esophagogastrostomy[ 5 cases], with colon interposition[3 cases], lower esophagectomy with esophagogastrostomy [10 cases], with total gastrectomy and esophagojejunostomy [1 case]. The operative mortality was 15.8%[3/19] and causes of death were respiratory failure in 2 cases, and 1 case of sepsis. Palliative procedures, such as feeding gastrostomy and jejunostomy[14 cases], bypass surgery without resection[ 4 cases], radiation therapy[ 3 cases] were done.
