담즙산의 생체 활성 물질로서의 역할 (Roles of Bile Acid as an Active Biological Substance)

  • The family of bile acids belongs to a group of molecular species of acidic steroids with very peculiar biological characteristics. They are synthesized by the liver from cholesterol through several complementary pathways and secreted into small intestine for the participation in the digestion and absorption of fat. The bile acids are mostly confined to the territories of the so-called enterohepatic circulation, which includes the liver, the biliary tree, the intestine and the portal blood with which bile acids are returned to the liver. In patients with bile acid malabsorption, the amount of primary bile acids in the colon is increased compared to healthy controls. Although the increase in the secondary bile acids including deoxycholic acid, is reported to have the potency to affect tumorigenesis in gastrointestinal tracts, there is no firm evidence that clinically relevant concentrations of the bile acids induce cancer. The list of their physiological roles, as well as that of the pathological processes is long and still not complete. There is no doubt that many new concepts, pharmaceutical tools and pharmacological uses of bile acids and their derivatives will emerge in the near future.

Antitumor Activity of Peptide Fraction from Traditional Korean Soy Sauce

  • Antitumor activities of a peptide fraction isolated from traditional Korean soy sauce (SSP) were investigated in vitro and in vivo using cancer cell lines and F9 teratocarcinoma-bearing BALB/c mice. SSP exerted a dose-dependent antiproliferative effect on P388D1 mouse lymphoma, F9 mouse teratocarcinoma, and DLD-l human colon cancer cells with $IC_{50}$ values of 11, 50, and $50\mug/ml$, respectively. Tumor growth in F9 teratocarcinoma-bearing BALB/c mice, orally administered with 80 and 200 mg/kg/day of SSPs, was inhibited 10.3% and 52.4%, respectively, and survival days increased by 11.9% and 22.1%, respectively, compared to the control group. The results of antitumor activities exerted by SSP in vitro and in vivo suggest the feasibility of using SSP as an antitumor agent.

Expression of Cyclooxygenase-2 (COX-2) in Colorectal Adenocarcinoma: an Immunohistochemical and Histopathological Study

  • Background: The aim of this study was to evaluate cyclooxygenase-2 (COX-2) immunoreactivity in colorectal adenocarcinomas and to find correlations with different pathological features. Materials and Methods: This study included 35 cases of colorectal carcinoma foir which surgical colectomy specimens were collected. Immunohistochemical staining of COX-2 (cyclooxygenase-2) is done by using the Streptavidin-biotin technique. Results: This work reveals that COX-2 is positive in most cases of colorectal carcinoma and negative in normal colon tissue with statistically non significant relations between COX-2 immunostaining and different pathological features. Conclusions: Our data suggest over expression of COX-2 protein in colorectal carcinoma in contrast to normal mucosa, with a possible role in cell proliferation in carcinogenesis.

무봉합 위십이지장 문합술 (Sutureless Gastroduodenostomy)

  • A gastroduodenostomy is the most physiological reconstruction after a distal gastrectomy. However, a gastroduodenostomy with either sutures or staples has many complications. These include bleeding, leakage and stenosis. A sutureless gastroduodenostomy with a biofragmentable anastomosis ring (BAR) in was used adenocarcinoma patients to prevent these complications from 1999. A BAR is composed of polyglycolic acid and Barium sulfate to allow for X-ray visualization. Hardy in first introduced the BAR in 1985. Since then, it has been used in an anastomosis of the colon or small bowel surgery but its use in a gastroduodenostomy is the first trial in the world. A 70 year male patient, old who received a subtotal gastrectomy (Billroth I), underwent a A sutureless gastroduodenostomy with a BAR. The gastroduodenostomy with the BAR was watertight and maintained the initial burst strength in the gastrografin X-ray study performed at the postoperative 1 week. The BAR began to fragment 3 weeks after the operation and disappeared from the digestive tract completely. The diameter of the anastomosis site was sufficient for passed foods. No other secondary changes from remained foreign bodies were found in the endoscopic examination. In a second operation to treat a primary hepatoma, there was no adhesive changes around the gastroduodenostomy site. In conclusion, a sutureless gastroduodenostomy with BAR is a safe, easy and efficient reconstructive method after a distal gastrectomy.

Dihydroartemisinine Enhances Dictamnine-induced Apoptosis via a Caspase Dependent Pathway in Human Lung Adenocarcinoma A549 Cells

  • Dictamnine (Dic) has the ability to exert cytotoxicity in human cervix, colon, and oral carcinoma cells and dihydroartemisinin (DHA) also has potent anticancer activity on various tumour cell lines. This report explores the molecular mechanisms by which Dic treatment and combination treatment with DHA and Dic cause apoptosis in human lung adenocarcinoma A549 cells. Dic treatment induced concentration- and time-dependent cell death. FCM analysis showed that Dic induced S phase cell cycle arrest at low concentration and cell apoptosis at high concentration in which loss of mitochondrial membrane potential (${\Delta}{\Psi}m$) was not involved. In addition, inhibition of caspase-3 using the specific inhibitor, z-DQMD-fmk, did not attenuate Dic-induced apoptosis, implying that Dic-induced caspase-3-independent apoptosis. Combination treatment with DHA and Dic dramatically increased the apoptotic cell death compared to Dic alone. Interestingly, pretreatment with z-DQMD-fmk significantly attenuated DHA and Dic co-induced apoptosis, implying that caspase-3 plays an important role in Dic and DHA co-induced cell apoptosis. Collectively, we found that Dic induced S phase cell cycle arrest at low concentration and cell apoptosis at high concentration in which mitochondria and caspase were not involved and DHA enhanced Dic induced A549 cell apoptosis via a caspase-dependent pathway.

Relationship between Colonic Polyp Type and the Neutrophil/Lymphocyte Ratio as a Biomarker

  • Aim: We designed this study to investigate the neutrophil lymphocyte ratio as a biomarker in distinguishing colonic polyps which are neoplastic or non-neoplastic. Materials and Methods: One hundred and twenty-five patients with colonic polyps were enrolled into the study. The following data were obtained from a computerized patient registry database: mean platelet volume (MPV), uric acid (UA), platelet count (PC), alkaline phosphatase (ALP), gamma-glutamyl transpeptidase (GGT) and the neutrophil to lymphocyte ratio (NLR). Exclusion criteria were active infectious disease, hematological disorders, and malignancies. Colonic polyps divided into two groups as neoplastic polyps (tubular adenoma, villous adenoma, tubulovillous adenoma) and non-neoplastic polyps (hyperplastic polyps, inflammatory pseudopolyps etc). The relationship between colonic polyp type and NLR was evaluated with statistical analysis. Results: There were 67 patients (53.6%) with neoplastic and 58 (46.4%) patients with non-neoplastic polyps. Mean NLRs of neoplastic and non-neoplastic groups were respectively $3.32{\pm}2.54$ and $2.98{\pm}3.16$ (P<0.05). Conclusion: Although sensitivity and specificity are not high, NLR may be used as a biomarker of neoplastic condition of colonic polyps.

  • 혈액에서 추출된 프로테옴 패턴(단백질 DNA 정보)는 인간 신체 기관의 병리학적 상태를 잠재적으로 반영하고 있다. 신체기관의 질병이나 이상은 이러한 프로테옴 패턴의 분석에 의해 식별될 수 있다고 알려져 있으며 프로테옴 패턴 정보를 분석하는 여러 가지 방법들이 현재 존재하고 있다. 본 논문에서는 SVM(Support Vector Machine)과 GA(Genetic Algoritm)의 융합에 근거하여 암 진단을 위한 디시전 모델의 효과적 학습(learning) 방법을 제안한다. <중략> 그 결과로서 개별적 kernel function 들보다 더 우수한 분류성능을 갖는 최적의 디시전 모델이 얻어졌다. 위암 데이터 셋 과 두 개의 일반 데이터 셋(대장암, 백혈병)을 사용한 컴퓨터 실험에서 제안된 방법이 다른 Kernel function 들에 비해 더 우수한 분류 성능을 보여주었다.

A Clustering Approach for Feature Selection in Microarray Data Classification Using Random Forest

  • Microarray data plays an essential role in diagnosing and detecting cancer. Microarray analysis allows the examination of levels of gene expression in specific cell samples, where thousands of genes can be analyzed simultaneously. However, microarray data have very little sample data and high data dimensionality. Therefore, to classify microarray data, a dimensional reduction process is required. Dimensional reduction can eliminate redundancy of data; thus, features used in classification are features that only have a high correlation with their class. There are two types of dimensional reduction, namely feature selection and feature extraction. In this paper, we used k-means algorithm as the clustering approach for feature selection. The proposed approach can be used to categorize features that have the same characteristics in one cluster, so that redundancy in microarray data is removed. The result of clustering is ranked using the Relief algorithm such that the best scoring element for each cluster is obtained. All best elements of each cluster are selected and used as features in the classification process. Next, the Random Forest algorithm is used. Based on the simulation, the accuracy of the proposed approach for each dataset, namely Colon, Lung Cancer, and Prostate Tumor, achieved 85.87%, 98.9%, and 89% accuracy, respectively. The accuracy of the proposed approach is therefore higher than the approach using Random Forest without clustering.

Cytotoxic Activities of Indigenous Plant Extracts in Cultured Human Cancer Cells

  • In continuous efforts for discovery of novel potent antitumor agents from natural products, fifty-seven methanolic extracts derived from indigenous Korean plants were primarily evaluated for in vitro cytotoxic activity in cultured human lung (A549) and colon (Col2) cancer cells. As a result, 16 plant extracts were found to be active against A549 cells and 15 extracts were active against Col2 cells in the criteria of $IC_{50}$<$50\;{\mu}g/ml$. In particular, the extracts of Calystegia soldanella $(IC_{50}$<$8.0\;{\mu}g/ml\;in\;A549;IC_{50}=27.4\;{\mu}g/ml\;in\;Col2)$, Heloniopsis orientalis $(IC_{50}=4.6\;{\mu}g/ml\;in\;A549; IC_{50}=4.5\;{\mu}g/ml\;in\;Col2)$, and Thuja koraiensis $(IC_{50}=1.2\;{\mu}g/ml\;in\;A549;IC_{50}=0.6\;{\mu}g/ml\;in\;Col2)$ showed a potent cytotoxic activity. Further study for the identification of active compounds from these lead extracts might be warranted.

Immunostimulating and Anticancer Activities of Hot-water Extracts from Acanthopanax senticosus and Glycyrrhiza uralensis

  • When 10 kinds of herbal medicines were fractionated into hexane, MeOH, cold-water, and hot-water extracts, hot-water extracts from Acanthopanax senticosus (AS), Glycyrrhiza uralensis (GU), Cichorium intybus (CI), and Polygonatum odoratum (PO) showed the potent intestinal immune system modulating activity (1.72-, 1.62-, 1.60-, and 1.53-fold of control at $100{\mu}g/mL$, respectively). Especially, hot-water extracts from AS (215% compared with the control) and GU (187%) also had macrophages stimulating activity and mitogenic activity of splenocytes (7.1- and 6.5-fold) at $100{\mu}g/mL$. In addition, the effects of hot-water extracts from herbal medicines on anticancer activities were studied in mice. Hot-water extracts from AS and GU enhanced cytotoxicity of natural killer cell against cancer cell, Yac-1 (37 and 34% cytotoxicity) at E/T ratio 100:1, and colon 26-M3.1 cancer cell lines had significantly inhibited (82.1 and 75.2%) in experimental lung metastasis. These results suggest that hot-water extracts from A. senticosus and G. uralensis can be used as biological response modifiers to stimulate immune system and inhibit tumor.
