Changes of Antioxidant Enzymes in Stevia Plants under Clinorotation, Shaking, and Low Temperature Stresses (스테비아(Stevia rebaudiana Bertoni)에 있어서 유사미소중력, 진동 및 저온처리에 의한 항산화 활성 변화)

  • A medicinal herb, Stevia rebaudiana Bertoni which is grown under physical stresses such as simulated microgravity, shaking, and low temperature for 4 days, showed fresh weight decrease of 3.6%, 21% and 8.7% compared with the respective control. On control plants, the radical scavenging value of DPPH represented 86% and 55%, respectively in the leaves and stems extracts. Relatively weak antioxidant activities of 22% and 27% were measured respectively in AA (ascorbic acid) and BHA (beta-hydroxyacetic acid) known as synthetic antioxidants. The radical scavenging effect of DPPH (2,2-diphenyl-1-picryl hydazal) in stevia plants under a simulated microgravity was observed to be consistently higher relative to the control, whereas those effects of shaking and low temperature treatments rapidly increased and then reduced after 6 hours in case of shaking process and 24 hours in case of low temperature treatment, which results had similar levels of scavenging effects to the control. The plants under simulated microgravity showed the highest level of activity with the value of 147% and the shaking and low temperature treatments showed the increases of SOD activity by 121% and 125%, respectively. From the above results, it is clarified that the simulated microgravity is more effective to the antioxidant activity than those of other abiotic stresses.

Effects of Pasteurization and Storage Temperatures on the Physicochemical Characteristics of Kiwi Juice (참다래 쥬스의 이화학적 특성에 미치는 살균 및 저장온도의 영향)

  • To investigate the effects of storage and pasteurization temperature on the quality of kiwi juice $(13.5^{\circ}Brix)$, the Kiwi juice was pasteurized at 65, 75 and $85^{\circ}C$ for 15 sec. The microbial, physicochemical measurements and sensory evaluations were conducted at the same condition during storage at 4 and $25^{\circ}C$ for 30 days. Most of the vegetative bacteria cells in kiwi juice were destroyed by heat treatment over $65^{\circ}C$, and they did not actively grow in kiwi juice after pasteurization. The D values of bacteria in kiwi juice by tubular type of heating exchange were 4.17, 1.47 and 0.81 sec at 65, 75 and $85^{\circ}C$, respectively. The growth of microorganisms during storage were not detected in the most samples. The amounts of vitamin C decreased as the pasteurization-storage-temperature and storage time increased. While reducing sugar increased as the pasteurization-storage temperature-storage time increased, it decreased rapidly after 20 days of storage. Hunter's color values L, a and b of stored kiwi juice were decreased at all storage conditions, browning reaction rate increased as the pasteurization-storage-temperature was decreased.

  • Meat tenderness is one of the most important factors in meat products because it plays a major role in the palatability of meat. To get information on the role of commercial meat tenderizer, the tenderizing ability of commercial meat tenderizer was measured with various substrates. The results obtained are as follows. 1. Content of crude protein in a commercial meat tenderizer was 4.9%. 2. Optimum temperature for proteolytic activity of meat tenderizer was $60{\sim}70^{\circ}C$. 3. Maximal activity of proteinase was obtained at pH $6{\sim}7$. 4. Proteolytic enzyme was activated by KCN, NaCN, EDTA. Thus, it was concluded that protease system of commercial meat tenderizer composed of plant origin proteinases. 5. Proteinase activity was completely inhibited by 10mM of N-Ethylmaleimide. 6. Commercial meat tenderizer showed stronger proteolytic activity on casein than on the water soluble fraction of meat protein, whereas it hydrolyzed the myofibrillar protein less efficiently.

Isolation of Chromoprotein and Its Amino Acids Composition in Korean Laver (한국산(韓國産) 김중의 색소단백질(色素蛋白質)의 분리(分離) 및 그 아미노산(酸) 조성(組成))

  • In this research chromoproteins, namely phycoerythrin and phycocyanin in Korean laver are isolated and purified and their characteristics and the contents of amino acid are investigated. Three kinds of laver, Yonpyungdo product, Jindo product, and Kohung product are used in the experiments. The results are as following: 1) 0.68g of phycoerythrin and 0.72g of phycocyanin are isolated from 50g of laver. 2) when observed by microscope (1,000 times amplification), the crystals of phycocrythrin have the shape of small needles and phycocyanin that of platelets. 3) Absorption spectra of purified phycoerythrin shows strong absorption peaks at 546, 526, and $496m{\mu}$,and phycocyanin shows strong absorption peaks at 610 and $546m{\mu}$. After the phycoerythrin and phycocyanin were kept at $4^{\circ}C$ for 24 and 48 hours. The peaks of phycoerythrin are slightly shifted and those of phycocyanin are decreased. 4) In amino acid analysis, sixteen kinds of amino acids are detected in phycoerythrin and phycocyanin. Amino acids found in the greatest amounts in phycoerythrin are glutamic acid, alanine, glycine and leucine. Essential amino acids are found to exist in phycocyanin in greater amounts than in phycoerythrin.

Study on the Chemical Change of General Composition, Fatty Acids and Minerals of Rapeseed during Germination (발아에 의한 유채의 일반성분, 지방산 및 무기물의 조성변화)

  • The objective of this study was to investigate the technical feasibility of producing toxicant-free rapeseed by germination. To this end, rapeseed (Brassica napus L.) was germinated at $25^{\circ}C$ for 120 hours, and the compositional changes of fatty acids and minerals were determined in every 24 hours during germination. Repeseed slightly decreased it's fat content during germination, whereas it showed a tendency to increase protein content. The contents of oleic, erucic, linoleic, linolenic, stearic, palmitic and arachidic acids in rapeseed before germination were found to be 24.6, 15.5, 10.6, 9.5, 1.0, 2.4 and 5.3% respectively. They were gradually decreased until 96 hours and showed a slight increase in 120 hours during germination. However, there was no significant difference in the variation of fatty acids composition in rapeseed during gemination. The content of minerals-phosphorous, calcium, magnesium, zinc, manganese, copper and iron-in rapeseed before germination were 6367, 3068, 2667, 80.3, 26.5, 15.9, and 94.4 ppm respectively and sodium was detected in trace. The mineral contents in rapeseed were gradually decreased in the early period of germination, but showed a slight increase in 120 hours.

Effects of High Hydrostatic Pressure Treatment on the Chemical Composition of Germinated Rough Rice (Oryza sativar L.) (고압처리가 발아벼의 화학성분 변화에 미치는 영향)

  • This study was performed to evaluate changes in the chemical composition of germinated rough rice with high hydrostatic pressure treatment (HPT). Rough rice was germinated at $37^{\circ}C$ over 6 days (control), and then subjected to HPT at 30 MPa for 24 h. The highest crude protein content was 9.54% in the control sample after 6 days of germination. Crude lipid content increased from 2.04-2.74% (control) to 2.27-3.10% (HPT). HPT samples showed higher values of total free sugar and glucose content than those of the control. The total amino acid value was not significant, but the essential amino acid content increased from 0.45-5.09 mg/g in the control to 1.57-5.30 mg/g in the HPT sample. The major fatty acids were found to be palmitic, oleic, and linoleic acid. The content of oleic acid decreases with HPT, whereas that of linoleic and linolenic acid increased slightly during the initial stages of germination. These results suggest that HPT after germination efficiently depolymerizes chemical components and enhances the content of essential nutrients.

Analysis of Antibiotic Resistant Patterns in Conjugant and Transformant of Three ESBL gene Harboring Klebsiella pneumoniae (세 가지의 ESBL 유전자를 가지고 있는 Klebsiella pneumoniae의 유전자접합체와 헝질전환체의 항생제내성분석)

  • To investigate the antibiotic resistant patterns of the bacteria producing ESBL, we isolated one organism of Klebsiella pneumoniae from a clinical laboratory in Busan. The organism that produces ESBL gene was detected by double disk synergy test and the presence of three ESBL genes (TEM-1, SHV-12, CTX-M-15) was confirmed by polymerase chain reaction and DNA sequencing analysis. To analyse the characteristics of three ESBL genes, we performed transconjugation, transformation and cloning experiment with the organism. The MIC of Klebsiella pneumoniae was revealed that ceftazidime, cefotaxime and ceftriaxone were $256\;{\mu}g/ml,\;128\;{\mu}g/ml\;and\;128\;{\mu}g/ml$ respectively. The MIC of conjugant (E. coli $RG176^{Na(r)}$) af was revealed that ceftazidime, cefotaxime and ceftriaxone were $256\;{\mu}g/ml,\;64\;{\mu}g/ml\;and\;128\;{\mu}g/ml$ respectively. The MIC of transformant (E. cofi $DH5{\alpha}$) was revealed that ceftazidime, cefotaxime and ceftriaxone were $128\;{\mu}g/ml,\;32\;{\mu}g/ml,\;and\;32\;{\mu}g/ml$ respectively, The MIC of cloned organism of SHV-12 gene (E. coli $DH5{\alpha}$) was revealed that ceftazidime, cefotaxime and ceftriaxone were $128\;{\mu}g/ml,\;8\;{\mu}g/ml,\;and\;32\;{\mu}g/ml$ respectively. The results indicated that MIC of conjugant was higher than MIC of transformant and also SHV-12 gene were not resistant against cefotaxime antibiotic.

Investigation of Single Nucleotide Polymorphisms in the Adipocyte Fatty-Acid Binding Protein (FABP4) Gene (FABP4 유전자의 단일염기 다형성에 관한 연구)

  • We found 8 single nucleotide polymorphisms (SNPs) in adipocyte fatty acid bonding protein (FABP4) gene as candidate gene of FAT1 locus on pig chromosome 4. With over 800 heads of major commercial pig breeds including Duroc, Landrace, Berkshire and Yorkshire, we analyzed SNPs of FABP4 gene to determine possible effects of FABP4 genotype to economically important traits. $400{\sim}800\;bp$ amplicons in FABP4 gene were used PCR-RFLP for each SNPs and we found that the frequency of some SNPs of this gene was different among the breeds. According to the statistical analyses to determine possible associations of each genotype with economic traits, it was found that subgroup with different genotypes showed significant differences in daily gain, backfat thickness, lean percentage and feed conversion ratio (P<0.05). Thus, as a Part of enhancing the selection competence related to swine growth rate and lean percentage, it is expected that FABP4 gene markers verified in this study will be useful to use for Korean commercial pig industry.

Effect of Dietary Benzoic Acid on Beneficial Microflora and Immune Response in the Intestine of Weaning Pigs (사료내 벤조산 첨가가 이유돼지의 장내 미생물 균총 및 면역체계에 미치는 영향)

  • We evaluated the effect of dietary supplements with benzoic acid on intestinal beneficial bacteria concentration and immune response of weaning pigs. Supplementation with benzoic acid at 0.5% or control diet for 35 days resulted in a higher Lactobacillus casei concentration in the cecum. Supplementation with benzoic acid at 0.5% increased concentration of L. plantarum in the cecum. Pigs with the control diet and 0.5% benzoic acid had significantly increased concentration of B. subtillis in the cecum compared to the antibiotic group, while the concentration of B. subtillis in the rectum increased in pigs given 0.3 and 0.5% benzoic acid (p<0.05). Compared with the control group, the level of interleukin-$1{\beta}$ mRNA showed a significant decrease in the proximal small intestine in pigs fed diets supplemented with benzoic acid at 0.5% or antibiotic. Feeding 0.5% benzoic acid resulted in a marked reduction in the expression of IL-6 mRNA in the middle small intestine (p<0.05). Supplementation with benzoic acid at 0.5% or antibiotic resulted in a lower level of tumor necrosis factor-mRNA in the middle intestine. Up to 0.5% benzoic acid may be included in weaning diets for improvement of intestinal beneficial bacteria, thus modulating genes of pro-inflammatory cytokines in the gastrointestinal tract.

Fiber Type Specific Expression of Toll-like Receptor4, IL-6, TNF-α, and Suppressor of Cytokine Signaling-3 after Acute Exercise in Rat Skeletal Muscles (일회성 유산소운동 후 쥐의 골격근에서 toll-like receptor4, IL-6, TNF-α 및 suppressor of cytokine signaling-3의 근섬유 형태 특이적 발현)

  • The purpose of this study was to determine whether a single bout of aerobic exercise affects the expression level of toll-like receptor4 (TLR4), IL-6, TNF-${\alpha}$, and suppressor of cytokine signaling-3 (SOCS-3) expression in rat hindlimb muscles depending on fiber types. To accomplish this, thirteen 7-wk Balb/c male mice were randomly assigned to an experimental group or a control group. The exercise protocol consisted of a single bout of treadmill exercise (inclination $10^{\circ}$, speed 17 cm/sec 10 min, 33 cm/sec 10 min, 50 cm/sec) and the animals were killed 24 hr after the exhaustion protocol. The level of TLR4, IL-6, TNF-${\alpha}$, and SOCS-3 mRNA expression was measured by quantitative real-time PCR in soleus and plantaris muscles. A single bout of aerobic treadmill exercise increased TLR4 mRNA expression in the soleus muscle (p<0.05), whereas plantaris TLR4 mRNA expression did not change. Additionally, acute exercise led to a significant increase in IL-6, TNF-${\alpha}$, and SOCS-33 mRNA in the soleus muscle, while transcripts of these genes were not affected by exercise in the plantaris muscle. In conclusion, expression level of several immune-related genes such as TLR4, cytokines, and SOCS-3 is regulated by acute exercise in a fiber type specific manner.
