The Usefulness of Selected Physicochemical Indices, Cell Membrane Integrity and Sperm Chromatin Structure in Assessments of Boar Semen Sensitivity

  • The present work describes experiments undertaken to evaluate the usefulness of selected physicochemical indices of semen, cell membrane integrity and sperm chromatin structure for the assessment of boar semen sensitivity to processes connected with pre-insemination procedures. The experiments were carried out on 30 boars: including 15 regarded as providers of sensitive semen and 15 regarded as providers of semen that is little sensitive to laboratory processing. The selection of boars for both groups was based on sperm morphology analyses, assuming secondary morphological change incidence in spermatozoa as the criterion. Two ejaculates were manually collected from each boar at an interval of 3 to 4 months. The following analyses were carried out for each ejaculate: sperm motility assessment, sperm pH measurement, sperm morphology assessment, sperm chromatin structure evaluation and cell membrane integrity assessment. The analyses were performed three times. Semen storage did not cause an increase in the incidence of secondary morphological changes in the group of boars considered to provide sperm of low sensitivity. On the other hand, with continued storage there was a marked increase in the incidence of spermatozoa with secondary morphological changes in the group of boars regarded as producing more sensitive semen. Ejaculates of group I boars evaluated directly after collection had an approximately 6% smaller share of spermatozoa with undamaged cell membranes than the ejaculates of boars in group II ($p{\leq}0.05$). In the process of time the percentage of spermatozoa with undamaged cell membranes decreased. The sperm of group I boars was characterised with a lower sperm motility than the semen of group II boars. After 1 hour of storing diluted semen, the sperm motility of boars producing highly sensitive semen was already 4% lower ($p{\leq}0.05$), and after 24 hours of storage it was 6.33% lower than that of the boars that produced semen with a low sensitivity. Factors that confirm the accuracy of insemination male selection can include a low rate of sperm motility decrease during the storage of diluted semen, low and contained incidence of secondary morphological changes in spermatozoa during semen storage and a high frequency of spermatozoa with undamaged cell membranes.

Genetic analyses of Acanthamoeba isolates from contact lens storage cases of students in Seoul, Korea

  • We conducted both the small subunit ribosomal DNA (SSU rDNA) polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) and mitochondrial (mt) DNA RFLP analyses for a genetic characterization of Acanthamoeba isolates from contact lens storage cases of students in Seoul, Korea. Twenty-three strains of Acanthamoeba from the American Type Culture Collection and twelve clinical isolates from Korean patients were used as reference strains. Thirty-nine isolates from contact lens storage cases were classified into seven types (KA/LS1 , KA/LS2, KA/LS4, KA/LS5, KA/LS7 KA/LS18, KA/LS31). Four types (KA/LS1 , KA/LS2, KA/LS5, KA/LS18) including 33 isolates were regarded as A. castellanii complex by riboprints. KA/LS1 type was the most predominant (51.3%) in the present survey area, followed by KA/LS2 (20.9%), and KA/LSS (7.7%) types. Amoebae of KA/LS1 type had the same mtDNA RFLP and riboprint patterns as KA/E2 and KA/E12 strains, clinical isolates from Korean keratitis patients. Amoebae of KA/LS2 type had the identical mtDNA RFLP patterns with A. castellanii Ma strain, a corneal isolate from an American patient as amoebae of KA/LS5 type, with KA/E3 and KA/E8 strains from other Korean keratitis patients. Amoebae of KA/LS 18 type had identical patterns with JAC/E1, an ocular isolate from a Japanese patient. Three types , which remain unidentified at species level, were not corresponded with any clinical isolate in their mtDNA RFLP and riboprint patterns. Out of 39 isolates analyzed in this study, mtDNA RFLP and riboprint patterns of 33 isolates (84.6%) were identical to already known clinical isolates, and therefore, they may be regarded as potentially keratopathogenic. These results suggest that contact lens wearers in Seoul should pay more attention to hygienic maintenance of contact lens storage cases for the prevention of Acanthamoeba keratitis.

  • Microbes and their toxins can be bioweapons that bioterrorists use them to commit bioterrorism and biocrime. Due to the potential and relative ease of the bioattack, life-threat pathogenic agents (bacteria, viruses, and toxins) as bioweapon revealed the need for a new field of microbial forensics. Microbial forensics is a new scientific discipline combining microbiology and forensic science, which is focused on characterization of evidence from a bioterrorism, biocrime, and an inadvertent release of biothreat agents. The sophisticated analytical tool and knowledge of microbial forensics can provide investigative leads and help determine who was responsible for the biocrime, the source of the bioweapon, and how and where the bioweapon was produced. Among the fields of microbial forensics, this paper will briefly describe evidence collection, handling, packaging, transportation, storage, analytical methods of evidence, and review microbial forensics as a response to bioterrorism and biocrime.

  • Bastard indigo, prevailing shrub species planted in erosion control work to constitute vegetation, is proved to be usefull for pulp material other than existing usage of green manure or fuel. Pulp made from bastard indigo is good enough for filler pulp though the quality of it is not remarkably excellent. (1) Sorts of paper possible to make from this pulp. Packing paper of medium grade. (general packing paper except heavy packing) (2) Traits of this pulp as for pulp material (A) It is more preferable than annual plants in these respects, ego collection, transport and storage of pulp material, and yield, freeness and chemical consumption of pulp. (B) Annual probable production of pulp material per ha from this plant is higher than that from long-term tree species or similar to that from fast growing species. (C) Its cultivation on eroded area is welcomed and consecutive annual production of material by copice method is also proved possible.

  • The GLP contains specific language concerning characterization of the test, control and reference substances used in toxicity studies. This paper will describe and discuss what types of documents are required to support test/reference substance characterization under GLP system. The purpose of this article is to present an overview of data needed in the characterization package that will adequately define the substance. Most sponsors use a certificate of analysis (COA) to communicate the test sub-stance characterization status information to the contracting research organizations. The COA should provide the test $material^{\circ}{\phi}s$ characterization results, substance storage requirements, expiration dates, verification of the collection of the retention sample, archival location of the data to support the characterization and GLP compliance status of the characterization.

To understand quality of information services, This articles deal with concept of quality on information related sector, quality management, the reason of importance of information quality, quality work of information services. There is little method to measure and evaluate quality of internet services. So it is necessary to study on internet quality, user's selection on its quality, and development on high quality site.

  • A learning resource is a SCO or a collection of on or more assets in the SCORM. A storage policy is required to search rapidly and reuse assets in e-learning environment. However there are not research results about it. In this paper, We propose a storing method for assets based on cluster and define the mathematical formulation of it. Also, we present criteria for assets evaluation and describe procedure to evaluate each asset. We show that the search based on proposed cluster storing method increase performance than the categorization search through performance evaluation.

  • Recently, as value for practical use of big data is evaluated, companies and organizations that create benefit and profit are gradually increasing with application of big data. But specifical and theoretical study about possible risk factors as introduction of big data is not being conducted. Accordingly, the study extracts the possible risk factors as introduction of big data based on literature reviews and classifies according to big data processing, data collection, data storage, data analysis, analysis data visualization and application. Also, the risk factors have order of priority according to the degree of risk from the survey of experts. This study will make a chance that can avoid risks by bid data processing and preparation for risks in order of dangerous grades of risk.

  • Deduplication technique can expand the lifespan and capacity of flash memory-based storage devices by eliminating duplicated write operations. The deduplication techniques can be classified into two approaches, i.e., online and offline approaches. We propose an offline deduplication technique that uses a lightweight hash algorithm, whereas the previous offline technique uses a high-cost hash algorithm. Therefore, the memory space for caching hash values can be reduced, and more pages can be examined for deduplication during short idle intervals. As a result, it can provide shorter write latencies compared to the online approach, and can show low garbage collection costs compared to the previous offline deduplication technique.
