Effect of Mulberry Extract Complex on Degenerative Arthritis In Vivo Models (In Vivo 실험모델에서 오디추출복합물의 퇴행성관절염 개선 효능 연구)

  • The objective of this research was to investigate the in vivo effects of treatment with mulberry extract complex (MEC) on cartilage degeneration and pain severity in an experimental model of rat degenerative arthritis. Monosodium iodoacetate ($2mg/50{\mu}L$) was injected into right knee joints of rats, followed by administration of MEC for 8 weeks at 400 mg/kg or 800 mg/kg of body weight. The experimental data show that treatment with MEC inhibited degradation of glycosaminoglycan and collagen in cartilage. On the other hand, concentrations of cartilage oligomeric matrix protein, C-terminal telopeptide-2, matrix metalloproteinase (MMP)-2, MMP-9, and MMP-13 in serum decreased in comparison with the control. The MEC at all dose levels could inhibit formation of xylene-induced ear edema. In this study, MEC demonstrated significant anti-arthritis activity, which is required for improvement of degenerative arthritis. Based on these results, MEC may be employed for the development of new health foods to ease symptoms of degenerative arthritis.

The effects of Far-infrared on Type II collagen induced mouse and the relative cytokine computerize programming (Emphasis on TNF-a and IL-1b) (원적외선이 관절염 흰쥐 모델에 미치는 영향과 관련 싸이토카인 예측 프로그래밍)

  • The purpose of this study was to observe the effects of Far-infrared on mice rheumatoid arthritis induced by type II collagen. The author peformed several experimental tests which were the hematoxylin-eosin tissue stain, the hind paw thickness, and ELSIA of cytokines(TNF-$\alpha$, IL-1$\beta$). On day 14 and 21, FIR-Tx. group decreased in synovial membrane thickness more than control II group FIR-Tx group decreased in hind paw thickness more than control II group. In the ELISA test of TNF-$\alpha$ concentration, control II group significantly increased in the concentration more than normal and FIR-Tx. group on day 7, 14, and 21. In the ELISA test of IL-$\beta$ concentration, FIR-Tx. group decreased in the concentration more than control II group on day 14 and 21. With J's Linear Function Data Process Program, researchers can get the estimated the concentration or the hind paw thickness from this experiment. The estimated results would be a good guide to therapists and researchers, planning Far-infrared application on the rheumatoid arthritis. These results indicated that far-infrared irradiating appeared to reduce the concentration of TNF-$\alpha$ and IL-1$\beta$ at mouse model of autoimmune arthritis. It would be considered that Far-infrared has an effect on relieving rheumatoid arthritis.

Dendritic Cell-based Immunotherapy for Rheumatoid Arthritis: from Bench to Bedside

  • Dendritic cells (DCs) are professional antigen presenting cells, and play an important role in the induction of antigen-specific adaptive immunity. However, some DC populations are involved in immune regulation and immune tolerance. These DC populations are believed to take part in the control of immune exaggeration and immune disorder, and maintain immune homeostasis in the body. Tolerogenic DCs (tolDCs) can be generated in vitro by genetic or pharmacological modification or by controlling the maturation stages of cytokine-derived DCs. These tolDCs have been investigated for the treatment of rheumatoid arthritis (RA) in experimental animal models. In the last decade, several in vitro and in vivo approaches have been translated into clinical trials. As of 2015, three tolDC trials for RA are on the list of ClinicalTrial.gov (www.clinicaltrials.gov). Other trials for RA are in progress and will be listed soon. In this review, we discuss the evolution of tolDC-based immunotherapy for RA and its limitations and future prospects.

Suppressive Effect of CYTG on Collagen Induced Arthritis(CIA) in DBA1/J Mice (CYTG 처방이 콜라겐 유발 관절염 모델에 미치는 효과)

  • To evaluate effect of CYTG on inhibiting the occurrence of arthritis, we performed the experiments including production of inflammatory cytokine and immunoglobulin in collagen arthritis model. The results were obtained as fellows. CYTG group showed inhibitory effect on arthritis incidence than control group for four weeks. Arthritis index of CYTG group reduced compared with control group. In CYTG group, production of cytokines which show suppressive effect on inflammation(IL-2, COX-2) was increased and which promotes inflammation(IL-10) was decreases in spleen. In CYTG group, production of immunoglobulin (IgG-RF) was reduced compared with control, and rate of CD3+CD69+T cell is lower in lymph node and CD4+CD25+ T cell is higher in lymph node and spleen. And synovial infiltration in the knee were observed in the controls (PBS-treated mice), whereas CYTG-treated mice exhibited significantly reduced histologic evidence of destruction and inflammation. So, the histopathological scoring average of CYTG group was 2.5 compared with control group(CIA mice) 4.5. It was thought that our data express high effect via immune system specially through the controling the inflammatory cytokines and immunoglobulins. CYTG could be usefully applied for the prevention and treatment of RA, and also is expected to be clinically helpful on the treatment of RA through modification.

Effect of Ulmus davidiana Planch herbal acupuncture Solution on proinflammatory cytokine IL-$1{\beta}$ and $TF-{\alpha}$ a production in collagen-induced arthritis of rats

  • Objective: We have evaluated UDHA into the joint for its effectiveness on immune responses to CII in the rat CIA. In an attempt to gain further insight into the mode of action of UDHA, we also investigated the effects of UDHA on the incidence and development of arthritis in rat CIA with 2 different regimens: (1) started prior to a primary immunization, (2) started on the day of a primary immunization. Methods : Male rats were immunized with an emulsion of $200\;{\mu}g/100g$ of CII and complete Freund's adjuvant (CFA). The rats were then given intraperitoneal(i.p) stimulation of Ulmus davidiana Planch herbal acupuncture(UDHA) or saline during the experiment. Lymph node cells were obtained from rats 14 days after immunization and cultured in vitro with CII. When compared with rats treated with saline as control, UDHA at doses of more than $20\;{\mu}g/100\;g$ rat once a day for 7 days inhibited the ability of inguinal lymph node cells to produce T cell cytokines interleukin-$1{\beta}$, tumor necrosis $factor-{\alpha}$ $(TNF-{\alpha})$. When rats were injected intraperitoneally with SRBC, hemaglutination titers in UD-treated and control rats did not differ significantly when low doses of UD was given to rats. However, i.p injection of UD at doses of more than $10\;{\mu}g/100\;g/day$ for 7 days slightly suppressed antibody production. Results : The present results show that treatment with UDHA can inhibit the onset and development of arthritis and the immune responses to collagen. Conclusion: Therapeutic i.p injection with UD affect the clinical course of the disease and the immune responses to CII.

The Effect to Inhibiting MIF Activation and Controlling of T Cell Specialization of Ulmus Davidiana Planchon Herbal Acupuncture on TypeII Collagen-induced Arthritis in Mice (유근피 약침이 제2형 Collagen 관절염에서 MIF 활성 억제 및 T세포 분화 조절에 미치는 영향)

  • Rheumatoid arthritis(RA) is a general, chronic, inflammatory and auto-immune disease and it can lead to joint edema, pain, stiffness which are caused by an inflammation in synovium covering our joints. Ulmus davidiana Planchon is a traditional herb used for the treatment on various inflammations, gastrointestinal trouble, ENT(ear, nose, and throat) disease, edema, cancer etc. and it works effectively on arthritis as well. In these study to search for the treatment efficacy of Ulmus davidiana Planchon in RA, I measure manifestation of cytokine gene in synoviocyte treated with Ulmus davidiana Planchon herbal acupuncture and in EL-4 cell, manifestation of cytokine gene cell related to T-cell. And after Ulmus davidiana Planchon herbal acupuncture treatment in Collagen induced arthritis(CIA) which has been known by a general model of RA, DBA mice, I observed foot thickness, general shape of synovium, early cytokine induce CIA and, generation and mutation of cytokine related to the control of T-cell specialization. It comes to conclusion as belows. 1. In synovium treated with Ulmus davidiana Planchon herbal acupuncture, there was the decrease in MIF mRNA does-dependently. Incase of CIA mice treated with Ulmus davidiana Planchon herbal acupuncture, there were the decrease in the damage in synovium and generation of the MIF which is related to induction of the early RA cytokine and IL-6 proinflammatory cytokine. 2. In case of EL-4 treated with Ulmus davidiana Planchon herbal acupuncture, there were decrease in the manifestation of the IL-2 mRNA, but the increase in the manifestation of the IL-4 does-dependently. 3. In the synovium of CIA mice treated with Ulmus davidiana Planchon herbal acupuncture, there were the decrease in generation of IL-2, IL-12 and CD-28, but the increase in generation of IL-4. These result suggest that Ulmus davidiana Planchon can block the process of the early RA by Inhibiting MIF activation, and mitigate Rheumatoid Arthritis by controlling Tcell specialization.

  • Head of Panax ginseng indicates its growth number of years and it has been widely used for supplying energy to weak person. However, the underlying mechanisms are not sufficiently reported. Thus, we inclined to study head of Panax ginseng in rheumatoid arthritis and inflammation. Rheumatoid arthritis (RA), an organ-specific inflammatory disease of human, is characterized by a chronic and destructive inflammatory reaction and possibly autoimmune in etiology. It is occurred when the synobial membranes of joints and many other tissues of the body is attacked which induces significant health problem in terms of numbers of sufferers (U.S. incidence 23.7/100,000), and the synobial inflammatory is dominated by activated macrophage.

Inhibitory Effect of Coicis Semen Composition on Inflammatory Responses in the Collagen-induced Arthritis Mouse Model

  • This study was peformed to investigate possible mechanisms underlying possible effect of Coicis Semen composition (CSC) on inflammatory diseases using in vivo model of RA in the mice. Results are summarized as follows. In production of inflammatory cytokines, INF-${\gamma}$ in the spleen and IL-6 in the serum were decreased by CSC treatment. TNF-${\alpha}$ in serum was significantly decreased, IL-4 in the spleen was significantly increased by CSC treatment. In production of rheumatoid factors, IgM and IgG were significantly decreased by CSC treatment. The present data suggest that CSC treatment can improve pathological damage by CIA. So we expect that CSC should be used as a effective drugs for not only rheumatoid arthritis but also another autoimmune disease. Therefore we have to survey continuously in looking for the effective substance and mechanism in the future.

Antiarthritic Activity and Inflammatory Mediators Modulation Effect of Traditional Ajmodadi Churna on Arthritis Experimental Model

  • Objectives: The study was designed to evaluate anti-arthritic activity of Ajmodadi Churna (AC) and its effect on Complete freund's adjuvant (CFA)-induced arthritis in Wistar rats. Methods: Arthritis was induced by injecting 0.2 mL CFA into sub plantar surface of left hind paw. Test sample AC-1 and AC-2, 200 and 400 mg/kg, respectively was given to the animals for 21 consecutive days. The increase in swelling was observed after induction of arthritis. The paw edema was measured on 0, 3, 7, 14 and 21 day using Vernier caliper after the induction of arthritis. The collected blood samples further used for the estimation of red blood cells (RBC), white blood cells (WBC), erythrocytes sedimentation rate (ESR), and hemoglobin (Hb), using hematology analyzer. Serum concentration of IL-6 and TNF-α were also measured using rat ELISA kits. Results: Results showed that a significant reduction in paw edema was observed in AC-2 treated rats. The paw edema was restored on day 21 was 4.48 mm for AC-2, which is near to the control group. The arthritis score in treated rats was found to be considerably lower than in the control group i.e. 0.83 for AC-2 and 1.50 for AC-1. A decrease in levels of RBC and hemoglobin were observed in arthritic rats. Inflammation was significantly reduced and serum levels of IL-6 and TNF-α were lowered after treatment with the test drug. Conclusion: It can be concluded from the study that AC possess significant anti-arthritic activity. Furthermore, this condition was linked to a reduction in abnormal humoral immune responses.
