Nitrification and Removal of Pathogenic Microorganisms Indicators by an upgraded RBC (생물막공정을 이용한 질산화 및 병원성 미생물의 제거)

  • Experiments on an upgraded RBC were conducted to investigate its removal efficiencies of $NH_4-N$, coliform group and coliphage in the municipal wastewater treatment. A bench scale RBC, made of stainless mesh media with 5mm high surface protrusions, was fed with settled municipal wastewater containing 13-25mg/l of $NH_4-N$ and 40-75mg/l of TOC at the hydraulic loading of $89l/m^2.d$ and $60l/m^2.d$. It was found that the nitrification effectively occured at bulk liquid TOC concentration of less 10mg/l. The coliform group and coliphage were more than 98% and 90%, respectively.

Study on the Sanitary Indicative Bacteria of Sea Water in Fish Preservatory Tanks during the Mid Summer (여름철 활어조해수(活魚槽海水)의 세균학적(細菌學的)인 연구(硏究))

  • The purpose of the present study is to obtain some fundamental data for hygienic control of sanitary indicative bacteria of sea water in fish preservatory tanks at popular swimming beaches in Busan including Songdo, Kwanganri, Haeundae and Songjeong from July to August in 1982. The geometric mean values of coliform group and fecal coliform MPN (most probable number) of sea water of Kwanganri were 27,300/100ml and 8,700/100ml respectively, which showed the highest population of the study regions. MPN's of the sea water of beaches and fish preservatory tanks showed 21,000/100ml and 8,800/100ml respectively at Kwanganri, which were also the highest numbers. The degree of the hygienic bacteriological populations of both the seawater and that of the fish tanks at a same region showed similar trend. The pollution caused by coliform group generally increased as viable cell counts increased. The types of coliform organisms isolated from seawater in fish presorvatory tanks by IMViC reaction were 42% Escherichia coli group, 2% Citrobacter freundii group, 44% Klebsiella aerogenes group and 12% others.

Impact of Land-based Pollution Sources on Seawater and Shellfish after Rainfall Event in the Jindongman Area (강우 발생에 의한 진동만해역의 육상오염원이 해수 및 패류에 미치는 영향 분석)

  • In this study, we evaluated the impacts of land-based pollution sources on seawater and shellfish in the Jindongman area after 20.5 mm and 90.6 mm rainfall events. We analyzed sanitary indicator microorganisms used in survey management, such as total coliform, fecal coliform, Escherichia coli, and male-specific coliphage in Waste water treatment plant (WWTP), major inland pollution source,s seawater, and shellfish for 4 days after rainfall events. Our results showed that the range of coliform group and fecal coliform was 1.8-49 and <1.8-4.5 MPN (most probable number)/100 mL, respectively, after rainfall events in WWTP discharge water. Furthermore, the radius of the calculated impacted area of major inland pollution sources ranged from 5 to 798 m after 20.5 mm of rainfall and 30 to 1,031 m after 90.6 mm of rainfall. The fecal coliform of seawater at 30 stations in the shellfish growing area and areas adjacent to four stations was <1.8-130 and from <1.8-79 MPN/100 mL, respectively. The E. coli level of shellfish at 7 stations in the shellfish growing area was <18-220 MPN/100 g.

Effects of Fermentation to Improve Hygienic Quality of Powdered Raw Grains and Vegetables Raw Grains and Vegetables Using Lactobacillus sp. Isolated from Kimchi (김치분리균주 Lactobacillus sp.를 Starter로 한 발효생식 제조에서의 위생미생물 살균효과)

  • Improvement of hygienic quality of powdered raw grains and vegetables by fermentation was investigated. Luc-tobacillus sp. isolated from kimchi was used as a starter. The cell counts of coliform group and SS enteric bacteria on the SS agar plate in raw grains and vegetables were 2.3$\times$103 cfu/mL and 8.6$\times$10$^3$ cfu/mL, respectively. The starlet, Lactobacillu sp., reached 10$^{7}$ cfu/mL after 48 hr in fermentation. At that time, the coliform group and enteric bacteria on the SS agar plate were gradually inactivated and eliminated after 60 hr of fermentation. During the fermentation process, pH of the suspension was lowered and acidity increased. Antimicrobial activity of the acidic supernatant of fermented raw grains and vegetal]les against the E. coli sp. and Salmonella sp. was higher than that of lactic acid solution or neutralized supernatant. Therefore, it was considered that antimicrobial effect of the fermented raw grains and vegetal]les might be accelerated by tile synergic effect of acid and bacteriocin, and liquid fermentation of powdered raw grains and vegetables will be effective for inactivation of hygienic microorganisms.

Growth Performance, Meat Quality and Caecal Coliform Bacteria Count of Broiler Chicks Fed Diet with Green Tea Extract

  • This study was conducted to investigate the effect of dietary green tea extract (GTE) on the performance, carcass and gastrointestinal tract (gut) traits, caecal coliform bacteria count, and pH and color (CIE $L^*$, $a^*$, and $b^*$) values of the breast muscle in broilers. A total number of 600 day-old broilers (Ross 308) was allocated to three treatments with four replicates containing 50 (25 males and 25 females) birds. The dietary treatments consisted of the basal diet as the control (0GTE) and diets with GTE at 0.1 (0.1GTE) or 0.2 (0.2GTE) g/kg. Body weights and the feed intake of broilers were measured at 1, 21 and 42 days, the feed intake was measured for different periods and the feed conversion ratio was calculated accordingly. At 42 day four birds per replicate were slaughtered for the determination of carcass and organ weights, caecal coliform bacteria count, and also quality of the breast muscle. The dietary GTE increased the body weight, feed efficiency, carcass weight and dressing percentage and decreased caecal coliform bacteria count of broilers (p<0.05). The 0GTE broilers consumed (p<0.01) less feed than the 0.1GTE birds in the entire experimental period. The relative abdominal fat weight and gut length of broilers in the 0.2GTE were tended to be lower (p<0.07) than those in the 0GTE group. The breast meat from 0.1GTE birds had a lower pH value when compared to that from 0GTE birds. The 0.1GTE broilers had lighter breast meat than 0GTE and 0.2GTE birds. The dietary GTE increased $a^*$ and $b^*$ values of the breast meat. Thus this product appeared to have a measurable impact on CIE color values of the breast meat in broilers. The results of the present study may indicate that the improved production results in the group with added GTE are directly connected with physiological mechanisms such as the regulation of the caecal micro-flora.

Bacteriological Quality of Sea Water at Swimming Beaches in Busan Area (부산시내 해수욕장해수의 세균학적 수질에 관한 연구)

  • This experiment was carried out to evaluate the sanitary quality of sea water at popular swimming beaches such as Haeundae, Kwanganri and Songjeong which are located in the suburbs of Busan Korea. One hundred and seventy five sea water samples were collected from the 25 stations during the both summer seasons in 1979 and in 1980. The sampling stations were established in the above 3 swimming beaches and the estuary of Suyeong river (See Fig.1.). To evaluate the sanitary quality, coliform group, fecal coliform, viable cell count and Vibrio parahaemolyticus were observed. The median value of coliform group MPN of the samples ranged 2,300 to 6,500/100m1 in Kwanganri, 670 to 910/100m1 in Haeundae and 430 to 460/100m1 in Songjeong swimming beach. Geometric mean of fecal coliform MPN was 3,200/100m1 in Kwanganri, 500/100m1 in Haeundae and 360/100m1 in Songjeong swimming beach. Composition of coliform was $23\%$ Escheri-chia coli group, $20\%$ Citrobacter freundii group, $41\%$ Klebsiella aerogenes group and $16\%$others. The density of Vibrio parahaemolyticus in Kwanganri was much higher than those of in other beaches by 5 to 6 times. Plate count of the samples in the estuary of Suyeong river was varied from 300 to $8.5\times10^6/ml$ during the study period. No remarkable difference was observed in bacterial density by tide in swimming beaches but the bacterial density of sea water at ebb tide was obviously higher than those of the samples at flood tide in the estuary of Suyeong river.

Bacterial Flora of East China Sea and Yosu Coastal Sea Areas 2. Horizontal Distributions of Bacteria Isolated from The Sea Area (여수 연안 및 동중국해의 세균상 2. 분리균의 수평분포)

  • The results identified for bacterial flora 174, Vibrio spp.132, and coliform group 183 strains isolated from the areas of last China Sea and Yosu coastal sea during from August 6th. to 14th. 1992 were as follow: 40 strains among the 74 strains of bacteria flora isolated from fast China sea area were Pseudomonas spp.$(54\%)$ and 60 strains among the 100 strains isolated from Yosu sea area were Enterobacteriaceae $(60\%)$. Four strains were Vibrio alginoliticus and one strain of V, parahaemolyticus among 5 strains of genus Vibrio isolated from last China Sea. While 54 strains were V. alginolyticus $(43\%)$ and V, parahaemolyticus $(17\%)$ among 127 strains genus Vibrio isolated from Yosu coastal sea area. Seventy nine strains among the 156 strains of coliform group isolated from Vosu sea area were Escherichia coli I $(51\%)$ and each one strain Citrobacter freundii I and II. 3 strains among 27 strains isolated from last China sea area were E. coli$(11\%)$ and 1 strain of C. freundii I. Coliform group was grouped into 16 types by IMViC system, $44^{\circ}C$, gelatin liquefaction test.

수질 장기관측자료를 활용한 우리나라의 지하수 수질변동 특성

  • Since 1995, MOCT(Ministry of Construction and Transportation) and KOWACO(Korea Water Resources Corporation) have established the National Groundwater Monitoring Network in South Korea and also MOE(Ministry of Environment) has operated Groundwater Quality Monitoring network. Until 2001, 202 monitoring stations by MOCT and 780 monitoring wells by MOE have been constructed, measured groundwater level and analyzed water samples. Groundwater quality analysis has been conducted two times a year during last 6 years for all monitoring wells. The quality data has about 15 components including pH, COD, Count of Coliform group, and etc.. Trend analysis has been peformed for 6 components(Coliform, pH, COD, NO$_3$-N, Cl and EC) of water quality which are analyzed more than 7 times for total monitoring wells. Two test methods have been used ; Sen's test and Mann-Kendall test. These trend tests have been done at the 0.05 significance level. By the result of Sen's test, Count of Coliform group has either upward or downward trends at 4.3 percent of the monitoring points. pH does at 5.6 percent, COD does at 8.6 percent, Nitrate-Nitrogen does at 13.2 percent, Chloride does at 13.4 percent, and. EC does at 11.6 percent of the monitoring points. The exact causes of the groundwater quality trends are difficult to specify. Notable downward trends in nitrate at many monitoring points may be the result of reduction on some contamination sources. Potential causes include diminished agricultural areas, improvements in sewage treatment and a decrease in atmospheric deposition. Increase in chloride at many monitoring points may be the result of increased non-point source pollution such as road salting and runoff from sprawling paved developments and suburbs.

A Microbiological Study on the Tap Water in Seoul (서울시 수도수 중의 위생물학적 조사연구)

  • This study was carried out to investigate Water Temperature, Residual Chlorine, Coliform Groups, and the Standard Plate Counts of Water Supply Areas provided by 7 Water Purification Plant (W.P.P) in Seoul from September 20, 1979 through October 20, 1979. The results were summarized below: 1) The mean water temperature of the 63 Water Samples was 19.8$\circ$C, the mean pH 7.18, and the mean residual chlorine concentration 0.52 ppm by each Water Supply Areas. There is no statistically significant differences between the WPP Areas, but there is significant differences between water supply areas. 2) 30(47.6%) out of the 63 Water Samples were Standard Plate Counts free and 33 Samples (52.4%) were contaminated by Standard Plate Counts. 30 (47.6%) out of 33 samples showed the existence of Standard Plate Counts less than 15 and the other 3 samples 15-30. 3) 2 (3.2%) out of the 63 Samples had the coliform. Those 2 Samples had 2 and 6 coliform group counts per 50ml respectively both of them were 0.1ppm in residual chlorine. 4) There is correlation among Water Temperature, pH, Residual Chlorine, Standard Plate Counts, and Coliform Groups. The Coefficient of Correlation(r) between Water Temperature & Residual Chlorine was 0.147, 0.240 between Water Temperature & Standard plate Counts and 0.215 between pH & Standard Plate Counts. These correlations are statistically no significant, But the correlation of coefficient between pH & Residual Chlorine was -0.291 which is showed significant correlation at p<0.05. The coefficient of correlation between Residual Chlorine & Standard Plate Counts was -0.441 which is showed Negative Correlation Statistically Significant difference at p<0.01.

Sanitary Characteristics of Seawater and Sediments in Tongyeong Harbor (통영항의 해수 및 저질의 위생학적 특성)

  • The bacteriological and physiochemical analysis of sea water and sediments in Tongyeong harbor was conducted to evaluate sanitary conditions. The samples were collected at 8 stations established once a month from June, 2008 to May, 2009. During the study period, the range of temperature was from 6.7 to $25.2^{\circ}C$, transparency ranged from 1.2 to 2.6 m, chemical oxygen demand ranged from 1.90 to 2.92 mg/L, dissolved oxygen ranged from 6.2 to 10.5 mg/L, dissolved nitrogen ranged from 0.052 to 0.098 mg/L, phosphate ranged from 0.044 to 0.065 mg/L, respectively. Seafood, if eaten raw, carries the risk of food poisoning. Seafood poisoning is often cause by pathogenic microorganism originating from fecal contamination, such as Salmonella sp., Shigella sp. and norovirus. Fecal coliforms are an important indicator of fecal contamination. Therefore, data on fecal coliform are very important for evaluating the safety of fisheries in coastal areas. So, we investigated the sanitary indicate bacteria. The coliform group and fecal coliform MPN's of sea water in Tongyeong harbor were ranged from < 1.8~22,000/100 mL (GM 164.9 MPN/100 mL) and < 1.8~7,900 MPN/100 mL (GM 33.7 MPN/100 mL), respectively. Total coliform were detected 97.0% in 96 of samples and 68.9% of total coliforms were fecal coliforms. These results similar to another seawater detection ratio of total coloforms and fecal coliforms. The Vibrios was isolated and identified with VITEK system. Four hundred eighty strains that were obtained from sea water samples in Tongyeong harbor Detection ratio Vibrio alginolyticus, 34.2%, Vibrio parahaemolyticus, 13.8%, Vibrio vulnificus 10.0%, and V. mimicus 12.5% respectively. Vibrio cholerae O1, was not detected. During the study period, the ranges of water content, ignition loss, COD, and acid volatile sulfates in sediments in Tongyeoung harbor were 41.0~57.4%, 7.8~10.5%, 6.51~9.30 mg/g, 0.04~0.09 mg/g, respectively. Heavy metals in sediment of Tongyeoung harbor were Cd, $0.10{\pm}0.05$; Cu, $4.79{\pm}8.20$; As, $1.95{\pm}0.17$; Hg, $0.10{\pm}0.07$; $Cr^{6+}$, $0.34{\pm}0.12$; Zn, $125.33{\pm}16.40$; Ni, $16.43{\pm}1.93$ mg/kg.
