This study was carried out to investigate the bacterial contamination of Ice Tea sold on the l street in Seoul area. For this, study 81 samples were collected on the street from July to September, 1985 and were examined on the following items. 1. Degree of bacterial contamination. 2. The relation of the occurrence of fecal coliform and salmonella. 3. The change of bacterial contamination in Ice Tea against temperature. As the results of this study, the following conclusion were obtained. 1. The mean count of total viable bacteria by standard plate count was $6.5{\times}10^3$/ml, the mean count of total coliform and fecal coliform by MPN method were $3.4{\times}10^2$/100ml, 5.5/100ml and those of fecal streptococci was $3.2{\times}10^2$/100ml. 2. The mean count of Staphylococcus aureus was 10.5/ml, the isolated rate of salmonella was 7.41%. 3. In relation to the occurrence of fecal colfform and salmonella, salmonella isolated that for values above $10^2$ fecal coliform 100ml. 4. In the change of bacterial contamination in Ice Tea against temperature, the number of total coliform and fecal coliform increased at $25{\circ}$C, decreased at $4{\circ}$C, but fecal streptococci increased at $25{\circ}$C and $4{\circ}$C.
A study was conducted to determine contamination status of fish sold at wholesale market in Seoul. A total of 79 samples (35 different kindry fish) were collected from the wholesale market. E. coli and coliform group bacteria were cultured and tested for sensitivity against antibiotics. The results are summarized as follows; 1. E. coli was isolated from 23 out of 79 samples (29.1%), and coliform groups from 53 out of 79 (67.1%). 2. Of coliform group, Citrobacter freundii was the most common and Enterobacter clacae was the next. 3.23 E. coli strains isolated from fishes were resistant to Oxacillin, Erythromycin and Lincomycin, meanwhile 23 E. coli strains were sensitive to Cefoperazone, Ceftazidime, Imipenem, and Ciprofloxacin.
Fecal coliform bacteria are indicators of fecal contamination and of the potential presence of pathogens associated with animal excreta, wastewater, sludge. This study examined the distribution of fecal coliform in sand of the playground at educational facilities and parks in Seoul. Fecal coliform were detected from 61 (22.1%) places out of 276 playgrounds. Escherichia (E) coli (75.4%) was found most frequently, followed by Cronobacter (Cro) sakazakii (8.7%), Klebsiella (K) pneumoniae (7.2%) and Actinobacter baumannii (4.3%). We also tested the bacteria for their susceptibility to 16 antibiotics using disk diffusion method. All the isolates were highly susceptible to cefepime, gentamicin, tetracycline, ciprofloxacin, enrofloxacin, nalidixic acid and sulfamethoxazole/ trimethoprim, but resistant to apramycin and cephalothin. E. coli and Cro. sakazakii were highly resistant to apramycin (100% and 83.3%), followed by cephalothin (51.9% and 50%) and ampicillin (19.2% and 16.7%). K. pneumoniae was highly resistant to ampicillin and apramycin (100%, respectively) but the remaining antibiotics were susceptible.
The aim of this study is to investigate microbial sanitary condition of public baths in Seoul, Korea. A total of 28 water samples were collected from 14 different public baths and sudatoriums. The prevalence of fecal indicator microorganisms such as total coliform, fecal coliform, and Escherichia coli was characterized. In addition, bacteria in water was membrane filtered by 0.45um nitrocellulose membrane, and the filter was analyzed by both cultivation and PCR amplification of partial 16S rRNA gene. The levels of chlorine were measured for each of water samples. More than 40% of 14 collected water samples, the concentrations of total coliform bacteria exceeded the water quality for bath water guideline. There was no significant correlation between chlorine residue and the presence of total coliform. Various microorganisms including pathogenic microorganisms were identified from cultivation and subsequent analysis of 16s rRNA gene sequences. Our results suggest that appropriate hygiene practice and continuous monitoring is needed for reducing health risk associated with public bathhouses.
식용이 간편한 즉석섭취용(ready-to-eat)샐러드 제품의 수요는 나날이 증가하고 있으나 국내 유통 샐러드의 미생물 오염수준은 기준 및 규격이 설정되어있지 않고 그 수준 또한 알려져 있지 않다. 따라서 소비가 집중되어 있는 대도시 지역을 중심으로 백화점, 대형할인점 또는 패밀리레스토랑 등에서 판매되는 즉석섭취용 샐러드 제품 120건을 대상으로 하여 coliform bacteria, E. coli, E. coli O157:H7, Staphylococcus aureus, Salmonella spp. Listeria monowtogenes 및 Bacillus cereus의 분리시험을 실시하고 일부 제품을 대상으로 일반세균수 및 세척효과를 조사하였다. 제품 120건 중 총 73건에서 coliform bacteria 등이 검출되어 60.8%의 검출율을 보였다. 5. coli가 17건, Staphylococcu aureu가 3건, Salmonella spp.가 1건에서 각각 검출되었고 기타 coliform bacteria는 59건에서 검출되었다. 샐러드는 양식방법에 따라 유기농샐러드와 비유기농샐러드로 나누었고, 종류는 야채로 구성된 야채샐러드와 튀긴 닭 등이 첨가된 혼합샐러드로 나누었으며 판매형태에 따라 완포장제품과 샐러드바 제품으로 나누었다. 양식방법 및 포장방법과 미생물오염과의 통계적 연관성은 없는 것으로 나타났다. 그러나 분리된 균주 수에 있어서는 야채샐러드와 혼합샐러드 간에 통계적으로 유의한 차를 보여 야채샐러드의 경우 좀 더 다양한 균종이 분리되는 것으로 나타났다 (p<0.005). E. coli는 일반 재배 야채샐러드의 경우 90건 중 10건에서 분리되었고 유기농재배 야채샐러드 의 경우 30건 중 7건에서 분리되었다. 식중독 원인균은 비유기농 재배 야채샐러드 4건에서 Staphylococcus aureu가, 1건에서 Salmonella arixonia가 검출되었으나 유기농재배 야채에서는 검출되지 않았다. 널리 판매되는 일부 제품 4종을 5회에 걸쳐 검사한 결과 총 호기성 세균수는 평균 4.8$\pm$0.19 log$_{10}$ cfu/g으로 조사되었다. 같은 제품을 생리식염수와 야채용세척제를 이용해 3분간 침지 후 세척효과를 본 결과 생리식염수는 5회, 야채용세척제는 1회 세척 시 95.5%의 세척효과를 내는 것으로 나타났다.다.
순환여과수조에 있어 수질 오염의 지표로서 대장균군의 수적인 변동을 측정하였다. 세균의 오염은 주로 어류의 먹이와 배설물 그리고 새로 교환되는 사육수의 기오염도에 원인이 있었다. 수조내의 세균량은 수질의 물리화학적인 요인에 의해서 조절되었으며, 이러한 요인들 중에서는 pH가 주된 것이었다. 수온 $20^{\circ}C$에 pH가 $5\~6$ 이하로 낮아질 경우에는 세균의 급격한 번식은 없었다. 수온이 $30^{\circ}C$ 이상 일 때 세균의 성장은 pH에 관계없이 현저하게 감소하였으며, 세균군은 그 이하의 온도에서 활발히 성장하는 것처럼 보였다.
This study was performed to compare finished water quality among three different processes. A detailed assessment of performance was carried out during the five months of operation. Finished water quality was evaluated on the basis of parameters such as Dissolved organic carbon (DOC), $UV_{254}$ absorbance, haloacetic acid formation potential (HAAFP), geosmin, 2-methylisoborneol (2-MIB), heterotrophic bacteria and total coliform bacteria. The treatment processes were Process 1 (coagulation-flocculation-sedimentation-sand filtration-ozone-GAC), Process 2 (coagulation-flocculation-sedimentation -microfiltration-ozone-GAC), and Process 3 (coagulation-flocculation-sedimentation- sand filtration-GAC), compared side by side in the pilot testing. Process 2 was found to have better removal efficiency of DOC, $UV_{254}$ absorbance, HAAFP and heterotrophic bacteria in comparison with process 1 and process 3 under identical conditions. Geosmin, 2-MIB and total coliform bacteria were not detected in finished water from each process.
Objectives: To estimate the microbial contaminant load discharged from livestock farms, we randomly selected livestock farmers of cattle, swine, and fowl and collected bacterial strains from domestic animals' feces and compost samples. Recently, as multi-antibiotic-resistant bacteria and super bacteria showing resistance to a variety of antibiotics have been reported one after another, the ecological and health hazard of antibiotic-resistant bacteria is emerging as an important issue. Methods: Monitored indicator microorganism constituents were totak coliform (TC), fecal coliform (FC), and aerobic bacteria. The multi-antibiotic-resistant bacteria were identified from investigated indicator microorganisms by 16S rRNA sequencing. Results: By microbiological analysis, the largest population of aerobic bacteria ($1.5{\times}10^5$ CFU/g) was found in cattle fecal compost, and total coliforms ($1.1{\times}10^7$ CFU/g) and fecal coliforms ($1.0{\times}10^5$ CFU/g) were found primarily in swine fecal compost, while the lowest population was found in fowl fecal compost. Among the 67 strains separated from aerobic bacteria, five strains expressing high antibiotic resistance were selected in each sample. We found the multi-antibiotic resistant strains to be Shigella boydii, Staphylococcus lentus, Acinetobacter sp. and Brevibacterium luteolum. Conclusions: These results suggest that increasing numbers of multi-antibiotic-resistant bacteria in the environment have a close relation to the reckless use of antibiotics with livestock.
This study evaluated the bacteriological safety of seawater and abalone produced in Uisin-myeon, Jindo-gun from September 2020 to December 2022 and calculated the effect of inland pollution sources on seawater and abalone. We surveyed 33 stations for seawater, 4 stations for abalone, and 7 stations for discharge water of major inland pollutants. We analyzed the coliform group, fecal coliform and Escherichia coli (E. coli) as bacteria indicators. In 924 seawater samples, the geometric mean of the coliform group and fecal coliform were <1.8-5.6 MPN/100 mL and <1.8-2.7 MPN/100 mL, respectively. The estimated 90th percentile range for the coliform group and fecal coliform were <1.8-42 MPN/100 mL and <1.8-8.4 MPN/100 mL, respectively. The fecal coliform and E. coli levels for 107 abalone were <18-460 MPN/100 g and <18-78 MPN/100 g respectively. Moreover, the analyzed standard plate count was <30-7,700 CFU/g. The fecal coliform level and diffusion area in 7 inland pollutants were <1.8-3,300,000 MPN/100 mL and 2-2,500 m, respectively. The bacteriological safety of seawater and abalone in Uisin-myeon, Jindo-gun, was demonstrated to be a designated area according to Korean standards, a conditionally approved area per US standards, and Class A according to EU standards.
To secure the biological safety of live fish and shellfish for raw consumption, the germicidal effects of electrolyzed seawater were evaluated. Upon direct exposure to electrolyzed seawater, coliform group bacteria were killed and decreased to undetectable levels after 1 day. The physicochemical characteristics of the seawater were stable during the test period. A byproduct of chlorine disinfection, trihalomethane, was not generated by the electrolysis of seawater. Vibrio parahaemolyticus infection in a live fish was effectively resolved by electrolyzed seawater and became undetectable after 12-36 h of treatment. Bioaccumulation of coliform group and fecal coliform bacteria in live oysters Crassostrea gigas was removed within 18 h of treatment. This study demonstrated that electrolyzed seawater is an effective and safe germicidal agent for the traditional retail market and can help to prevent outbreaks of foodborne disease associated with the consumption of raw fish and shellfish.
