In this paper, experimental and numerical study on a combined sheet metal forming and plate forging of a seal part of a passenger car's hub bearing is conducted to develop the new process of which target is to remove machining process by plate forging and to achieve near-net shape manufacturing. The previous process of a sheet metal forming inevitably needed a machining process for making stepped sheet after conventional sheet metal forming in a progressive way. The stepped sheet is intended to be formed by plate forging in this study. Through the systematic way of developing the combined forming process using solid elements based-elastoplastic finite element method (FEM), several conceptual designs are made and an optimized process design in terms of geometric dimensioning and tolerance of straightness of the thin part is found, which is exposed to bending in metal forming of axisymmetric part. The predicted straightness measured by the slope angle of the tilted thin region is compared with the experimental straightness, showing that they are in a good agreement with each other. Through this study, a systematic approach to optimal process design, based on elastoplastic FEM with solid elements, is established, which will contribute to innovating the conventional small-scaled sheet metal forming processes which can be dealt with by solid elements.
A cDNA clone encoding phytocystatin was isolated from Brassica rapa seedlings, through rapid amplification of cDNA ends (RACE). This gene (name as Brcpi1; GenBank accession no.: EF079953) had a total length of 881 bp with an open reading frame of 609 bp, and encoded predicted polypeptide of 203 amino acid (aa) residues including a putative N-terminal signal peptide. Other relevant regions found its sequence included the G and PW conserved aa motifs, and the consensus LARFAV sequence for phytocystatins and the reactive site QVVAG. The BrCPI1 protein shared 95, 94, 81, 80 and 78% identity with other CPI proterins isolated from Brassica oleracea (BoCPI-1), Arabidopsis thaliana (AtCY SB), Glycine max (GmCPI), Oryza sativa (OsCYS-2) and Zea may (ZmCPI) at amino acid level, respectively. Southern blot analysis showed that Brcpi1 was a low copy gene. Expression pattern analysis revealed that Brcpi1 was a tissue-specific expressing gene during reproductive growth and strongly expressed at mature seedling stages. Furthermore, overexpression of Brcpi1 in transgenic Arabidopsis was enhanced tolerance to salt and cold stresses. Meanwhile the juvenile seedling of Brcpi1 transgenic plants was not affected by various concentrations ABA in MS medium. Taken together, the results showed that Brcpi1 functioned as a cysteine protease inhibitor and it exhibited a protective agent against diverse types of abiotic stress, which induced this gene in a tissue- and stress-specific manner.

월동기간 동안 zoysiagrass[Zoysia matrella(L.) Merr]와 creeping bentgrass(Agrostis palustris Hunds)의 저온 스트레스에 대한 탄수화물 대사 반응을 구명하기 위해 11월부터 이듬해 4월까지의 식물체내의 탄수화물 대사산물의 변화를 비교 분석하였다. Creeping bentgrass의 녹색도와 뿌리 성장은 온도변화와 거의 평행하게 변화하였으나 zoysiagrass는 뿌리 성장에 거의 변화가 없이 3월 말까지 담황색을 나타냈다. 11월부터 이듬해 1월까지 zoysiagrass에서 glucose와 sucrose 함량은 크게 높았고, 1월에서 4월까지 starch를 제외한 모든 탄수화물 화합물들의 감소의 정도가 현저하게 낮았다. Reeping bentgrass에서 fructan의 depolymerization이 2월 이후 단당류와 이당류의 감소와 일치했으나 zoysiagrass에서는 starch 가수분해가 11월부터 일찍이 시작되었다. 이상의 결과들은 creeping bentgrass와 비교 했을때 zoysisagrass 뿌리의 높은 비구조적 탄수화물의 농도가 저온 내성과 밀접한 관계가 있음을 잘 나타내어 주고 있다.
External morphology of three species of damselfishes, Chromis notata, C. analis and C. fumea were studied based on the samples collected from the Korean waters from 1993 to 1994. In morphology of the black spot at base of pectoral fins, C. notata has a large triangluar spot at Cheju population and a small circular spot at Sin - am. In the length of pelvic fin, C. notata of Korea can be classified into 3 groups. First group has a pelvic fin that extends to the origin of anal fin, second extends to the anus, third not extend to the anus. C. analis has higher body depth than C. notata and C. fumea in % of standard length. C. fumea has a minute serrate preopercle and no black spot at base of pectoral fins. In distribution, C. notata has a more widly distriubtion area than C. analis and C. fumea. Therefore, C. notata has more tolerance of cold waters than C. analis and C. fumea.
The aim of this study is selection of the male-sterile plant for inhibiting transgene flow through gamma-irradiation ($^{60}Co$) at the pollination and fertilization cycle of herbicide-tolerant genetically modified (GM) zoysiagrass (Zoysia japonica Steud.). High frequencies of plant mutations were obtained about 18% from $M_1$ generation at the doses (10 to 50 Gy). We also found that some $M_1$ plants showed male-sterile plants using de-husked seeds and comparison of stainable pollen using $KI-I_2$ solution. Besides the effects of irradiation on pollination and fertilization cycle, various other mutations like dwarf, cold tolerance, increasing grains and mass were observed. Four of dwarfism plants were selected through comparison of morphological characteristic between control and mutants during 4 years. These results demonstrated that the gamma-irradiation on pollination and fertilization cycle is very effective to induce the various mutations, and the male-sterile mutants are useful for controlling transgene flow and developing of high quality turfgasses.
Korean ginseng (Panax ginseng) has long been cultivated as an important economic medicinal plant. Owing to the seasonal and long-term agricultural cultivation methods of Korean ginseng, they are always vulnerable to various environmental stress conditions. ABSCISIC ACID (ABA) is an essential plant hormone associated with seed development and diverse abiotic stress responses including drought, cold and salinity stress. By modulating ABA responses, plants can regulate their immune responses and growth patterns to increase their ability to tolerate stress. With recent advances in genome sequencing technology, we first reported the functional features of genes related to canonical ABA signaling pathway in P. ginseng genome. Based on the protein sequences and functional genomic analysis of Arabidopsis thaliana, the ABA related genes were successfully identified. Our functional genomic characterizations clearly showed that the ABA signaling related genes consisting the ABA receptor proteins (PgPYLs), kinase family (PgSnRKs) and transcription factors (PgABFs, PgABI3s and PgABI5s) were evolutionary conserved in the P. ginseng genome. We confirmed that overexpressing ABA related genes of P. ginseng completely restored the ABA responses and stress tolerance in ABA defective Arabidopsis mutants. Finally, tissue and age specific spatio-temporal expression patterns of the identified ABA-related genes in P. ginseng tissues were also classified using various available RNA sequencing data. This study provides ABA signal transduction related genes and their functional genomic information related to the growth and development of Korean ginseng. Additionally, the results of this study could be useful in the breeding or artificial selection of ginseng which is resistant to various stresses.

Objectives: Insulin resistance (IR) is major cause of type 2 diabetes (T2D), and adipokines (e.g., adiponectin, leptin, and resistin) play an important role in insulin sensitivity. Medicinal plants are frequently used for T2D treatment. This study investigates the effect of Artemisia annua L. (AA) extracts on adipokines in mice with high-fat-diet (HFD)/streptozotocin (STZ)-induced T2D. Methods: We divided 60 mice into 12 groups (n = 5 per group): control, untreated T2D, treated T2D, and 9 other groups. T2D was induced in all groups, except controls, by 8 weeks of HFD and STZ injection. The treated T2D group was administered 250 mg/kg of metformin (MTF), while the nine other groups were treated with 100, 200, and 400 mg/kg of hot-water extract (HWE), cold-water extract (CWE), and alcoholic extract (ALE) of AA (daily oral gavage) along with 250 mg/kg of MTF for 4 weeks. The intraperitoneal glucose tolerance test (IPGTT) was performed, and the homeostasis model assessment of adiponectin (HOMA-AD) index and blood glucose and serum insulin, leptin, adiponectin, and resistin levels were measured. Results: Similar to MTF, all three types of AA extracts (HWEs, CWEs, and ALEs) significantly (p < 0.0001) decreased the area under the curve (AUC) of glucose during the IPGTT, the HOMA-AD index, blood glucose levels, and serum insulin, leptin, and resistin levels and increased serum adiponectin levels in the MTF group compared to the T2D group (p < 0.0001). The HWEs affected adipokine release, while the CWEs and ALEs decreased leptin and resistin production. Conclusion: Water and alcoholic AA extracts have an antihyperglycemic and antihyperinsulinemic effect on HFD/STZ diabetic mice. In addition, they decrease IR by reducing leptin and resistin production and increasing adiponectin secretion from adipocytes.

The amount of required forage is increasing by 20% every year in South Korea, but the cultivation area for forage production is limited. The yield ability of triticale forage is the highest among the winter forage crops including rye and the crop has cold tolerance within the average low temperature of -10℃ in January. Therefore, this study analyzed effects of sowing and harvesting times on feed value and quality for efficiently using and supplying triticale as livestock feed. Seed of the triticales, 'Joseong' was sown in 2021 fall (October) and 2022 spring (March). The triticales were harvested according to growth stages: seedling stage, booting stage, heading stage, 10 days after heading, and 20 days after heading. Moisture contents of each harvested triticales were adjusted to about 60%, and then the triticales were fermented for 40 days at room temperature under anaerobic conditions as silage. We have analyzed pH and organic acid to determine the feed value and quality of each silage. The contents of lactic acid in silage ofthe triticale harvested at the seedling stage of both fall and spring-sown (1.61%, 1.63%) were the highest among all of the silages; the booting stage (0.75%, 1.33%), the heading stage (0.50%, 0.69%), 10 days after the heading stage (0.31%, 0.42%), and 20 days after heading stage (0.22%, 0.40%). Such as the contents of lactic acid in the silages, and the pH value of the silages The pH value in both the fall- and spring-sown became lower as the triticale was grown up: seedling stage (7.05, 6.85), booting stage (6.21, 6.75), heading stage (6.18, 6.28), 10 days after heading stage (6.22, 6.17), and 20 days after heading stage (6.15, 5.81). Taken together, the results showed that the feed value and quality of triticale silage were more affected by harvesting time than sowing time.

Plant reproduction associated with crop yields is highly vulnerable to global climate change components such as high and cold temperatures. The objectives of this study were to determine the effects of season-high exposure to temperature treatments in pollen morphology on Adzuki bean (V. angularis) and Mung bean (V. radiate). V. angularis and V. radiata were treated at high temperatures in the high temperature gradient greenhouse designed to cause temperature deviation. The pollen shapes treated at high temperature were compared by an electron scanning microscope. As a result, it was confirmed that the number of abnormal pollens morphology at high temperature was the least in V. radiata, and V. angularis was vulnerable to high temperatures. Also, it was found that the number of abnormal pollen morphology atT4 (Con +5~6℃) varied according to the cultivars of V. angularis. Therefore, the differences in Vigna species or cultivars with thermo-tolerance in pollen morphology to high temperature are projected to occur in the changeable future climate.
Our group had isolated Bifidobacterium breve strain BS2-PB3 from human breast milk. In this study, we sequenced the whole genome of B. breve BS2-PB3, and with a focus on its safety profile, various probiotic characteristics (presence of antibiotic resistance genes, virulence factors, and mobile elements) were then determined through bioinformatic analyses. The antibiotic resistance profile of B. breve BS2-PB3 was also evaluated. The whole genome of B. breve BS2-PB3 consisted of 2,268,931 base pairs with a G-C content of 58.89% and 2,108 coding regions. The average nucleotide identity and whole-genome phylogenetic analyses supported the classification of B. breve BS2-PB3. According to our in silico assessment, B. breve BS2-PB3 possesses antioxidant and immunomodulation properties in addition to various genes related to the probiotic properties of heat, cold, and acid stress, bile tolerance, and adhesion. Antibiotic susceptibility was evaluated using the Kirby-Bauer disk-diffusion test, in which the minimum inhibitory concentrations for selected antibiotics were subsequently tested using the Epsilometer test. B. breve BS2-PB3 only exhibited selected resistance phenotypes, i.e., to mupirocin (minimum inhibitory concentration/MIC >1,024 ㎍/ml), sulfamethoxazole (MIC>1,024 ㎍/ml), and oxacillin (MIC >3 ㎍/ml). The resistance genes against those antibiotics, i.e., ileS, mupB, sul4, mecC and ramA, were detected within its genome as well. While no virulence factor was detected, four insertion sequences were identified within the genome but were located away from the identified antibiotic resistance genes. In conclusion, B. breve BS2-PB3 demonstrated a sufficient safety profile, making it a promising candidate for further development as a potential functional food.
