This experiment was carried out to breed the cold-tolerant variety of Italian ryegrass (Lolium multiflorum Lam.) at the Grassland and Forage Crops Division, National Livestock Research Institute, RDA at Suwon, Korea, from 1999 to 2005. The new variety 'Kogreen' is a diploid variety with a green leaf color and had semi-erect habit in late autumn and erect growth habit in early spring. The heading date of 'Kogreen' was on May 4, and for 'Florida 80' was May 2, both are considered as early-heading variety. 'Kogreen' was wider by 0.7mm in flag leaf width, longer by 2 cm in flag leaf length and shorter by 1cm in plant length than the control variety, Florida 80. Stem width of 'Kogreen' was similar to that of 'Florida 80' and 'Hwasan 101'. cold tolerance of 'Kogreen' was better than that of 'Florida 80'. Dry matter(DM) yield (12,884kg/ha) of 'Kogreen' was 3% less than that of 'Florida 80'. in vitro dry matter digestibility was 74.7% and total digestible nutrient was 65.1%, which are 3.6% and 1.5% higher than those of 'Florida 80', respectively. Acid detergent fiber and neutral detergent fiber of 'Kogreen' were 30.1% and 54.1% which are 1.9% and 1.3% lower than those of 'Florida 80', respectively.

This experiment was carried out to breed a cold-tolerant variety of Italian ryegrass (Lolium multiflorum Lam.) in Grassland and Forage Crops Division, National Institute of Animal Science, RDA, Suwon from 1999 to 2006. ew variety, Kowinmaster as a diploid variety was light-green in leaf color and had medium and semi-erect growth habit in late autumn and early spring, respectively. Kowinmaster was 13th May in heading date as a medium-maturing variety. Especially, Kowinmaster was similar to Florida 80 in flag leaf width, longer 3cm in flag leaf length and longer 6cm in plant length than those of control variety, Florida 80, respectively. Stem width of Kowinmaster was similar to that of Florida 80. Cold tolerance of Kowinmaster was better than that of Florida 80. Dry matter yield (9,521 kg/ha) of Kowinmaster was 2% more than that of Florida 80. Feed value of Kowinmaster was 12.1% in crude protein, 70.9% in In vitro dry matter digestibility and 32.9% in acid detergent fiber that were similar to those of Florida 80, respectively, but in neutral detergent fiber, Kowimaster was 56.0% that was 1% lower than that of Florida 80.
To understand the responses of grapevines in response to cold stress causing the limited growth and development, differentially expressed genes (DEGs) were screened through transcriptome analysis of shoots from 2 grapevine cultivars ('Campbell Early' and 'Muscat Baily A') kept at -$2^{\circ}C$ for 4 days. In gene ontology analysis of DEGs from 'Campbell Early', there were 17,424 clones related with biological process, 28,954 with cellular component, and 6,972 with molecular function genes in response to freezing temperature. The major induced genes included dehydrin xero 1, K-box region and MADS-box transcription factor family protein, and MYB domain protein 36, and inhibited genes included light-harvesting chlorophyll B-binding protein 3, FASCICLIN-like arabinoogalactan 9, and pectin methylesterase 61 in 'Campbell Early' grapevines. In gene ontology analysis of DEGs from 'Muscat Baily A', there were 1,157 clones related with biological process, 1,350 with cellular component, and 431 with molecular function gene. The major induced genes of 'Muscat Baily A' included NB-ARC domain-containing disease resistance protein, fatty acid hydrozylase superfamily, and isopentenyltransferase 3, and inhibited genes included binding, IAP-like protein 1, and pentatricopeptide repeat superfamily protein. All major DEGs were shown to be expressed differentially by freezing temperature in real time-PCR analysis. Protein domain analysis using InterPro Scan revealed that ubiquitin-protein ligase was redundant in both tested grapevines. Transcriptome profile of shoots exposed to cold can provide new insights into the molecular basis of tolerance to low-temperature in grapevines, and can be used as resources for development new grapevines tolerant to coldness.

This experiment was carried out to breed the cold-tolerant variety of Italian ryegrass (Lolium multiflorum Lam.) in Grassland and Forage Crops Division, National Livestock Research Institute, RDA, Suwon from 1997 to 2002. Hwasan 104 as a tetraploid variety was dark green in leaf color and had semi-prostrate and medium growth habit in late autumn and early spring, respectively. Hwasan 104 was 19th May in heading date as a late-heading variety. Especially, Hwasan 104 was broader in flag leaf and longer 7 cm in plant length, and thicker in stem width than those of control variety, Hwasan 101. Cold tolerance of Hwasan 104 was better than that of Hwasan 101 registered as a cold-tolerant variety. Dry matter (DM) yield of Hwasan 104 was $5\%$ more than DM 9,348 kg/ha of Hwasan 101. In vitro dry matter digestibility and total digestible nutrient of Hwasan 104 were 74.3 and $63.1\%$ which are 3.3 and $1.5\%$ lower than those of Hwasan 101, respectively. Acid detergent fiber and neutral detergent fiber of Hwasan 104 were 32.6 and $55.8\%$ which are 1.9 and $2.4\%$ higher than those of Hwasan 101, respectively.
In order to obtain the basic data for clarifing the mechanism of cold tolerance in crops, we analyzed various biochemical changes according to the Uniconazole treatment in Brassica seedling. Peroxidase activity in the root fraction of Brassica seedling was about 3 to 4 times higher than that in hypocotyl fraction, while catalase activity in those fractions showed opposite trend to the peroxidase activity. The content of hydrogen peroxide in root fraction was higher than that of hypocotyl fraction as being a reciprocal proportion with catalase activity. Especially in all fractions, peroxidase· activity of 'Sandongchae' (B. campestris) seedling, known as cold tolerant, was two-fold higher than that of cold sensitive rape(B. napus). The elongation rate of hypocotyl after germination was faster in B. napus than in B. campestris. The application of Uniconazole at 0.3 to 1.0 ppm to B. napus suppressed 43 to 46% of hypocotyl elongation and increased 65 to 73% of peroxidase activity in hypocotyl fraction. The shortening rate of hypocotyl length due to Uniconazole treatment was positively correlated with the increasing rate of peroxidase activity in hypocotyl fraction. Superoxide dismutase was not induced upon Uniconazole treatment and has only 3 isozymes in any fractions. Its activity was observed in the order of cotyledon>root>hypocotyl fraction.
In 1980, rice was considerably damaged by abnormal low temperature. In this paper, to determine the effect of low temperature on the growth and nutrition of rice, the experiment were carried out: varietal response to low temperature in the region with different elevations. Regional differences of heading response to low temperature were observed among varieties. The difference of days between regions were bigger in tongil lines than Japonica lines. Especially, Milyang 42 and Hangangchalbeo might belong the cold suceptible group, since the varieties were severely delayed their heading in the high mountainous region as comparred to plain region. The delay of heading with low temperature was brought out grain sterility, and fertility and ripening ratio is influenced cold tolerance and elevations, and it's decreased yield. Varieties with higher grain sterility by low temperature have higher total nitrogen content, but tended to have lower potassium and phosphate contents in the flag leaf. High content of total nitrogen, low contents of potassium and silicate were observed in the sterilized grain chaff an the opposite result were noted in the ferilized grain chaff at the ripening stage. The results reveal that the balance of these mineral element may play an important role in ripening and possibly cold tolerance.
Background: Haenyeo is a woman who has the job of collecting seafood in the Jeju Sea at an average temperature of 13℃-14℃. The purpose of this study was to examine the cold acclimatization and occupational characteristics of Haenyeo through biomarkers such as orexin and irisin related to heat generation in the body. Methods: Twenty-one Haenyeo and 25 people with similar age, body type, and body mass index were selected as the control group (Control G). In the cold exposure experiment, a climate chamber was set to 5℃ and both feet were immersed in a 15℃ water tank for 30 minutes. Tympanic temperature (Tty) and skin temperature (Tsk) were measured, and the mean body temperature (mTb) was calculated. Blood samples were collected before and immediately after the examination. Orexin and irisin levels were analyzed. Results: Orexin levels were elevated after cold stimulation from 12.17 ± 4.44 to 12.95 ± 4.53 ng/mL (Haenyeo group [Haenyeo G], p < 0.01) and 10.37 ± 3.84 to 11.25 ± 4.02 ng/mL (Control G, p < 0.001). Irisin levels were elevated after cold stimulation from 4.83 ± 2.28 to 5.36 ± 2.23 ng/mL (Haenyeo G, p < 0.001) and 3.73 ± 1.59 to 4.18 ± 2.04 ng/mL (Control G, p < 0.001). The difference between Haenyeo G and Control G values in orexin and irisin appears not only in pre-exposure but also in post-exposure (p < 0.05). Conclusions: Our experimental results suggest that Haenyeo G were relatively superior in cold tolerance to Control G under cold exposure conditions. Haenyeo's cold acclimatization is due to the basic differences in pyrogens regarding body temperature control such as orexin and irisin. This means that Haenyeo are advantageous for cold survival.

The IGRINS (Immersion GRating INfrared Spectrometer) is a high resolution wide-band infrared spectrograph developed by Korea Astronomy and Space Science Institute (KASI) and the University of Texas at Austin (UT). Immersion grating is a key component of IGRINS, which disperses the input ray by using a Silicon material with a lithography technology. Opto-mechanical mount for the immersion grating is important to keep the high spectral resolution and the optical alignment in a cold temperature of $130{\pm}0.06K$. The optical performance of immersion grating can maintain within the de-center tolerance of ${\pm}0.05mm$ and the tip-tilt tolerance of ${\pm}1.5arcmin$. The mount mechanism utilizes the flexure and the kinematic support design to satisfy the requirement and the operation condition. When the IGRINS system is cooled down to a cold temperature, three flexures compensate the thermal contraction stress due to the different material between the immersion grating and the mounting part(Aluminum 6061). They also support the immersion grating by an appropriate preload. Thermal stability is controlled by a copper strap with proper dimensions and a heater. Generally structural and thermal analysis was performed to confirm the mount mechanism. This talk presents the opto-mechanical mount design of the immersion grating of IGRINS.

The diamondback moth, Plutella xylostella, overwinters in some protected areas in Korea. Using a sex pheromone trap, the adults were monitored since the occurrence of the overwintering populations. In Andong, P. xylostella exhibited four adult peaks in a year. Biological characters, such as cold tolerance, insecticide susceptibility, and developmental rate, were analyzed and showed a significant variation among different local overwintering populations. Population genetic variation was assessed with molecular markers, in which the initial high genetic variation among the overwintering populations decreased with the progress of seasons. These results suggests that there may be a significant migration of P. xylostella to decrease the genetic variation among the different local populations that are different in biological characters.
OsDREB1D, a special DREB (dehydration responsive element binding protein) homologous gene, whose transcripts cannot be detected in rice (Oryza sativa L), either with or without stress treatments, was amplified from the rice genome DNA. The yeast one-hybrid assay revealed that OsDREB1D was able to form a complex with the dehydration responsive element/C-repeat motif. It can also bind with a sequence of LTRE (low temperature responsive element). To analyze the function of OsDREB1D, the gene was transformed and over-expressed in Arabidopsis thaliana cv. Columbia. Results indicated that the over-expression of OsDREB1D conferred cold and high-salt tolerance in transgenic plants, and that transgenic plants were also insensitive to ABA (abscisic acid). From these data, we deduced that this OsDREB1D gene functions similarly as other DREB transcription factors. The expression of OsDREB1D in rice may be controlled by a special mechanism for the redundancy of function.
