Bombesin Immunorectivtiy in Suprachiasmatic Nucleus in Rat and Mongolian Gerbil after Colchicine Treatment (Colchicine 투여 후 흰쥐와 모래쥐 시각교차위핵내 bombesin에 대한 면역조직화반응)

  • This experiment was carried out to investigate the bombesin immunoreactivity in suprachiasmatic nucleus in rat and Mongolian gerbil hypothalamus after colchicine treatment and analyze the morphological difference between rat and Mongolian gerbil which is focused for experimental animal model of neuronal and circulatory diseases. The results were as followings. 1. The shape of suprachiasmatic nucleus was triangle in rat, but oval or kidney-shape in Mongolian gerbil 2. The suprachiasmatic nucleus devided into ventrolateral portion and dorsomedial portion in rat, but dorsolateral portion and ventromedial portion or superior portion and inferior portion in Mongolian gerbil. 3. The area of suprachiasmatic nucleus of rat was greater than one of Mongolian gerbil. 4. The bombesin immunoreactivity showed after colcichine treatment in rat and Mogolian gerbil suprachiasmatic nucleus. 5. The bombesin immunoreactivity was stronger in ventrolateral portion than in dorsomedial portion of suprachiasmatic nucleus in rat, but in ventromedial or inferior portion than in dorsolateral or superior portion of suprachiasmatic nucleus in Mongolian gerbil. 6. The bombesin immunoreactivity showed at the oval, ellipsoid or triangular neurons and varicose nerve terminals in ventrorateral portion of rat, and only nerve terminals in dorsomedial portion of rat suprachiasmatic nucleus. But the bombesin immunoreativity didn't show at neurons of Mongolian gerbil suprachiasmatic nucleus.

Induction of Valiant of Cyrtomium caryoptideum var. coreanum Nakai by Chemical Mutagenesis In vitro and RAPD Analysis (기내에서 화학돌연변이원 처리에 의한 참쇠고비의 변이주 유기 및 RAPD 분석)

  • With the aim of inducing mutation in fern Cyrtomium caryoptideum var. coreanum, rhizome segments of In vitro-grown cultures were treated with chemical mutagens such as EMS, NMU and colchicine. Based on regeneration ratios, sensitivities for each treatments were assessed and also optimum treatment condition of each mutagens was explored. Optimum concentration for EMS treatment was considered to be 20 to 50mM and for NMU 5 to 10mM. NMU was found to be more effective in inducing chlorophyll and morphological variations than EMS. The RAPD were performed to check the genetic modification of phenotypical variants. As a result, polymorphic DNA band patterns between wild type and variants were observed by two 10-mer primers.

  • Familial Mediterranean fever (FMF) is the most common Mendelian autoinflammatory disease, characterized by uncontrolled activation of the innate immune system that manifests as recurrent brief fever and polyserositis (e.g., peritonitis, pleuritic, and arthritis). FMF is caused by autosomal recessive mutations of the Mediterranean fever gene, MEFV which encodes the pyrin protein. Although FMF predominantly affects people from Mediterranean and Middle Eastern ethnic origins, 3 cases of FMF have been reported in Korea since 2012. We report another case of FMF in Korea in which the patient presented with a month-long fever without serositis. After treatment with colchicine was initiated, the patient's symptoms quickly subsided. The response to colchicine was helpful for diagnosis. We compare the FMF genotypes in Korea with in other countries. Studying FMF cases in Korea will help establish the best MEFV exons to use for screening and diagnosis of Korean FMF.

Induction and RAPD Analysis of Mutant Plants by Chemical Mutagens in Gentiana axillariflora Leveille (큰용담 (Gentiana axillarifloa L,) 기내배양에서 화학돌연변이원 처리에 의한 돌연변이주 유기 및 RAPD 분석)

  • In order to induce the mutants of Gentiana axillariflora Leveille, nodes were cultured on Schenk and Hilderbrandt (SH) medium containing TDZ 2 mg/L, BAP 2mg/L, GA3 0.5 mg/L and 0.1 mg/L NAA and each mutagen of ethylmethanesulfate (EMS), colchicine, N-methyl-N-nitrosourea (MNU), and sodium azide (NaN$_3$) through filtration. Comparision of morphological characteristics and survival rate in each mutant plants differed depending on mutagen sources and their concentrations. When EMS were treated on nodes, the regenerated plants was thin and albino, regenerated shoots appeared 'erectoides type' and get twisted. The case of colchicine were treated on nodes, the survival rate was from 84% to 97% at ail concentration after 30days but the rate of survival was decreased about 50% at 200 $\mu$M after 60days. The treatment of NaN$_3$200 $\mu$M was not survived. The survival rate was extremely decreased in MNU treatment at 500$\mu$M, according to concentrations two types of leaf characteristic were obtained. Type I of leaf characteristic was modified from oblanceolate to oboid at leaf shape and type II of leaf characteristic was modified from light green to dark violet at leaf color. RAPD analysis was carried out to check the genetic modification of regenerated plants by mutagen treatments. Three polymorphic DNA fragments out of thirty-seven obtained by RAPDs were observed in regenerated plants using 5 decamer primers.

A STUDY OF THE LOW LEVEL RADIATION EFFECT ON THE MITOTIC INDEX OF THE BASAL CELLS IN THE BUCCAL POUCH OF HAMSTERS (저선량방사선이 햄스터의 협낭 기저세포 분열지수에 미치는 영향에 관한 연구)

  • The purpose of this study was to investigate the effects of the low level irradiation on the mitotic index of the basal cells in the buccal pouch of hamsters{golden hamster: APG strain). After colchicine was administrated to the hamsters through the intraperitoneal, the low level radiation{5460mR) was exposed on the buccal pouch of hamsters. The mitotic index of the basal cells was estimated 2 hours after irradiation. The results were as follows: 1. The mean mitotic index of the control group was 4.32. 2. The mean mitotic index of the irradiated group was 2.46. 3. T-test of data in the irradiated group showed significant difference from the mitotic index in the control group. These results suggested the lowered mitotic index of the irradiated group resulted from the low level irradiation.

  • An autotetraploid, Sawonppong 28 was derived from a thick-leaf and highly productive mulberry variety, Huyeoprok, by colchicine solution treatment on growing point and its characteristics were as follows. The number of somatic chromosomes of the derived variety was 2n=56 of an autotetraploid and green tip sprouting stage was medium budding variety with triangular oval shape and thick leaves contained rich chlorophyll. The length of branches was comparatively short and the number of branches was also small. However the branches were thick and non-budding length percentage at basal part of branch was high and cold hardiness was weak.

  • Apoptosis is the normal physiological process of cell death essential for the maintenance of homeostasis. The function of nicotinamide adenine dinucleotide (NAD) and adenine diphosphate (ADP) ribosylation (transfer of ADP-ribose to proteins) reactions in modifying apoptosis have recently been of great interest. Recently. CD38. a type 2 transmembrane glycoprotein expressed in hematopoietic and non hematopoietic cell lines. has been reported to possess NAD glycohydrolase activity (Han. 1999) and PC-1 and CD38 NADase regulates T cells by inhibition of phosphodiesterase/pyrophosphatase activity of PC-1 by its association with glycosaminoglycan (Hozada et al., 1999). Sindhuphak et al. (2000) has reported that cervical cancer cells can be differentiated from normal cells by using FTIR (Fourier-Transformed Infrared) technique. which has characterized shifts to be due to the phosphodiester bond in nucleic acid. protein amide I&II. carbohydrate and glycogen bands. Mechanisms how phosphodiester bond shift in cervical cancer cells as compared to control cells remain to be elucidated. Suramana et al. (2000) as well as Lohitnavy and Sinhaseni (1998) have studied methomyl and colchicine effects in rat splenocytes. Lactate Dehydroge-nase Isozymes 3 (LDH3) and LDH4 were observed to increase transiently and subsided in plasma of rats exposed to 6~8 mg/kg methomyl after 48 hours. Phosphodiester bond shift of nucleic acid. detected by FTIR. was also reported (Suramana et al., 2000). We report here, after analysis of bond shift patterns. a similar bond shifts detected by FTIR spectrum observed in human cervical cells and splenocytes of rats exposed orally to 2~8 mg/kg methomyl as well as rats exposed to colchicine 2~6 mg/kg orally.

Effects of 17-DMAG Administration on Autophagy Flux in Mouse Skeletal Muscle (17-DMAG이 마우스 골격근에서 autophagy flux에 미치는 영향)

  • The purpose of this study was to determine if heat shock proteins are involved in autophagy in skeletal muscle. We used the autophagy flux strategy, which is an LC3 II/p62 turnover assay conducted with and without an autophagy inhibitor, to determine whether 17-DMAG (an Hsp90 inhibitor/Hsp72 activator) stimulates autophagy in skeletal muscle. We treated C2C12 cells with 17-DMAG (500 nM) for 24 hr with and without the autophagy inhibitor (Bafilomycin A1, 200 ng/ml), and we injected C57BL/6 mice i.p. with 17-DMAG (10 mg/kg) daily for 7 days with and without colchicine as an autophagy inhibitor (0.4 mg/kg/day, administered on the last 2 days). C2C12 myotubes and tibialis anterior muscles were harvested for analysis of mTOR-dependent autophagy signaling pathway proteins and autophagic marker proteins (p62 and LC3 II) by Western blot analysis. The blots showed that 17-DMAG upregulated hsp72 and decreased Akt protein levels and S6 phosphorylation in C2C12 cells. However, an in vitro autophagic flux assay demonstrated that 17-DMAG did not increase LC3 II and p62 protein concentrations to a greater extent than Bafilomycin A1 treatment alone. Similarly, 17-DMAG increased Hsp72 protein levels and decreased the expression of Akt and the phosphorylation of S6 in mouse skeletal muscle. However, unlike the response seen in C2C12 myotubes, the p62 protein levels were significantly decreased in 17-DMAG-treated mouse skeletal muscle (~50%; p<0.05). The LC3 II protein levels in 17-DMAG-treated mice were increased ~2-fold more when degradation was inhibited by colchicine (p<0.01). This suggests that 17-DMAG stimulates basal autophagy in skeletal muscle but is not found in C2C12 myotubes.
