Effect of Phytoecdysteroid on Pure Breed Performance of Silkworm Bombyx mori L.

  • Phytoecdysteroids with moulting hormone (MH) activity induce different responses in silkworms when used on different day of final instar, which can be manipulated for maximum benefit like early and uniform spinning behaviour, reducing crop loss and to increase cocoon yield. The results showed that application of this hormone on seed crop viz., CSR2, CSR4 and BL44 and BL67 in early stage of 5$^{th}$ instar i.e., at 72 hrs and 96 hrs though induced early and uniform spinning behaviour, there was an adverse effect by 9-36% on the economic characters like cocoon yield, cocoon weight, cocoon shell weight and also on fecundity etc. Application of this hormone in late stage of $5^th$ instar i.e., at the onset of spinning showed non-significant variations in some of the characters like cocoon weight, cocoon shell weight, cocoon shell ratio and fecundity. The physiological implications of phytoecdysteroid in hastening the maturation events and synchronization of spinning activities in different breeds are discussed.

Induction of Non-Diapause Eggs and Manifestation of Quantitative Characters by Low Temperature Incubation of Eggs in the Silkworm, Bombyx mori L.

  • Induction of non-diapause eggs and manifestation of quantitative characters were studied in different seasons by low temperature incubation of eggs of a low yielding diapause with coloured cocoons silkworm breed, RDI of Bombyx mori. Hundred percent of nandiapause eggs were induced when the egg incubation was carried out at 15$^{\circ}C$ followed by incubation initially at $25^{\circ}C$ and then at $15^{\circ}C$. The diapause-nondia-pause relation was found to be reversible. Analysis of variance study demonstrated significant variation in all the quantitative characters whereas significant variation due to different incubation treatments was observed for larval span, fifth instar larval span, cocoon yield/10,000 larvae by weight, cocoon weight, cocoon shell weight, and cocoon shell ratio. study on hybrid vigour skewed significant heterosis over mid parent value for four economic characters viz., cocoon weight, cocoon shell weight, cocoon shell ratio and filament length in a Fl hybrid between RD1 with diapause eggs and Japanese type bivoltine $NB_{4} D_{2}$.

  • Strength, elongation and elasticity are essential properties with regard to the tensile properties of raw silk thread. The reasonable harmony of the above three physical characters appears to be the specific nature of the raw silk thread and the dynamical functions are determined by the cocoon filament forming the raw silk. From such viewpoint, the author has studied the tensile property of the thread when the cocoon filament is stretched and transformed. The results obtained are as follows: 1) The stretching strength of the inner layer filament is stronger than that of the outer and medium layer ones. 2) The stretching strength of the filaments is influenced by the moisture-heat denaturalization of cocoon layer sericin in the drying process. 3) The more transformed functions by the stretching become, the lower the elasticity index of the cocoon filament was shown. 4) When cocoon layer sericin is denatured by moisture and heat, elasticity index of the filament is decreased.

Dissolution of degummed Antheraea yamamai silkworm cocoon

  • Dissolution of Antheraea yamamai silkworm cocoon was carried out in various solvent systems with various dissolving conditions including dissolution salts, salt concentration, dissolving temperature, and time. General chaotropic salt for Bombyx mori silk fibroin does not work for A. yamamai silkworm cocoon. Lithium bromide 9.3 M at $100^{\circ}C$ also does not work to dissolve wild silkworm cocoon. However, 9 M of lithium thiocyanate treatment at $100^{\circ}C$ induced 100% dissolution of wild silkworm cocoon. But it could not be dissolved lower than $60^{\circ}C$. Like lithium thiocyanate, less than $60^{\circ}C$ treatment with molten calcium nitrate 4 hydrate could not dissolve wild silkworm cocoon. As the dissolution temperature increased up to $100^{\circ}C$, the solubility of wild one was reached over 90%. SDS-PAGE showed broad tailing stream pattern that means the molecule of wild silk was depolymerized with dissolution temperature and time. From the above results, the best chaotropic salt for A.yamamai silkworm cocoon is calcium nitrate 4 hydrate.

The Test of Combining Ability and Heterosis on the Silkworm(Bombyx mori) Breeding (누에 견.사질에 관한 잡종강세 및 조합능력검정)

  • The study was conducted to obtain the genetic information on heterosis and combining ability of the quantitative characters for F1 hybrid breeding in silkworms. Six parental varieties and each set of 30 diallel crosses in F1's were used as materials, and bred on the randomized complete block design with three replications. Fourteen characters were observed with the twenty samples in each tray. The data were analyzed for (1) heterosis and combining ability in F1 hybrid. The heterosis in the weight and the length of cocoon showed positively high at 24.51%, and 23.4%, respectively and the weight of the whole cocoon as well as the weight of the whole cocoon layer showed a siginificant heterosis ranging from 15.56% to 15.71% and from 17.14% to 19.01%, but the fifth and the total instar period showed negative heterosis. It was found that the combination between, C70XRomogua and N9 X Romogua showed highly a negative heterosis on the fifth instar period and for the cocoon weight. The female of N9+Sansuian and the male of Romogua X Sansurian have a high heterosis effect, on the cocoon shell weight, and Sansurian X Romogua(reciprocal) on the length and the weight of cocoon filament with no regard to sexuality. The significant maternal and cytoplasmic effect on heterosis of the cocoon weight and the cocoon shell weight were observed with the combinations, N9 X C5, N63 X C70 and on the length of the cocoon filament with the combinations, Sansurian X N63, Sansurian X C5, Sansurian X C70 and N9 X C70, N63 X C70 on the weight of cocoon filament. As mean squared of GCA, SCA and RCA were significant with these combining ability for all characters resulted from additive and non-additive altogether and there is a significant difference between reciprocals. Sansurian showed a negative GCA effect on the fifth and total larval duration, but the higher positive GCA effects took places with varieties N9 and C5 on the length, width, weight of cocoon, cocoon shell weight, percentage of cocoon shell weight, length and weight of cocoon filament, percentage of raw-silk with no regard to both generations and silkworm sexuality. The values of SCA between the cross combinations varied generation-wise and sex-wise. It was shown that SCA value for the fifth instar period was highly negative for Sansurian X C70, Romogua X C70, Sansurian X C5, Romogua X C5, but it was positive effect on the cocoon weight, cocoon shell weight with N9 X C5, and C70 X Sansurian, on the length of cocoon filament with N9 X C5, Romogua X Sansurian on the weight of cocoon filament between Romogua and N63 and on the percentage of raw-silk between the combination of Sansurian X Romoga.

Studies on the Operation Control with Automatic Silk Reeling Process to be responded for Korean Silk Cocoon (한국원료견질에 적응하는 자동조사 공정관리의 구명에 관한 연구)

  • These studies were attempted to find out the optimum silk reeling system by use of automatic silk reeling machine to increase raw silk yield and reeling efficiency with various silk reelable cocoons. The obtained results are as follows; 1. The mean silk reelability ratio(X)of the Korean cocoons during the last ten years was 61 per cent, beside 64.7 per cent in autumn cocoon and 57.3 per cent in spring cocoon. However, the ratio variation of autumn cocoons was larger than that of spring cocoons. 2. A positive correlation between cocoon filament breaks during its process and silk reelability levels was shown to be significant. The cocoons of both poor and good reelability evidenced "J" shape distribution on the filament break graph by the order of reeling cocoon end. Many bave breaks were found at the inner shell of the cocoons, or in case of poor reelability cocoons. 3. The morphology of broken cocoon ends during the process was classified into A, B, C, D, E and F types, The occurrence of B type was majority, but that of F type was minority among them. 4. In case of the cocoon cooking, H-type ion-exchanged soft water was better for good reelability cocoons, Na and H-type ion exchanged neutral soft water for those of fair reelability, and alkaline (Na-type) soft water for those of poor reelability, respectively. 5. The modification of cooking water by mixing the above different types of water (50% Na-type and 50% H-type passed by standard natural water; 75% Na-type and 25% H-type passed by hard natural water; 25% Na-type and 75% H-type passed by soluble natural water) made higher yield of raw silk with tess breaks of thread. 6, In case cocoon ends groping water included sodium hexametaphosphate as much as 800 ppm. the groping efficiency and raw silk yield of cocoon was improved. The effect was pronounced in case of poor reelability cordons. 7. The most reasonable cocoon cooking and silk reeling condition for automatic silk reeling process were observed to be rather incomplete cook with good reelability cocoons and optimum cook with poor reelability cocoons succeeded by the reeling bath temperature of 45$^{\circ}C$, 8. The reasonable silk reeling velocities were observed to be about 150m per min. for good reelability cocoons, 120m per min. for fair reelability ones and 90 to 120m per min. for poor reelability ones. 9. In order to improve the raw silk yield of cocoons and reeling efficiency, the cocoon stand-by-ratio for reeling should be kept at the level of 40 per cent for good reelability cocoons or at 60 per cent for poor reelability ones beside necessary end found cocoon condition.

Induction and Characterization of Precocious Trimolting Silkworm, Bombyx mori (조숙 3면잠의 유발과 실용형질)

  • For fine-denier cocoon filament production, the precocious trimolting silkworms were induced by the treatments with both the imidazole compound "KK-42" hating anti-juvenile hormone activity and high temperature, and their growth, dietary efficiency, cocoon and cocoon filament qualities were compared with those of normal tetramolters as control. 1. The percentage of precocious trimolters was higher in the application of KK-42 than the treatment of high temperature. The effective concentration of KK-42 was 10$\mu\textrm{g}$/1arva. The high temperature treatment for 48 hour was more effective than 36 hour treatment, and silkworm larvae fed on mulberry leaves were more sensitive than artificial diet to induce trimolters. 2. The larval duration of the trimolters induced by KK-42 was 5.17 day shorter than that of normal tetramolters in mulberry leaves rearing, and the increasing pattern of body weight in the 4th inster larvae of trimotlers was similar to the 5th instar larvae of normal tetramolters. 3. The qualities such as cocoon weight, cocoon layer weight and cocoon layer ratio of precocious trimolter induced by KK-42 were much lower those of normal tetramolters: the cocoon weight and cocoon layer ratio were 0.78g, 14.2cg and 18.4% in mulberry leaves rearing, and 0.86 cg, 10.3cg and 12.3% in artificial diet rearing, respectively. 4. The size of cocoon and cocoon filament was smaller in the precocious trimolter, both KK-42 and high temperature, as compared with that of the normal tetramolters. 5. The efficiency of cocoon layer production of the precocious trimolters by the KK-42 was lower than that of the normal tetramolters: amount of the cocoon layer production per 1g of dry mulberry leaves ingested was 7.97cg in the precocious trimotors, while 9.20cg in the normal tetramolters.

  • In the silkworm fed on ginseng, the body weight and the cocoon weight were not only heavier, but also the ratio of a cocoon layer to a cocoon weight was higher than those in the control. Especially, the heavier pupae were appeared. The ginseng may be therefor available in silkworm improvement.

Analysis of Gene for Major Quantitative Characters in Silkworm, Bombyx mori (가잠의 주요 양적 형질에 관한 유전분석)

  • In the experiment, gene actions were analyzed for seven silkworm varieties in order to obtain basic information on their genetic improvement by diallel crosses. The results obtained were summarized as follows : in analysis of variance, significantly maternal and reciprocal effects were observed for all character except female cocoon shell percentage and female pupal weight. Over-dominance was shown by Vr-Wr graphic analysis in three characters such as female cocoon weight, female and male pupal weight, and partial dominance in five characters such as male cocoon weight, cocoon shell percentage in both female and male. Component of genetic varience analyzed for five characters such as male cocoon weight, cocoon shell weight, cocoon shell percentage in both female and male showed that additive effects were higher than dominant effects, and female cocoon weight showed dominant effects higher than additive effects. Heritability of narrow sence was higher more than 0.51 and heritability of broad sence was higher more than 0.77 in all characters except for female pupal weight. The directions of dominance were negative for female cocoon shell weight and somewhat higher for female and male cocoon shell percentage. Meanwhile cocoon weight, pupal weight in both female and male showed positive in the direction of dominance.

Line X Tester Analysis for Economic Characters in the Bivoltine Silkworm, Bombyx mori L.

  • In a line X tester crossing programme (24 lines 2 testers) the general combining ability (GCA) and specific combining ability (SCA) effects were analyzed for five economic characters in the bivoltine silkworm, Bombyx mori L. The results showed desired GCA effects in 934D1 (9500), 934B (9789) and 934A1 (9855) for cocoon yield per 10,000 larvae brushed by number. Likewise, the lines found to be superior based on GCA effects for other characters were as follows; 931D (14.040 Kgs), 935E (17.023 Kga.), 934D1 (15.643 Kgs.) and 934B (15.687 Kgs.) for cocoon yield by weight: 931D (1.717 g) and 930E (1.796 g) for single cocoon weight; 932B (0.330 g) for single shell weight: 931D (1.717 g) and 930e (1.796 g) for single cocoon weight; 932B (0.330 g) for single shell weight; 932B (18.7%), 933A (18.86%) and 935A (19.89%) for shell ratio. SCA effects showed the superiority of 932D$\times$KA (9822 cocoon yield per 10,000 larvae brushed by number); 932A$\times$NB4D2 (16.933 Kgs. cocoon yield per 10,000 larvae brushed by weight); 931C$\times$KA (1.911 g single cocoon weight); 934$\times$NB4D2 (0.371 g single shell weight and 21.0% shell ratio). The analysis indicated non-additive gene action for all the five characters.
