Neuroprotective effect of Aster yomena (Kitam.) Honda against hydrogen peroxide-induced oxidative stress in SH-SY5Y cells

  • Oxidative stress is one of the contributors of neurodegenerative disorders including Alzheimer's disease. According to previous studies, Aster yomena (Kitam.) Honda (AY) possesses variable pharmacological activities including anti-coagulant and anti-obesity effect. In this study, we aimed to determine the neuroprotective effect of ethyl acetate fraction from Aster yomena (Kitam.) Honda (EFAY) against oxidative stress. Therefore, we carried out 3-(4,5-dimethylthiazol-2-yl)-2,3-diphenyl tetrazolium bromide, lactate dehydrogenase (LDH), and 2',7'-dichlorofluorescin diacetate assays in SH-SY5Y neuronal cells treated with hydrogen peroxide (H2O2). H2O2-treated control cells exhibited reduced viability of cells, and increased LDH release and reactive oxygen species (ROS) production compared to normal cells. However, treatment with EFAY restored the cell viability and inhibited LDH release and ROS production. To investigate the underlying mechanisms by which EFAY attenuated neuronal oxidative damage, we measured protein expressions using Western blot analysis. Consequently, it was observed that EFAY down-regulated cyclooxygenase-2 and interleukin-1β protein expressions in H2O2-treated SH-SY5Y cells that mediated inflammatory reaction. In addition, apoptosis-related proteins including B-cell lymphoma-2-associated X protein/B-cell lymphoma-2 ratio, cleaved caspase-9, and cleaved-poly (ADP-ribose) polymerase protein expressions were suppressed when H2O2-treated cells were exposed to EFAY. Our results indicate that EFAY ameliorated H2O2-induced neuronal damage by regulating inflammation and apoptosis. Altogether, AY could be potential therapeutic agent for neurodegenerative diseases.

Effect of Climate Change Characteristics on Operation of Water Purification Plant (정수장 운영에 영향을 미치는 기후변화 요인 분석)

  • Climate change has a broad impact on the entire water environment, and this impact is growing. Climate adaptation in water supply systems often involves quantity and quality control, but there has been a lack of research examining the impacts of climatic factors on water supply productivity and operation conditions. Therefore, the present study focused on, first, building a database of climatic factors and water purification operating conditions, and then identifying the correlations between factors to reveal their impacts. News big data was analyzed with keywords of climatic factors and water supply systems in either nationwide or region-wide analyses. Metropolitan area exhibited more issues with cold waves whereas there were more issues with drought in the Southern Chungcheong area. A survey was conducted to seek experts' opinions on the climatic impacts leading to these effects. Pre-chlorination due to drought, high-turbidity of intake water due to rainfall, an increase of toxins in intake water due to heat waves, and low water temperature due to cold waves were expected. Pearson correlation analysis was conducted based on meteorological data and the operating data of a water purification plant. Heavy rain resulted in 13 days of high turbidity, and the subsequent low turbidity conditions required 3 days of high coagulant dosage. This insight is expected to help inform the design of operation manuals for waterworks in response to climate change.

Production of porcine fibroblasts carrying a vector enforced specific expression of CD73 to endothelial cells (돼지 혈관내피세포 특이적 CD73 발현 벡터가 도입된 돼지 섬유아세포 생산)

  • Nucleotide metabolism in endothelium is variable between different species. Recent studies demonstrated that this variability could contribute coagulation dysfunction, even though organs of the alpha 1,3-galactosyltransferase gene knockout pig were transplanted into the primate. CD73 (ecto-5'-nucelotidase) is an enzyme at cell surface catalyzing the hydrolysis of adenosine triphosphate to adenosine, which plays role on a substance for anti-inflammatory and anti-coagulant. Thus, overexpression of CD73 in endothelial cells of the pig is considered as an approach to reduce coagulopathy. In this study, we constructed a human CD73 expression vector under control of porcine Icam2 promoter (pIcam2-hCD73), which is expressed specifically at endothelial cells, and of CMV promoter as a control (CMV-CD73). First, we transfected the CMV-CD73 vector into HEK293 cells, and then confirmed CD73 expression at cell surface by flow cytometry analysis. Next, we transfected the pIcma2-CD73 and CMV-CD73 vectors into primary porcine fibroblasts and endothelial cells. Consequence was that the pIcma2-CD73 vector was expressed only at the porcine endothelial cells, meaning that the pIcam2 promoter lead to endothelial cell-specific expression of CD73 in vitro. Finally, we nucleofected the pIcam2-hCD73 vector into passage 3 fibroblasts, and enforced hygromycin selection of 400mg/ml. We were able to obtain forty three colonies harboring pIcam2-CD73 to provide donor cells for transgenic cloned porcine production.

Characteristics of Stormwater Treatment in Construction Site (건설 현장 내 비점오염원 처리 특성 평가)

  • Total suspendid solid (TSS) of non point source pollutants in construction site are in higher concentration than others (BOD, COD etc). Also, the TSS concentration is very sensitive to the rainfall intensity in early stage of construction. There are two methods for treatment of non point source pollutants, which are temporary treatment facility and filtering one. But they have disadvantages. Temporary facility system has very low efficiency and filtering system consumes high energy and takes up large footprint. This study shows how prefabricated flocculation/coagulation system is developped to cover the above weakness and evaluation of the system performance in construction site. The prefabricated flocculation/coagulation system has very high treatment efficiency comparing with temporary and filtering system and takes small footprint. Therefore, it expects that the system leads to prevention of pollution near construction site and reduction of public grievance. Proper coagulant dosage and sludge circulation facility application, controlling the height of sludge interfacial are necessary to maximize the system efficiency.

On-line Monitoring of the Flocs in Mixing Zone using iPDA in the Drinking Water Treatment Plant (정수장 응집혼화공정에서의 응집플럭 연속 모니터링)

  • This study evaluated the flocs forming characteristics in the mixing zone to increase the coagulation effect in the drinking water plant. As a measuring tool of formed flocs, on-line particle dispersion analyzer (iPDA) was used in Y drinking water plant. To evaluate the forming flocs, many parameters such as poly amine, coagulant dosing amount, raw water turbidity, and pH was applied in this study. During the periods of field test, poly aluminium chloride (PACl) as a coagulant was used. With the increase of the raw water turbidities, poly amine was also added as one of aids for increasing in coagulation efficiency. The turbidity and pH of raw water was ranged from 7 to 9 and from 25 to 140 NTU, respectively. The increasing of raw water turbidity brought the bigger floc sizes accordingly. From a regression analysis, $R^2$ value was 0.8040 as a function of T, raw water turbidity. Floc size index (FSI) was obtained from a correlation equation as follows; FSI = 0.9388logT - 0.3214 Also, polyamine gave the bigger flocs the moment it is added to the coagulated water in the rapid mixing zone. One of parameters influencing the floc sizes was the addition of powdered active carbon(PAC) in the mixing zone. In case of higher turbidity of raw water, $R^2$ value was 0.9050 in the parameters of [PACl] and [PAC]; FSI = $0.0407[T]^{0.324}[PACI]^{0.769}[PAC]^{0.178}$ On-line floc monitor was beneficial to evaluate the flocs sizes depending on the many parameters consisting raw water properties, bring the profitable basic data to control the mixing zone more effectively.

Influence of Dispersed and Anaerobic Bacteria in Aerobic Paper-making Wastewater Treatment (호기적 제지폐수 처리공정중에의 분산 혐기성 미생물의 영향)

  • Tn order to be helpful to control dispersed microorganisms for stabLlization of wastewater treatment in a paper-makillg process, dominant strains were isolated aerobically and anaerobically. and identified and physiological characteristics were also analyzed. Pseudomonas carboxydohydrogena, Cardiobacten'm hominis, lvIicrococcus lylae, XanfomonCls campestris p" juglandis, Micrococcus diversus, and Comamonas terrigencl as aerobic dominants, and Streptococcus bovis and Prevotella buccae as anaerobIc dominants were identified fi'om the supernatent of the primary settling tank. It seemed that microflora in the treatment process would consist of many kinds of microorganisms, whose dominant would change easily according to environmental conditions, They all grew well at $37^{\circ}C$ and at different initial medium pH's. Especially, some of them required sulfate ion for their growth, which came from a chemical coagulant of aluminium sulfate in the primary settling tank. Interestingly. many anaerobes grew well even in the aerobic wastewater treatment process and seemed to have some functions. Population of anaerobes increased three times in the supematant of primary settling tank and ten times in Lhe bottom sludge of primary settling tank than in the prime wastewater. Therefore, these anaerobes contributed to the producH tion of offensive gases, which would make some microorganisms not precLpitate and be buoyant.

The Effect of Coagulant on Filtration Performance in Submerged MBR System (침지형 MBR 공정에서 응집제가 여과성능에 미치는 영향)

  • The purpose of this study was to investigate effect of coagulation on filtration performance of membrane in submerged MBR system and influence of continuous aeration to reduce fouling of membrane surface on coagulated floc. For this purpose, aeration tank sludge of MBR system was compared with jar-test sludge. The experimental results were analysed in terms of floc size and SRF (Specific resistance of Filtration). The more alum was added, the more content of floc below $10{\mu}m$ reduced and SRF decreased. But compared with jar-test results, it was found that effect of coagulation on MBR floc was reduced. Operation time of membrane in alum added MBR was longer than that in control MBR. But operation time was not proportional to alum dose. It was thought that the result was reason that floc below $10{\mu}m$ was not reduced sufficiently by shear force of continuous aeration. Moreover it was founded that if alum is added more than proper dose, it brings filtration resistance to increase.
