In order to compare greenhouse gases emission from different animal manures and to explore how different animal manures effect on soil mineralization, three kinds of materials, cattle, goat and chicken manure were amended to soil for 14 days incubation as CtS (cattle manure-amended soil), GS (goat manure-amended soil) and ChS (chicken manure-amended soil). Cumulative $NH_3$ emissions in all treatments were rapidly increased until day 7 and then it was slightly increased in three manure-amended soils but maintained in control until day 14. GS had the highest $NH_3$ emission at $0.14mg\;kg^{-1}$ during the entire experimental period. Emissions of $CO_2$ were highly increased by 7.8-, 9.0- and 12.4-fold in CtS, GS and ChS, respectively, compared to control at day 14. A significant increase of $N_2O$ emission in all treatments occurred within 5 days and then it was slightly increased until day 14. $N_2O$ emission was 2-fold higher in all manure-amended soils than that of control. Compared to day 1, inorganic N ($NH_4{^+}$ plus $NO_3{^-}-N$) content was highly increased in all four treatments at day 14. The increase rate was the highest in CtS treatment. Net N mineralization was increased by 4.0-, 2.4- and 2.9-fold in CtS, GS and ChS, respectively, compared to control. These results indicate that increase of $NH_3$, $CO_2$ and $N_2O$ gas emissions was positively related to high N mineralization.
King oyster mushrooms(Pleurotus eryngii) are the second biggest mushroom for exporting in Korea but their browning and soft rot is the main factors of claim during long distance transportation. Fresh king oyster mushrooms were treated with $CO_2$ at 30, 50% for 3 hours at $5^{\circ}C$ prior to storage at $20^{\circ}C$ and $5^{\circ}C$. There was no difference on respiration rate after $CO_2$ treatment. However exposure to $CO_2$ for 3h prior to MA packing maintained the firmness and delayed color(hunter L and b value) change of mushrooms during storage. Especially an incubation in high $CO_2$ at 30% significantly reduced soft rot and browning symptoms resulting in one week extension of shelf-life during storage at $5^{\circ}C$ compared to control and 50% $CO_2$ treatment.
Ten college women were treated with either corn oil(CO< as a source of C18:2, $\omega$6) or fish oil concentrates(FO, as a source of C20:5+C22:6, $\omega$3) with experimental diet for 7 days and then returned to normal home made diet. Kinetic changes of plasma and RBC lipids, tocopherol, MDA levels and hemolysis were observed at day-0, -3, -7, -8, -10, -14 and -21 of experimental periods. Plasma and RBC tocopherol contents were significantly increased at day-3 and -7 by tocopherol tocopherol supplement in dietary oil, but there was no significant difference between CO and FO diet with large dose of tocopherol supplement. After stop experimental regimen at day-7, plasma tocopherol content was sustained at high level until day-10 but drastically decreased at day-14 and remained at low level at day-21. However, RBC tocopherol level was not greatly responded to the dietary intake of tocopherol and was varied in the very narrow range. MDa levels of plasma and RBC were not responded in the same way as tocopherol content of plasma. MDa content of RBC was very low compared to that of plasma. RBC hemolysis by incubation in hypotonic solution was negatively correlated to plasma tocopherol level and was not correlated to the level of MDa in either plasma or RBC. There were no significant change in the levels of plasma cholesterol, HDL-chol, triglyceride, phospholipid, and lipoprotein pattern throught experimental periods.
V parahaemolyticus possessed an extracellular alkaline pretense activity during the stationary growth phase. Various factors such as nitrogen sources, the concentration of NaCl and metal ions were investigated for optimizing the production of alkaline pretense from V. parahaemolyticus ATCC 17802. Among the nitrogen sources tested skim milk showed the distinct increase of the activity and the activity was the highest at 2% in final concentration after 60 hours incubation. The addition of NaCl and metal ions did not increase the alkaline pretense activity. CoC$_2$, CuC1$_2$, and HgCl rather highly inhibited alkaline protease production.
Serratia marcescens co-cultured with various phytopathogenic fungi, including Rhizopus stolonifer, Helminthosporium allii, Pyricularia oryzae, Fusarium oxysporium and Collectothricom cassiicola, in an LB- agar medium containing 1.5% swollen chitin, significantly inhibitied fungal growth. Fungal hyphae grew rapidly outward from the culture dish center, but the hyphal extensions of the pathogenic fungi were significantly inhibited in a perimetric contact area with S. marcescens. This was especially evident in pathogenic fungi which have a high chitin content in their cell walls. The extracellular chitinase activities of S. marcescens were increased seven fold by the addition of 1.5% swollen chitin to the LB-broth, compared to chitinase activities in a culture medium without chitin. The type of induction was dependent on the various forms of chitin used. When the culture supernatant of S. marcescens or the chitinases of Streptomyces griceus purchased from Sigma Chemical Co., were incubated with the mycelium of F. oxysporium, the mycelium gradually burst as cultivation time progressed and completely lysed after incubation for 2 days. On the other hand, E. coli extract did not hydrolyze the F. oxysporium mycelium at all. These data showed that the chitinolytic activities of S. marcescens play important roles in the biochemical control of phytopathogenic fungi.

The productivity and the precision machining are the major concerns of the machine tools, and the importance of those features will be even more emphasized with the needs of the modern industries. In this paper the dynamic properties of a lathe have been investigated through the frequency analysis test and the computer simulation to minimize the transmission of vibration generated during the machining process. The modifications have been applied to the original structure to suppress the vibration transmission and further for the structure to achieve less vibrations and higher precision machining. The result shows that the machine can have much less vibrations with simple design alterations for minor expenses, the result can also be readily applied to many similar machines for improved stability.
In vitro maturation of denuded porcine immature oocytes can be enhanced by co-incubation with spermatozoa even before fertilization. This study was to determine whether the addition of spermatozoa into the culture medium could influence the nuclear maturation of porcine cumulus-enclosed germinal vesicle (GV) oocytes. Cumulus-oocyte complexes (COCs) were collected from follicles of 3- to 5-mm diameter. Porcine COCs were cultured in tissue culture medium containing spermatozoa. After 48 h culture, oocytes were examined for evidence of GV breakdown, metaphase I, anaphase-telophase I, and metaphase II. The proportion of oocytes reaching at metaphase II was significantly (P < 0.05) increased in the oocytes cultured in media containing spermatozoa compared to those in media without spermatozoa (52.3% vs 12.5%). No difference in the percentage of metaphase II was observed among the different periods of spermatozoa exposure and among the spermatozoa from different species. The proportion of oocytes reaching metaphase II was significantly different between high and low concentrations of spermatozoa. The present study suggests that manunalian spermatozoa contain a substance(s) that improves nuclear in vitro maturation of porcine cumulus-enclosed GV oocytes. Enhancing effect of spermatozoa for in vitro maturation of oocytes is a highly dose-dependent.
It was attempted in this study to investigate the effect of ginseng saponin on several glycolytic enzymes of yeast cell and the following results were obtained. The amount of $CO_2$formed during the incubation of yeast cells in medium containing saponin fraction of Panax ginseng C.A. Meyer was greater than that of control cells and found that the $CO_2$ formation was greatest when the cells were grown in the medium containing 10$^{-3}$% of the saponin fraction, at which the uptake of inorganic phosphate and glucose consumption were also increased. Radioactivity study of several glycolytic intermediates of yeast cells cultured in the medium containing [U-$^{14}$C]-glucose showed that the radioactivity of fructose 6-phosphate of test cells was as much as 1.6times that of control group. On the other hand, the radioactivity of pyruvate of test cells was considerably decreased compared to control. Investigation of the effect of ginseng saponin on yeast hexokinase, phosphoglucose isomers, pyruvate kinase and perverted decarboxylase in vitro showed that the maximum activities of the above enzymes were observed when the concentration of ginseng saponin was 10-$^{-5}$% in the reaction mixture. It seemed that the ginseng saponin stimulated both glycolytic enzymes such as hexokinase, phosphoglucose isomers and perverted decarboxylase significantly.
Biodegradable microspheres made from poly-lactide-co-glycolide polymers have been considered as a new delivery system for single-dose vaccine. Purified tetanus toxoid (TT) was encapsulated in poly-lactide(PLA) and poly-lactide-co-glycolide (PLGA) microparticles using a solvent evaporation method in a multiple emulsion system (water-in oil-in water). The morphology of 77-loaded microparticles was spherical and the suface of them was smooth. The particle size was in a range of 2-10. Protein loading efficiency was 68-97.8%. PLGA (85:15) microparticle showed the highest efficiency. Protein release pattern was influenced by polymer molecular weight and composition. The release rate of PLA(Mw 100,000) microsphere was higher than any other microspheres. In consequence of the hydrolysis of PLGA(50:50) microspheres, environmental pH decreased from 7.4 to 5.0. The PLA, PLGA (75:25) and PLGA (85:15) microshperes showed no significant pH change. The antigenicity or n in microshperes was assayed by indirect sandwich ELISA using equine polyclonal tetanus antitoxin for capture antibody and human polyclonal tetanus antitoxin for primary antibody. The antigenicity of TT in PLA (Mw 100,000), PLGA(50:50, Mw 100,000) and PLGA (75:25, Mw 73,300) after 30 days incubation showed 54, 40.9 and 76.7%, respectively.

The finite element method (FEM) is widely used to evaluate the seismic performance of pile-supported buildings. However, there are problems associated with modeling the pile end resistance using the FEM, such as the dependence on the mesh size. This paper proposes a new method of modeling around the pile tip to avoid the mesh size effect in two-dimensional (2D) analyses. Specifically, we consider the area of influence around the pile tip as an artificial constraint on the behavior of the soil. We explain the problems with existing methods of modeling the pile tip. We then conduct a three-dimensional (3D) analysis of a pile in various soil conditions to evaluate the area of influence of the soil around the pile tip. The analysis results show that the normalized area of influence extends approximately 2.5 times the diameter of the pile below the pile tip. Finally, we propose a new method for modeling pile foundations with artificial constraints on the nodal points within the area of influence. The proposed model is expected to be useful in the practical seismic design of pile-supported buildings via a 2D analysis.
