Ginseng, a traditional herbal drug, has been used in Eastern Asia for more than 2000 years. Various ginsenosides, which are the major bioactive components of ginseng products, have been shown to exert numerous beneficial effects on the human body when co-administered with drugs. However, this may give rise to ginsenoside-drug interactions, which is an important research consideration. In this study, acassette assay was performed the inhibitory effects of 12 ginsenosides on seven cytochrome P450 (CYP) isoforms in human liver microsomes (HLMs) using LC-MS/MS to predict the herb-drug interaction. After incubation of the 12 ginsenosides with seven cocktail CYP probes, the generated specific metabolites were quantified by LC-MS/MS to determine their activities. Ginsenoside Rb1 and F2 showed strong selective inhibitory effect on CYP2C9-catalyzed diclofenac 4'-hydroxylation and CYP2B6-catalyzed bupropion hydroxylation, respectively. Ginsenosides Rd showed weak inhibitory effect on the activities of CYP2B6, 2C9, 2C19, 2D6, 3A4, and compound K, while ginsenoside Rg3 showed weak inhibitory effects on CYP2B6. Other ginsenosides, Rc, Rf, Rg1, Rh1, Rf, and Re did not show significant inhibitory effects on the activities of the seven CYPs in HLM. Owing to the poor absorption of ginsenosides after oral administration in vivo, ginsenosides may not have significant side effects caused by interaction with other drugs.
This study was per-formed to screen lactic acid bacteria poultry for the probiotic use. Among the previously obtained acid tolerant, 139 strains, 111 strains were selected with MRS medium containing 0.3% oxgall. 34 strains of 111 was re-selected by Gram-staining and acid producing ability. These strains was identified by MIDI Sherlock Microbial Identification System. Among the identified 34 strains Lactobacillus fermenum YL-3 was selected for the final pro-biotic use because of the good growth and high survival rate at pH 2.0. 60%, 50% and 40% cells of Lactobacillus fermentum YL-3 survived at pH 3.0, 2.5 and 2.0, respectively. More than $10^{7}$ / CFU/ml survived when exposed with the number of $10^{8}$ CFU/ml at pH 2.0 after 12 hr. L.fermenum YL-3 maintained growth in MRS broth containing 0.3, 0.5, 1.0 and 2.0% oxgall for 24 hr. L.fermenum YL-3 showed an inhibitory effect against pathogenic strains of Sal. enteritidis and E. coli O157:H7. In mixed culture with L.fermenum YL-3 Sal. enteritidis lost ability com-pletely in 15 hrs and E. coil O157:H7 in 16 hrs.
During the lifecycle of a nuclear power plant many organizations are involved in KOREA. Korea Plant Engineering Co. (KOPEC) participates in the design stage, Korea Hydraulic and Nuclear Power (KHNP) operates and manages all nuclear power plants in KOREA, Dusan Heavy Industries manufactures the main equipment, and a construction company constructs the plant. Even though each organization has a digital data management system inside and obtains a certain level of automation, data sharing among organizations is poor. KHNP gets drawing and technical specifications from KOPEC in the form of paper. It results in manual re-work of definition and there are potential errors in the process. A data warehouse based on a neutral model has been constructed in order to make an information bridge between design and O&M phases. GPM(generic product model), a data model from Hitachi, Japan is addressed and extended in this study. GPM has a similar architecture with ISO 15926 "life cycle data for process plant". The extension is oriented to nuclear power plants. This paper introduces some of implementation results: 1) 2D piping and instrument diagram (P&ID) and 3D CAD model exchanges and their visualization; 2) Interface between GPM-based data warehouse and KHNP ERP system.
The purpose of this study was to pelletize to calendula and salvia seeds with peat moss as basic material for the efficient greening and vegetating of slopes and damaged areas. Also the pelletizing was compressed by spherical types that mixed basic fertilizer of N.;300 mg/l, P.;200 mg/l, K.;400 mg/g and plant growth regulator of A and NAA each 300PPM. Soil and soil surface seeding methods were researched to find the growing state of germination percent, germination date, germination force, length of leaf, number of leaf, width of leaf, length of plant, and etc. Comparing with controlled pelletizing, peat moss and +GA pelletizing treatments resulted in higher from two to three times as following growing states : length of leaves, number of leaves, width of leaves, length of plants, length of roots, fresh weights, and ratio of germination. Especially the two treatments above showed four more times effects than the +NNA treatment. Also their germinating date germinating force were earlier and stronger. The surface seeding method was superior to soil seeding.
본 연구는 흑미(복합)발효물 및 분획물의 항산화 성분(총페놀, 총플라보노이드 및 proanthocyanidin)을 측정하였고 또한, 다양한 in vitro 모델을 이용하여 항산화 활성을 평가하였다. 항산화 활성은 DPPH 라디칼 소거능, ABTS 라디칼 소거능, reducing power 및 FRAP을 이용하였고 아질산염 소거능도 평가하였다. 저분자 분획물은 흑미(복합)발효물 및 고분자 분획물에 비해 유의적으로 높은 항산화 활성을 나타내었다. 흑미(복합)발효물 및 분획물 중에서, 저분자 분획물의 총페놀, 총플라보노이드 및 proanthocyanidin 함량은 각각 $109.2{\pm}2.9$ mg GAE/g, $39.4{\pm}0.8$ mg RE/g 및 $32.9{\pm}1.4$ mg CE/g으로 흑미(복합)발효물 및 고분자 분획물에 비해 높은 함량을 보였고 이들 항산화 성분들은 저분자 분획물의 항산화 활성과 유의적인 상관관계를 보였다. 건강증진에 대한 소비자들의 높은 관심도를 고려해 볼 때, 흑미(복합)발효물 및 분획물은 건강기능식품, 건강증진 및 의약품 소재로서 이용이 가능할 것으로 사료된다.
Biodegradable polymeric microspheres were studied for their usefulness as carriers for the delivery of vaccine antigens. However, protein antigen could be denatured during microencapsulation processes due to the exposure to the organic phase and stress condition of cavitation and shear force. Therefore this study was carried out to re-evaluate the degree of protein denaturation during microencapsulation with poly(lactide-co-glycolide) (PLGA) copolymer. PLGA microspheres containing ovalbumin (OVA), prepared by W/O/W multiple emulsification method, were suspended in pH 7.4 PBS and incubated with shaking at $37.5^{\circ}C$. Drug released medium was collected periodically and analyzed for protein contents by micro-BCA protein assay. In order to evaluate the protein integrity, release medium was subjected to the analyses of SDS-PAGE and size exclusion chromatography (SEC). And enzyme-linked immunosorbent assay (ELISA) was introduced to measure the immunoreactivity of entrapped OVA and to get an insight into the three-dimensional structure of epitope. The structures of entrapped protein were not affected significantly by the results of SDS-PAGE and SEC. However, immunoreactivity of released antigen was varied, revealing the possibility of protein denaturation in some microspheres when it was evaluate by ELISA method. Therefore, in order to express the degree of protein denaturation, antigenicity ratio (AR) was obtained as follows: amount of immunoreactivity of OVA/total amount of OVA released ${\times}100(%)$. ELISA method was an efficient tool to detect a protein denaturation during microencapsulation and the comparison of AR values resulted in more accurate evaluation for immunoreactivity of entrapped protein.
Nitric oxide (NO) is a small molecule (mol. wt. 30 Da) and oxidative free radical. It is uncharged and can therefore diffuse freely within and between cells across membrane. Such characteristics make it a biologically important messenger in physiologic processes such as neurotransmission and the control of vascular tone. NO is also highly toxic and is known to acts as a mediator of cytotoxicity during host defense. NO is synthesized by nitric oxide synthase (NOS) through L-arginine/nitric oxide pathway which is a dioxygenation process. NO synthesis involves several participants, three co-substrates, five electrons, five co-factors and two prosthetic groups. Under normal condition, low levels of NO are synthesized by type I and III NOS for a short period of time and mediates many physiologic processes. Under condition of oxidant stress, high levels of NO are synthesized by type II NOS and inhibits a variety of metabolic processes and can also cause direct damage to DNA. Such interaction result in cytostasis, energy depletion and ultimately cell death. NO has the potential to interact with a variety of intercellular targets producing diverse array of metabolic effects. It is known that NO is involved in hemodynamic regulation, neurogenic inflammation, re-innervation, management of dentin hypersensitivity on teeth. Under basal condition of pulpal blood flow, NO provides constant vasodilator tone acting against sympathetic vasoconstriction. Substance P, a well known vasodilator, was reported to be mediated partly by NO, while calcitonin-gene related peptide has provided no evidence of its relation with NO. This review describes the roles of NO in dental pulp in addition to the known general roles of it.
Background: The prognostic factors for patients with full-thickness rotator cuff tears (RCTs) include tear size, muscle atrophy and fatty infiltration. However, the influence of early coexisting degenerative changes on RCT outcomes is unappreciated. The purpose of this study was to calculate the impact that pre-existing partial glenohumeral cartilaginous changes have on patients undergoing arthroscopic RCT repair. Methods: A study of 54 patients undergoing arthroscopic RCT repair was undertaken. The presence of co-existing patches of glenohumeral degenerative cartilaginous changes and RCT size was recorded at surgery. Pre- and postoperative outcomes were assessed using traditional (Oxford Shoulder Score [OSS], 5-level EuroQol-5D [EQ-5D-5L] questionnaire and EuroQol visual analog scale [EQ-VAS]) and patient-centric re-formatted prisms. Outcomes were assessed as an entire dataset, and sub-group analysis was performed according to the grade of co-existing arthritis and tear size. Results: Significant improvements (p<0.05) in clinical outcomes were recognized when assessed using either the traditional or reformatted prisms (average % improvements in OSS, EQ-5D-5L and EQ-VAS were 47%, 33% and 43%, respectively; average improvements in pain, function, and psychological well-being were 48%, 33% and, 29%, respectively). Positive gain was noted in all sub-groups of arthritic grading and tear size. Conclusions: Good clinical outcomes can be achieved following RCT repair even in the presence of local partial degenerative cartilage changes and advancing tear size. These benefits are patient-centered but require RCT repairability.
Acyl-coenzyme A (CoA):diacylglycerol acyltransferase 2 (DGAT2) catalyzes the last stage of triacylglycerol (TAG) synthesis, a process that forms ester bonds with diacylglycerols (DAG) and fatty acyl-CoA substrates. The enzymatic role of Dgat2 has been studied in various biological species. Still, the full description of how Dgat2 channels fatty acids in skeletal myocytes and the consequence thereof in glucose uptake have yet to be well established. Therefore, this study explored the mediating role of Dgat2 in glucose uptake and fatty acid partitioning under short interfering ribonucleic acid (siRNA)-mediated Dgat2 knockdown conditions. Cells transfected with Dgat2 siRNA downregulated glucose transporter type 4 (Glut4) messenger RNA (mRNA) expression and decreased the cellular uptake of [1-14C]-labeled 2-deoxyglucose up to 24.3% (p < 0.05). Suppression of Dgat2 deteriorated insulin-induced Akt phosphorylation. Dgat2 siRNA reduced [1-14C]-labeled oleic acid incorporation into TAG, but increased the level of [1-14C]-labeled free fatty acids at 3 h after initial fatty acid loading. In an experiment of chasing radioisotope-labeled fatty acids, Dgat2 suppression augmented the level of cellular free fatty acids. It decreased the level of re-esterification of free fatty acids to TAG by 67.6% during the chase period, and the remaining pulses of phospholipids and cholesteryl esters were decreased by 34.5% and 61%, respectively. Incorporating labeled fatty acids into beta-oxidation products increased in Dgat2 siRNA transfected cells without gene expression involving fatty acid oxidation. These results indicate that Dgat2 has regulatory function in glucose uptake, possibly through the reaction of TAG with endogenously released or recycled fatty acids.
연구 목적: 본 연구 목적은 서로 다른 재료로 제작된 지대주를 이용하여 동적하중을 가했을 때 풀림토크와 침하량에 대한 임플란트-지대주 연결부의 안정성에 대하여 알아보는 것이다. 연구 재료 및 방법: 원추형 내부연결 구조를 갖는 임플란트에 이용되는 세 가지 지대주, 티타늄 합금 지대주(Cement abutment, Osstem Co., Seoul, Korea), 귀금속 UCLA 지대주(UCLA Goldcast abutment, Osstem Co., Seoul, Korea), 비귀금속 UCLA 지대주(CCM Metalcast abutment, 3M ESPE, Seefeld, Germany)를 사용하였다. 귀금속 UCLA 지대주와 비귀금속 UCLA 지대주를 티타늄 지대주와 유사한 형태로 납형을 형성 한 후 각각 제 3형 금합금(E-3, Heesung catalyst Co., Seoul, Korea)와 니켈-크롬 합금인(Rexillium $III^{(R)}$, $Pentron^{(R)}$, San Diego, USA)를 사용하여 주조 하였다. 임플란트와 지대주를 디지털 토크 컨트롤러를 이용하여 30 N cm로 체결하였고 전하중 상실을 보상하기 위하여 10분 후 다시 같은 힘으로 체결 하였다. 디지털 토크 게이지를 사용하여 풀림토크를 측정하였고 디지털 마이크로미터를 이용하여 임플란트/지대주 길이를 측정하였다. 그 후 동적 하중 피로 시험기에 지대주를 장착한 후 최대 250 N, 최소 25 N, 주기 14 HZ의 sine형의 하중을 장축과 평행하게 가하였다. 하중 후 임플란트/지대주의 길이를 재측정 하고, 전후 길이변화량을 이용하여 침하량을 계산하였으며 풀림토크를 측정하였다. 결과: 하중 전후의 길이를 비교하였을 때 세 군 모두에서 유의한 차이를 보였다($P$<.05). 군 간의 침하량을 비교하였을 때 통계적으로 유의한 차이를 보이지는 않았다. 하중 전후의 풀림토크를 비교하였을 때 역시 세 군 모두에서 유의한 차이를 보이며 감소하였다 ($P$<.05). 하중 전 풀림토크와 하중 후 풀림토크를 군간 비교하였을 경우에는 통계적으로 유의한 차이를 보이지는 않았다. 결론: 원추형 내부 연결구조를 갖는 임플란트에서 동적하중은 임플란트의 침하량과 풀림토크에 영향을 미치는 것으로 보이나 지대주간 차이는 나타나지 않았다. 비귀금속UCLA 지대주이 사용은 귀금속 UCLA 지대주와 마찬가지로 기능적 하중에서 안정적일 것으로 사료된다.
