Sepsis induces variation of intestinal barrier function in different phase through nuclear factor kappa B signaling

  • The intestinal barrier function disrupted in sepsis, while little is known about the variation in different phases of sepsis. In this study, mouse models of sepsis were established by caecal ligation and puncture (CLP). The H&E staining of sections and serum diamine oxidase concentration were evaluated at different timepoint after CLP. TUNEL assay and EdU staining were performed to evaluate the apoptosis and proliferation of intestinal epithelium. Relative protein expression was assessed by Western blotting and serum concentrations of pro-inflammatory cytokines was measured by ELISA. The disruption of intestinal barrier worsened in the first 24 h after the onset of sepsis and gradually recovered over the next 24 h. The percentage of apoptotic cell increased in the first 24 h and dropped at 48 h, accompanied with the proliferative rate of intestinal epithelium inhibited in the first 6 h and regained in the later period. Furthermore, the activity of nuclear factor kappa B (NF-κB) presented similar trend with the intestinal barrier function, shared positive correction with apoptosis of intestinal epithelium. These findings reveal the conversion process of intestinal barrier function in sepsis and this process is closely correlated with the activity of NF-κB signaling.

Interferon-β alleviates sepsis by SIRT1-mediated blockage of endothelial glycocalyx shedding

  • Sepsis is a life-threatening multi-organ dysfunction with high mortality caused by the body's improper response to microbial infection. No new effective therapy has emerged that can adequately treat patients with sepsis. We previously demonstrated that interferon-β (IFN-β) protects against sepsis via sirtuin 1-(SIRT1)-mediated immunosuppression. Another study also reported its significant protective effect against acute respiratory distress syndrome, a complication of severe sepsis, in human patients. However, the IFN-β effect cannot solely be explained by SIRT1-mediated immunosuppression, since sepsis induces immunosuppression in patients. Here, we show that IFN-β, in combination with nicotinamide riboside (NR), alleviates sepsis by blocking endothelial damage via SIRT1 activation. IFN-β plus NR protected against cecal ligation puncture-(CLP)-induced sepsis in wild-type mice, but not in endothelial cell-specific Sirt1 knockout (EC-Sirt1 KO) mice. IFN-β upregulated SIRT1 protein expression in endothelial cells in a protein synthesis-independent manner. IFN-β plus NR reduced the CLP-induced increase in in vivo endothelial permeability in wild-type, but not EC-Sirt1 KO mice. IFN-β plus NR suppressed lipopolysaccharide-induced up-regulation of heparinase 1, but the effect was abolished by Sirt1 knockdown in endothelial cells. Our results suggest that IFN-β plus NR protects against endothelial damage during sepsis via activation of the SIRT1/heparinase 1 pathway.

Identification of Novel Salt Stress-responsive Genes Using the Activation Tagging System in Arabidopsis (애기장대에서 activation tagging system을 이용한 새로운 고염 스트레스 반응 유전자의 동정)

  • Abiotic stresses limit the growth and productivity of plants. Cellular adaptation to abiotic stresses requires coordinated regulation in gene expression directed by complex mechanisms. This study used the activation tagging system to identify novel salt stress-responsive genes. The study selected 9 activation tagging lines that showed salt stress-tolerant phenotypes during their germination stages. Thermal asymmetric interlaced-PCR (TAIL-PCR) was used to identify the T-DNA tagging sites on the Arabidopsis genome in selected activation tagging lines, including AT7508, AT7512, AT7527, AT7544, AT7548, and AT7556. RT-PCR analysis showed that ClpC2/HSP93-III (At3g48870), plant thionin family (At2g20605), anti-muellerian hormone type-2 receptor (At3g50685), vacuolar iron transporter family protein (At4g27870), and microtubule-associated protein (At5g16730) were activated in AT7508, AT7512, AT7527, AT7544, and AT7556, respectively. Interestingly, in AT7548, both the genes adjacent to the T-DNA insertion site were activated: Arabinogalactan protein 13 (AGP13) (At4g26320) and F-box/RNI-like/FBD-like domains-containing protein (At4g26340). All of the seven genes were newly identified as salt stress-responsive genes from this study. Among them, the expression of ClpC2/HSP93-III, AGP13, F-box/RNI-like/FBD-like domains-containing protein gene, and microtubule-associated protein gene were increased under salt-stress condition. In addition, AT7508, AT7527, and AT7544 were more tolerant to salt stress than wild type at seedling development stage, functionally validating the screening results of the activation tagging lines. Taken together, our results demonstrate that the activation tagging system is useful for identifying novel stress-responsive genes.

Non-Invasive Environmental Detection using Heat Shock Gene-Green Fluorescent Protein Fusions

  • Three 'stress probe' plasmids were constructed and characterized which utilize a green fluorescent protein (CFP) as a non-invasive reporter to elucidate Escherichia coli cellular stress responses in quiescent or 'resting' cells. Facile detection of cellular stress levels was achieved by fusion of three heat shock stress protein promoter elements, those of the heat shock transcription factor ${\sigma}^{32}$, pretense subunit ClpB, and chaperone DnaK, to the reporter gene $gfp_{uv}$. When perturbed by chemical or physical stress (such as heat shock, nutrient (amino acid) limitation, addition of IPTG, acetic acid, ethanol, phenol, antifoam, and salt (osmotic shock), the E. coli cells produced GFPuv which was easily detected from within the cells as emitted green fluorescence. A temporal and amplitudinal mapping of these responses was performed, demonstrating regions where quantitative delineation of cell stress was afforded.

  • FR과 ATM은 서로 다른 트래픽 파라메타를 사용하기 때문에 연동하기 위해서는 트래픽 파라메타를 서로 변환하는 기능이 필요하다. 변환식 FR과 ATM을 경유하면서 종단 단말간의 서비스 품질이 보장되고 또한 망 자원을 효율적으로 사용할 수 있어야 한다. 변환식에는 여러 조합이 고려된다. 즉 FR/ATM 연동 방식에서는 망 연동과 서비스 연동으로 구분되고, 망 연동에서 다중화 방식에 따라 1-새-1과 N-to-1 방식이 가능하다. 또 FR 트래픽은 ATM에서 CBR, VBR, GFR 등의 ATC로 대응시킬 수 있고, 각 ATC는 CLP에 따른 대역의 조합 구성이 가능하다. ATM 대역 계산에서도 FR 연결의 최대 대역을 AR 또는 CIR+EIR로 사용할 수 있다. 기존 ITU-T 방식은 서비스 연동만으로 제한하였고, ATM Forum 방식은 가능한 방법을 나열함으로써 연동 시스템 간의 기능 차이에 의한 선택이 어렵다. 또한 변환식을 실제로 적용하기 위해서는 기존 방식에서 고찰되지 않은 많은 문제점들이 있다. 따라서 본 고에서는 기존 방식을 고찰하고, 최근의 연구 및 기술 동향을 반영한 변환 방식을 제안하고, 가상 AR 적용과 운용시의 변환식 선택 및 고려할 점 등에 대하여 기술한다.

Performance Analysis of the Multiplexer for Shaped Video Traffic Sources (쉐이퍼를 사용한 비디오 트래픽원의 다중화기 성능 분석)

  • This paper considers the problem of performance analysis for the multiplexer, when homogeneous periodic on-off sources are statistically multiplexed. Periodic on-off source model is defined that on-period and off-period are repeated by once in a deterministic periodic time and in on-period, cell arrives with deterministic time interval. In order to reflect periodicity of source model, we consider two multiplexing situation, such as random multiplexing and synchronized multiplexing. In both case, we obtain the overload-period distribution in the multiplexer, and an approximate method using the overload period distribution is suggested for obtaining the CLP(Cell Loss Probability) in the buffer of the multiplexer. A numerical example using MPEG-I real traffic samples and the results are also presented.

  • In this paper, we propose an enhanced usage parameter control algorithm, which is one of the preventive traffic control method in ATM networks and implemented it with VLSI Proposed algorithm is based on the cell loss priority bit in the ATM cell header. This algorithm can solve the measurement phasing problem in cell conformance testing in ATM networks. Proposed algorithm can minimize the cell loss ratio of high priority cell(CLP=0) and resolve the burstiness of cells which may be introduced in traffic multiplexing and demultiplexing procedure. The result of performance evaluation shows that the performance of proposed algorithm is better than that of ITU-T usage parameter control algorithm.

  • In order to build and manage an ATM network effectively under several types of control methods, it is necessary to estimate the performance of the equipments in various viewpoints, especially of ATM multiplexer. As for the method to model the input stream into the ATM multiplexer, many researches have been done to characterize it by, such as, fluid flow, MMPP(Markov Modulated Poisson Process), or MMDP (Markov Modulated Deterministic Process). We introduce an MRP(Markov Renewal Process) to model the input stream which has proper structure to represent the burst traffic with high correlation. In this paper, we build a model for aggregated heterogeneous ON-OFF sources of ATM traffic by MRP. We make discrete time MR/D/1/B queueing system, whose input process is the superposed MRP and present a performance analysis by finding CLP(Cell Loss Probability). A simulation is done to validate our algorithm.

Protective Effect of Decursinol on Mouse Models of Sepsis: Enhancement of Interleukin-10

  • The effects of decursinol on various models of sepsis were investigated. Intra-peritoneal pretreatment of mice with various doses of decursinol ($1{\sim}100$ mg/kg) effectively suppressed lethality induced in three mouse models of experimental sepsis, i.e., lipopolysaccharide (LPS)/D-galactosamine (GalN), high-dose LPS (20 mg/kg), and cecal ligation and puncture (CLP). Intra-peritoneal pretreatment of mice with decursinol (50 mg/kg) markedly enhanced the LPS/GalN -induced increase of plasma interleukin-10 (IL-10) levels, without affecting plasma TNF-${\alpha}$, IL-6 and IL-12 levels. These results suggest that decursinol could be effective for prevention or treatment of sepsis.

  • Sepsis remains the leading cause of morbidity and mortality following trauma. Although hepatocellular dysfunction occurs during trauma and sepsis. the mechanism responsible for this remains unclear. We investigated the role of Kupffer cells in the alterations in microsomal drug metabolizing function during trauma and sepsis. Rats were subjected to trauma by femur fracture (FFx). After 72h, polymicrobial sepsis was induced by cecal ligation and puncture(CLP). (omitted)
