Clitocybe aurantiaca 균주가 생산하는 주름개선소재 clitocybin A의 대량 발효생산 및 MMP-1 발현저해활성 (Fermentation Process for Mass Production of Clitocybin A, a New Anti-Wrinkle Agent from Clitocybe aurantiaca and Evaluation of Inhibitory Activity on Matrix Metalloproteinase-1 Expression)

  • 본 연구에서는 국내 자생버섯의 일종인 Clitocybe aurantiaca KCTC11143BP 균주가 생산하는 주름개선 화장품 신소재 clitocybin A의 발효생산 최적조건, 추출정제조건, 세포독성 및 자외선 조사에 따른 MMP-1 발현 저해활성을 규명하였다. C. aurantiaca 균주를 5 L 발효조를 이용하여 회분식으로 배양하였을 경우, PD 액체배지보다 YM 액체배지에서 양호하게 생육하는 것을 확인하였다. 300 L 용량의 대형 발효조에서 modified된 YM 배지를 이용한 유가식 배양으로 14일간 배양한 결과, 12.5 kg/120 L의 총 균체량을 얻었다. 발효 추출물로부터 항노화 소재인 clitocybin A를 추출정제를 한 후 HPLC를 실시하여 배양 4일째부터 clitocybin A가 생산되고 있음을 확인하였다. Clitocybin A 화합물의 세포독성을 확인하기 위하여 MTT assay를 실시한 결과, 100 ${\mu}g/ml$ 농도로 처리하였을 경우 $134.6{\pm}10.4%$로 오히려 세포가 증식하여 안전한 소재임을 확인하였다. 반면, 대조군으로 사용한 oleanolic acid는 25 ${\mu}g/ml$ 농도에서도 강한 세포독성을 나타냈다. 또한, clitocybin A는 100 ${\mu}g/ml$ 처리시 33.1%의 MMP-1 발현 저해활성 보여 주름개선 기능성화장품 소재로 매우 우수한 화합물임을 확인하였다.

Clitocybin A의 모유두 세포증식 효능 (Effect of Clitocybin A on the Proliferation of Dermal Papilla Cells)

  • The present study was conducted to evaluate the hair growth-promoting effect of Clitocybin A from mushroom Clitocybe aurantiaca with dermal papilla cells (DPCs), which play important roles in the regulation of hair cycle. Clitocybin A significantly increased the proliferation of immortalized rat vibrissa DPCs. Flow cytometry analysis revealed that Clitocybin A promoted cell-cycle progression through G0/G1 to S phase in immortalized rat vibrissa DPCs. In addition, Clitocybin A increased the level of cell cycle proteins such as cyclin D1, phospho-pRB, and phospho-CDK2. To elucidate the molecular mechanisms of Clitocybin A on the proliferation of DPCs, we examined the activation of wnt/${\beta}$-catenin signaling which is known to regulate hair follicle development, differentiation and hair growth. Clitocybin A activated wnt/${\beta}$-catenin signaling via the increase of phospho(ser552)-${\beta}$-catenin, phospho(ser675)-${\beta}$-catenin and phospho(ser9)-$GSK3{\beta}$. Furthermore, Clitocybin A markedly increased the activation of extracellular signal-regulated kinase (ERK). These results suggest that the Clitocybin A may induce hair growth by proliferation of DPCs via cell-cycle progression as well as the activation of Wnt/${\beta}$-catenin signaling and ERK pathway.

ROS Scavenging and Anti-Wrinkle Effects of Clitocybin A Isolated from the Mycelium of the Mushroom Clitocybe aurantiaca

  • Clitocybin A, an isoindolinone from Clitocybe aurantiaca, was investigated to assess its anti-wrinkle properties, through reactive oxygen species (ROS)-scavenging and elastase inhibitory activities, procollagen synthesis, and matrix metalloproteinase-1 (MMP-1) expression, in human primary dermal fibroblast-neonatal (HDF-N) cells. Clitocybin A exhibited no significant cytotoxicity up to 10 ppm in HDF-N cells, with cell viability and cell proliferation activity greater than 94.6% and 91.9%, respectively. Strong and concentration-dependent ROS radical scavenging activities of clitocybin A were observed following irradiation with UVB at $30mJ/cm^2$. Furthermore, clitocybin A treatment of cells at 0.1, 1, and 10 ppm exhibited decreased elastase activity, in a concentration-dependent manner, by 1.97%, 6.6%, and 8.31%, respectively, versus the control group. The effects of clitocybin A on procollagen synthesis and MMP-1 expression were investigated. Clitocybin A treatment of cells at 1, 5, and 10 ppm increased procollagen synthesis, by 67.9%, 74.4%, and 112.9%, respectively, versus the control group. At these concentrations, MMP-1 expression decreased significantly following UV irradiation. Together, these findings suggest that clitocybin A may be an effective ingredient for use in anti-wrinkle cosmetic products.

Clitocybin D, a Novel Human Neutrophil Elastase Inhibitor from the Culture Broth of Clitocybe aurantiaca

  • Clitocybin D, a novel human neutrophil elastase inhibitor, was isolated from the culture broth of Clitocybe aurantiaca. This compound was purified by solvent extraction, silica gel column chromatography, Sephadex LH-20 column chromatography, and preparative HPLC. The compound was determined to be 4-(4,6-dihydroxy-3-methoxy-3H-isoindol-1-yl)-benzoic acid on the basis of 1D and 2D NMRs and MS spectroscopic analysis. Analysis of the human neutrophil elastase (HNE) inhibitory activity of the isolated compound revealed that it showed significant HNE inhibitory activity with an $IC_{50}$ value of $17.8{\mu}M$.
