Runoff Analysis using ModClark Model (ModClark 모형을 이용한 유출 해석)

  • The purpose of the present study is examining the changes of runoff characteristics and extracting hydrologic parameters by applying ModClark model on grid divided watershed. Bocheong stream basin in Geum River system, one of the representative watersheds of IHP projects, is selected. Hydrology-based topographical informations are calculated using GIS data in the HEC-GeoHMS V1.1 extension in Arcview 3.2. The ModClark model requires precipitation data in a gridded format. The gridded data must be recorded in the HEC Data Storage System file format. Therefore, kriging method was used to interpolate the point values to create a grid that gives each cell over the entire watershed a precipitation value. Hec-DSSVue program was used to create DSS file for the rain gage data. The completed HEC-HMS model was calibrated for use in simulating three measured storm events and cell size of 10000m, 5000m, 2000m, 1000m was chosen for the application. As the result of applying distributed rainfall-runoff model to analyze relatively good agreement for peak discharge, runoff volume and peak time.

  • Measuring of the dissolved oxygen is widely used for the environmental control of natural waters, sewage waste treatment, medical and biochemical studies, soil husbandry, food and drug process control, and prevention of corrosion in boilers. Especially, a power plants need special management for preventing accidents from corrosion, therefore, it is essential to measure the concentration of dissolved oxygen in real-time. In this paper we present a method of measuring dissolved oxygen very accurately up to pub units. This method, called polarograph method, is based on the measures of the electric current generated by the oxidation process in cathode and de-oxidation process in anode, assuming that the amount of the current is proportional to the density of dissolved oxygen.

A Study on the Microfabricated Clark-type Sensor for Measuring Dissolved Oxygen (용존 산소 측정용 초소형 Clark-type 센서에 대한 연구)

  • This paper presents a microfabricated Clark-type sensor which exactly can measure dissolved oxygen in the cell containing solution. We designed, fabricated, and characterized a microfabircated Clark-type oxygen sensor for measuring dissolved oxygen. The microfabricated oxygen sensor consists of 3-electrodes on a glass substrate, a FEP (Fluorinated ethylene propylene) oxygen-permeable membrane, and PDMS (Polydimethylsiloxane) reservoir for storing sample solution. Thin-film Ag/AgCl was employed as a reference electrode and its durability was verified by obtaining a stable open circuit potential for 2 hours against a commercial Ag/AgCl electrode and a stable cyclic voltammetry curve. Selectivity, response time, and linearity of the fabricated oxygen sensor were also verified well by cyclic voltammetry and amperometry depending. The fabricated oxygen sensor showed a 90% response time of 40sec and an excellent linearity with a correlation coefficient of 0.994.

Decision of GIS Optimum Grid on Applying Distributed Rainfall-Runoff Model with Radar Resolution (레이더 자료의 해상도를 고려한 분포형 강우-유출 모형의 GIS 자료 최적 격자의 결정)

  • Changes in climate have largely increased concentrated heavy rainfall, which in turn is causing enormous damages to humans and properties. Therefore, the exact relationship and the spatial variability analysis of hydrometeorological elements and characteristic factors is critical elements to reduce the uncertainty in rainfall -runoff model. In this study, radar rainfall grid resolution and grid resolution depending on the topographic factor in rainfall - runoff models were how to respond. In this study, semi-distribution of rainfall-runoff model using the model ModClark of Inje, Gangwon Naerin watershed was used as Gwangdeok RADAR data. The completed ModClark model was calibrated for use DEM of cell size of 30m, 150m, 250m, 350m was chosen for the application, and runoff simulated by the RADAR rainfall data of 500m, 1km, 2km, 5km, 10km from 14 to 17 on July, 2006. According to the resolution of each grid, in order to compare simulation results, the runoff hydrograph has been made and the runoff has also been simulated. As a result, it was highly runoff simulation if the cell size is DEM 30m~150m, RADAR rainfall 500m~2km for peak flow and runoff volume. In the statistical analysis results, if every DEM cell size are 500m and if RADAR rainfall cell size is 30m, relevance of model was higher. Result of sensitivity assessment, high index DEM give effect to result of distributed model. Recently, rainfall -runoff analysis is used lumped model to distributed model. So, this study is expected to make use of the efficiently decision criteria for configurated models.

Sesquiterpene Lactones from Saussurea lappa and Their Cell Proliferation Effects on Human Breast Cell Lines (목향(木香)으로부터 분리된 Sesquiterpene 화합물의 암세포 생육저해에 대한 활성)

  • Saussurea Radix, the dried roots of Saussurea lappa Clark (Compositae), has been used in oriental traditional medicine for aromatic stomachic. Present study was carried out for the anticancer effect of Saussurea Radix. Bioassay-directed fractionation of Saussurea Radix led to the isolation of two sesquiterpenes, dehydrocostuslactone (1) and costunolide (2). The structures of 1 and 2 were elucidated by spectral methods (MS, IR, $^1$H and $^{13}$C NMR). These compounds showed a potent cell proliferation activity against human breast cancer cell MCF-7 and MDA-MB-453.

Experimental Study on Aerodynamic Characteristics of Morphing Airfoil Configuration (모핑 에어포일 형상의 공력특성 실험연구)

  • The present paper is the preliminary study of the development of a morphing aircraft wing and investigates experimently the aerodynamic characteristics of a base airfoil and a morphing airfoil. The wind tunnel tests are conducted for a base Clark-Y airfoil, an airfoil with a mechanical flap, and a morphing airfoil. Lifts, drags, and pitching moments are measured by using a three-axis load cell and they are calibrated by considering solid blockage and wake blockage. The wind tunnel tests are conducted for various air speeds, Reynolds' numbers, and angles of attack. The experimental results show that the aerodynamic characteristics of the morphing airfoil in lift-drag and lift-pitching moment are better than those of the airfoil with a mechanical flap.

Survival Association and Cell Cycle Effects of B7H3 in Neuroblastoma

  • Objective : The function of B7H3, a member of the B7 family of proteins, in neuroblastoma (NB) remains poorly characterized. Here we examine the expression pattern of B7H3 in clinical NB specimens and characterize the phenotype of B7H3 knock-down in NB cell line. Methods : Immunohistochemical (IHC) staining was carried out to assess the expression of B7H3 in clinical NB specimens. Survival association was analyzed using five Gene Expression Omnibus (GEO) datasets (GSE85047, GSE45480, GSE62564, GSE16476, GSE49710). Clonogenic survival and flow cytometry were performed after B7H3 knockdown to assess the cellular proliferation and cell survival in vitro. Impact of B7H3 silencing on NB growth was examined in vivo using the SH-SY5Y xenograft model. Results : On IHC staining, B7H3 was widely expressed in clinical NB specimens. Analysis of the transcriptional profiles of five GEO datasets clinically annotated NB specimens revealed that decreased B7H3 expression was associated with improved overall survival. B7H3 knockdown suppressed the proliferation of the SH-SY5Y NB model in vitro and in vivo. Cell cycle analysis revealed that B7H3 silencing induced G1/S arrest. This arrest was associated with the suppression of E2F1 expression and induction of Rb expression. Conclusion : Our results demonstrate that B7H3 expression correlate with clinical survival in NB patients. Preliminary studies suggest that B7H3 may mediate the G1/S transition.

Effects of Ginseng Extract on Excitable Cell Membrane Potential (인삼추출물이 흥분성세포의 막전압에 미치는 영향)

  • Studies have been conducted to test the effect of Ginseng alcohol extract on the membrane potentials of frog skeletal muscle. The gastrocnemius muscle was isolated and placed in a chamber containing the Clark-frog Ringer solution. Membrane potentials were recorded using microelectrodes filled with 3 M KCI and muscle was electrically stimulated to obtain action potential. Changes in both the action potential and the resting membrane potential were observed after adding an appropriate amount of Ginseng alcohol extract in the perfusing Ringer solution. The results obtained from 346 muscle cells are summarized as follows : 1) The average resting membrane potential of the normal frog gastrocnemius muscle cell was -92.8 mV and the peak of the action potential reached at 29.8 mV. 2) Both the resting membrane potential and the peak of the action potential decreased by Ginseng alcohol extract, the effect being proportional to the dose of Ginseng alcohol extract. 3) The resting membrane potential and the peak of the action potential continuously decreased until about 40 min after Ginseng addition and leveled off thereafter. The potentials recovered to its original value after Ginseng was washed out. 4) The resting membrane potential was more sensitive to the Ginseng alcohol extract than was the action potential. These results strongly suggest that Ginseng alcohol extract increases both the $Na^+$ and $K^+$ permeability in the skeletal muscle cell membrane.

Co-Expression of Putative Cancer Stem Cell Markers, CD133 and Nestin, in Skin Tumors

  • Background: Cancer stem cells (CSC) are populations of cells responsible for tumor initiation, progression and therapeutic resistance in many cancers. In the present study, we aimed to investigate the expression pattern and clinical significance of two CSC markers, CD133 and Nestin, in a series of skin tumors. Materials and Methods: One hundred and thirteen paraffin blocks from skin cancers including 16 (14%) cases of melanoma, 37 (33%) of squamous cell cancer (SCC) and 60 (53%) of basal cell cancer (BCC) were collected and assembled in a tissue microarray (TMA). The samples were immunohistochemically examined for the expression of CD133 and Nestin. Expression of these markers was also correlated with clinicopathological parameters. Results: A significant difference was observed in the expression of CD133 and Nestin in melanomas, SCC and BCC (p value=0.001). Furthermore, the level of expression was significantly higher in the melanomas compared to the SCC and BCC tumors. Expression of CD133 in the melanoma was significantly associated with increased tumor invasiveness (p value=0.05), a higher rate of metastasis (p value=0.04) and the presence of ulceration (p value=0.02). Increased expression of Nestin was observed in metastatic melanoma (p value=0.04), while no statistically significant correlation was found with other clinicopathological parameters including Breslow thickness, Clark level and ulceration. Conclusions: Elevated expression levels of CD133 and Nestin in the melanomas are associated with advanced disease, with more aggressive and metastatic skin tumors. Therefore, these markers could be potential therapeutic targets for malignant tumors of the skin.
