Monoamine Oxidase B Inhibitors from the Fruits of Opuntia ficus-indica var. saboten

  • Three varieties of methyl citrate and 1 -methyl malate were isolated from the fruits of Opuntia ficus-indica var. saboten Makino through in vitro bioassay-guided isolation for the inhibition on monoamine oxidase(MAO). The $IC_50$ values for MAO-B of 1-monomethyl citrate, 1,3-dimethy citrate, trimethyl citrate and 1-methyl malate were 0.19, 0.23, 0.61 and 0.25 mM, respectively. However, on MAO-A, their inhibitions showed only marginal activity.

  • We quantitated the major chemical components of ramie leaves (Boehmeria nivea L.) powder. The proximate compositions (all w/w) was 5.42% moisture, 28.15% crude protein, 6.95% crude fat, 15.27% crude ash, and 54.79% carbohydrate, respectively. The total, insoluble, and soluble dietary fiber contents were $39.66{\pm}1.84g/100g$, $20.32{\pm}2.02g/100g$, and $19.34{\pm}2.84g/100g$, respectively. The major free sugars were glucose, galactose and lactose. Seventeen amino acids were isolated. Essential amino acids constituted 44.65% of the total. When free amino acid levels were evaluated, 25 kinds of components were detected, of which 18.15% were essential. Only caproic acid methyl ester and pentadecanoic acid were detected when fatty acid examined.   • Phytochemical investigation of leaves, barks and roots of Nauclea vanderguchtii led to the isolation of sixteen compounds, which includes one citric acid derivative (2), one alkaloid (16), one peptide derivative (3), and twelve triterpenes (1, 4 - 13). These compounds were identified as rotundanonic acid (1), 2-hydroxy-1,2,3-propanetricarboxylic acid 2-methyl ester (2), asperphenamate (3), lupeol (4), stigmasterol (5), betulin (6), betulenic acid (7), stigmasterol 3-O-β-D-glucopyranoside (8), quinovic acid 3β-O-α-L-rhamnoside (9), α-amyrin (10), 3-oxoquinovic acid (11), ursolic acid (12), hederagenin (13), rotundic acid (14), clethric acid (15), and naucleficine (16) by the analysis of their NMR spectroscopic data including 2D NMR spectra and by comparison of their spectroscopic data reported in the literature. Compounds 1 and 3 were isolated for the first time in the genus Nauclea, and compound 2 was isolated for the first time from the Rubiaceae family. Complete NMR assignations for 1 have been published for the first time.
