Long-Term Exposure of Sildenafil Citrate on Sperm Parameters in Rat

  • Sildenafil citrate (SIL) a phosphodiesterase 5 inhibitor (PDE5I) has been used for long time as a first line oral drug for erectile dysfunction. Though it has beneficial effects on erectile organ it also has some adverse effects in other cells and/or tissues related to reproductive system when exposed to longer duration. The objective of the present study is to evaluate the long term effect of SIL on sperm parameters in Wistar albino rat. The animals are divided into two groups, for group I - rats were treated with saline (vehicle alone) and group - II oral administration of 5 mg/kg b.w. of SIL was administrated orally once in a day for 120 days. At the end of the trial period animals were sacrificed and epididymal sperm were subjected to various analysis. Results showed significant reduction in sperm count, motility, viability and morphologically intact sperm in long term PDE5I exposed animals when compared to control. Acrosomal status and fertility test also showed significant reduction in long term PDE5I exposed animals. The present study clearly indicated that long term SIL has shown to induce alteration in sperm quality and quantity, leading to decline in fertility rate. Indicate that SIL impinge on spermatogenesis as well as epididymal function. Understanding the molecular down-stream events involved in long-term exposure to PDE5 inhibitor can be valuable to supervise on related infertility issues and to suggest corrective measures.

Study on the reducibility of substituted $LaFeO_3$ (치환된 $LaFeO_3$의 환원반응성에 대한 연구)

  • $LaFeO_3$ and substituted $LaFeO_3$ mixed oxides were prepared by Citrate and Cyanide method in air $850^{\circ}C$/24h. These oxides of orthorhombic perovskite were characterized by XRD and IR, but substituted $LaFeO_3$ with 0.5mol Cu at B site was not obtained single phase. Also, reduction reaction of un-substituted $LaFeO_3.17$ were two steps but each site substituted oxides were three steps reactions. These means that new reduction step of each site substituted oxides were atributed tot dopant.

  • We prepared the Au-coated $TiO_2$ (Au/$TiO_2$) nanoparticles by deposition-precipitation (DP) method with and without bases (urea or NaOH) and investigated the effects of pH on the preparation of Au/$TiO_2$ nanoparticles for various kinds of bases. For the DP method without bases, the Au nanoparticles in the diameter of about 50 nm were generated in the solution by the reduction reaction with trisodium citrate and they did not deposit on the surface of $TiO_2$. For the DP method with bases, Au precursors deposited on the surface of $TiO_2$ and then reduced to the Au nanoparticles in the diameter of 4-5 nm on the surface of $TiO_2$ by the reaction with trisodium citrate.

A Study of the Electroless Ni-W-B Depsition on Alumina Ceramics (Alumina Ceramics상의 무전해 Ni-W-B 도금에 관한 연구)

  • Effects of bath composition on electroless deposition of Ni-W-B from sulphate solution were invesrigated in terms of deposition kinetics, electro resistivity and composition of deposit film. The microstruigated and crystataine structure of the films were also studied using a scanning electron microscope and X-ray diffractometer. The deposition rate increased linearly with increasing the concentration of nickel sulphate in bath solution, wheras the rate decreasing with sodium citrate. The rate was also affected by sodium tungstate, which was maaximum at the concentration of 0.06 M/1 in sodium tungstate, The content of W in the deposit increased with increased with increasing the sodium citrate had on opposite effect on the composition of W and B in the deposit. The crystal change film from armorphous to cryatallicne nature by heat treatments was proved by the reduction of specific resistance and X-ray diffration.

  • Hexavalent chromium (Cr(VI)) is recognized to be carcinogenic and toxic and registered as a contaminant in many drinking water regulations. It occurs naturally and is also produced by industrial processes. The reduction of Cr(VI) to Cr(III) has been a central topic for chromium remediation since Cr(III) is less toxic and less mobile. In this study, fermentative Fe(III)-reducing bacterial strains (Cellu-2a, Cellu-5a, and Cellu-5b) were isolated from a groundwater sample and were phylogenetically related to species of Cellulomonas by 16S rRNA gene analysis. One selected strain, Cellu-2a showed its capacity of reduction of both soluble iron (ferric citrate) and solid iron (hydrous ferric oxide, HFO), as well as aqueous Cr(VI). The strain Cellu-2a was able to reduce 15 μM Cr(VI) directly with glucose or sucrose as a sole carbon source under the anaerobic condition and indirectly with one of the substrates and HFO in the same incubations. The heterogeneous reduction of Cr(VI) by the surface-associated reduced iron from HFO by Cellu-2a likely assisted the Cr(VI) reduction. Fermentative features such as large-scale cell growth may impose advantages on the application of bacterial Cr(VI) reduction over anaerobic respiratory reduction.

  • The $La_{0.8}Ca_{0.2}CoO_3$ prepared by a citrate process was shown to have higher oxygen reduction current density and specific activity than $LaCoO_3$, $La_{0.6}Ca_{0.4}CoO_3$. In the cyclic voltammogram, an oxygen desorption peak of a $La_{0.8}Ca_{0.2}CoO_3$+carbon electrode was larger than that of a only carbon electrode. $La_{0.8}Ca_{0.2}CoO_3$ sintered at $900^{\circ}C$ for 5 hours was shown high oxygen reduction current density because of the particle size distribution and sintering effect.

  • Exfoliated graphite oxide (EGO) was prepared by graphite oxide in an aqueous solution of TMAOH. The hybrid graphene/Zn-Al LDH material was fabricated by the hydrothermal reduction of the solution of EGO, $Zn(NO_3)_2{\cdot}6H_2O$, $Al(NO_3)_3{\cdot}9H_2O$, urea, and trisodium citrate. That is, metal ions were absorbed on the surface of EGO, and Zn-Al LDH material was randomly dispersed on the surface of graphene along with a reduction process of EGO to graphene by hydrothermal treatment. The composition, morphology, and thermal property of the obtained graphene-based hybrid material were studied by FE-SEM, EDX, TEM, FT-IR, XRD, TGA, and DSC.

Factors Influencing Siderophore Production by Plant Growth Promoting Rhizopseudomonas Strains (식물생장촉진 근권 Pseudomonas의 Siderophore 생성에 관여하는 요인)

  • This study demonstrates the influence of environmental conditions, other than iron, on pyoverdin production by fluorescent Pseudomosonas. In slightly acidic pH conditions(pH 6), cell yield was reduced while the siderophore production per cell yield was increased. The optimum temperatures for the siderophore production and cell yield was $19^{\circ}C$ and $28^{\circ}C$ for 7NSK2 and $12^{\circ}C$ and $19^{\circ}C$ for ANP15. The carbon and nitrogen balance showed that at low C : N ratio of the growth medium (higher nitrogen concentration), both cell yield and siderophore production was reduced. Use of different carbon sources revealed that citrate as a carbon source facilitated iron uptake and resulted in a significant reduction in siderophore production. However, at the late exponential phase, the iron content in the cell biomass was not significantly different from those grown in glucose or succinate. From these results it can be suggested that the environmental factors other than iron may also influence siderophore production by fluorescent pseudomonas.

  • The influence of (-0-Hydroxycitrate(HCA), shown to be a competitive inhibitor of adenosine 5-triphosphate(ATP) citrate lyase, on food intake and body weight, serum triglyceride and cholosterol level, in vivo rates of fatty acid and cholesterol synthesis, and fat cell number and size was investigated. 3 groups of female, 5 weeks old Sprague Dawley rats, 8 animals each, were ad libitum meal-fed or pair-fed(3 hours from 10 : 00 to 13 : 00) AIN based high glucose diet for a total period of 8 weeks. Providing normolipidemic rats orally with 400mg of HCA formula containing approximately 20mg of HCA 1 hour prior to daily feeding schedule significantly depressed in vivo hepatic rates of fatty acid and cholesterol synthesis in the liver and adipose tissue. Serum triglyceride and cholesterol levels were significantly reduced by HCA. At the end of treatment period, the rats administered with HCA resulted in a significantly reduction in body weight gain. The reduction in weights was attributable to a significant decrease in fat cell size with a smaller extent, but not significant, reduction in fat cell number. Rats receiving HCA demonstrated less food intake than the controls ; however, this decreased caloric intake was not fully responsible for the HCA induced depression of hepatic and adipocytic lipogenesis, since experiment using pair-fed cojntrol rats showed, less magnitude but similar results. Both a anorectic and an antilipogenic properties of HCA seem to be responsible for this weight reduction activity of HCA. The outcome of this study suggest that metabolic regulation may be a feasible approach to the control of obesity and hyperlipidemia.

Ultrafine Copper Nanoparticles Exhibiting a Powerful Antifungal/Killing Activity Against Corticium Salmonicolor

  • In this paper ultrafine copper nanoparticles (CuNPs) were prepared from copper salt via chemical reduction method with sodium citrate dispersant and polyvinylalcol (PVA) capping polymer. The colloidal CuNPs were characterized by using UV-Visible spectroscopy, Transmission Electron Microscopy (TEM), and X-ray Diffraction (XRD) techniques. Our obtained results indicated that the CuNPs were produced ranging from 2 to 4 nm in diameter. The colloidal solution at 7 ppm of CuNPs exhibited a powerful antifungal activity against Corticium salmonicolor (C. Salmonicolor). Fungal killing assays showed colloid solutions containing 10 ppm of CuNPs killed entirely the cultured fungus. A highly killing activity against the fungus was also performed when the CuNPs were sprayed on pink disease-infected rubber trees. These positive results may offer a great potential to produce CuNPs-based eco-fungicide for pink disease.
