Effects of Squalene on the Immune Responses in Mice(I): Humoral Immune Responses of Squalene

  • Effects of squalene on humoral immune system in mice were investigated. Squalene exhibited significant increases in the circulating leukocyte counts and relative spleen and thymus weights of the mice. However, the relative liver weight was slightly decreased. Hemagglutination titers (HA) were signficantly enhanced by squalene while Arthus reaction was not affected. Splenic plaque forming cells (PFC) were also greatly increased by squalene, especially at doses of 50 and 100 mg/kg of it.

  • Propyl gallate used as an antioxidant was examined for its effects on murine immune system. As immunotoxicology assay parameters, we adopted circulating leukocytes and immunoorgan weights for pathotoxicology, delayed hypersensitivity and colloidal carbon clearance for cell mediated immuntity. Propyl gallate's effects were observed as follows; 1) Propyl gallate decreased circulating leukocyte counts, dose dependently. 2) Propyl gallate decreased delayed hypersensitivity reaction. 3) Phagocytic index were similar in the test and control groups.

  • Erythrosine used as a colouring agent in drugs, cosmetics and foods in Korea, was examined for its effects on murine immune system and methemoglobin formation. As immunotoxicologic assay parameters, we adopted circulating leukocytes and immunoorgan weights for pathotoxicology, IgM plaque forming cells and arthus reaction for humoral immunity, delayed hypersensitivity reaction of cell mediated immunity and carbon clearacnce for macrophage function. Erythrosine's effects were observed as follows; 1. Ery throsine showed no significant effects on circulating leulocyte counts and relative immunoorgan weight. 2. Erythrosine diminished IgM plaque forming cells. 3. Erythrosine decreased arthus reaction, in the dose dependent manner. 4. Erythrosine had no significant effect on delayed hypersensitivity. 5. Phagocytic and corrected phagocytic index were not affected. 6. Methemoglobin content was similar in the test and control groups.

Delta Neutrophil Index as an Early Marker of Sepsis in Burn Patients (화상환자에서 패혈증의 조기 예측인자로서의 DNI)

  • Purpose: The immature granulocyte count has been reported to be a marker of infection and sepsis. The difference in leukocyte subfractions (delta neutrophil index, DNI) in ADVIA 2120 reflects the fraction of circulating immature granulocytes in the blood. This study evaluated the clinical utility of DNI as a severity and prediction marker in critically ill patients with burn sepsis. Methods: One hundred and sixty nine patients admitted to the burn care unit were studied. DNI (the difference in leukocyte subfractions identified by myeloperoxidase and nuclear lobularity channels) was determined using a specific blood cell analyzer. Results: Seventy one patients (42 %) were diagnosed with burn sepsis. DNI was significantly higher in patients with burn sepsis than in patients without (P<0.01). Delta neutrophil index was a better indicator of burn sepsis than C-reactive protein, lactate, white blood cell count, HCO3, base excess, lactate, procalcitonin (odds ratio, 6.31; confidence interval 2.36~16.90; P<0.01). And the receiver operating characteristic curves showed that delta neutrophil index, AUC 0.795 (95% confidence interval, 0.721~0.869; P<0.05) was a better predictor of burn sepsis than lactate, procalcitonin, white blood cell, base excess and abbreviated burn severity index. Conclusion: Delta neutrophil index may be used as a early marker of patients with burn sepsis.

In Vivo Immunotoxicities of Butylated Hydroxyanisole (BHA) and Butylated Hydroxytoluene (BHT) in Male Mice

  • The effects of butylated hydroxyanisole and butylated hydroxytoluene on the immune status in normal male were evaluated. They exhibited significant decrease in the circulating leukocyte counts. Relative spleen and thymus weights were slightly decreased, but not stratistically significant. These were, however, significant liver hypertrophies in theier exposed mice. Splenic IgM PFCs per one million cells in 1/20 LD50 BHA and BHT exposed mice were significantly reduced IgM PFCs per spleen were similar tothose of control, except in 1/20 LD50 BHA exposed mice, where they were significantly suppressed. The precise nature of the inhibition is not clear. Direct cytotoxicity is not responsible for the depressed antibody response, even following relatively high doses of them, because the changes in spleen cellularity are not significant. Both substances, however, did not show any effects on the arthus reaction and delayed hypersensitivity reaction induced by heat aggreagted bovine serum albumin, and in vivo phagocytosis of colloidal carbon. In the light of the present results, in vivo antibody response as well as in vitro, may be sensitive to BHA of the present results, in vivo antibody response as well as in vitro, amy be sensitie to BHA and BHT. Further elucidation of the precise nature of antibody suppression in their exposed mice, is warranted.
