We identified the echinostome metacercariae in Chinese mystery snails, Cipangopaludina chinensis malleata, from Xiengkhuang Province, Lao PDR with morphologies of adult worms recovered. Total 20 snails were examined with artificial digestion method and then the collected metacercariae were orally infected to a mouse and a rat. Adult worms recovered from experimental animals were observed with a light microscope and a SEM. The metacercariae were round, 125×123 ㎛ in average size, with a moderately thick cyst wall, collar spines distributed in the head collar and excretory granules in 2 canals of excretory tube. Adult flukes (3-week-old in a rat) were elongated, ventrally curved and 5.310×1.023 mm in average size. Head collar distinct, bearing 43 collar spines with 5 end group ones on each side. Oral sucker subterminal, prepharynx very short, pharynx well developed, and esophagus relatively short. Cirrus sac well developed, with a saccular seminal vesicle, and ventral sucker very large. Ovary round and on the median line of the body. Testes tandom and elongated. Eggs operculated, elliptical and 90×57 ㎛ in average size. In the SEM observation, the head crown prominent, with 43 collar spines resembled with horns of younger stag. Scale-like tegumental spines were densely distributed on the surface between the head collar and ventral sucker, and their densities were decreased posteriorly. Conclusively, the metacercariae detected in C. chinensis malleata from Lao PDR were identified as those of Echinostoma macrorchis based on the morphological characteristics of adult worms.
The ultrastructure of the tentacular pigment cells and the origin of the pigment granules in the Chinese mystery snail, Cipangopaludina chinensis malleata Reeve, are studied with electron microscope. Tentacular pigment cells of the Enail are the melanophores which contain electron dense melanosomes(melanin pigment granules) . Melanophores are distributed among the connective tissues but otter kind of dermal chromatephores are not observed. The epidermal melanin units are observed in the epithelial tissues of the tentacles. Among the several kinds of epithelial cells, only the epithelial supporting cells contain these pigment granules. Synthesis of the pigment materials is from the rough ER and pigment granules are finally being packaged and released by the Golgi complexes but limiting membrane of these granules are presumed to be originated from the smooth ER.
Water quality and plankton community dynamics after the introduction of two domestic freshwater shellfishes (Unio douglasiae Griffith & Pidgeon and Cipangoplaudina chinese malleata Reeve), were monitored daily in enclosures, which constructed in the tidal zone of eutrophic agricultural reservoir (Shingu reservoir, Korea) for one week between July 31 and August 6, 2007. This biomanipulation study to improve the water quality of eutrophic lake, comprised eight enclosures (duplicate x four kinds), enclosure had no mussels (Control), stocked only with U. douglasiae at density of 60 individuals (UD), stocked only with C. chinese malleata at density of 60 individuals (CCM), and combined-stocked with 30 individuals of UD and 30 individuals of CCM (MIX), respectively. Our results clearly indicate that UD strongly decreased the concentration of chlorophyll-a and increased the water transparency, whereas CCM and MIX (included CCM) did increase algal density or decrease transparency, due perhaps to the disturbance of C. chinese malleata. Therefore, the field application of shellfish to enhance the water quality of eutrophic agricultural reservoir, should consider the grazing characteristics of biological control agents, especially active moving animals like snail.
There are groups of large monopolar cells containing numerous secretory inclusions in the muscular and connective tissue. These cells contain the electron-lucent vesicles in the size of $0.5{\sim}0.9{\mu}m$, which are secreted to the exterior of the tentacle through their cytoplasmic processes projected to the epithelial layer. Secretory material can be seen accumulating in swollen portions of the granular endoplasmic reticulum. It is postulated that this material is transported to the Golgi-complex and thus the limiting membrane of the inclusions is derived from the Golgi-membrane. An electron lucent material secreted from these cells seems to be similar to fibrous electron lucent material filled in the brush border of the epidermis.
Heaty metals in mollusks and in sedimints collected from five stations along Mangyong river area in Jeonra Bug Do were examined during 1988 through 1989. Concentration of heavy metals in Cipangopaludina chinensis malleata and Anodonta woodiana were highly correlated with that in sediments, And concentration of heavy metals in both of millusks and sediments from industrialized area was higher than the other stations. Above results suggested that Mangyong river was polluted with heavy metals and heavy metals were accumulated in molluscan soft bodies.
Upo wetland is the largest inland wetland in Korea as Ramsar Convention Area. The purposes of the study were to investigate the levels of heavy metals (Cd, Cr, Cu, Ni, Pb and Zn) in the sediment and Cipangopaludina chinensis malleata from three sites of the wetland and to assess the potential of the gastropod as a bioindicator for heavy metal levels. The gastropods were dissected into shell and soft tissue without the digestive and excretive organs. The levels of Cd, Cu and Pb were below the guideline of Soil Environment Conservation Act and the heavy metals except Cr were slightly different among the sites. Cd was higher in Upo site ($0.32{\mu}g/g$) than Sajipo site ($0.28{\mu}g/g$). Cu and Zn showed the highest value in Sajipo as $43.5{\mu}g/g\;and\;39.8{\mu}g/g$, respectively while the concentrations of Pb and Zn were the highest in Upstream as $58.8{\mu}g/g\;and\;138{\mu}g/g$, respectively. In the soft tissues and shells of the gastropod, the overall common trend in the concentrations of the heavy metals was revealed with the following order: Zn > Cu > Cr > Ni > Pb > Cd and Ni > Zn > Cr > Cu > Pb > Cd, respectively. Although the soft tissues exhibited higher concentrations of the heavy metals except Ni than the shell in the gastropod, the levels of Cd and Pb in the gastropod were generally below the restrictive values set up by Korea Food & Drug Administration. From Duncan's Multiple Range Test (DMRT) results, the concentrations of Pb and Zn in the sediments among the sites were reflected on the soft tissue (Pb) and the shell (Pb and Zn) of the gastropod in the same order. The lower value of coefficient of variation (CV) in Pb concentration of the shell than in that of the soft tissue supports the usefulness of the shell as a bioindicator for Pb pollution. Although the CV value in the shell was a little higher than in the soft tissue, DMRT results and the stability of incorporated Zn into the shell support the use of the shell of the gastropod as a potential bioindicator for long-term contamination of Zn.
The tentacular retractor muscle has many arrays of muscle fiber bundles under the epithelial layer. Most of muscle fiber bundles are arranged in parallel to the longitudinal axes of muscle fibers and a small number of them perpendiculary to them. These smooth muscle cells are filled with compactly arranged myosins and actins. These microfilaments, when the tentacle is protracted, keep abreast with straight for-ward-lined shapes while these microfilaments, when it is retracted, with curved shapes. The foldings in the sarcolemma of the muscle cell, when the tentacle is retracted, lead to the formation of normal subsurface tubules along with which a few mitochondria are included. It is thought that the formation of the sarcolemmal differentiation like the subsurface tubules has a close relation with the protraction and retraction of the tentacle. Mitochondria are found throughout the muscle cell, and sarcoplasmic reticulum (SR) developed greatly in the exoplasm close to the sarcolemma and associated with the cell membrane. Dense bodies are distributed irregularly and thin filaments are scattered around the thick filament in cross-sections, but the thin filaments may be arranged in complete or partial orbits around thick filaments. Complete orbits are infrequent.
