  • This study was conducted to investigate growth characteristics to develop the technique to select resistant cultivar by wet injury at an early stage through the automatic irrigation maintaining 30, 20, 10 kPa respectively using native variety, Chunpoong, Yunpoong, Gumpoong and Sunun. The aerial growth was decreased at 10 kPa compared to 30 kPa. In addition, the survival rate was decreased by 66.6%, 62.3%, 33.8% at 30, 20, 10 kPa, respectively. The survival rate of Chunpoong and Gumpoong were higher than others at 10 kPa. While root growth characteristics such as root length, root weight, number of lateral root and side root were tended to decrease, root diam was no significant or increased. And the more humid condition is, the more the incidence rate of rusty root and rough skin were tended to increase. The epidermal thickness of Chunpoong and Gumpoong was increased but the figures of native variety, Yunpoong and Sunun were decreased at 10 kPa compared to 30 kPa. But, the tissue stiffness of root was decreased at 10 kPa compared to 30 kPa.

  • The Korean ginseng, Panax ginseng C. A. Meyer is a popular medicinal herb in Araliaceae. Genetic map in crops provides valuable information for breeding, genetic and genomic researches. However, little information is available for construction of genetic map in ginseng. Up to now, we have produced large amounts of expressed sequence tags (ESTs) from four ginseng cultivars (37Mb, 49Mb, 39Mb, 47Mb from Gopoong, Gumpoong, Chunpoong and Yunpoong respectively using pyrosequencing technique and 5Mb from normalized full-length cDNA library of Chunpoong) to obtain comprehensive information of gene expression, and constructed EST database including ESTs from public database. Till now, we designed 261 SSR primer sets using EST sequences and identified 106 intergenic polymorphic markers. And 44 of the 106 showed polymorphisms among panax ginseng cultivars. Among 44 markers, 27 SSR polymorphic markers were inspected to 51 $F_2$ population from Yunpoong x Chunpoong, which showed good at the fitness of Mendellian segregation ratio 1:2:1. To enrich the number of markers, and thus construct high resolution genetic map which can be used as frame map for further genome sequencing. we are planning to develop large scale EST-derived SNP markers which are available in the F2 population. This study provides genetic information as well as foundation for ginseng researches such as genetics, genomics, breeding, and the final goal for whole genome sequencing. This study was supported by Technology Development Program for Agriculture and Forestry, Ministry for Food, Agriculture, Forestry and Fisheries, Republic of Korea (Grant No. 609001-051SB210).

  • In order to investigate the diversity of endophytes, fungal endophytes in Panax ginseng Meyer cultivated in Korea were isolated and identified using internal transcribed spacer (ITS) sequences of ribosomal DNA. Three cultivars of 3-year-old ginseng roots (Chunpoong, Yunpoong, and Gumpoong) were used to isolate fungal endophytes. Surface sterilized ginseng roots were placed on potato dextrose agar plates supplemented with ampicilin and streptomycin to inhibit bacterial growth. Overall, 38 fungal endophytes were isolated from 12 ginseng roots. According to the sequence analysis of the ITS1-5.8S-ITS2, 38 fungal isolates were classified into 4 different fungal species, which were Phoma radicina, Fusarium oxysporum, Setophoma terrestris and Ascomycota sp. 2-RNK. The most dominant fungal endophyte was P. radicina in 3 cultivars. The percentage of dominant endophytes of P. radicina was 65.8%. The percentage of colonization frequency of P. radicina was 80%, 52.9%, and 75% in Chunpoong, Yunpoong, and Gumpoong, respectively. The second most dominant fungal endophyte was F. oxysporum. The diversity of the fungal endophytes was low and no ginseng cultivar specificity among endophytes was detected in this study. The identified endophytes can be potential fungi for the production of bioactive compounds and control against ginseng pathogens.

  • Expressed sequence tags (EST) are help to quickly identify functions of expressed genes and to understand the complexity of gene expression. In order to analyze gene expression of the leaf development in Panax ginseng, which is one of the most important medicinal plant, expressed sequence tags (EST) analysis was carried out. We constructed a cDNA library using the immature leaf of Chunpoong. Partial sequences were obtained from 3,170 clones. The ESTs could be clustered into 1,624 (56.1%) non-redundant groups. Similarity search of the non-redundant ESTs against public non-redundant databases of both protein and DNA indicated that 1,137 groups show similarity to genes of known function. These ESTs clones were divided into sixteen categories depending upon gene function. Most abundant transcripts in immature ginseng leaf were photosynthesis related protein, such as chlorophyll a/b binding protein LHCII type I (128), chlorophyll a/b binding protein (53), ribulose-1,5-bisphosphate carboxylase (41), and photosystem I psaH (26). The EST data from immature leaf generated in this study is useful in dissecting gene expression in leaf organ of ginseng.

  • Expressed sequence tags (EST) are help to quickly identify functions of expressed genes and to understand the complexity of gene expression. To assist genetic study of the root development in Panax ginseng, which is one of the most important medicinal plant, expressed sequence tags (EST) analysis was carried out. We constructed a CDNA library using the 4-year Chunpoon root. Partial sequences were obtained from 3,841 clone. The ESTs could be clustered into 2,056 (64%) non-redundant groups. Similarity search of the non-redundant ESTs against public non-redundant databases of both protein and DNA indicated that 1,498 groups show similarity to genes of known function. These ESTs clones were divided into eighteen categories depending upon gene function. The most abundant transcripts were major latex protein (41), ribonuclease 2 (36), metallothionein 2(35). Our extensive EST analysis of genes expressed in 4-year Chunpoong root not only contributes to the understanding of the dynamics of genome expression patterns in root organ development but also adds data to the repertoire of all genomic genes.

  • Recently, there has been increased attention to the development of new plant cultivars with enhanced resistance to biotic and abiotic stress. To develop ginseng cultivars with such traits, systematic breeding programs and comprehensive field studies are prerequisites. In this study, we applied a pure-line selection method to identify a ginseng cultivar with enhanced stress resistance. Phenotypic and agronomic characteristics, seed yield, and physiological responses to biotic and abiotic stresses were investigated according to the guidelines of the International Union for the Protection of New Varieties of Plants (UPOV). In the newly developed 'Kowon' cultivar, the time of emergence, flowering, and berry maturity were intermediate between those of the controls, 'Yunpoong' and 'Chunpoong'. The stem length of 'Kowon' was intermediate, whereas the root length was shorter and the main root diameter was greater than those of 'Chunpoong'. In local adaptability tests conducted in three regions, the yield of 'Kowon' was $666kg{\cdot}10a^{-1}$; 27% and 4% higher than that of 'Chunpoong' and 'Yunpoong'. Diseases such as Alternaria blight, Phytophthora blight, mulberry mealybug, and nematode infestation did not occur in 'Kowon'; and it also exhibited moderate resistance to damping-off and anthracnose. In these cases, yellow spots occurred on aerial parts and the rusty skin of the root, and it exhibited moderate resistance at high temperatures. Our study demonstrates that 'Kowon', which has a high root weight and enhanced biotic/abiotic stress resistance, is a superior cultivar that could increase farmers' income.

  • Little is known about the genetics or genomics of Panax ginseng. In this study, we developed 70 expressed sequence tagderived polymorphic simple sequence repeat markers by trials of 140 primer pairs. All of the 70 markers showed reproducible polymorphism among four Panax species and 19 of them were polymorphic in six P. ginseng cultivars. These markers segregated 1:2:1 manner of Mendelian inheritance in an $F_2$ population of a cross between two P. ginseng cultivars, 'Yunpoong' and 'Chunpoong', indicating that these are reproducible and inheritable mappable markers. A phylogenetic analysis using the genotype data showed three distinctive groups: a P. ginseng-P. japonicus clade, P. notoginseng and P. quinquefolius, with similarity coefficients of 0.70. P. japonicus was intermingled with P. ginseng cultivars, indicating that both species have similar genetic backgrounds. P. ginseng cultivars were subdivided into three minor groups: an independent cultivar 'Chunpoong', a subgroup with three accessions including two cultivars, 'Gumpoong' and 'Yunpoong' and one landrace 'Hwangsook' and another subgroup with two accessions including one cultivar, 'Gopoong' and one landrace 'Jakyung'. Each primer pair produced 1 to 4 bands, indicating that the ginseng genome has a highly replicated paleopolyploid genome structure.

  • The principal objective of this study was to develop a discrimination method using SSR markers in Korean ginseng cultivars. Five cultivars--Chunpoong, Yunpoong, Gopoong, Sunpoong, and Kumpoong--were evaluated by nine markers out of 22 SSR markers. A total of 23 alleles were detected, ranging from 1 to 4, with an average of 2.6 alleles per locus, and an averages of gene diversity (GD) of 0.480. Nine markers were tested in order to distinguish among five Korean ginseng cultivars. Two markers out of nine SSR markers, GB-PG-065 and GB-PG-142, were selected as key markers for discrimination among Korean ginseng cultivars. Two genotypes were detected in GB-PG-065. Chunpoong and Kumpoong shared the same allele type, and Yunpoong, Gopoong, and Sunpoong shared another identical allele type. In the case of GB-PG-142, a specific allele type differentiated from those of other four cultivars was observed only in Sunpoong cultivar. Consequently, the SSR markers developed in this study may prove useful for the identification of Korean ginseng cultivars and the development of ginseng seed management systems, as well as tests to guarantee the purity of ginseng seeds.

  • Background : This study was carried out to investigate the changes to fatty acid, mineral, and ginsenosides contents in ginseng seed when they were stratified for different length of time and to determine whether variety had any effects on the changes. The aim was to improve the ginseng seed stratification process. Methods and Results : The ginseng varieties used were Geumpoong, Chunpoong, Yunpoong, and K-1. Stratifying periods treated on ginseng seed were 0, 20, 40, 60, 80, and 100 days. The main fatty acids of ginseng seed were oleic acid (C18 : 1, n9c) with a content of 78.40 - 79.20% followed by linoleic acid (C18 : 2, n6c). The main mineral in the seeds was potassium (K), at 1208.2 -1337.6 mg/100 g. The main ginsenosides in ginseng seed were ginsenoside Re and Rb1. Increasing the length of the stratification periods led to increases in oleic acid content (60 - 80 days), however after this the content declined. In contrast, linoleic acid content fell as the stratification period increased. K, P, Mg, Ca and Na content rose as the stratification period increased. The ginsenoside Re content of Chunpoong and K-1 cultivar seeds also rose as the stratification period increased which meant that total ginsenoside content increased. However, ginsenoside Re content rose in Geumpoong and Yunpoong seeds, but total ginsenoside content decreased as the stratification period increased. Conclusions : Some beneficial compound in ginseng seed rose as the stratification period increased. Therefore, ginseng seed stratification could improve the food value of ginseng.
