Chromosome numbers of eight Carex taxa in Korea (Cyperaceae)

  • In the flora of Korea, Carex L. is one of the most species-rich genera. Among nearly 157 Carex taxa, less than 30 have had chromosome numbers reported. We report the meiotic chromosome numbers of eight Carex taxa from Korean populations, which include the first count for C. accrescens Ohwi (n = 37II) and the first chromosome investigations of Korea populations for three taxa: C. bostrychostigma Maxim. (n = 22II), C. lanceolata Boott (n = 36II), and C. paxii Kuk. (n = 38II). In most species, chromosome counts observed in the study are included in the variation ranges of previous chromosome numbers. However, C. bostrychostigma Maxim. (n = 22II) and C. planiculmis Kom. (n = 29II) are assigned new chromosome numbers. Carex is known to have holocentric chromosomes, lacking visible primary constrictions and exhibiting great variance in its chromosome number. Further investigations of the diversity of Carex chromosomes will provide basic information with which to understand the high species diversity of the genus.

Trichoderma속의 염색체(染色體)에 관한 연구(硏究) (Chromosomal Studies on the Genus Trichoderma)

  • ATCC로 부터 구입한 11종(種)과 우리나라의 토양에서 분리 동정한 6종(種)의 Trichoderma를 실험(實驗) 재료(材料)로 하였다. 2% malt extract agar 배지(培地)에서 배양(培養)하면서 균사의 영양핵에서의 핵분열을 관찰하였다. 염색(染色)은 Giemsa staining solution으로 염색(染色)하였고 그들의 염색체(染色體) 수(數)를 세었다. 염색체의 형태(形態)는 다른 균류와 마찬가지로 점(點) 모양(dot-like)의 것이 대부분(大部分)이었다. 염색체(染色體)의 크기는 매우 작아서 $0.2{\sim}0.3\;{\mu}$정도이다. 염색체(染色體)의 수는 $5{\sim}10$개로서 n=5인 종(種)이 2종(種)(Trichoderma pseudokoningil, T. saturnisporum)이고, n=6인 종(種)은 8종(種)(T. koningil, T. viride, T. hamatum, T. longibranchiatum, T. sp. No.40, T. sp. No.58 T. sp. No.237, T. sp. No.241)으로 가장 많았다. n=7인 종(種)은 2종(種)(T. reesei, T. harzianum, T. polysporium, T. virigatum, T. sp. No.196)으로 관찰되었다. 이상의 17종(種)을 비교 검토하여 보면 Trichoderma속의 기본(基本) 염색체(染色體) 수(數)는 $4{\sim}10$개 사이에 있을 것으로 생각되며 그 중에서 가장 많은 수(數)를 나타탠 n=6을 기본(基本) 염색체수(染色體數)로 추론하였다.

  • 일본산메추리(Coturnix coturnix japonica)의 핵형연구를 위하여 정소조직 및 백혈구, 초기태아조직들로서 염색체 분석을 수행하였다. 이들 염색체의 수는 정상 이배체(2n)가 78개로 나타나고, 계2 감수분열 중기상태의 정모세포의 관찰에서 반수체(n)가 39개로 나타났다. 한편 8개의 대형염색체 및 성 염상체에 대한 형태적 분석은 동원체지수 및 상대적 길이비로서 측정하였다. 이용된 여러 조직간의 염색체의 형태적 차이는 거의 없는 것으로 나타났으나 4번 염색체템서 p-alm의 존재유무에 따라 상당한 변이를 나타내었다.

Chromosome Counts from the Flora of Korea with Emphasis on Apiaceae

  • Seventy-three chromosome counts are reported from populations in 70 species from Korea, of which 34 counts in 32 species belong to Apiaceae. New species counts are in Acanthopanax chiisanensis(2n=48), Hydrocotyle japonica(2n=ca.96), H. maritima (n=12), Libanotis coreana (2n=22), Lindera sericea (n=12), Quercus myrsinaefolia (n=12), Rhamnus yoshinoi (n=12), Salix hulteni (n=19), Symplocos chinensis for. pilosa (n=11) and Vaccinium oldhami (n=12). New chromosome level for Asperula odorata (n=11), Cryptotaenia japonica (2n=16) and Sium ninsi (2n=18) is also provided. The taxonomic implications of certain of these counts are discussed.

대학생들과 과학교사들의 염색체 행동의 이해에 관한 연구 (A Study on the College Science Students' and Science Teachers' Understaning of Chromosome Behavior)

  • The purpose of this study is to investigate college science students' and science teachers' understanding of chromosomal behavior in the context of cell division. The research problems were as follows: 1. What is the level of college science students' understandings of chromosomal behaviors? 2. What is the level of science teachers' understandings of chromosomal behaviors? 3. What is the level of understanding by grade and major area? The sample consisted of 28 sophomore, 17 junior and 23 senior biology students; and 23 middle school science teachers and 14 high school biology teachers. The instrument of the study was a short answer required paper and pencil test. The results of the study were as follows: 1) About 15 percent of the sample could not count the number of chromosome in a cell in appropriate. 2) Seventy percent of the students, and 80 percent of the teachers identified homologous chromosomes as ones with the similar shape and size, and 30 percent of the whole sample could not pair two homologous chromosomes. 3) About 70 percent of the students and 30 percent of the teachers could not mark corresponding allele on chromosome. 4) Biology major students showed higher understanding of overall chromosomal behaviors than non Biology students. Based upon the results, some implications were made. The major one was a development of a teaching model in which students can improve the ability to connect chromosome theory to mendelian genetics.

Genome-Wide Association Study Identifies Candidate Loci Associated with Platelet Count in Koreans

  • Platelets are derived from the fragments that are formed from the cytoplasm of bone marrow megakaryocytes-small irregularly shaped anuclear cells. Platelets respond to vascular damage, contracts blood vessels, and attaches to the damaged region, thereby stopping bleeding, together with the action of blood coagulation factors. Platelet activation is known to affect genes associated with vascular risk factors, as well as with arteriosclerosis and myocardial infarction. Here, we performed a genome-wide association study with 352,228 single-nucleotide polymorphisms typed in 8,842 subjects of the Korea Association Resource (KARE) project and replicated the results in 7,861 subjects from an independent population. We identified genetic associations between platelet count and common variants nearby chromosome 4p16.1 ($p=1.46{\times}10^{10}$, in the KIAA0232 gene), 6p21 ($p=1.36{\times}10^{-7}$, in the BAK1 gene), and 12q24.12 ($p=1.11{\times}10^{-15}$, in the SH2B3 gene). Our results illustrate the value of large-scale discovery and a focus for several novel research avenues.

  • Chronic Myeloblastic Leukemia(CML) is characterised by excessive production of neoplastic myeloid cell, reciprocal translocation between chromosome 9 and 22 called 'Philadelphia chromosome'. This one case of CML with Intracerebral Hemorrhage(ICH) patient is thought of Kidney-Yin and Yang deficiency, by the first clinical symptoms, at admission. So the nourishing Yin and Tonifying kidney, warming and tonifying kidney-yang is used. After the incresing of WBC count, the nourishing Yin and Tonifying kidney, invigorating-Yin and reducing fire is used for treatment of the essence-of-life and blood deficiency, fever due to Yin deficiency.

Early Selection of Double Flowers Based on Cotyledon Shape in Cut Stock (Matthiola incana L.) Flowers

  • Selection of double-flowered plants at the seedling stage is one of the main purposes of stock breeding programs. Eight stock cultivars of Matthiola incana L. named 'Nobel', 'Cinderella', 'Pacific', 'Avalanche', 'Midblue', 'Lavender', 'Goddess' and 'Esfahan', with different percentage of double-flowered plants were used for examining the relationship with three morphological types of cotyledons. The results of a chi-square test indicated that in heart-shaped (HC) and cup-shaped cotyledon (CC) populations, the number of plants with double flowers was much more than that of single flowers and CC seedlings rarely produced single flowers. Therefore, increasing the number of CC seedlings can improve the percentage of double flowers. The highest and lowest numbers of CC seedling were observed in high double and single flower cultivars, respectively. Single flower cultivars showed the maximum count of dumbbell-shaped cotyledons. Chromosome pairing of these cultivars was evaluated using the squash technique. Aneuploid cells were found in 'Nobel' and 'Goddess' cultivars, which showed the highest percentage of CC seedling. Based on morphological measurements, the highest value of inflorescence size was observed in the seedlings with cup-shaped cotyledons.

우리나라 미기록 식물: 고깔닭의장풀(Commelina benghalensis L.)과 큰닭의장풀(C. diffusa Burm. f.) (Two new records for the Korean flora: Commelina benghalensis L. and C. diffusa Burm. f. (Commelinaceae))

  • 우리나라 미기록 종인 닭의장풀과(Commelinaceae)의 고깔닭의장풀(Commelina benghalensis L.)과 큰닭의장풀(Commelina diffusa Burm. f.) 2종이 제주도의 저지대에서 채집되었다. 고깔닭의장풀은 불염포의 밑 부분이 합생하여 깔때기 모양을 하고 폐쇄화를 갖는다는 점에서 나머지의 다른 종들과 구분이 된다. 큰닭의장풀은 삭과가 3개의 과피편으로 되어 있고, 불염포상 총포편이 피침형이면서 심장저 또는 원저라는 점에서 나머지 다른 종들과 구분된다. 염색체수는 고깔닭의장풀이 2n = 2x = 22(2배체)이며, 염색체 크기가 1.25-2.70 ${\mu}m$로 매우 작았고, 큰닭의장풀은 2n = ca. 100 이상으로 정확한 수를 알 수 없었다. 이 종들은 지금까지는 아시아와 아프리카의 열대 및 아열대에 분포하여 우리나라보다 남쪽에만 분포하는 것으로 알려져 왔다.

쉬리, Coreoleuciscus splendidus (Cyprinidae)의 세포유전학적 연구 (Cytogenetic Analysis of Korean Shinner, Coreoleuciscus splendidus (Cyprinidae))

  • 우리나라 고유 담수 어종인 쉬리(Coreoleuciscus splendidus; Cyprinidae)를 대상으로 세포크기, 염색체 수 분석, 핵형 분석, 세포당 DNA 함량 조사 등 세포유전학적 연구를 실시하였다. 쉬리 암,수의 염색체 modal number는 모두 2n = 48로 나타났으나 암,수간 형태가 다른 성염색체(sex chromosome)가 관찰되었다. 쉬리 암컷은 10쌍의 중부염색체, 6쌍의 차중부염색체, 8쌍의 차단부염색체 그리고 XX 염색체로 나타났고, 반면 수컷은 10쌍의 중부, 6쌍의 차중부 및 8쌍의 차단부염색체와 함께 XY 성염색체를 나타냄으로써 전형적인 XX-XY의 성 결정 기작(sex determination mechanism)을 갖는 것으로 나타났다. 또한 쉬리는 1쌍의 Ag-NOR을 차단부 상동염색체에 갖고 있었고, 쉬리의 세포당 평균 DNA 함량은 flow cytometry 분석을 통해 2.4 pg/cell로 나타났다. 적혈구 세포 크기를 분석을 통해 핵 용적을 평가한 결과 암수 모두 $28\;{\mu}m^3$를 나타내었다. 본 세포유전학적 분석 결과를 통해 본 어종은 진화과정 중 여러 잉어과 어류에서 관찰되는 배수성에 의한 진화 보다는 유전자 재조합 등 점 돌연변이가 진화가 기본 기작이었을 것으로 판단된다.
