Karyological Analysis of Somaclonal Variation in Callus Cells of Allium victorialis var. platyphyllum (산마늘의 캘러스세포에서 체세포군 변이의 핵학적 분석)

  • Calli obtained from basal disc explants of Allium victorialis var.platyphyllum wre grown in three kinds of nutrient media (MS, BDS, and B5), and the frequencies of mitotic index and the chromosomal aberrations were analysed. The mitotic index varied from 0.55% to 1.01% with respect to culture media and ages. The mitotic irregularities like micro-, bi- and multi-nuclei, chromosome bridge and laggards were noted in each types of calli. The chromosome number variations observed in metaphase stage were identified aneuploid and tetraploid. Structural variations such as dicentric chromosomes, centromere breakage and small chromosomes were observed. Relationship between basal medium and chromosomal variability was not observed in this study. But, in BDS medium, NAA and BA had a more effect on number variation than kinetin.

Karyotype of Dwarf Loach, Kichulchoia brevifasciata (Pisces: Cobitidae) from Korea (미꾸리과 소형어류 좀수수치, Kichulchoia brevifasciata의 핵형)

  • The karyotype analysis of Kichulchoia brevifasciata was carried out, known as an end-angered small endemic cobitid fish of Korea. Five females and one male were collected from Geum-san-myeon, Goheung-gun, Jeollanam-do, Korea and its gill, kidney and digestive tract tissues were treated by flame-drying technique. The karyotype was found to be 2n= 48 from 33 cells; 16 meta-submetacentric and 32 subtelo-telocentric chromosomes; 64 FN (fundamental number). This karyotype showed a remarkably different karyotype formula from Niwaella multifasciata which is closely related to this species.

New diploid populations of Chrysanthemum indicum L. (Asteraceae) from Korea

  • Chrysanthemum indicum (Asteraceae) is a perennial plant belonging to the genus Chrysanthemum. The basic chromosome number of Chrysanthemum sensu stricto is x = 9, and it consists of a series of polyploids ranging from diploid to decaploid. However, C. indicum, which occurs in Korea, is known to consist of only tetraploids, except for two diploid populations that are sympatric with C. zawadskii and C. boreale. During the collection of plant materials as part of a study to ascertain the diversity of Chrysanthemum in Korea, we found new diploid populations (2n = 18) of C. indicum in the southern region of Korea and describe them here in detail.

  • A comparative study on 'Dang-gui' and 'Kang-whal' the two species of Angelica growing in Korea was carried out about the morphology and cytology. The somatic chromosome number of Cham dang-gui, Ill dang-gui, Badi-namul was found to be 2N=22. The somatic chromosome number of Kang-whal was found to be 2N=110, that of Chili kang-whal to be 2N=44, that of Mul kang-whal to be 2N=22. Stoma of species of Dang-gui were same in size. Those of Chili kang-whal and Kang-whal were strikingly larger than the other. Chili kang-whal and Kang-whal were verified to be polyploid in the genus Angelica.

Interspecific Hybridization between Oryzias sinensis and O. latipes from Korea (한국산 송사리 두 종의 교잡실험)

  • Cytogenetic studies and hybridization experiment using two populations of Oryzias latipes from Korea were conducted in order to examine their systematic positions. In the chromosome number of specimens examined. O. sinensis had 46 with a pair of large metacentrics and O. latipes had 48 chromosomes without a pair of large metacentrics. However, the diploid chromosome number of the hybrid between O. latipes and O. sinensis was 47 and they had a large metarcentric chromosome in their karyotype. In the $F_2$ hybrids, distribution of chromosome number was variable among individuals. Hatching rates showed little difference between control and hybrids. However, hatchiabilties of $F_2$ hybrids were decreased by 83.3% as compared with $F_1$ offspring. Incidence of abnormal larvae was increased by 15.3% as compared with their parents. These results indicate that the two karyotype populations formerly regarded as two subspecies would be two distinct allopatric sibling species O. sinensis, and O. latipes.

Comparison of Morphological and Chromosomal Characteristics and Cross Breeding of the Two Types Korean Mandarin Fish, Siniperca scherzeri (쏘가리와 황쏘가리(Siniperca scherzeri)의 염색체와 외부형태 비교 및 교배 실험)

  • This study was carried out to confirm the taxonomical status by comparison of morphological and chromosomal charateristics and cross breeding in the two types of black and yellow body color of Korean mandarin fish, Siniperca sherzeri from Han River, Korea. Black and yellow types of Korean mandarin fish were similar in having 90-98 lateral line scale pores, 13 dorsal fin rays and XII XIII spines, 9 anal fin rays and 15 pectoral fin rays, but yellow type was differ from blacks type by yellow body color pattern. Diploid chromosome and arm number (fundamental number, NF) of the two types were the same to 2n=48 and NF=52. Karyotypes in the two types are consisted of 2 pairs submetacentric chromosome and 22 pairs of acro and/or telocentric chromosome. The black type females of Siniperca scherzeri were artificially crossed with yellow type males and black type males, respectively. The progenies from each cross breeding produced the normal individuls that possessed with parent types in the body color pattern. These data suggest that two types of Korean mandarin fish were same species, and yellow type was mutant by albino.

  • Somatic chromosome numbers for 10 taxa and karyotypes analysis for 6 taxa of Korean Vicia were investigated. Somatic chromosome numbers of treated taxa were 2n = 12, 14 or 24 and therefore they proved to be diploid or tetraploid with basic chromosome numbers of x = 6 or 7. The chromosome number of V. hirticalycina (2n = 2x = 12) was reported for the first time in this study. The chromosome numbers of nine taxa were the same as in previous studies; V. angustifolia (2n = 2x = 12), V. cracca (2n = 4x = 24), V. hirsuta (2n = 2x = 14), V. tetrasperma (2n = 2x = 14 + 2B), V. amurensis (2n = 2x = 12), V. chosenensis (2n = 2x = 12, 12 + 2B), V. unijuga (2n = 4x = 24), V. unijuga f. minor (2n = 4x = 24), V. venosa var. cuspidata (2n = 4x = 24). The karyotypes of V. cracca, V. amurensis, V. hirticalycina, V. unijuga, V. unijuga f. minor, V. venosa var. cuspidata were observed as 2 m + 8 sm + 2 st, 2 m + 2 sm + 2 st, 3 m + 1 sm + 2 st, 4 m + 6 sm + 2 st, 4 m + 6 sm + 2 st, 4 m + 8 sm, respectively.

Karyotype Analysis and Physical Mapping of rDNAs Using Bicolor-FISH in Tiarella polyphylla D. Don (헐떡이풀의 핵형분석과 Bicolor FISH를 이용한 물리적 지도 작성)

  • Tiarella polyphylla D. Don(Saxifragaceae) is a perennial herb and distributed in China, Japan, Taiwan and Korea. Especially, it only grows in Ulleung island of Korea. It has been using for asthma, bruise and audition troubles with main components of some Triterpenoids and seven oleanolic Saponins. There is only known its chromosomal number rarely and cytogenetic study was not done. From this study, karyotype analysis and chromosomal localization of 5S and 45S rDNAs using bicolor-FISH(fluorescence in situ hybridization) were carried out. The somatic metaphase chromosome number was 2n=2x=14 and the size of chromosomes ranged $1.66{\sim}3.50{\mu}m$. The chromosome complement consisted of four pairs of submetacentrics(chromosomes 1, 2, 3 and 6), two pairs of subtelocentrics(chromosomes 5 and 7) and one pair of telocentrics(chromosome 4). We also observed NOR(nucleolus organizer region) on the chromosome 4. In bicolor-FISH, one pair of 55 and 45S rDNA sites was detected on the centromeric region of chromosome 3 and short arm of chromosome 4, respectively. Bicolor FISH was very useful tool for the localization and identification of rDNAs on the chromosomes in Tiarella polyphylla.

Identification of a Copy Number Variation on Chromosome 20q13.12 Associated with Osteoporotic Fractures in the Korean Population

  • Osteoporotic fractures (OFs) are critical hard outcomes of osteoporosis and are characterized by decreased bone strength induced by low bone density and microarchitectural deterioration in bone tissue. Most OFs cause acute pain, hospitalization, immobilization, and slow recovery in patients and are associated with increased mortality. A variety of genetic studies have suggested associations of genetic variants with the risk of OF. Genome-wide association studies have reported various single-nucleotide polymorphisms and copy number variations (CNVs) in European and Asian populations. To identify CNV regions associated with OF risk, we conducted a genome-wide CNV study in a Korean population. We performed logistic regression analyses in 1,537 Korean subjects (299 OF cases and 1,238 healthy controls) and identified a total of 8 CNV regions significantly associated with OF (p < 0.05). Then, one CNV region located on chromosome 20q13.12 was selected for experimental validation. The selected CNV region was experimentally validated by quantitative polymerase chain reaction. The CNV region of chromosome 20q13.12 is positioned upstream of a family of long non-coding RNAs, LINC01260. Our findings could provide new information on the genetic factors associated with the risk of OF.

Sexing and Cell Cycle Induction Hanwoo Fetal Fibroblast Cells (한우 섬유아세포의 성 판별 및 세포주기 유도 분석)

  • For somatic cell nuclear transfer in Hanwoo, fetal fibroblast cell lines were established from 35, 50, 70 and 90-day fetuses of Korean native cattle. The sex of these fetal fibroblast cells were analyzed by PCR using Y-specific primers and confirmed that two cell lines were female and the other two cell lines were male. Karyotyping of these cell lines indicates that the chromosome numbers of fetal fibroblast cells were not affected by passage number and more than 80% of fetal fibroblast cells have normal chromosome number. To evaluate Go stage in cell cycle of fetal fibroblast cells, Western blotting was performed to detect the expression level of PCNA which is known to be expressed in all cell cycle stages except G$_{0}$ stage. Following serum starvation or confluent culture for 7 days, fetal fibroblast cells were effectively reached to G$_{0}$ stage. The cell cycle was resumed after culture of these Go stage-fetal fibroblast cells with normal medium. These results indicates that fetal fibroblast cells originated from Hanwoo were successfully isolated and culture system and induction of cell cycle of these cells were established for somatic cell nuclear transfer in Hanwoo.woo.
