The Study of G- Banding Chromosome in Silkie (오골계의 염색체 분염법 (G-banding)에 따른 핵형분석에 관한 연구)

  • This experiment was carried out to identify the chromosomes of silkie. It was many difference from other breeds in morphology and characteristics. In this experiment, chromosomal analysis was used early embryos. In aspect of morphological chromosomes, chromosomal size and shape are similar to other breeds. The chromosomes of silkie were shown to morphlogy as follows. They were identified that chromosome #l and #2 were grouped as submentacentric, #3, #5 and #6 were telocentric #4 and #7 were acrocentric and #8 was metacentric chromosome. Zㆍsex chromosome was shown 5th, W-sex chromosome was 8th to 9th and they were metacentric chromosome, respectively. Each chromosome through the G-banding was shown the 3 dark bands in 1 p2, distinct light band in 1p1, dark band in 2p2, broad light band in 3pl, dark band from centromere and distal part in 4th chromosome and dark band in 5pl. Z-sex chromosome was shown dark at p-arm distal part.

Suppressing Effects of Tannic Acid on UVB induced Chromosome Aberrations in Chinese Hamster Lung Cells (자외선(UVB)에 의한 염색체이상과 Tannic acid의 방어효과)

  • We observed the frequency of chromosome aberrations induced by UVB irradiations, and the suppressing effect of tannic acid on chromosome aberrations induced by UVB irradiations in CHL cells, which is a phenolic compound, a hydrolysate of tannin and a components of green tea. UVB doses used for the frequency of chromosome aberrations were from 0.2 to 1.6 KJ/m$^2$ and tannic acid concentrations were from 1.16 $\mu$g/ml to 37.50 $\mu$g/ml. For the observation of suppressing effect of tannic acid on UVB-induced chromosome aberrations, UVB dose was 1.6 KJ/m$^2$ and tannic acid concentrations were 1.0, 2.0, 4.0 $\mu$g/ml. In our study, tannic acid was treated for 24 hours in CHL, cells after UVB irradiation without S9 mix or for 6 hours with S9 mix. From this study, we obtained the following results : (1) The frequency of chromosome aberrations UVB induced were dose-dependently increased. (2) The tannic acid did not induce chromosome aberrations in cultured Chinese hamster cells. (3) UVB-induced chromosome aberrations were suppressed by tannic acid at every concentration from 1.0 $\mu$g/ml to 4.0 $\mu$g/ml with or without metabolic activation. These results suggest that the tannic acid acts as an inhibitor to UVB-induced clastogenicity of the cultured cell.

  • Fluorscence in situ hybridization with chromosome-specific probe has been shown to be a valid and rapid method for detection of chromosome rearrangements induced by chemical and physical agents. This method is useful for quantifying structural aberrations, expecially for stable ones, such as translocation and insertion, which are difficult to detect with conventional method in human lymphocyte. In order to use the FISH method as a biodosimeter for monitoring human population exposed to various chemical and physical agent, baseline level of chromosome rearragement was established. Blood from forty four healthy adults were collected and analysed with whole chromosome-specific probes by human chromosome 1,2 and 4. The frequencies of stable translocation were 2.45 per 100 cell equivalent and those of insertion, color juction, acentric and dicentric were 0.32, 3.28, 0.23 and 0.27 per 100 cell equivalent respectively. The frequencies of chromosome rearragements increased with age in both sexes except for dicenrics. From above result, stable aberrations accumulate with age and it may reflect integrated lifetime exposure of adverse environment.

  • Based on the co-linearity in the Triticeae, comparative RFLP analysis of 2M chromosome of Ae. comosa Sibth et Sm. was performed with 2MS and 2M additional lines of Triticum aestivum L. cv. Chinese Spring. Among the wheat RFLP probes conserved in the short arms of wheat chromosome 2, those above psr912 were located on the long arms of 2M in Aegilops comosa. The rest probes on the short arm and all the probe sequences on the long arm of group 2 chromosome in wheat were conserved on the equivalent chromosomal position in Aegilops comosa. So, it is apparent that some chromosomal segment from the short arm had been transferred to long arm while reconstructing 2M chromosome relative to wheat chromosomes. The break-point was located between psr912 and psr131 of the short arm. This rearrangement of chromosome 2M might be a molecular evidence of the M genome speciation from an ancestral type.

Deletion or Duplication Syndromes of Chromosome 22: Review

  • Chromosome 22 is an acrocentric chromosome containing 500-600 genes, representing 1.5%-2% of the total DNA in cells. It was the first human chromosome to be fully sequenced by the Human Genome Project. Several syndromes involving the partial deletion or duplication of chromosome 22 are well descibed, including 22q11.2 deletion syndrome, 22q11.2 duplication syndrome, 22q11.2 distal deletion syndrome, Phelan-McDermid syndrome caused by a 22q13 deletion or pathogenic variant in SHANK3, and cat-eye syndrome caused by a 22 pter-q11 duplication. This review aims to provide concise information on the clinical characteristics of these syndromes. In particular, the similarities in features among these syndromes, genetic basis, and standard detection techniques are described, providing guidance for diagnosis and genetic counselling.

  • A karyotype of cytotype BB plant in Scilla scilloides Complex was established and the frequency of B-chromosomes were investigated. Chromosome complements of BB genome were composed of five pairs of subtelocentric and four pairs of metacentric chromosomes. Chromosome 1 has satellite with nucleolar organizer. Polymorphism was found in chromosome 2. The karyotype of cytotype BB will be available for analysis of genome composition in various cytotypes of S. scilloides Complex. The frequency of B-chromosome was 78.6%. Numbers of B-chromosome ranged from 1 to 4 and plants with 2B-chromosomes were predominant (57.2%). Two type of B-chromosomes, F and F', were found; F is a large iso-chromosome and F' a small one.

  • Benzene is a widespread human carcinogen, inducing leukemia and hematotoxicity. Exposure to benzene metabolites has been shown to cause genetic damage, including aneusomy and chromosome aberrations. Fluorescence in situ hybridization(FISH) procedure was used to determine if the benzene metabolite, 1, 2, 4-benzenetriol(BT), hydroquinone(HQ) and trans, trans-muconic acid(t,t-MA) induced specific chromosomal change in HL-60 cells. Treatment with BT, HQ and t,t-MA resulted in the induction of monosomy 8 and 21 in HL-60 cells in a dose-dependent manner. All of these metabolites also induced trisomy 8 and 21, but no correlation between frequencies of trisomy and concentration was found. Translocations between chromosome 8 and another unidentified chromosome [t(8:\ulcorner)], and between chromosome 21 and another unidentified chromosome [t(8:21)] were found. However, translocation between chromosome 8 and 21 [t(8:21)] was not found. Results indicate that the benzene metabolites, BT, HQ and t,t-MA, induce chromosome specific numerical and structural aberrations, and the fluorescence in situ hybridization (FISH) approach may be a useful and powerful technique for detection of aneuploidy.

  • The chromosome number was identified and fluorescence in situ hybridization(FISH) mapping of 5S and 45S rDNAs were conducted for C. minima and C. lanceolata in the genus Codonopsis from Jeju island. In this study, we have confirmed that the somatic metaphase chromosome number determined as 2n=2x=16 was the same as the findings from the previous studies. While the conventional staining method makes it rather difficult to distinguish satellite chromosomes due to high degree of variability, FISH analysis produced the exact number and location of 5S and 45S rDNAs. Both species in the genus Codonopsis have a pair of 5S rDNA and their gene loci were observed on chromosome 3. Although two pairs of 45S rDNAs (one on chromosome 1 and the other on chromosome 8) were identified in both species, the 45S rDNA signals on chromosome 8 in C. minima were significantly weaker than those on chromosome 1. In addition, the 45S rDNA signals on chromosome 1 in C. lanceolata showed that the chromosome is non-homologus. In this study, we have determined cytogenetic characteristics of C. minima and C. lanceolata according to their gene replication patterns.

Application of Chromosome Manipulation, DOP-PCR and AFLP Methods to Isolate Sex-Specific DNAs from Rumex acetosa L.

  • Rumex acetosa L. is a dioecious flowering plant with well developed sex chromosome system: 2n = 12 + XX in the female plants and 2n = 12 + XY1Y2 in the male plants. To isolate sex-linked DNA, we carried out chromosome micromanipulation, followed by DOP-PCR, AFLP of the PCR products, reverse Southern hybridization and sequence analysis. From 500 AFLP specific clones, 13 X-chromosome and 5 Y-chromosome specific clones were obtained. Except one clone RADAX-239 ($\underline{R}umex\;\underline{a}-\underline{D}OP-PCR-\underline{A}FLP-\underline{Y}-chromosome\;specific$), all clones appear to be R. acetosa plant-specific sequences and non-coding sequences. Southern blot analysis using these clones could not discriminate genomic DNAs either from male or female plants. Results of this study imply that both autosome-origin and degeneration of sex chromosomes are prevalent in plant systems.

Chromosome numbers of eight taxa of Aconitum L. in Korea and their systematic significance (Ranunculaceae)

  • Various aneuploidy and polyploidy have been reported in the genus Aconitum L. (ca. 300 species worldwide, Ranunculaceae), and there is a demonstrated association between major lineage diversification and polyploidy. This study reports chromosome counts of eight Aconitum from Korea, including the first counts for A. japonicum Thunb. subsp. napiforme ($H. L{\acute{e}}v.$ & Vaniot) Kadota (2n = 32) and A. longecassidatum Nakai (2n = 16). The study also includes chromosome numbers for two taxa on the Critically Endangered species list in Korea. Among Korean native species, chromosome numbers in Aconitum subgenus Aconitum range from 2n = 16 to 2n = 64 with diverse levels of polyploidy (2x, 4x, and 8x), whereas Aconitum subg. Lycoctonum exhibits only diploids (2n = 16). Greater chromosome number diversity in subg. Aconitum than subg. Lycoctonum might explain higher species diversity within the former subgenus (more than 250 species worldwide). Investigating chromosome number diversity of Aconitum in a phylogenetic framework will be a critical step to understand species richness of the genus.
