염색체 마이크로어레이를 이용한 표지염색체의 분자세포유전학적 특성 (Molecular Cytogenetic Characterization of Supernumerary Marker Chromosomes by Chromosomal Microarray)

  • 목적: 표지염색체(supernumerary marker chromosome, SMC)는 유래한 염색체에 따라서 임상 증상이 다양하다. 본 연구는 염색체 마이크로어레이를 이용하여 SMC의 기원을 밝히고 각 증례마다 분자세포유전학적 특성과 임상 표현형을 분석하고자 하였다. 대상 및 방법: 염색체 검사에서 SMC가 검출된 환자들 중에서 15번 염색체 유래를 제외한 4명의 환자에서 CGH 기법의 올리고 뉴클레오티드 염색체 마이크로어레이를 시행하였다. 결과: 3명의 환자에서 유래된 염색체 부위를 확인할 수 있었다. 증례1은 1q21.1-q23.3에서 16.1 Mb의 SMC를 가졌고, 증례2는 19p13.11-q13.12에서 21 Mb, 증례3은 22q11.1-q11.21과 22q11.22-q11.23의 두 구간에서 각각 2.5Mb와 2.0Mb로 재배열된 4.5 Mb의 SMC를 나타내었다. 결론: 증례1은 1q21.1 중복증후군을 포함하여 광범위한 임상표 현형을 나타내었다. 증례2는 아스퍼거 증후군과 유사한 정신행동 이상 소견은 19p12-q13.11, 청력장애와 사시는 19p13.11, 그 외 증상은 19q13.12의 유전자와 연관 가능성이 높다. 증례3은 묘안 증후군 type I 및 22q11.2 미세중복증후군과 비교했을 때 항문폐쇄는 22q11.1-q11.21, 그 외 증상들은 22q11.22-q11.23과 연관성을 시사하였다. 고해상도 염색체 마이크로어레이 분석은 SMC의 유래를 확인할 수 있고 유전형-표현형 상관성을 이해함으로써 유전상담에 도움이 된다.

Identification of Microsatellite Markers Linked to Photoperiod Insensitive Gene Ppd-D1a in Wheat

  • To facilitate breeding of lines with either the Ppd-D1a or ppd-d1a, we screened 342 $F_2$ progenies from a cross between Laura (photoperiod insensitive, Ppd-D1a) spring wheat and SWP5304 (photoperiod sensitive, ppd-d1a) for their time to heading under 10 hour day length, and with a set of 37 microsatellite primers previously mapped to chromosome 2D. Bulk segregant analysis was used to identify tow linked microsatellite loci. The Ppd-D1a locus was flanked by Xgwm484 with 13.7 cM distance and Xgwm455 with 27 cM. These markers may be useful in selection of the desired photoperiod sensitivity in segregating populations grown in Northern latitude.

  • Selection of oocyte cryopreservation method is a prerequisite factor for developing an effective bank system. Compared with slow freezing method, the vitrification has various advantages such as avoiding intracellular ice crustal formation. In our previous, we attempted to employ a vitrification method using ethylene glycol and an electron microscope grid for cryopreservation of mouse oocytes. However, A high incidence of spindle and chromosome abnormalities was detected in thawed oocytes after vitrification. We examined whether the addition of a cystoskeleton stabilizer Taxol $^{TM}$, to the vitrification solution could promote the post-thawed survival and subsequent development of stored oocytes. More oocytes developed to the 4-cell (44.7% vs. 69.7%), 8-cell (31.8% vs. 64.2%), morula (24.7% vs. 54.3%), and blastocyst (20.3% vs. 49.2%) stages after the addition of Taxol$^{TM}$ to the cryoprotectant than after no addition. 21 and 26 mouse pups were born after transfer of blastocyst derived from oocytes vitrified without and with Taxol. The addition of Taxol to vitrification solution greatly promoted post-thaw preimplantation development of ICR morose oocytes.tes.

Williams 증후군 환아의 치의학적 소견에 대한 증례 보고 (WILLIAMS SYNDROME : TWO CASES)

  • Williams syndrome is a rare genetic disorder with a frequency of one per 20,000~50,000 live births. It is caused by a deletion of one elastin allele located within chromosome subunit 7q11.23(long arm). This syndrome is frequently accompanied by disorders such as congenital heart disease, facial anomalies, mental retardation, and so on. The characteristic facial appearance includes full lips, rounded cheeks, broad forehead, periorbital fullness, flattened bridge of nose, small nose with anteverted nostril, long filtrum and low-set ears. In oral features, hypodontia, high prevalence of dental caries, microdontia, enamel hypoplasia, delayed eruption, and malocclusions have been found. Most adult patients with Williams syndrome lack social adaptability and lead seclusive lives, however, young patients are rather very friendly and talkative, and seem smarter than their actual intellectual quotients. They also tend to favor staying with grown-ups rather than mixing with their peers, and tend to present problematic temper tantrum during dental treatment.

Prevalence and Characterization of Methicillin-Resistant Staphylococcus aureus in Raw Meat in Korea

  • A total of 2,858 meat samples collected during 2003-2008 in Korea were investigated, and methicillin-resistant Staphylococcus aureus (MRSA) isolates were isolated from 1.0% (9/890) of beef, 0.3% (4/1,055) of pork, and 0.3% (3/913) of chicken meat samples, respectively. MRSA isolates showed the two sequence types (STs), ST72 from beef and pork and ST692 from chicken meat. MRSA isolates from beef and pork were Panton-Valentine leukocidin-negative, staphylococcal cassette chromosome mec type IVa strain with ST72, which is the most prevalent type of communityacquired MRSA in Korea. An identical pulse-field gel electrophoresis pattern was detected among 10 of 16 MRSA isolates: 9 strains from beef (n=5) and pork (n=4) in 2008, and one strain from beef in 2005.

Transient neonatal diabetes mellitus caused by a de novo ABCC8 gene mutation

  • Transient neonatal diabetes mellitus (TNDM) is a rare form of diabetes mellitus that presents within the first 6 months of life with remission in infancy or early childhood. TNDM is mainly caused by anomalies in the imprinted region on chromosome 6q24; however, recently, mutations in the ABCC8 gene, which encodes sulfonylurea receptor 1 (SUR1), have also been implicated in TNDM. Herein, we present the case of a male child with TNDM whose mutational analysis revealed a heterozygous c.3547C>T substitution in the ABCC8 gene, leading to an Arg1183Trp mutation in the SUR1 protein. The parents were clinically unaffected and did not show a mutation in the ABCC8 gene. This is the first case of a de novo ABCC8 gene mutation in a Korean patient with TNDM. The patient was initially treated with insulin and successfully switched to sulfonylurea therapy at 14 months of age. Remission of diabetes had occurred at the age of 16 months. Currently, the patient is 21 months old and is euglycemic without any insulin or oral hypoglycemic agents. His growth and physical development are normal, and there are no delays in achieving neurological and developmental milestones.

Cloning and Expression of Human KCNE1 Gene

  • KCNE1 is the causal gene of long QT syndrome. KCNE1 gene is located in chromosome 21. In compliance with this KCNE1 gene the proteins come out. KCNE1 is responsible for $K^+$ channel which maintains the normal function of the heart muscle for contraction. Affected individuals manifest prolongation of the QT interval on electrocardiongrams, a sign of abnormal cardiac repolarization. The clinical features of LQT result from episodic cardiac arrhythmias, such as torsade de pointes and ventricular fibrllation. Blood DNA was isolated and kept in $4^{\circ}C$ refrigerator. The KCNE1 gene was amplified by PCR method and about 414 bp band was identified by agarose gel electrophoresis. PCR products were inserted into pGEX-4T-1 vector in order to express KCNE1 protein after treatment with IPTG SDS-PAGE was carried out and the protein band which was about 47 kDa was clearly odserved. Results of this study would contribute to the detailed understanding of KCNE1 protein function and to designing better treatment of Long QT symdrome.

Genome-wide Examination of Chromosomal Aberrations in Neuroblastoma SH-SY5Y Cells by Array-based Comparative Genomic Hybridization

  • Most neuroblastoma cells have chromosomal aberrations such as gains, losses, amplifications and deletions of DNA. Conventional approaches like fluorescence in situ hybridization (FISH) or metaphase comparative genomic hybridization (CGH) can detect chromosomal aberrations, but their resolution is low. In this study we used array-based comparative genomic hybridization to identify the chromosomal aberrations in human neuroblastoma SH-SY5Y cells. The DNA microarray consisting of 4000 bacterial artificial chromosome (BAC) clones was able to detect chromosomal regions with aberrations. The SH-SY5Y cells showed chromosomal gains in 1q12~ q44 (Chr1:142188905-246084832), 7 (over the whole chro-mosome), 2p25.3~p16.3 (Chr2:18179-47899074), and 17q 21.32~q25.3 (Chr17:42153031-78607159), while chromosomal losses detected were the distal deletion of 1p36.33 (Chr1:552910-563807), 14q21.1~q21.3 (Chr14:37666271-47282550), and 22q13.1~q13.2 (Chr22:36885764-4190 7123). Except for the gain in 17q21 and the loss in 1p36, the other regions of gain or loss in SH-SY5Y cells were newly identified.

아이소발레린산혈증 신생아에서 발견된 새로운 돌연변이 (Neonatal Onset Isovaleric Acidemia with Novel Mutation)

  • Isovaleric acidemia is autosomal-recessively inherited and an inborn error of metabolism caused by abnormal leucine metabolism due to the genetic defect of IVD (Isovaleryl-CoA dehydrogenase). IVD corresponds to mitochondrial matrix enzyme that acts on converting isovaleryl-CoA into 3-methylcrotonyl-CoA in the leucine catabolism. The IVD gene is located at Chromosome 15q14-q15, particularly between base pair 40,405,485 and base pair 40,435,948. It consists of 12 exons and has been reported to cause over 50 diseases so far. We conducted IVD gene test on the patient with acute isovaleric acidemia and confirmed a new type of mutation for the first time. As a result of analyzing the IVD gene sequence, we found out that c.129T>G(p.Asn43Lys) and c.1033A>G(p.Asn345Asp) mutations exist as heterozygosity at Exon 1 and Exon 10 respectively, novel mutation.

A diagnosis of hypochondroplasia by next generation sequencing

  • Achondroplasia and hypochondroplasia are the two most common forms of short-limb dwarfism. They are autosomal dominant diseases that are characterized by a rhizomelic shortening of the limbs, large head with frontal bossing, hypoplasia of the mid-face, genu varum and trident hands. Mutations in the fibroblast growth factor receptor-3 (FGFR3) gene, which is located on chromosome 4p16.3, have been reported to cause achondroplasia and hypochondroplasia. More than 98% of achondroplasia cases are caused by the G380R mutation (c.1138G>A or c.1138G>C). In contrast, the N540K mutation (c.1620C>A) is detected in 60-65% of hypochondroplasia cases. Tests for common mutations are often unable to detect the mutation in patients with a clinical diagnosis of hypochondroplasia. In this study, we presented a case of familial hypochondroplasia with a rare mutation in FGFR3 identified by next generation sequencing.
