한국산 석패와 7종의 염색체 연구 (Chromosomal Study of Seven Species of Unionidae ( Bivalvia : Palaeoheterodonta ) in Korea)

  • The chromosome numbers and the karyotypes of seven species in Unionidae are reported, using air drying in gonad. In seven species, the chromosome number of 38(2n) was counted. The mitotic chormosomes of A. arcaeformis flavotincta, A. woodiana and L. gottschei hd 7 pairs of metacentric and 12 pairs of submetacenrtic chromosomes, U. douglasiae had 6 pairs of metacentrics, 13 pairs of submetacentrics, U. douglasiae sinuolatus had 4 metacentric pairs and 15 submetacentric pairs, L. acrorhyncha had 5 metacentric pairs and 14 submetaacentric pairs, and S. triangularis had 5 mdtacenrtric pairs, 13 submetacentric pairs and 1 pair of subtelocentric chromosomes. The size of chromosomes of A. woodiana was the longest in length and L. gottschei was the shortest. The sexual difference of chromosomes was not observed.

Effects of gamma-irradiation on the infectivity and chromosome aberration of Clonorchis sinensis

  • Effects of gamma irradiation on the worm survival and chromosomal aberration of Clonorchis sinensis were studied. The metacercariae irradiated with various amounts of gamma radiation (ranging from 5 Gy to 50 Gy) were fed to rats, and the effects were compared with those of non-irradiated controls. Recovery rates of adult worms in irradiated groups were reduced gradually as increasing of the irradiation doses. No worm was recovered from rats which were fed with 50 Gy irradiated metacercariae. The chromosome number was 2n = 56 in all worms from all experimental groups. However, the groups irradiated with 20 Gy, 25 Gy or 30 Gy showed variations in the chromosome number, depending on different cells in the same individual. Radiation doses used in this study did not appear to induce chromosome aberrations, however, irradiation with 30 Gy showed slightly reduced chromosome size.

Identification of QTLs Affecting Physical Traits of Cooked Rice

  • This study was conducted to ascertain the chromosomal locations and effect of quantitative trait loci (QTL) associated with the physical traits of rice (Oryza sativa L.) eating quality. One hundred sixty four recombinant inbred lines (MGRILs) of F$_{11}$ were derived from the cross between Milyang 23 (Tongil type) and Gihobyeo (japonica type). They were evaluated for six physical traits of cooked rice. Transgressive segregation was observed for all examined traits. Significant QTL were detected (LOD$\geq$2.0) in three traits, including single QTL for adhesiveness, gumminess, and chewiness of cooked rice, respectively. Phenotypic variation explained by each QTL ranged from 6.3% to 14.6%. However, no significant QTL was detected for hardness, cohesiveness, and elasticity of cooked rice. Pleiotropic effects of single QTL on different traits are observed.d.

  • Animal experiments need numerous kinds of animal which are suitable for every research. About 300 mouse strains are developed up to the present, but they do not give satisfaction to every researchers. So we must build up the methods of breading animals which are newly developed and of maintenance of characteristics which were developed before. To maintain experimental animal is not only proceeding the generation but also increasing the animal populations, it needs geneticai control. Genetic factors which influence to reproduction are very important to maintain colony. These factors include lethal gene, chromosomal abberation, sterility gene, etc.. With the recent development of transgenic technology, scientists now can deliberately creat numerous specific animal models. To know how to manage the colony which has genetic defect on reproduction and transgenic mice is one of the key to study in vitro fertilization.

  • Reciprocal exchanges of DNA in sister chromatids (SCEs) are induced by various carcinogens and mutagens, although the quantitative relationship between the number of mutations and SCEs induced varies among chemicals. Nevertheless, the analysis of SCEs production by various agents often proposed as a sensitive and quantitative assay for mutagenicity and cytotoxicity. Mercury, even if which has no evidences for mutagenicity and carcinogenicity, is reported to exert some cytotoxic effects, such as chromosomal aberrations or bad influences to ovulation and reproduction in experimental animals, etc.. In this study, tests for sister chromatid exchanges have been carried out on normal human lymphocytes in whole blood culture to add mercury chloride ($HgCl_2$) or methylmercury chloride ($CH_3\;HgCl$) for 72 hr. The results indicate the dose-dependent relationship between the frequencies of SCEs and the concentrations of $HgCl_2,\;CH_{3}HgCl$ and 5-bromo-2'-deoxyuridine (BrdU). Lymphocyte proliferation has depressed in the higher concentration of mercury.

  • Sophoricoside was isolated as the inhibitor of IL-5 bioactivity from Sophora japonica (Leguminosae). It has been reported to have an anti-inflammatory effect on rat paw edema model. To develope as an anti-allergic drug, genotoxicity of sophoricoside was investigated in bacterial and mammalian cell system such as Ames bacterial test, chromosomal aberration assay and single cell gel electrophoresis (Comet) assay.(omitted)

  • Previous study demonstrated that the restriction barrier of Streptomyces griseus is almost completely bypassed by the Streptomyces-E. coli shuttle vectors passed through the E. coli GM161 strain and methylated with AluI and HpaII methyltransferases. The same DNA methylation of the genomic DNA fragments cloned the nonreplicative vectors generated integrative transformation and gene disruption of their chromosomal counterparts at high efficiencies in S. griseus. This result indicated that the efficiency of gene disruption depends on the efficient transfer of the incoming DNA into bacterial hosts.

  • EGCG [(-)-epigallocatechin-3-gallate], a major component of green tea has been considered as a major antioxidant constituent. In addition to having been considered for cancer treatment as a chemopreventive and chemotherapeutic agent, EGCG has recently been attributed an anti-proliferative effect. We re-examined the latter finding in this study and added specific focus on the ability of EGCG to induce apoptosis in human osteogenic sarcoma (HOS) cells. Antiproliferative action of EGCG $(IC_{50}=35.3{\pm}6.0{\mu}g/mL)$ appeared to be linked to apoptotic cell death based on morphological changes, chromosomal DNA degradation, and an increase in the $sub-G_1$ apoptotic cell population. Treatment of HOS cells with EGCG gradually activated caspase-3, an established inducer of apoptotic cell death.

  • The objective of this study was to determine genotoxic potential of Sophora Japonica Linne Seed Extract(SE). The bacterial reverse mutation test set the treatment levels of SE at 0, 312.5, 625, 1250, 2500, 5000 $\mu\textrm{g}$/plate using Salmonella typhimurium strains (TA1535, TA1537, TA98, TA100) and Escherichia coli WP2uvrA(pKM101). Actin-related protein BAF53 is essential for the formation of replication foci

  • It has been suggested that chromatin is organized into the stable structures that provide fundamental units of chromosome architecture in interphase mammalian cells. The stable structures of chromatin can be visualized as replication foci when replicating DNA is labeled with thymidine analogs. Previously, we showed that the chromosome territory expanded after BAF53 knockdown. In this study, we found that BAF53 is required for the formation of replication foci. DNA replication was not impaired in BAF53 knockdown cells, suggesting that the decrease in the number of replication foci is due to disintegration of replication foci, but not suppression of DNA replication. The attractive forces that maintain structural integrity of replication foci could be disrupted by BAF53 knockdown, and it may be responsible, at least in part, for the expansion of chromosome territories after BAF53 knockdown.
