Species, doses and routes extrapolation can be sucessfully carried out by using a physiologically-based pharmacokinetic (PBPK) approach. And PBPK approach to assess risk of hazardous chemicals is reasonable whatever the exposure scenarios are happened. Both partitioning coefficients of chemical between tissue and blood and enzymatic metabolic rate constants are key parameters to build up the PBPK model. In this study, we tried to estimate in vitro metabolic rate constants using a special apparatus instead to measure the in vivo constants which are used to PBPK simulation since the in vitro tests are less expensive and more convenient than in vivo tests. For the purpose, we designed and tested the new system to measure continuously the headspace concentration of VOC. The newly designed system is composed with a diffusion chamber which generates gaseous substrate, a reaction vessel with a recirculating pump to establish a closed system, an autbmatic sampler from a gas phase, a gas chromatography to analyze the headspace. In addition, a cold water condenser is attached between the reaction vessel and pump to reduce the content of gaseous moisture which interferes with chemical analysis. To validate the newly developed methodology, in vitro metabolic rate constants of trichloroethylene (TCE) as a prototype VOC were estimated by simulating observed results with an ACSL program. The simulated results are consistent to those estimated by the other research groups. This finding suggests that our newly designed closed system may be a useful apparatus to estimate in vitro metabolic rate constants for VOC.

The process for bioethanol production from lignocellulosic biomass was studied through process simulation using PRO/II. Process integration was conducted with concentrated acid pretreatment, hydrolysis process, SMB (simulated moving bed chromatography) process and pervaporation process. Energy consumption could be minimized by the heat recovery process. In addition, material and energy balance were calculated based on the results from the simulation and literature data. A net production yield of 4.07 kg-biomass and energy consumption value of 3,572 kcal per 1 kg ethanol were calculated, which is indicating that 26% yield increase and 30% energy saving compared to the bioethanol production process with dilute-acid hydrolysis (SRI report). In order to make it possible, sugar conversion yield of cellulose and hemi-cellulose is to be reached up to 90% and fermentation of xylose needs to be developed. In order to reduce the energy consumption up to 30%, the concentration of acid solution after being separated by 5MB should exceed 20%. If acid/sugar separation by SMB process is to be practical, the bioethanol process designed in this study can be commercially feasible.
Lipase-catalyzed esterification of structural butanol isomers and n-butyric acid was investigated in supercritical carbon dioxide. The experiments were performed in a high pressure cell for 5 hrs with a stirring rate of 150 rpm at 323.15 K and 130 bar. The Candida Antarctica lipase B (CALB) was used in whole system as a catalyst. The experimental results were analyzed by GC-FID using a INNOWax capillary column. The conversion yield and the tendency of the esterification in supercritical carbon dioxide were compared with estimated results by molecular dynamics simulation. Based on the Ping-Pong Bi-Bi mechanism with competitive inhibition, each step of the reaction was optimized; using this result the transition state was predicted. Conformational preference of isomers was also analyzed using molecular dynamics simulations. This kind of approach will be further extended to the prediction of enzyme-catalyzed reactions using computers.

Peptides with bioactive effects are being researched for various purposes. However, there is a lack of overall research on pork-derived peptides. In this study, we reviewed the process of obtaining bioactive peptides, available analytical methods, and the study of bioactive peptides derived from pork. Pepsin and trypsin, two representative protein digestive enzymes in the body, are hydrolyzed by other cofactors to produce peptides. Bicinchoninic acid assay, sodium dodecyl sulfate-polyacrylamide gel electrophoresis, chromatography, and in vitro digestion simulation systems are utilized to analyze bioactive peptides for protein digestibility and molecular weight distribution. Pork-derived peptides mainly exhibit antioxidant and antihypertensive activities. The antioxidant activity of bioactive peptides increases the accessibility of amino acid residues by disrupting the three-dimensional structure of proteins, affecting free radical scavenging, reactive oxygen species inactivation, and metal ion chelating. In addition, the antihypertensive activity decreases angiotensin II production by inhibiting angiotensin converting enzyme and suppresses blood pressure by blocking the AT1 receptor. Pork-derived bioactive peptides, primarily obtained using papain and pepsin, exhibit significant antioxidant and antihypertensive activities, with most having low molecular weights below 1 kDa. This study may aid in the future development of bioactive peptides and serve as a valuable reference for pork-derived peptides.

21-hydroxylase deficiency (21-OHD), most common form of congenial adrenal hyperplasia, is categorized into classical forms, including the salt-wasting (SW) and the simple virilizing (SV) types, and nonclassical (NC) forms based on the severity of the disease. Newborn screening for 21-OHD has been performed in Korea since 2006. $17{\alpha}$-hydroxyprogesterone (17-OHP) is a marker for 21-OHD and is measured using a radioimmunoassay or a fluoroimmunoassay. Premature and low birth weight infants are likely to give false positive 17-OHP findings, therefore, cutoff values for these infants should be determined based on gestational weeks or birth weight. ACTH simulation test is helpful when the 17-OHP shows equivocal increase, and it is gold standard for diagnosis of NC type. Recently, liquid chromatography linked with tandem mass spectrometry was developed for rapid, highly specific, and sensitive analysis of multiple analytes. Molecular analysis of CYP21A2 is useful for confirming diagnosis of mild SV or NC type, predicting prognoses, and genetic counseling. In order to make newborn screening for 21-OHD more efficient, early detection of boy with SW type, early determination of girl with ambiguous genitalia, detection of NC type, and overcoming of false positive in premature and low birth weight infants should be considered. Above all, early treatment should be started when the patient is suspected as having 21- OHD clinically before confirming the diagnosis to prevent adrenal crisis. Here, author reviewed recent articles of guideline and proposed guideline for 21-OHD.
Objective: To examine the effects of Rhodobacter sphaeroides (R. sphaeroides) supplementation as a direct-fed microbial (DFM) on rumen fermentation in dairy cows and on coenzyme Q10 (CoQ10) transition into milk, an in vitro rumen simulation batch culture and an in vivo dairy cow experiment were conducted. Methods: The characteristics of in vitro ruminal fermentation were investigated using rumen fluids from six cannulated Holstein dairy cows at 2 h post-afternoon feeding. A control treatment was included in the experiments based on a typified total mixed ration (TMR) for lactating dairy cows, which was identical to the one used in the in vivo study, plus R. sphaeroides at 0.1%, 0.3%, and 0.5% TMR dry matter. The in vivo study employed six ruminally cannulated lactating Holstein cows randomly allotted to either the control TMR (C-TMR) treatment or to a diet supplemented with a 0.5% R. sphaeroides culture (S-TMR, dry matter basis) ad libitum. The presence of R. sphaeroides was verified using denaturing gradient gel electrophoresis (DGGE) applied to the bacterial samples obtained from the in vivo study. The concentration of CoQ10 in milk and in the supernatant from the in vitro study was determined using high performance liquid chromatography. Results: The results of the in vitro batch culture and DGGE showed that the concentration of CoQ10 significantly increased after 2 h of R. sphaeroides supplementation above 0.1%. When supplemented to the diet of lactating cows at the level of 0.5%, R. sphaeroides did not present any adverse effect on dry matter intake and milk yield. However, the concentration of CoQ10 in milk dramatically increased, with treated cows producing 70.9% more CoQ10 than control cows. Conclusion: The CoQ10 concentration in milk increased via the use of a novel DFM, and R. sphaeroides might be used for producing value-added milk and dairy products in the future.
